Identification of Chrysanthemum Genetic Resources Resistant to Chrysanthemum Stunt Viroid (CSVd) (국화왜화바이로드 저항성 국화 유전자원 선발)

  • Chrysnathemum stunt viroid (CSVd), a small, single-stranded, infectious RNA, has caused a severe problem in chrysanthemum in the world. In Korea, since CSVd was firstly observed in 1997, the disease has been spread throughout the whole country rapidly. In spite of the seriousness, few methods for control of CSVd have been known without prevention. The use of resistant cultivars is one of the most efficient approaches for overcoming CSVd disease in chrysanthemum cultivation. This study was carried out to identify chrysanthemum genetic resources for the resistance to chrysanthemum stunt viroid (CSVd). A total of 192 commercial cultivars including 167 spray and 25 standard chrysanthemum were screened with CSVd through grafting inoculation method. In most of the inoculated cultivars, typical disease symptoms, stunting of plant height, reduced flower size, and flower color bleaching, were induced. Several cultivars, however, were symptomless or showed delay in symptom expression. Of 192 chrysanthemum cultivars, two cultivars with less than 5% of the average reduction rate of plant height, 'Chiwerel' and 'Jeongheungdaesin', were rated as resistant. And six cultivars less than 20% inculding 'Inga' were rated as moderate resistant. The remaining 184 cultivars with a high level of reduction were rated as susceptible. We expect that these genetic resources can be used in crossbreeding programs for developing CSVd resistant cultivars of chrysanthemum.

  • To examine the effects of ozone (O$_3$), one of the major air pollutants in the city area, on genetic changes in Cornus controversa Hemsl., we compared genetic structures between sensitive (S) and tolerant (T) tree groups of C. controversa fumigated with ozone using isozyme markers. The genetic structures were measured in terms of allele and genotype frequencies determined at ave polymorphic enzyme loci. Marked genetic differences between the two groups were detected at three loci (Lap-2, Mdh-1 and Skdh-1). Genetic parameters, genetic multiplicity, genetic diversity and heterozygosity showed that the tolerant group retained greater genetic variation than did the sensitive group. Results of the study were congruent with the general expectation that the more heterozygous individuals and/or populations exhibit higher resistance to various stress factors.

Inheritance of Fenvalerate Resistance in the Diamondback Moth, Plutella xylostella Linne(Lepidoptera: Yponomeutidae) (배추좀나방의 Fenvalerate에 대한 저항성 유전)

  • The mode of inheritance of resistance to fenvalerate in the diamondback moth (Plutella xylostella L.) was studied using insecticide susceptibility and mixed function oxidase tests. Tehre were no differences in the concentration-mortality relationships between $F_1$ progenies $(S_{female}\timesR_{male},\;R_{female}\timesS_{male})$ obtained from reciprocal crosses with the susceptible and fenvalerate-selected strains(R), indicating the absence of sex-linked inheritance. Degree of dominance of the $F_1$ progenies $(S_{female}\timesR_{male},\;R_{female}\timesS_{male})$ were -0.50 and -0.46, respectively, in the insecticide susceptibility test and -0.85 and -0.81, respectively, in the mixed function oxidase test. These results suggest that inheritance of fenvalerate resistance is controlled by and incompletely recessive autosomal gene.

Current status of peach genomics and transcriptomics research (복숭아 유전체 및 전사체 최근 연구 동향)

  • In this review, we summarized the trends of genomics and transcriptomics research on peach, a model species of Rosaceae. Peach genome maps have been developed from various progeny groups with many next-generation sequencing (NGS) based single nucleotide polymorphism markers. Molecular markers of qualitative traits and quantitative trait loci (QTL) such as fruit characteristics, blooming date, and disease resistance have been analyzed. Among many characteristics, markers related to flesh softening and flesh adhesion are useful for marker assisted selection. Through comparative genomics, peach genome has been compared to the genome of Arabidopsis, Populus, Malus, and Fragaria species. Through transcriptomics and proteomics, fruit growth and development, and flavonoid synthesis, postharvest related transcriptomes and disease resistance related proteins have been reported. Recently, development of NGS based markers, construction of core collection of germplasm, and genotyping of various progenies have been preceded. In the near future, accurate QTL analysis and identification of useful genes are expected to establish a foundation for effective molecular breeding.

Evaluation of Early Generations of Crosses for Incorporation of Resistance to Phytophthora Blight into Sweet Pepper (감미종(甘味種)고추에 역병저항성(疫病抵抗性)을 도입(導入)하기 위한 교잡(交雜) 초기세대(初期世代) 검정(檢定))

  • A leading sweet pepper cultivar, Keystone Resistant Giant #3, was crossed with a line with resistance to Phytophthora capsici, PI201232, for incorporation of the resistance and to study the inheritance of resistance to the disease. Seedlings of parents, $F_1$, $F_2$ and backcross populations of a cross between Keystone Resistant Giant #3 and PI201232 were inoculated with zoospore suspension of P. capsici at 36 days after seeding. Most of the $F_1$ seedlings survied the inoculation and this suggested that resistance is dominant over susceptibility. The number of survived plants in $F_2$ population was, however, much less than the killed. All the plants in a backcross to Keystone Resistant Giant #3 were killed. Therefore, the observed numbers did not fit the expected ratio for segregation of one or two dominant alleles as previously reported. The resistance to P. capsici appeared to be inherited in a quantitative mode in evaluation of root rot. Resistant individuals in $F_2$ population were selected and a breeding program for incorporation of the resistance to P. capsici by backcross method is continued.

연초 1대잡종 품종 육성과 이용 방향

  • 일대잡종육종이란 현대의 다양한 육종기술 중 하나로 교배 제1세대의 잡종강세현상을 곧바로 이용하는 것으로 여러 작물에서 실용적으로 이용되고 있다. F$_1$잡종종자를 생산하려면 자가수분을 통제해야 하는데 웅성불임을 이용하면 자친의 화분제거 노력이 생력되어 경제적인 종자생산이 가능하다. 연초는 자식성 작물로 잡종강세는 높지 않으나 종간 교잡에 의한 cms가 만들어진 이후 버어리종 등에서는 60년대부터 직적형질에 가깝게 유전되는 우성 단일유전자의 지배를 받는 N. logniflora의 역병(race 0)저항성을 도입한 웅성불임(cms) F$_1$잡종이 활발하게 이용되어 왔다. 최근에는 cms F$_1$잡종의 이용영역이 넓어져 우리나라의 경우 N. glutinosa에서 유래한 우성 단일인자 TMV저항성을 도입한 황색종 cms F$_1$잡종 KF 114가 주 재배품종이 되었고 버어리종에서는 VAM에서 유래한 열성단일유전자의 지배를 받는 PVY저항성이 도입된 KB 111의 확대재배가 예상되고 있다. 본 글에서는 cms F$_1$잡종의 국내외 이용 사례를 살펴보고 cms F$_1$잡종의 육성과 이용방향, cms F$_1$잡종 종자생산에 관하여 기술하였다. 그리고 병저항성뿐 아니라 알칼로이드함량, 염록소함량, DVT나 챤-abienol 및 SE함량 등 질적형질에 가깝게 유전되고 유전양식이 명확하게 밝혀진 화학성분에 대해서도 F$_1$잡종 육성의 대상 형질로 논의하였다.

Molecular Mapping of the Blast Resistance Loci in the Durable Resistance Japonica Rice Cultivar, Palgong (도열병 내구 저항성 자포니카 벼품종 팔공의 저항성 관련 유전좌위 분석)

  • Rice blast caused by the fungus Magnaporthe grisea (anamorphic: Pyricularia oryzae) is an important disease in rice and development of resistant varieties to blast is one of the most important goals in rice breeding programs. A japonica rice variety, Palgong, has shown resistance to the Korean blast pathogen since it was developed in 1996. Nine blast resistance quantitative trait loci (QTLs) in Palgong alleles were identified on chromosomes 2, 4, 7, and 11. Four QTLs of qBn2.3, qBn4.2, qBn11.1, and qBn11.2 explained 28-56.7% of total phenotypic variation, while five QTLs of qBn2.2, qBn2.4, qBn4.1, qBn7.1, and qBn7.2 explained 9.7-18.8%. In a previous study, one to four resistance genes were located on the loci qBn2.2, qBn2.3, qBn4.2, qBn11.1, and qBn11.2, however, resistance genes were not located on the loci qBn2.4, qBn4.1, and qBn7.1. A major QTL, qBn11.2, explaining 56.7% of total phenotypic variation was related to the durable resistance of Palgong. Additionally, rice stripe virus resistance of Palgong was assumed to be based on the Stvb-i gene, which is located on a major QTL qBn11.2.

Isolation of Mutants Susceptible to Rice Blast from DEB-treated Rice Population (DEB 처리에 의해 유도된 벼 돌연변이 집단으로부터 도열병 감수성 돌연변이 분리)

  • Rice blast, which is caused by the fungus Magnaporthe grisea, is one of the most destructive diseases of rice. To identify genes involving in the signal transduction pathways that mediate rice blast resistance, we screened over 2,000 mutant lines of a highly resistant variety RIL260 that were generated by using a DEB (1, 3-Butadiene diepoxide) treatment method. In the mutant population, the frequency of albino plants was 6.7%, indicating that this population has a high frequency of mutations in the genome. The primary screening identified 29 mutant plants that exhibit a complete or partial loss of the resistance to rice blast. Among them, M5465, the most susceptible line, was subsequently examined by DNA gel-blot experiments using DNA molecular markers of Pi5(t) that has been previously identified as a durable resistance locus in RIL260. The result revealed that a large deletion and rearrangement of genomic DNA occurred in the Pi5(t) locus. The results suggest that DEB can be used as an efficient mutagen to induce large scale mutations in the rice genome. The isolated mutants should be useful for elucidating the Pi5(t)-mediated signaling pathways of rice blast resistance.

Complete genome sequence of Enterococcus faecalis strain DM01, a potential starter culture candidate for soybean fermentation (콩발효 종균후보 Enterococcus faecalis strain DM01의 유전체 염기서열)

  • Enterococcus faecalis strain DM01 was isolated from meju, a traditional Korean fermented soybean product. The strain did not exhibit resistance to eight antibiotics (ampicillin, chloramphenicol, ciprofloxacin, erythromycin, gentamicin, penicillin G, tetracycline, and vancomycin), biofilm formation, and hemolytic activity. The genome of strain DM01 consists a single circular 2,785,968-bp chromosome with a G + C content of 37.68%. The complete genomic sequence of strain DM01 provides genetic information to support the absence of antibiotic resistance, biofilm formation, and hemolytic activity.

Development of Resistance Evaluation Method for Powdery Mildew (Leveillula taurica) in Capsicum spp. (고추 흰가루병 저항성 평가방법 개발)

  • Pepper powdery mildew causes increasingly economical damage due to increased cultivation of pepper in greenhouses. To assess resistance of pepper resources against pepper powdery mildew, we developed a novel evaluation formula for pepper resistance against powdery mildew. The evaluation formula named S index is as follows; (number of the highest disease leaf/top leaf number)-(number of the lowest disease leaf/top leaf number). Positive correlation (81%, P = 0.01) between S index and authentic disease leaf rate was observed from the pepper plants infected by powdery mildew. Various pepper species from our genetic resources were evaluated to identify pepper varieties conferring resistance against powdery mild using S-index. Capsicum frutescens accessions 3CA131 and C. baccatum accessions 3CA0162, 3CA174, 3CA176 showed high resistance to powdery mildew, but none of C. annuum was resistant. Results suggest that S-index proposed in this study is useful to assess resistance evaluation of powdery mildew in chili pepper breeding.
