Effects of Dietary Supplementation of Yeast Pichia farinosa on Performance, Intestinal Microflora, and Fecal $NH_{3}$ Emission in Laying Hens (효모 Pichia farinosa의 첨가 급여가 산란계의 생산능력, 장내미생물 변화 및 분의 암모니아 발생량에 미치는 영향)

  • A feeding trial was conducted to study the effects of a live yeast, Pichia farinosa culture(PF), on the production performance and intestinal microflora in laying hens. One hundred and sixty ISA Brown layers, 21 weeks of age, were randomly allotted to four dietary treatments, with four replicates per treatment. Dietary treatments consisted of four levels (0, 0.1, 0.3, and 0.5%) of PF added to a com-soybean meal based diet. Egg production, egg weight, feed intake and fred conversion ratio(FCR) were measured. Egg qualifies were examined at 25th and 29th weeks of age. A metabolism trial was conducted following the feeding trial, during which intestinal microflora, nutrient digestibility and fecal NH3 gas emission were measured. Egg production of birds fed 0.1 and 0.3% PF were significantly higher than those from birds fed 0 and 0.5% PF(P<0.05). Daily egg mass of 0.3% PF increased significantly compared to that of 0% PF. There was no difference in egg weight among all treatments. Feed conversion ratio was significantly improved as the PF level increased. No significant difference was found in eggshell quality and Haugh unit at both 25 and 29 weeks of age. Viable count of ileal Lactobacillus spp. increased significantly as the rf level increased. However, the total number of yeast and anaerobes in ileum were similar among all treatments. Cecal Lactobacillus spp. and yeast counts showed no difference among all treatments. Fecal NH$_{3}$ gas emission of layers fed PF decreased significantly by the PF supplementation. From the result of this experiment, it could be concluded that dietary supplementation of the live yeast Pichia farinosa improves the laying performance and decreases the fecal ammonia gas emission.

The Changes of $\alpha$-galactosidase Activities and Stachyose and Raffinose Contents During Fermentation of Soybeans (대두의 발효에 따른 $\alpha$-Galactosidase활성 및 Stachyose, Raffinose 함량 변화)

  • Changes in the contents of stachyose and raffinose were determined during soybean fermentation. ${\alpha}$-Galactosidase activities were also monitored in soybean and its fermented products. The stachyose contents were 31.8239 mg/g of soybean, 4.2217 mg/g of Meju, and 2.1184 mg/g of Doenjang. The raffinose contents were 2.6914 mg/g of soybean, 1.7413 mg/g of Meju, and negligible of Doenjang. ${\alpha}$-Galactosidase activities was distinct in soybean and Meju. They were 14.5954 units/mg protein of soybean, 13.1489 units/mg protein of Meju, and 1.9157 units/mg protein of Doenjang. The results suggested that the decrease of stachyose and raffinose contents in fermented soy products were due to the ${\alpha}$-galactosidase activity.

  • Jet vane is an useful component which is installed at the end of a nozzle for the purpose of the posture control and the secure controlling stability during the initial launching of a rocket. During several seconds from its initial launching moment, the JV driving part is heated due to the direct contact of the vane with the combusted gas and the vane is ablated mechanically or chemically. In this study, as the fundamental study for the thermal analysis of jet vane, the heat transfer into a jet vane which is located in the uniform supersonic flow field is calculated. For this, boundary layer integral method and finite difference method are used simultaneously. Based on the thermal boundary conditions derived from the analysis, the transient heat conduction in the vane is also calculated.

  • This study was conducted to investigate the influence of feeding various Lactobacillus on production performance, nutrients digestibility, intestinal microflora, and fecal $NH_{3}$ gas emission in laying hens. Three hundred and sixty ISA Brown layers, 21 weeks of age, were randomly allotted to nine treatments, with low replicates per treatment. Nine treatments consisted of Control(no Lactobacillus), Lactobacillus crispatus avibrol(LCB: KFCC-11195), Lactobaciilus reuteri avibro2(LRB: KFCC-11196), Lactobacillus crispatus avihenl(LCH: KFCC-11197), Lactobacillus vaginalis avihen2(LVH: KFCC-11198). Each Lactobacillus was added at two levels ($10^{4}$and $10^{7}$ cfu/g diets). Egg production, and egg weight were measured daily. A metabolism trial was conducted following the 12-week feeding trial, during which egg qualities, intestinal microflora and fecal $NH_{3}$ gas emission were examined. Egg production and daily egg mass improved significantly by the addition of various Lactobacillus(P<0.05), of which effect was more notable during the latter part of the feeding trial. But, no significant differences were found among Lactobacillus strains and between two levels of supplementation. Egg weight and feed intake showed no difference among all treatments. Feed conversion ratio of birds fed lactobacillus was significantly improved compared to that of the Control(P<0.05), but not different among lactobacillus treatments. Digestibility of crude protein, ether extract and crude ash improved significantly in lactobacillus treatments(P<0.05). However, there were not statistically different by adding levels and strains. Total counts of Lactobacillus spp. in ileum of layers fed Lactobacillus were significantly higher than that of the control, but no consistent trend was found in cecum. There were no significant differences in intestinal yeast and anaerobes counts among all treatments. The Lactobaciilus supplementation did not exert my effect on the eggshell quality and Haugh unit. Fecal $NH_{3}$ gas emission decreased significantly in Lactobacillus treatments, and showed no difference between the two supplementation levels. From the result of this study, it could be concluded that dietary supplementation of Lactobacillus, regardless of their species, Improves the laying performance and decreases the fecal ammonia gas emission. The proper level of supplementation appears to be $10^{4}$ cfu/g of diet.

Impact of Feeding Multiple Probiotics on Productivity, Intestinal Microflora and Fecal Noxious Gas Emission in Broiler Chicks (혼합생균제 급여가 육계의 생산성, 장내 미생물 및 계분 유해가스 발생에 미치는 영향)

  • This experiment was conducted to investigate the influence of dietary supplementation of two different types of multiple probiotics in broiler chicks. Four hundred one day old male broiler chicks(Ross ${\times}$ Ross) were raised in the floor pen with five treatments(0, A: 0.10, 0.20%, B: 0.10, 0.20% of probiotics), consisting of 5 replicates for 5 weeks. Weight gain, feed intake and feed conversion were measured weekly, Total Lactobacillus, yeast, E. coli, Salmonella were examined from ileum and cecum at the end of experiment. Sera protein, cholesterol and ND vaccine titer were measured and fecal COD, CO$_2$ and NH$_3$ were detected in 24 hours after collection. Weight gain of chickes fed both types of probiotics increased for the first three weeks and showed significantly higher for the rest two weeks. Feed intake tended to be high in probiotics treatments compared with contro(P<0.05)1. Feed conversion improved significantly in B types of 0.2% probiotics compared with control. Total number of Salmonella was not consistent in ileum among treatments, whereas E. coli tended to be decreased in both types of probiotics compared with control. NH$_3$ and CO$_2$ emission of feces were decreased in both types of probiotics, but it was not statistically different(p>0.05). COD of chicks fed both probiotics was lower than control. There were no significantly different breast meat(P>0.05). Total cholesterol decreased in probiotics groups regardless of it's type. ND antibody vaccine titer was prone to be a small increments.

Effects of Packaging Treatment on Quality of Fresh-cut Mushrooms (Agaricus bisporus Sing.) during Storage (신선편이가공 양송이의 포장방법에 따른 품질변화)

  • The effects of packaging material and method on quality of fresh-cut mushrooms (Agaricus bisporus Sing.) were investigated in terms of weight loss, surface color, phenolics, vitamin C and sensory characteristics during storage at $5^{\circ}C$. The fresh-cut mushrooms were subjected to passive, gas exchange and vacuum packaging conditions at st. Polyethylene film (PE), polypropylene film (PP), anti-fogging film (AP) and pen orated film (PF) were used for the passive packaging. The mixed gas of $5\% CO_2/\;2\%O_2$ (MA1) and $10\%CO_2/\;2\%O_2$(MA2) were applied for the gas exchange packaging. The respiration rate of sliced mushroom was 1.27 times higher than intact mushroom at $5^{\circ}C$. Gas concentrations in the passive packaging were $1-2\%\;O^2\;and\;5-16\%m$ during storage of sliced mushrooms for 14 days at $5^{\circ}C$, and levels of the gases were different by the films used The mushroom in perforated film (PF) showed the highest weight loss of $4.56\%$. Anti-fogging film (AF) was somewhat effective for prevention of the weight loss compared with other films after 14 days storage. The mushrooms in MA1 and MA2 packages showed lower delta L value than in other films. PE packaging mitigated decrease of free and bound phenolics during storage. The mushrooms in MA2 kept better quality in sensory aspect, and then in MA1, PE and PP in order during storage at $5^{\circ}C$.

Comparison of Methane Production in Korean Native Cattle (Hanwoo) Fed Different Grain Sources (곡류 사료원별 육성기 한우 장내발효에 의한 메탄가스 배출량 비교)

  • Methane production during anaerobic fermentation in the rumen represents an energy loss to the host animal and induces emissions of greenhouse gases in the environment. Our study focused on comparison in methane production from growing Korean native steers fed different grain sources. Six Hanwoo steers (BW = $180.6{\pm}3.1$ kg) were fed, on a DM basis (TDN 2.80 kg), 40% timothy and 60% barley concentrate (Barley) or corn concentrate (Corn), respectively, based on the Korean Feeding Standards. Each period lasted 18 days including a 14-day adaptation and a 4-day measuring times. The steers were in the head hood chamber system (one cattle per chamber) during each measuring time to measure heat and methane production per day. Different grain sources did not affect digestibilities of dry matter, crude protein, crude fiber, crude fat, NDF, ADF and nitrogen-free extract. The mean methane concentrations per day were 202.0 and 177.1 ppm for Barley and Corn, respectively. Methane emission averaged 86.8 and 77.7 g/day for Barley and Corn, respectively. Methane emission factor by maintenance energy requirement for the growing steers fed barley based concentrate was higher than the steers fed corn based concentrate (Barley vs. Corn, 31.7 kg $CH_4\;head^{-1}\;yr^{-1}$ vs. 28.4 kg $CH_4\;head^{-1}\;yr^{-1}$). Thus, methane conversion rate was 0.065 (6.5%) and 0.055 (5.5%) for Barley and Corn, respectively.

가금에서 분리된 유산균의 생리적 특성 및 급여효과

  • These studies were conducted to evaluate the Properties of lactic acid Producing bacteria(LAB), isolated from broiler and laying hens cecum and select the optimum strains to improve the performance, environment of poultry house, immunity, and intestinal microflora of broiler and laying hens. In experiment I , 23 LAB strains were isolated from broiler and laying hens cecum as a colony form. Six strains were selected by acid tolerance, bile salt tolerance, viability, enzyme release, antagonism, and antibiotics susceptibility. In Experiment II, selected LABs from Ex. 1 were conducted to investigate the effects of feeding various Lactobacillus on performance, nutrients digestibility, intestinal microflora, villi development and observation of epithelium surface, blood chemicals and fecal noxious gas of broiler chicks. One thousand eighty one day old broiler chicks were fed into Lactobacillus crispatus avibrol(LCB), Lactobacillus reuteri avibro2(LRB), Lactobacillus crispatus avihen1(LCH), and Lactobacillus vaginalis avihen2(LVH) at the level of 10$^4$ and 10$\^$7/cfu/g diet. Weight gam of chicks fed Lactobacillus tended to increase from the first week and was higher from 50 to 100g in Lactobacillus treatments than control. Feed intake and feed conversion were not statistically different of all treatments. Dry Matter digestibility of Lactobacillus treatments was prone to improve compared to that of control, but was not significantly different. Protein and Ca digestibility were also tended to improve in Lactobacillus treatments relative that of control. Lactobacillus treatments showed improved tendency in crude ash and fat compared to those of control, whereas phosphorus digestibility was not consistency. Nutrients digestibilities of bird fed LCH were superior to those of other treatments, It showed significantly higher in Ca and P digestibility than control(P〈0.05). Total Lactobacillus spp. of birds fed various Lactobacillus was significantly higher in illeum for five weeks(P〈0.05), but was not different at cecum. Yeast was thought to be not completely attached to intestinal lumen for one week. However, total number of yeast was significantly increased in cecum and illeum of three weeks old chicks (P〈0.05). The number of anaerobes exhibited to tendency the increase in Lactobacillus treatments from one week old of age at both ileum and cecum.

Effects of Dietary Supplementation of Mixed Probiotics on Production Performance and Intestinal Environment in Broiler Chicken (혼합 생균제의 사료 내 첨가 급여가 육계의 생산성과 장내 환경에 미치는 영향)

  • This study was conducted to investigate the effects of dietary supplementation of the mixture of probiotics (MP) on growth performance, size of small intestine, cecal microflora and ammonia concentrations in broiler chicks. A total of 700, one-day-old male broiler chicks were randomly allotted to four treatments with seven replications having 25 birds per pen. The birds were fed one of the four experimental diets; containing no antibiotics nor MP (negative control, NC), containing antibiotics without MP(positive control, PC), negative control with MP 0.1% and negative control with MP 0.2% for 5 weeks. During overall experiment, birds in PC and MP treatments had higher final BW and daily BW gains than birds in NC treatments; however, the significance was not identified. The feed conversion ratio of the chicks fed the diet containing MP was significantly improved as compared to those fed the NC diets. The weights of jejunum were increased by the MP (p<0.05), but weights of duodenum, ileum and length of small intestine were similar among the groups. Birds in PC treatment showed lower populations of total microbes and lactic acid bacteria than other groups (p<0.05), cecal ammonia concentrations of the chicks fed the diet containing MP were significantly decreased as compared to those of NC and PC (p<0.01). In conclusion, MP added to the broiler diets improved the feed conversion rate and reduced cecal ammonia concentration.

Porduction and Enzymatic Characteristics of ${\alpha}-Galactosidase$ from Aspergillus niger (Aspergillus niger에 의한 ${\alpha}-Galactosidase$의 생산 및 효소적 특성)

  • ${\alpha}$-Galactosidase from Aspergillus niger as a possible enzyme for removal of flatulence factors in soybean foods was produced the highest in 120 hours in either Czapeck-Dox liquid medium or wheat bran solid medium. The most efficient carbon and nitrogen sources in Czapeck-Dox medium were raffinose and sodium nitrate, respectively, whereas the addition of the sources showed negative effects in wheat bran. pH optima for enzyme activity and stability were 4.0-5.0 and 3.5-6.5, respectively, and optimum temperature for stability was $40-50^{\circ}C$. Upon reaction on p-nitrophenyl-${\alpha}$-D-galactoside, Michaelis constant was 0.42 mM and maximum velocity was 152 ${\mu}moles$ substrate/minute/kg solid medium. Mercuric chloride acted as a strong noncompetitive inhibitor and p-chloromercuribenzoate, even in low concentration, acted as a competitive inhibitor. Crude ${\alpha}$-galactosidase hydrolyzed raffinose and stachyose completely, giving spots of monosaccharides only on thin-layer chromatogram.
