Quality Characteristics and Antioxidant Effects during Makgeolli Fermentation by Purple Sweet Potato-rice Nuruk (자색고구마-쌀누룩으로 제조된 막걸리 발효 중 품질 특성과 항산화 효과)

  • Makgeolli is the oldest Korean traditional wine made from rice, and contains 6-8% alcohol. Also, nuruk is traditionally prepared by exposing moistened ground wheat or rice to the air, and there contained airborne microorganism. In this study, purple sweet potato (PSP) was used as a raw material in the rice nuruk- and makgeolli-making processes. PSP rice nuruk (RN) was made with rice and 0-10% (w/w) PSP, and the characteristics of makgeolli were studied during fermentation. The pH during fermentation decreased from 4.27 (0% PSP-RN), 4.59 (2.5%), 4.50 (5.0%), 4.46 (7.5%), and 4.43 (10.0%) of initial fermentation day to 4.12, 3.82, 3.67, 3.71, and 3.66, respectively, at day 7. The sugar contents increased from $0.4^{\circ}Bx$ of the initial day to $8.0-8.4^{\circ}Bx$, and the levels of alcohol increased to 13.0% (0% PSP-RN), 13.2% (2.5%), 12.8% (5.0, 7.5, and 10.0%) by day 7, respectively. In addition, the contents of soluble anthocyanins and phenolics increased from 0.001-0.121 and 0.2-0.27 g/L on the initial fermentation day to 0.002-0.166 and 1.65-1.87 g/L, corresponding to increases in levels of antioxidant activities of 20-50%, respectively.

Quality Characteristics of Puffed Snacks (ppeongtuigi) with Purple Sweet Potato Flours Using Different Puffing Conditions (자색 고구마 분말을 첨가한 팽화과자(뻥튀기)의 팽화 조건에 따른 품질 특성)

  • The effect of puffing conditions on the physical properties and sensory evaluations of a puffed snack (ppeongtuigi) made of purple sweet potato flour was investigated. The snacks consisted of coated artificial rice with purple sweet potato flour with water, tempered to 7%, 9% and 11% moisture. The coated material was puffed at 236, 241, or $246^{\circ}C$ for 4, 5, or 6 s. The $L^*$ value decreased as the heating time increased. The $a^*$ and $b^*$ values, specific volume, and hardness increased with increasing heating temperature and heating time. Puffed snacks produced under conditions of higher moisture (7%), higher heating temperature ($246^{\circ}C$), or longer heating time (5 s) showed high scores for appearance, color, taste, and overall acceptability in sensory evaluations.

Effect of High Voltage Pulsed Electric Fields on Extraction of Purple Sweet Potato Pigment (자색 고구마 색소의 추출에 미치는 고전압 펄스 전기장의 영향)

  • High voltage pulsed electric fields (PEF) is a promising technology for the nonthermal extraction of effective components from biological materials. Plant cells were ruptured with PEF at ambient or refrigerated temperature for a short treatment time of second or microsecond. Treatments of coarsely ground purple sweet potato (PSP) with PEF(30 kV/cm, 500 Hz) resulted in maximum extraction yield of 65% as compared with 45-50% for control. An increase in electric field strength (from 10kV/cm to 35kV/cm) and frequency (from 100Hz to 500Hz) resulted in increased amount of extracted pigments, but treatment time is not affected on pigment extraction. Starch granules were not detected and large intracellular spaces were visible between the cells on light and scanning election microscopy of PEF treated PSP. This result suggests that PEF have potential to use on extraction of pigments from plant cells.

Concentration of Pigment Extracted from Purple Sweet Potato by Nanofiltration (막분리에 의한 자색고구마 색소의 농축)

  • Purple sweet potato pigment extract was concentrated using both membrane separation method and vacuum concentration method. The pigment extract (anthocyanin content 1.6 g/L) was concentrated $({\times}25)$ after 5 hr of continuous operation of a nanofiltration to get anthocyanin content of 10.6 g/L. Total solid content also increased continuously while the flux decreased continuously during the concentration process. Degradation index (DI) changes of concentrated pigment solution were insignificant during the whole concentration process which is indicating that the nanofiltration method does not affect color degradation of anthocyanin pigment. For the comparison test, the same pigment extract was concentrated using a rotary vacuum evaporator at temperatures of 40 and $60^{\circ}C$. At both temperatures, pigment content increased in a similar manner during concentration $({\times}5)$. However, DI value at $60^{\circ}C$ increased while that at $40^{\circ}C$ did not change appreciably. Total color difference value changed only slightly by nanofiltration and $40^{\circ}C$ while changed significantly by $60^{\circ}C$. These indicate that a membrane filtration method is more effective in concentrating purple sweet potato pigment extract than a vacuum concentration method by high temperature.

Antioxidant Activities of Ipomoea batatas L. Lam. (Purple Sweet Potato) Extracts Cultured in Korea (국내산 자색고구마(Ipomoea batatas L. Lam.) 추출물의 항산화활성 평가)

  • In this study, the antioxidative effects for the extracts of purple sweet potato (Ipomoea batatas L. Lam.) were investigated. The purple sweet potato was extracted with 70% ethanol and the ethyl acetate fraction was obtained from the extracts. The yields of extract and ethyl acetate fraction were 39.2% and 3.49% per dried powder, respectively. To confirm the antioxidative effects of the extracts, free radical scavenging activities (1,1-diphenyl-2-picrylhydrazyl), total antioxidant capacity by luminol-dependent chemiluminescence assay and the protective effects against reactive oxygen species (ROS) in erythrocytes were measured. Free radical scavenging activities ($FSC_{50}$) of the 70% ethanol extract and ethyl acetate fraction were $90.16{\mu}g/mL$ and $7.69{\mu}g/mL$, respectively. The free radical scavenging activity of ethyl acetate fraction was higher than that of (+)-${\alpha}$-tocopherol ($8.98{\mu}g/mL$). Total antioxidant capacities ($OSC_{50}$) of the 70% ethanol extract and ethyl acetate fraction were $5.75{\mu}g/mL$ and $1.92{\mu}g/mL$, respectively. The capacity of ethyl acetate fraction was similar to L-ascorbic acid, known as a prominent water soluble antioxidant ($1.50{\mu}g/mL$). The cellular protective effects of the ethyl acetate fractions on the $^1O_2$-induced cellular damage of human erythrocytes were increased in a concentration dependent manner ($5{\sim}50{\mu}g/mL$). The ${\tau}_{50}$ value in $5{\mu}g/mL$ was 45.6 min which was higher than that of (+)-${\alpha}$-tocopherol in all concentrations. These results indicate that the ethyl acetate fraction of purple sweet potato (I. batatas) has the excellent antioxidative capacity and could be applicable to anti-aging cosmeceutical ingredients for skin aging inhibition.

Acrylamide content and quality characteristics of sweet potato chips using different cultivars (품종에 따른 고구마 칩의 아크릴아마이드 함량과 품질 특성)

  • To select suitable cultivars to produce sweet potato (SP) chips instead of potato chips, ten Korean cultivars, Sinjami, Yeunjami, Sinhwangmi, Juhwangmi, Sincheonmi, Sinyulmi, Gunhwangmi, Dahomi, Daeyumi, and Pungwonmi were used. The acrylamide content, quality characteristics, and oxidation stability of SP chips were investigated. Acrylamide content was shown to be under 0.48 mg/kg in case of all cultivars except Yeunjami (1.07 mg/kg). The color values significantly differed among the cultivars and were maintained like those of raw roots. The hardness, brittleness, and fracturability of Juhwangmi and Sinhwangmi chips showed the lowest values, while those of Daeyumi were the highest. According to the sensory evaluation data, the quality characteristics of chips were influenced by not only hardness, fracturability, and crunchiness, but also the flavor taste. Pungwonmi chips showed the highest score, followed by Gunhwangmi and Dahomi chips. Therefore, Pungwonmi, Dahomi, and Gunhwangmi are suggested as suitable cultivars to make SP chips, because of their high overall quality and very low acrylamide content.

Changes in the Physiological Activities of Four Sweet Potato Varieties by Cooking Condition (고구마 네 가지 품종의 조리방법에 따른 생리활성 변화)

  • The present study was performed to investigate antioxidant, anticancer, and antimicrobial activities of four Korean sweet potato variaties and to identify the changes in these biological activities under different cooking conditions. Total polyphenol content was 3.8-73.6 mg/g in 80% ethanol extracts of sweet potatoes. The polyphenol content was highest Sinjami variety (p < 0.05). Radical scavenging activity against DPPH and $ABTS^{{\cdot}+}$ was high in Sinjami (p < 0.05) and the ethanol extract from Sinjami also showed effective superoxide dismutase (SOD)-like activity, which decreased significantly by steaming and roasting (p < 0.05). Ethanol extracts from the four sweet potato variaties did not inhibit cancer cell growth in MCF-7 or HepG2 cells at concentrations of 1, 10, and $100\;{\mu}g$/mL. Of the investigated sweet potato variaties, only Sinjami exhibited strong antimicrobial activity against Escherichia coli, Staphylococcus aureus, and Salmonella typhimurium. The antimicrobial activity of Sinjami against E. coli, St. aureus, and S. typhimurium decreased following steaming and roasting (p < 0.05). These results indicate that the Sinjami Korean sweet potato had higher polyphenol content, radical scavenging activity, SOD-like activity, and antimicrobial activity than those of the other variaties and consuming raw Sinjami might be beneficial for maintenance of biological activities.

Antioxidant Activities of Extracts Prepared from Sweet Potatoes with Different Flesh Colors (육질색 종류별 고구마 추출물의 항산화 활성)

  • Antioxidant activities of ethanol extracts from three types of sweet potatoes with distinctive flesh color (white, orange, and purple) were investigated. Total phenolic content of purple-fleshed sweet potato (PF) was always 2- to 5-fold higher than those of white-fleshed (WF) and orange-fleshed sweet potato (OF). 2,2-diphenyl-1-picryl-hydrazyl free radical scavenging activity of 70% ethanol extracts was highest ($EC_{50}$ = 0.179 mg/mL) for PF and lowest ($EC_{50}$ = 2.127 mg/mL) for OF. And then, reducing power was also highest ($EC_{50}$ = 0.236 mg/mL) for PF and lowest ($EC_{50}$ = 1.587 mg/mL) for WF. Antioxidant activities of 70% ethanol extracts were closely related to the amount of total phenolics. These results imply that antioxidant activity of PF is 6.7 to 11.9 times higher than those of WF and OF.
