Analysis of characteristics of natural resins using organic residue analysis organic materials attached on wooden coffins from Singok-dong Site, Uijeongbu (유기잔존물 분석을 활용한 천연 수지 특성 분석 - 의정부 신곡동 유적 출토 목관 부착 유기물을 중심으로 -)

  • Organic residues analysis is an analysis method that reveals the types of organic material samples by using the characteristic that main components constituting substances are different depending on the species of animals and plants. In this study, scientific analysis of the organic residues attached to wood coffins in the Joseon Dynasty Hoemyo excavated from the site of Singok-dong, Uijeongbu was used to identify the types of remnants and to use them as information to restore the uses of organic materials and the way they lived in the past. As a result of FT-IR analysis of the residue attached to the inside of the wood, it was estimated to be a natural plant resin material. In addition, as a result of analysis by GC-MS to confirm the characteristic factors of natural resins, diterpenoids (abietane) and pimaran (pimarane), such as dehydroabietic acid and pimaric acid (diterpenoid) compounds, and saturated and unsaturated fatty acid components were detected together. Diterpenoid compounds are components mainly found in Pinaceae resins. It is confirmed in the literature that rosin, a representative material of Pinaceae resin, was used as an adhesive material. Considering the situation where an organic material remained at the joint of the wood, the organic material attached to the wood is judged to be an adhesive material made of Pinaceae resin. In addition, the fatty acid component detected together is a component derived from plant oil, and it is presumed to be made by mixing rosin and oil as recorded in previous studies. This study confirms that organic residues remain in the burial environment without losing their characteristics. It is expected that scientific analysis of organic residues will be conducted in the future to accumulate information necessary for the interpretation of past living culture.

Prehistoric subsistence and pottery use in the ancient Korean Peninsula: New evidence from organic geochemical analysis of potsherds (토기 내 잔존유기물을 활용한 한반도 선사·고대의 토기 사용과 식생활에 관한 연구)

  • This study focuses on the understanding of human subsistence and pottery use during ancient times on the Korean peninsula through lipid analysis of potsherds from several major prehistoric settlement sites. Ancient human subsistence has been one of the long-standing topics in Korean archaeology. However, since the high acidity of sediments does not allow long-term preservation of organic remains, we still lack some critical information related to the prehistoric diet. Pottery contains relatively well-preserved organic remains created during past cooking events. Though pottery is one of the most studied material cultures in Korean archaeology, almost no attention has been given to analyzing the pottery itself. This is a surprising omission and represents a serious gap in our understanding of prehistoric technology and subsistence. The analysis of ancient lipids extracted from the pottery matrix using GC-MS and isotope analysis can contribute to our understanding of the true nature of past subsistence strategies. Potsherd samples for the analyses in this study were collected from six prehistoric and early historic settlement sites located in the central part of the Korean peninsula. The results showed that subsistence strategies differed by both location and time period. For example, at Jungdo, an inland open-air Bronze Age settlement site in Chuncheon City, we were able to see the presence of terrestrial mammals. At Gahak-dong, Gwangmyeong City, marine resources were utilized, as the location of the site is not far from the coastline. At the early historic site of Guwol-dong, Incheon City, we were able to detect dairy products. The results of this study suggest that there was utilization of a wider range of resources among ancient dwellers in the central part of the Korean peninsula.

Cropping Systems for Vegetable Peanut and Environmental Effect of Residue Incorporation in Soil (풋땅콩 작부체계와 수확 후 잔존 유기물의 친환경적 효과)

  • A new demand for vegetable peanut (Arachis hypogaea L.) in Korea has increased farmers interest in growing vegetable peanut. Compared to grain peanut production, vegetable peanut production enables the growth period to be shortened by 20 or 30 days and farmers to adopt various cropping systems and to return crop residues in the soil. With the purpose of establishing desirable cropping systems for sustainable vegetable peanut production, three field experiments were conducted from 2000 to 2001 at Milyang, the southeastern part of Korea. Main focuses were given into the effect of cropping systems for vegetable peanut production on each crop's yield and soil sustainability. The cropping systems investigated were single vegetable peanut, peanut-radish-green barley, peanut-barley, and peanut-garlic cropping system, with or without crop residue incorporation in the soil. Among the cropping systems investigated for sustainable vegetable peanut production, peanut-only and peanut-radish-green barley cropping systems showed vulnerable to diseases and lodging while peanut-barley and peanut-garlic cropping systems showed higher stability in response to diseases and lodging, consequently leading to higher yield potential of vegetable peanut production. In the peanut-barley cropping system, both barley and peanut residues returned to the soil played an important role in soil improvement as well as in significantly increased grain yield of peanut and barley. A particular notice was taken to the pronounced increase in Trichoderma population and the amount of nitrogen mineralization induced by the returned barley residue. Soil structure, compactness, pH, and fertility were positively influenced by the returned crop residues, which apparently increased sustainability in vegetable peanut production systems.

Algal Contribution to the Occurrence of Refractory Organic Matter in Lake Paldang, South Korea: Inferred from Dual Stable Isotope (13C and 15N) Tracer Experiment (팔당호 난분해성 유기물에 대한 조류기원 유기물의 기여)

  • While a fairly large amount of organic matter is produced daily via phytoplankton photosynthesis in Lake Paldang, South Korea, knowledge of the role of algal-derived organic matter (OM) as a refractory OM source is not adequate. To understand the contribution of algal-derived OM to the refractory pool, biodegradation experiment and $KMnO_4$ oxidation experiment were conducted for 60 days using $^{13}C$ and $^{15}N$ labeled natural phytoplankton assemblage. The assemblage was collected from Lake Paldang on May 20, 2010. The photosynthetically produced total organic carbon ($TO^{13}C$), particulate organic carbon ($PO^{13}C$), and particulate nitrogen ($P^{15}N$) remained at 26%, 20%, and 17% of the initial concentrations, respectively, in the form of non-biodegradable organic matter. In addition, 12% and 38% of $PO^{13}C$ remained after $KMnO_4$ treatment on Day 0 and 60, respectively. These results indicate that photosynthetic products could be an important source of refractory organic matter after microbial degradation. Moreover, the microbially transformed algal-derived OM could contribute to the oxidation rate of the chemical oxygen demand.

Analysis of organic Residues from Open-Air Hearths at Nambuk-dong, Yongyu-do (용유도 남북동유적 야외노지에 대한 잔존지방분석)

  • The archaeological site of Nambuk-dong, Yongyu Island, Incheon which goes back to the Neolithic making it difficult to identify character of site, To get information for identifying utility and character of Open-Air Hearths within the finds, an organic residues analysis was conducted on soil and stone sample, sterol was analysis to determine the originality of archaeological sample, then percentage of fatty acid was identify the species of sample. As a result, it is assumed that No. 9, 10, 12 finds once had shellfish-related materials remain, No. 32 find seemed to have animal materials, No. 42, 43 finds were related to plant. In conclusion, it seems that open-air hearths of Yongyu Island had utility mainly related to making seafood.

Analysis of adhesive material for joining pottery fragments excavated from Duurlig Nars, Mongolia (몽골 도르릭나르스 유적 토기의 접합에 사용한 물질 분석)

  • The adhesive material was found for joining pottery fragments from Duurlig Nars, Mongolia estimated in AD 1C. In this study, analysis of natural substance for joining fragments of pottery was performed using gas chromatograph and mass spectrometer. As a result, it was identified triterpenoid substances, such as lupeol and betulin which were known to constituents of birch bark tar. It was suggested that Mongolian used adhesives made by birch bark tar for joining pottery fragments. Therefore if organic materials of ancient objects are systematically researched, it can provide significant evidence related to the way of life of ancient people.

Organic Residues Analysis of Oil Bottle of Goryeo Dynasty Excavated from the Soejeoul Site, Geumneung-dong, Chungju (충주 금릉동 쇠저울유적 출토 고려시대 유병의 유기물 분석)

  • Organic residues are substances derived from diverse natural sources. Recent scientific analysis of organic residues has yielded important information in restoring the lifestyles of ancient peoples. In this study, the organic material contained within the celadon oil bottle of the Goryeo dynasty, excavated from the Soejoul site in Geumneung-dong, Chungju, was analyzed using Fourier-transform infrared spectroscopy (FT-IR) and gas chromatograph-mass spectrometer (GC-MS). The results showed that the organic materials in the bottle were plant-derived oils. In particular, polyunsaturated fatty acids and phytosterols were detected using GC-MS analysis. Sesamin components were also identified. Sesamin, which is a characteristic component of sesame seeds, is a lignan and an antioxidant. As the organic residues in the oil bottle were derived from sesame seeds, it is presumed that sesame oil was stored in the bottle.

Changes of the Soil Physic-Chemical Properties and Rice Productions with Organic Materials used in Organic Culture (벼 유기재배에 있어서 유기자재처리에 따른 토양 및 수량 특성 변화)

  • 벼 유기재배시 토양양분공급용으로 이용되고 있는 유기자재(금수강산골드)를 대조로 하고 식물성유기 자재(쌀겨팰렛), 동물성유기자재, 식물성과 동물성이 혼합된 유기자재를 질소 성분량(7kg/10a)을 기준으로 하여 이앙 20일전에 전량 기비로 시비하고 경운한 다음 동진1호를 시험품종으로 하여 2년연속 유기 자재와 벼를 재배하면서 일어나는 토양의 이화학적 특성과 벼 생육 및 특성의 변화를 시기별로 조사하였다. 시험 전 토양의 화학성은 전반적으로 유기물은 높고 인산함량은 매우 낮은 조건의 토양이었다. 관행유기자재(금수강산골드)는 20일경에 50% 무기화율을 보였으나, 식물성자재 40~60일경, 동물성자재와 혼합자재(식물성+동물성)는 60~80일경에 47~52% 무기화 정도를 나타내 식물성 자재의 무기화 속도가약 20일정도 빨랐다. 토양 중의 유기물 잔존함량은 식물성자재 > 혼합자재 > 동물성자재 > 관행 순이었으며, 토양 중의 전 질소 잔존함량의 경우 관행유기자재는 처리초기부터 빠르게 감소하는 특성을 보이나, 식물성자재와 혼합 자재는 시비초기와 거의 비슷한 수준을 유지하였고, 동물성 자재는 서서히 감소되는 경향이었다. 토양 물리성은 액상과 공극율 다소 증가되는 경향이었으며 식물성과 혼합유기자재처리구가 컸으며, 토양 유효 입단 형성력에 있어서도 유사한 경향이었다. 벼 수량 특성은 관행유기자재보다 1년차에는 3~9%의 낮았으나, 2년 연속처리를 할 경우 관행유기자재를 처리할 때와 동일한 생산성을 기대할 수 있었다. PME와 $\beta$-Glucosidase의 효소활성은 관행유기자재 < 식물성자재 < 동물성자재 < 혼합유기자재의 순으로 높은 경향을 볼 수 있었다.

Assessment of Inactivation for Campylobacter spp. Attached on Chicken Meat (계육에 오염된 Campylobacter 균의 불활성화 평가)

  • The inactivation efficiency of Campylobacter jejuni were assessed in vitro and in vivo using confocal laser microscopy and flow cytometry. C. jejuni cells were inactivated with $1\%$ (w/v) trisodium phosphate (TSP) and the live cells and inactivated cells were distinguished by staining with LIVE/DEAD BacLight Bacteria Viability fluorescent probe. After treatment of TSP for 5 min, most of C. jejuni cells turned to coccoid form from original spiral shape. C. jejuni cells lost total cell viability in the absence of organic nutrients but did not lost total cell viability in the presence of organic nutrients. In vivo test, C. jejuni cells turned to viable but non-culturable (VBNC) form after TSP treatment and remained alive on chicken skin. C. jejuni cells attached on chicken meat would transform to coccoid form by sanitizer treatment, but could possibly be alive by the benefits of organic nutrients present in chicken meat.

Studies on Establishment of Application Method of Organic Matter in Rice-Barley Cropping System (벼-보리 작부지대(作付地帶) 유기물연용시용법(有機物連用施用法) 연구(硏究))

  • The fertilizer experiment was conducted to establish a pattern of a continuous application of organic matter in rice-barley cropping system. Fine textured gray lowland soil was used to grow the crops. The amount of nitrogen absorption and the yield of the crops were the highest in barnyard manure 1,000kg/10a. A correlation among the content of humus and nitrogen, the amount of nitrogen absorption, and the yield was significant while it was not significant among the the nitrogen content of the soil fertility, the total nitrogen, and the yield of crops. The result of this research suggests that revelation of available nitrogen after the decomposition of the organic matter depending on climate and regions should be studied for the organic matter application in due season.
