Control of Cucumber Downy Mildew Using Resistant Cultivars and Organic Materials (저항성 품종과 유기농업자재를 활용한 오이 노균병 방제)

  • We selected eight resistant cultivars including 'Heukryongsamcheok', 'Heukgeumsolsamcheok' and 'Gangryeoksamcheok' showing high suppressive effect against cucumber downy mildew (CDM) through plastic film house and field trials in spring and autumn season in 2015. Of them, these three varieties, 'Heukryongsamcheok', 'Heukgeumsolsamcheok' and 'Gangryeoksamcheok' were used to evaluated suppressive effect against CDM by comparing disease severity (area under disease development progress curve, AUDPC) with those of three susceptible varieties under plastic film house condition. AUDPC of three resistant cultivars was in the range of 10.9 to 23.6, meanwhile those of three susceptible cultivars was in the range of 286.7 to 290.3. Consequently, we confirmed that cultivation of the selected resistant cultivars can reduce CDM in vivo. When disease severity (diseased leaf area) was investigated on a resistant cultivar ('Heukryongsamcheok') sprayed without any organic materials and susceptible cultivars sprayed single-, two times- and three times with Bordeaux mixture, sulfur-loess mixture and $Kocide^R$ (copper hydroxide), respectively, it was the lowest in the resistant cultivar plot compared to susceptible cultivar plots sprayed with any organic materials. In addition, we evaluated control effect of the selected resistant cultivar by comparing that of organic materials including lime sulfur mixture, Bordeaux mixture and garlic extract. As a result, Bordeaux mixture showed the highest control effect against CDM. AUDPC of Bordeaux mixture, resistant cultivar, lime sulfur, garlic extract and untreated control was 3.9, 10.6, 95.6, 24.9, and 258.7, respectively. Based on the above-mentioned results, we think that the resistant cultivars and Bordeaux mixture can be effectively used to control CDM as one of control measures under the farmhouse condition.

Effects of Crop Loads on Vine Growth and Fruit Quality of 'Jinok' Grape in Unheated Plastic House (포도 '진옥' 품종의 무가온 하우스 재배시 착과량이 수체생육 및 과실품질에 미치는 영향)

  • This study was conducted to investigate optimal crop loads of 'Jinok' grape for unheated plastic house culture. The crop loads of 'Jinok' grapes were managed to be 1.3, 1.8, 2.2, 2.4, and 2.6t per 10a from 2012 to 2014. We measured vine growth and berry qualities. Crop loads were not significantly affected on plant height, trunk diameter, shoot length, and the internode length of 'Jinok'. However, the maturation of berries was delayed when the crop load was higher. And the harvest date was earlier about three weeks in an unheated plastic house compared to in an open field. The average berry weight was decreased by the higher crop load although higher crop loads made higher yields showing the lowest weight at 342g with the treatment of 2.6t per 10a and the highest weight at 363g with the treatment of 1.3t per 10a. Also, the soluble solids content showed a tendency that higher crop loads brought to lower degree Brix. The contents of P, K, Ca, and Mg in grape leaves and shoots were not significantly different by crop loads. To sum up, when crop loads were under the 2.4t per 10a, the berries were harvested as a marketable fruit having $15^{\circ}Brix$ in the cultivar 'Jinok' grape. This result could help to increase grower's benefit having improved quality of fruit for the sustainable production by the established cultivation techniques for the newly developed cultivar 'Jinok'.

Effect of Nitrogen Fertigation by Soil Testing on the Growth and Yield of 'Campbell Early'(Vitis labrusca L.) Grapevine in Field Cultivation (노지 포도재배에서 토양검정시비량을 이용한 질소관비가 수체의 생육과 수량에 미치는 영향)

  • Optimum nitrogen fertigation level by soil testing was determined on the growth and yield of eleven-year-old 'Campbell Early' (Vitis labrusca L.) grapevine in a sandy loam soil from 2005 to 2007. Fifty percent of the annual application of the nitrogen rate (195 kg/ha/yr) was top-dressed as basal fertilizer in all treatments, and the remainders were drip-irrigated with fertigation rate at 25 (12.5% of total N, N 1/4 level of the remainder), 50 (25% of total N, N 1/2 level), and 100 mg/L (50% of total N, N 1 level) in intervals of twice (2.1 mm/times) a week for 12 weeks, and the effect of N drip fertigation was compared to control which the N remainder was applied with surface application as an additional fertilizer. The results showed that chlorophyll content reading in SPAD value and N contents of leaves increased as nitrogen fertigation level increased. Also observed was the growth of the internode and stem diameter of shoots which were longest at N 1/2 level among the treatments conducted both in 2005 and 2006. It was also noted that yield of the fruit was different every year, where average yield for three years was recorded highest in N 1/4 level, and lowest in N 1 level compared to control(surface application). Soluble solid content and titratable acidity of fruit juice were also not significant during the treatments, the maturation of fruits tended to be retarded in N 1 level. The study proved that N 1/4 (N 25 mg/L) levels of fertigation based on soil testing was most efficient in obtaining optimum yield and also, fertigation of grapevine at open field condition reduces the use of nitrogen fertilizer.

Studies on Improvement of Quality Wine of "Vitis amurensis RUPRECHT" ("개량머루" 과실주의 품질향상에 관한 연구)

  • New Wild Grape berry weighs approximately 3.7g, which is one fourth of the weight of the general grape wine (12.2g). The pH of New Wild Grape wine is around 3.34, which is lower by 0.3 than that (3.62) of general grape wine. It contains higher organic acids, so it has stronger sour taste. The sugar content of New Wild Grape is 17° Brix, which is higher than that (15° Brix) of general grape wine. The color of New Wild Grape is blackish, owing to the lower lightness, redness, and yellowness compared with those of general grape wine. Depending on the mixing with or without oak tree, there is little difference in the pH, organic acid, reducing sugar, and amount of alcohol after ripening for 1 year. In taste, New Wild Grape wine mixing with oak tree is evaluated better than that of the wine without oak tree. Especially, in the case of the wine mixing with oak tree's sawdust, the quality in color, taste, and smell became improved. After fermentation, the pH of New Wild Grape Wine ripened with oak tree charcoal increased from 3.82 to 3.86, as it gets more oak tree charcoal, and the organic acid decreased from 0.91㎖ to 0.86㎖. However, there is no difference in alcohol amount. Lightness in color tended to be reduced, whereas redness tended to be increased. Adding 3% of oak tree charcoal made the taste and smell improved. When fermenting New Wild Grape Wine with mixing 1% of oak tree's sawdust or 3% of oak tree charcoal, there is little difference in the pH and the organic acid, whereas there is a little difference in those of New Wild Grape Wine without addition of oak tree charcoal. As the addition of oak tree charcoal increased, the lightness and redness became higher. When fermenting New Wild Grape Wine with mixing 1% of oak tree's sawdust or 3% of oak tree charcoal in oak tree barre, the taste became improved.

Flower and Microspore Development in 'Campbell Early' (Vitis labruscana) and 'Tamnara' (V. spp.) Grapes ('캠벨얼리'와 '탐나라' 포도의 꽃과 소포자 발달)

  • The majority of cultivated varieties of grape have perfect flowers that are clustered in an individual inflorescence. Grape flower has a single pistil, five stamens, a protective flower cap (calyptra), and a calyx. After fertilization, an individual flower develops into a single berry. Although there are a number of reported studies focusing on berry formation, berry enlargement, and sugar accumulation in grape, the morphological studies of flower, including gametophyte morphogenesis and structural change in floral organs, have not yet been studied in detail. In this study, we investigated the flower structure and development characteristics of grape using microscopy and defined the floral development stages 9 to 13 based on microspore or male gametophyte development stage from tetrad to mature pollen. We used seeded diploid table grapes 'Campbell Early' (Vitis labruscana) and 'Tamnara' (V. spp.) as plant materials. At floral development stage 9, pollen mother cells develop to tetrads. During floral development stages 10 to 11, unicellular microspore develop to mid bicellular pollen. At the end of floral stage 12, male gametophyte develops to mature tricelluar pollen. In floral stage 13, the flower cap falls off and flower bud opens. During floral development stages 9 to 12, there were no major changes in calyx length, whereas the length of the flower cap continuously increased. The flower cap-to-calyx length ratio was 2.0, 3.0, 4.5, and 6.5 at floral stages 9, 10, 11, and 12, respectively. The flower cap-to-calyx length ratio was consistent in the two grape cultivars, suggesting that the ratio is a morphological character representing floral development stage. This study provides a reference for determining floral development stage of the two grape cultivars. It will be useful for the determination of optimum time for microspore culture needed to generate doubled haploid lines and appropriate gibberellic acid treatment needed to induce parthenocarpic fruit development in 'Tamnara' grape.

Development of the paper bagging machine for grapes (휴대용 포도자동결속기 개발연구)

  • The research project was conducted to develop a paper bagging machine for grape. This technology was aimed to highly reduce a labor for paper bagging in grape and bakery. In agriculture labor and farm population has rapidly decreased since 1980 in Korea so there was so limit in labor. In particular there is highly population in women and old age at rural area and thus labor cost is so high. Therefore a labor saving technology in agricultural sector might be needed to be replaced these old age with mechanical and labor saving tool in agriculture. The following was summarized of the research results for development of a paper bagging machine for grape. 1. Development of a new paper bagging machine for grape - This machine was designed by CATIA VI2/AUTO CAD2000 programme. - A paper bagging machine was mechanically binded a paper bag of grape which should be light and small size. This machine would be designed for women and old age with convenience during bagging work at the field site. - This machine was manufactured with total weight of less than 350g. - An overage bagging operation was more than 99% at the actual field process. - A paper bagging machine was designed with cartridge type which would be easily operated between rows and grape branches under field condition. - The type of cartridge pin was designed as a C-ring type with the length of 500mm which was good for bagging both grape and bakery. - In particular this machine was developed to easily operated among vines of the grape trees. 2. Field trials of a paper bagging machine in grape - There was high in grape quality as compared to the untreated control at the application of paper bagging machine. - The efficiency of paper bagging machine was 102% which was alternative tool for the conventional. - The roll pin of paper bagging machine was good with 5.3cm in terms of bagging precision. - There was no in grape quality between the paper bagging machine and the conventional method. - Disease infection and grape break was not in difference both treatments.

Difference of Starch Characteristics of Sweetpotato (Ipomoea batatas (L.) Lam) by Cultivated Regions (재배지역에 따른 고구마(Ipomoea batatas (L.) Lam) 품종의 전분 특성 차이)

  • We investigated out the effect of cultivation region on the physicochemical characteristics of starch in six sweetpotato cultivars. The following sweetpotato cultivars were analyzed: Daeyumi, Shingeonmi, Jinhongmi, Shinyulmi, Yulmi, and Yeonhwangmi. Samples were cultivated in Muan, Iksan, Nonsan, Boryeong and Hamyang. The soil texture was found to be sandy loam in Muan, Iksan, and Boryeong, sandy clay loam in Nonsan, and loam in Hamyang. The starch content of the sweetpotato was higher in Muan than in Hamyang. The amylose content was 22.3-30.9%, and the highest amylose content was found in samples from Iksan. Rapid viscosity analysis showed that the samples from Hamyang had the lowest values of pasting temperature, while samples of the Daeyumi cultivar had the highest values. Thermal analysis with a differential scanning calorimeter showed that the Muan samples had the highest values of onset temperature, maximum peak temperature, and completion temperature, and the samples from Hamyang had the lowest values. There was no difference between the cultivation regions or the cultivar in the X-ray pattern of the starch or its appearance in scanning electron micrographs. Therefore, the results of this study confirm that cultivation region and cultivar play an important role in determining the quality of sweetpotato and the physicochemical characteristics of sweetpotato starch.

Changes in Components of Salted Eggplants (Chukyang) during Storage (축양품종 염절임 가지의 저장중의 성분 변화)

  • This study was carried out to investigate the changes in components of salted eggplant (chukyang) during storage. The contents of total and reducing sugar were decreased during storage, and the tendency was lower in the salted eggplants of vacuum packing storage than in the salted eggplants of rice bran immersion storage. The organic acids of salted eggplants were acetic acid, citric acid, lactic acid, and malic acid. And acetic and malic acid contents were much higher than the others. The content of acetic acid was increased during storage, but malic acid was decreased. Major amino acids of eggplants were valine, aspartic acid, glutamic acid, glycine, and alanine. The content of total amino acid was 1,488.18 mg/100 g in eggplant packed vacuum film at 20 days of storage, and higher than those in fresh eggplant. The content of total amino acid in eggplant immersed in wet rice bran was 745.42 mg/100 g at 20 days of storage, and decreased during storage. The contents of aspartic acid, alanine, cystine, and proline in free amino acids of salted eggplants were higher than other amino acids. The contents of phosphoserine, taurine, ${\gamma}$ -aminoisobutyric acid and hydroxyproline were higher than others. Posphoserine content was decreased during storage, but ${\gamma}$ -aminoisobutyric acid content was increased during storage.

Effects of Antioxidant and Blood Flow Improvement of Grape Leaf Extract and Resveratrol from Vitis romaneti (남항(Vitis romaneti) 유래 포도잎 추출물과 Resveratrol의 항산화 및 혈행개선 효과)

  • The purpose of this study is to investigate the effects of 80% ethanol grape leaf extract (VGLE) and resveratrol (VGLR) from Vitis romaneti on antioxidant of red blood cells (RBC) of rat and efficacy of blood flow improvement in a rat model of topical ferric chloride ($FeCl_3$)-induced carotid artery damage. RBC oxidative hemolysis and plasma lipid peroxidation induced by the aqueous peroxyl radical generator 2,2'-azobis(2-amidinopropane) dihydrochloride were significantly suppressed by VGLE or VGLR in a dose-dependent manner. The $FeCl_3$ treatment seriously damaged the carotid artery: the walls of the artery and blood flow. However, VGLE or VGLR administration has ameliorated the blood flow and suppressed thrombus in blood vessels. These results suggest that VGLE or VGLR ameliorates the oxidative stress of RBC and thrombosis against blood vessel damage.

Occurrence of Bunch Rot Disease Caused by Aspergillus tubingensis on Shine Muscat Grape (Aspergillus tubingensis에 의한 샤인머스켓 포도송이썩음병(가칭)의 발생)

  • During the year 2018, the symptoms of bunch rot on Shine Muscat (Vitis vinifera L.) were observed in Kimcheon-si, Gyeongbuk province in Korea. The disease appears on the Shine Muscat as a black rot due to prolific fungal sporulation after it has invaded into the Shine Muscat which look completely empty and dryness. Colonies of these fungi are present on the Shine Muscat skin from fruit setting and increase in amount from early season to harvest, while become peak at ripening stage. To isolate the causal agent, small fragments (2 to 3 mm) of decayed tissue from the lesion margin were placed onto potato dextrose agar (PDA) plates. Fungal colonies on PDA produced dense white aerial mycelium and then covered with dark black conidial heads. These heads were large and radiate, and vesicles were globose (2.12-32.0×2.0-3.1 ㎛). Based on morphological and cultural characteristics, this fungus was identified as Aspergillus tubingensis. To confirm its identity, the internal transcribed spacer, β-tubulin, and RNA polymerase II was sequenced for molecular identification. BLAST search indicated 99% identity with A. tubingensis. The pathogenicity test on healthy grape of Shine Muscat produced bunch rot, as the original symptoms. To select effective fungicides for the control of brunch rot, an in vitro antifungal activity of seven fungicides were evaluated against the growth of A. tubingensis. Five fungicides (dipenoconazole, tebuconazole, metconazole, iminoctadine, and captan) exhibited significantly strong suppression of the mycelial growth of A. tubingensis.
