A edible mushroom, Hypsizygus marmoreus is commercially cultivated. However, the researches of cultivation and physiological characteristics were not conducted in Korea. In this study, we conducted on artificial cultivation of H. marmoreus and elucidated the effect of light on the mycelial growth and fruit body development using LED light sources with different wavelength; blue (peak wave length 460nm), green(peak wave length 530nm), yellow(peak wave length 590nm), red(peak wave length 630nm), and white as positive control. Mycelial growth of H. marmoreus strains were inhibited about 30~40% in inhibition ratio under the illumination with blue, green, yellow LED light. However, red LED light was not inhibited. Elongation of stipe was effective under the long wave length such as yellow and red light. Abnormal fruit body was produced under the long wavelength and dark. However, development of pileus was effective under the short wavelength such as green and blue light. Also, as a result of cultivation with mixed light for high quality and harvest, many effective numbers and yields of fruiting bodies were obtained in the mixed treatment of blue and white light, and pileus developed well.

This study was performed to develop new Hypsizygus marmoreus cultivars that have enhanced functional materials and improved physiological characteristics with mutagenesis by gamma ray irradiation. Protoplasts of H. marmoreus brown strain HYM-056 were irradiated by gamma ray for mutagenesis, and then 2,000 clones of mutants were randomly selected and the fruiting bodies were induced by bottle culture. Among them, 157 isolates with fast-growing, heavy and many fruiting body-producing were selected. The isolates were cultured in plastic bottle containing rice bran, barley hulls and fir sawdust to form the fruiting bodies. About 100 days after inoculation, characteristic of fruiting bodies were investigated. The isolates were divided into 6 groups based on color, shape and size of pileus, and length, diameter, number and weight of stipe. In addition, the genetic variation of the isolates was analyzed by URP-PCR fingerprinting.
This study was conducted to established method of culture for Paecilomyces japonica using an egg. Mycelia grew favorably at the temperature of 22~26$^{\circ}C$ on eggs. 5.1g of dry matter basis(average 7.2cm of longer and 199.6 of numbers) of artificial fruiting bodies were harvested at 60 days after inoculation from one of egg. Commercial fruiting bodies of Paecilomyces japonica from silkworms was used for comparative nutriental contents. Cordycepin contents of fruit bodies of Paecilomyces japonica cultivated on eggs and silkworms were not significantly different. Crude fat contents of fruiting bodies of Paeilomyces japonica cultivated from eggs was significantly higher than from silkworms(P<0.05). Mn and Cu contents of fruiting bodies of Paecilomyces japonica cultivated from silkworms were significantly higher than from eggs(P<0.05), but Na, Mg, Fe and Zn contents were significantly higher from eggs(P<0.05). Glycine, Arginine and Proline contents in the fruiting bodies of Paecilomyces japonica cultivated from silkworms were tend to higher than from eggs, but Serine, cystein. methionine, isoleucine and phenylalanin were tend to higher from eggs. These results were made possible that possible mass production of artificial fruiting bodies of Paecilomyces Japonica cultivated on eggs.
This study was performed to obtain basal data for development of new functional food by using mushroom Pholiota spp.. Among several kinds of extracts, water extracts of Pholiota adiposa ASI 24027 showed the highest solid yield of 58.7%. Crude protein of Pholiota adiposa ASI 24015 and ${\beta}$-glucan contents of Pholiota adiposa ASI 24005 were 25.0% and 0.66%, respectively. K and Mg were also contained plentifully in the fruiting body of Pholiota adiposa ASI 24004 (3,649 mg and 138 mg/100g dried fruiting body, respectively). Amino acid and total fatty acid contents were similar between almost all of Pholiota spp..
Interspecific heterokaryons were obtained between auxotrophic mutants of Pleurotus ostreatus and Pleurotus florida by polyethylene $glycol-Ca^{++}$ induced fusion of somatic protoplasts. The fusion products produced colonies of dense growing mycelium after 10-14 days culture on hypertonic Mushroom Minimal Medium. When they were transferred to Mushroom Minimal Medium plates, the sectors showed normal vegetative morphology but the colonies of irregular shape varied in growth rate. All of the colonies produced fruit body of normal pilei. Some heterokaryon colonies bearing none or only a small amounts of basidiospores were isolated. Fusion products, generally, gave higher basidiocarp yields than the parents.
To compare the morphological and cultural characteristics of Cordyceps militaris, forming conditions of artificial fruiting body production were investigated in this experiment. The stroma shapes of artificial fruiting body were confirmed to club and/or stick. However, both shapes were same color as a orange. Perithecia of stroma was semi-egg shape and was ranged from $30{\sim}90{\times}90{\sim}130\;{\mu}m$ and it's asci was narrowly cylindric. Ascospore of perithecia was belong to filiform and multiseptate. The conidia of C. militaris was centurally grown and shaped with globose, long clavate type, floccose, centrally raised during anamorph stage. When it was cultured between glass and PP bottle, production of artificial fruiting body, pinheading ratio and total yield were higher in PP bottle. The optimum illumination was 1,000 lx for the those production. The culture medium of barley, wheat and hulled rice showed higher artificial fruiting body compared to that of silkworm. Pinheading and yield of it's isolates was decreased at more than three subsequent transculture.
Myxococcus xanthus is a Gram negative, rod-shaped, soil bacterium that displays a social behaviors, and multicellular development upon nutrient deprivation. The csgA gene encoding a cell surface protein is essential for developmental behaviors including rippling, aggregation, fruiting body formation and sporulation. csgA mutants show normal vegetative growth, but lack all these developmental phenotypes. Expression of the CsgA (C-signal) specific genes are eliminated or dramatically reduced in csgA mutants. In order to identify components of C-signal transduction pathway, second site mutations were introduced into csgA mutants and were identified which can fully or partially restore development of csgA mutants (Rhie, H. G. et. al. 1989. J. Bacteriol. 171, 3268-3276). One of such csgA suppressor mutations, socD500 restores only sporulation to csgA mutants at 15$^{\circ}C$. The socD500 mutaion however eliminates the three basic developmental requirements, starvation, high cell density and a solid surface. Only sporulation, not accompanied with fruiting body formation is induced simply by shifting the temperature of vegetatively growing cells from $32^{\circ}C$ to $15^{\circ}C$. Spores induced by socD500 mutation is not as thick as that of wild-type fruiting body. In socD500 genetic background, two of ten C-signal dependent genes, $\Omega$DK4506 and $\Omega$DK4406 are more highly expressed in growing cells at $15^{\circ}C$. These results indicate that the socD500 mutation may be partly involved in the regulation of expression of two C-signal dependent genes and genes for sporulation in this transduction pathway.

The effect of the nitrogen content of sawdust medium (1.2~1.8%) and the restriction stage temperature (2, 4, and 6℃) on the growth and storage characteristics of Flammulina velutipes (winter mushroom) were investigated. With increased nitrogen content, the growth days shortened and the yield of the fruiting body increased. The effect of restriction temperature on the growth of the fruiting body differed depending on the nitrogen content. No difference in restriction temperature was evident for T1 with a low nitrogen content of 1.28%. In medium with a nitrogen content ≥1.55%, the yield and length of the pileus and stipe increased as the restriction temperature decreased. The weight loss ratio according to the storage period did not change according to the nitrogen content in the medium. A low weight loss ratio of 1.50 to 1.93% was observed at a restriction temperature <4℃. When T3 with high nitrogen content in the medium was treated at a restriction temperature of 4℃, the fruiting body color values after 31 days of storage were 84.81 (L) and 6.3 (ΔE). This color change was minute compared to that after other treatments. The sensory characteristics score was 5.2 after 31 days of storage, and the quality remained acceptable for consumption.
Ribonucleic acid contents and mononucleotides distribution from the mycelium and fruit bodies of Ganoderma lucidum were studied. P.E.I. cellulose TLC and HPLC were applied in this study. The obtained results are as follows; The levels of ribonucleic acids from the young basidiocarp mycelium were higher than those of mature basidiocarp. Guanosine 5'-monophosphate and xanthosine 5'-monophosphate were found in both young basidiocarp mycelium and mature basidiocarp. The levels of guanosine 5'-monophosphate and xanthosine 5'-monophosphate from the young basidiocarp were higher than those of the mature basidiocarp. However, inosine 5'-monophosphate was not detected.
Isozyme band patterns were investigated by isoeletric focusing Esterase and Leucine amino peptidase of Pleurotus spp. in Korea. Esterase patterns of mycelia and fruitbody were distinquished. However, those of primordia, cap and stem were similar. Interspecies differences of Pleurotus ostreatus, P. cornucopiae and P. florida of Esterase zymogram were found. Species identification by electrophoretic zymogram may be a role as an additional taxonomic tool.
