Amount of Telomeric DNA on Pig Lymphocytes by Quantitative Fluorescence in situ Hybridization (양적형광접합보인법(Q-FISH)에 의한 돼지 백혈구 세포의 텔로미어 함량 분석)

  • The amount of telomeric DNA was quantified across different breeds(Landrace, Duroc, Yorkshire and Berksire), at different ages(90 days old and 180 days old) and among sexes(male and female) in pigs raised at the Performance Testing Station of Korea Swine Association, Jinkyo, Korea. The telomeric DNA amount was quantified by Quantitative Fluorescence In Situ Hybridization(Q-FISH) using a porcine telomeric DNA probe on interphase nuclei of lymphocytes. Analysis revealed that the amount of telomeric DNA on the pig lymphocytes was found to decrease with age. The quantity of telomeres significantly differed among breeds at 90 days of age. The colored breeds such as Berkshire and Duroc had higher amount of telomeric DNA than the Yorkshire and Landrace breed. In addition, the amount of telomeric DNA in male lymphocytes was significantly higher than that of females. In the correlation coefficients between the telomere quantity and their productive traits; average daily gain, loin percent and index value were positively correlated, whereas body length, feed efficiency and back fat thickness correlated negatively. However, the correlation coefficients were very low and not significant. Therefore, this study suggests that the amount of telomeres on lymphocytes can be considered as a physiological marker but not as a productive marker in pig.

Effects of Feeding High Quality Roughage (Timothy Hay) during Growing Period on Growth Performance and Carcass Characteristics of Hanwoo Steers (육성기의 양질조사료(티머시 건초)급여가 거세한우의 성장 및 도체특성에 미치는 영향)

  • Effects of feeding high quality roughage (timothy hay) in growing period on growth performance and carcass characteristics during 24 month (5 month from 28 month to) in twenty of Hanwoo steers were investigated. The hanwoo steers were randomly allotted to 2 treatments (10 heads/treatment) ; rice straw group (control) and timothy hay group (treatment). The final body weights of rice straw group and timothy hay group on 28 month were 667.70 kg and 681.75kg, respectively. The daily gain of total period was slightly higher in timothy hay group (0.83kg ) than in rice straw group (0.81 kg). The daily roughage intake in the growing period was tended to be higher in timothy hay group (4.27kg ) than rice straw group (3.80 kg). The carcass grades (yield grade and quality grade) and Longissimus dorsi area were significantly higher in timothy hay group ($85.62 cm^2$, 2.02 and 2.20 point) than in rice straw group ($81.29 cm^2$, 2.00 and 1.60 point respectively) (p<0.05), and were affected by the feeding with high quality roughage in the growing period.

Effects of High Energy Density Diet on Pork Quality (고에너지 수준의 사료급여가 돈육 품질에 미치는 영향)

  • In a trial involving 120 pigs, the growth performance of finishing pigs and proximate composition, physicochemical properites and fatty acid composition of loin muscle were investigated by feeding the high-energy density diet and low-energy density diet. The treatments included feeding 1) the low-energy density diet (3,290 cal/kg DE, 14.50% CP, 0.70% lysine and 0.78% Ca) for 38 days and 2) the hight-energy density diet (3,350 cal/kg ME, 15.0% CP, 0.70% lysine and 0.78% Ca) for 27 days. The average daily gains were significantly higher for pigs fed the high-energy density diet than those fed the low-energy density diet, but average daily feed intake and feed conversion rate were significantly lower than those fed the low-energy density diet (p<0.05). The cooking loss and yellowness of pork were significantly (p<0.05) lower for pigs fed the low-energy density diet than those fed the high-energy density diet. There were no significant differences in carcass characteristics, proximate composition and fatty acid composition of pork. In conclusion, the results of the experiment suggested that the high-energy density diet for pigs tended to improve the growth performance, but the cooking loss and yellowness of pork were higher for pigs fed the high-energy density diet than those fed the low-energy density diet.

Effects of Corn Silage on Growth Performance, Carcass Characteristics, and Gene Expression of Longissimus dorsi Muscle Biopsy of Hanwoo Steers (옥수수 사일리지가 거세한우의 성장, 도체특성 및 등심 유전자 발현에 미치는 영향)

  • This study was conducted to investigate effects of corn silage on final fattening performance, carcass characteristics, and gene expression of Hanwoo Longissimus dorsi muscle biopsy. Twenty one steers with initial body weight of control 291.5±6.5kg, corn silage 291.7.0±17.3kg were used for 18 months of fattening period. Average daily gain of corn silage tended to increase compared to control in early fattening period(p=0.092). Feed conversion ratio of corn silage was higher than control in early fattening(p=0.005). The animals in corn silage increased A grade 23% in meat quantity than the control. Myogenic gene expression on the Longissimus dorsi biopsy were compared between corn silage and control. The level of myosin heavy chain(MHC) I, IIX mRNA were greater than the control in the whole period(p<0.05). The level of Peroxisome proliferator-activated receptor γ (PPAR γ) mRNA was greater than the corn silage(p<0.05). Inconclusion, corn silage will be possible to use as an alternative concentrate feeding system.

Studies on the Meat Production and Woolskin Processing of Sheep and Korean Native Goats for Increasing Farm Income as a Family Subsidiary Work (농가부업(農家副業)의 소득향상(所得向上)을 위한 양육생산(羊肉生産) 및 모피가공(毛皮加工)에 관(關)한 연구(硏究))

  • The purpose of the study was to find out possible ways for increasing farm income through the sheep and Korean native goats farming, and to investigate meat productivity, wool productivity; woolskin utility, physiological characteristics and correlation between economical college animal farm of the Chungnam National University and sample farms in the suburbs of Dae jeon City were selected for feeding 20 heads of Corriedale wethers and another 20 heads Korean native kids as research materials for the periods of 5th May-26th November, 1977. The data such as growth rate, carcass, viscera weight, blood picture and plamsa components, hebage intake and economic traits were obtained and analysed. The result of the study are summarized as follows: 1. Meat production and quality 1) After 196days of feeding, the body weight of sheep and Korean native goats was increased by two times of those at the beginning of the trial, i.e. 20kg and 8kg respectively. 2) There was no significance of growth rates of sheep in housing and grazing. 3) The growth rate of Korean native goats were excellent at the mountainous areas of Gong ju-Gun where infectious diseases were not found 4) Accroding to the body measurements of 18-month-old sheep, percentages of hip height, body length, rump length, chest depth, chest width, hip width, chest girth and forearm circumference to the withers height were 103,%, 104%, 33%, 44%, 31%, 23%, 135% and 15% respectively, and those of hip height, body length, chest depth and chest girth of 8-month-old native goats to the withers height were 106%, 109%, 46% and 122,% respecitively. As a result, it was found that the percentage of hip height, body length and chest depth of Korean native goats were higher than those of sheep while that of the chest girth of goats was lower. 5) In the carcass data, 47, $52{\pm}2.27%$ of carcass percentage, $34.61{\pm}1.62%$ of lean meat, $26.07{\pm}2.51%$ of viscera, $9.75{\pm}1.4%$ of bone, and $20.95%{\pm}2.14%$ of woolskin for sheep, and $45.58{\pm}5.63%$ of carcass percentage, $27.62{\p}3.81%$ of meat, $34.86{\pm}4.16%$ of viscera, $11.66{\pm}1.83%$ of bone, $3.63{\pm}1.61%$ of skull and $9.26{\pm}2.41%$ of woolskin for native goats were obtained. 6) The contents of moisture, crude protein, crude fat and crude ash in native goat meat were much similar in both plots of housing and grazing. It was, however, known that the contents of moisture and protein were higher in grazinrg than in housing, while fat content was lower in grazing plots. 7) The weights of visceral organs shown similar tendency for both of sheep and native goats. For the weights of liver, heart, kidney and spleen, significance was not reconized among the treatments. Those of rumen, reticulum, small and large intestine were heavier in grazing than in housing, while the amount of visceral fat was heavier in housing. 2. Wool productivity and woolskin 1) The wool production of sheep for 7 months was $3.88{\pm}1.02kg$, and wool percentage, staple length, straighten length, wool growth per day and number of crimps were $9.27{\pm}1.48%$, 8. $47{\pm}1.00cm$, $10.63{\pm}0.99cm$, $0.40{\pm}0.04cm$ and $2.78{\pm}0.40$ respecitively. 2) The tensile strength and tear strength of woolskin treated by alum tanning were highest on the skin obtained from rump, i.e. $1,351kg/mm^2$ and $2,252kg/mm^2$ respectively, and they are in order of loin and shoulder. 3. Utilization and improvement of pasture. 1) The difference of herbage intake of native goats was not recognized between grazing and tethering, but the intake in the afternoon was s lightly higher than that in the morning. However the hervage intake of sheep was superior in grazing and in the afternoon. 2) The cultivation effect was lower in the native goat plots due to their cultivation abilities, in other words, the establishment rates of pasture by hoof cultivation were 60.25% in the goat plots and 77.35% in the sheep plots. 4. Correlation among economical traits. 1) The correlation between live weight of sheep and daily gain was higher. On the other hand, the correlation between other traits was not significant except that live weight, daily gain and lean meat percentage to the length of thoracic vertebrae. The live weight of native goats and meat production were highly correlated, and high correlation was also found between weights of carcass and meat. However, negative correlation was shown between viscera weight and live weight as well as daily gain. 2) The correlatoin between fleece weight of sheep and other traits such as live weight, daily gain and fleece percentage is very high at the 1% siginficant level, and this means that rapid-growth individuals can produce much fleece. 3) The correlation between the factors such as weights of live body, lean meat and viscera of sheep and body measurements, i. e. chest girth and body length was highest, and weights, of carcass and lean meat was highly correlated to chest width and depth. It will be therefore reasonable that the meat productivity estimates will have to be made on the basis of chest girth and body length. The meat production traits of native goats were highly correlated to the most of body measurement data, and the correlation coefficient between chest girth and weights of live body, carcass, lean meat and bone percentage was very high, i. e. 0.992-0.974 in particular. The correlations of meat production traits to chest depth, forearm circumference, body length were 0.759-0.911, 0.759-0.909 and 0.708-0.872 respectively. Therefore, the meat production of native goats will have to be estimated on the basis of chest data. 5. Blood picture and plasma components. 1) The number of erythrocyte and MCHC of native goats were $12.93{\times}10^6/mm^3$ and 36.14%, and those of sheep were $10.68{\times}10^6/mm^3$ and 36.26 respectively. The values of native goats were significantly higher than those of sheep. 2) The hemoglobin concentration, PVC, MCV and MCR of native goats were 10.92 g/100ml, $23.40{\mu}^3$ and 10.94 pg, and those of sheep were 11.73 g/100ml, 36.25 ml/100ml, $33.97{\mu}^3$ and 30.2 ml/100ml 8.43 pg respectively. The values of native goats were significantly lower those of sheep. 3) The number of leukocytes of native goats was significantly higher than that of sheep, that is, $11.64{\times}10^3/mm^3$ in native goats and $9.32{\times}10^3/mm^3$ in sheep. 4) In differential count of leukocyte, neutrophil was significantly high in native goats while lympocyte in sheep. On the other hand, the basophil, eosinophil and monocyte were not significant between native goats and sheep. 5) The amounts of total protein and glucose in the plasma of native goats were 6.2g/100ml and 53.6mg/100ml, and those of sheep were 5.6g/100ml and 45.7mg/100ml, which means that the values of native goats were significantly higher that those of sheep. The amount of total-lipid of native goats(127.6mg/100ml) was significantly than that of sheep(149.6mg/100ml). 6) The amount of non-protein nitrogen, cholesterol, Ca, P, K, Na and Cl were not different between native goats and sheep. 6. Economic analysis. 1) The gross revenue of a farm which fed native goats and sheep was 4,000won per head and the optimum size for feeding them in a farm as a subsidiary work is 5-10 heads. 2) Since there was no difference between housing and grazing, they can be fed in group for farm's subsidiary work. 3) They can be also fed by youths and house wives in the suburbs of cities, because labour requirement is estimated as only two hours per days for feeding 5 heads of native goats and sheep.

Studies on the Efficacy of the Newly Developed Automatic Liquid Feeder for 7-day Old Early Weaned Piglets (7일령 조기 이유자돈에 대한 액상사료 자동급이기 이용효과)

  • This study was conducted to test the efficacy of the automatic liquid feeder(designated as NLRI) developed by National Livestock Research Institute, Korea for 7-day old early-weaning piglets. The other automatic liquid feeder imported from USA(designated as IALF) was used for a comparative purpose. A total of sixty piglets of 7 days of age were allotted to three treatments. The control group of 20 piglets was raised by their own sows until 21 days of age, while others were shifted to automatic liquid feeders of either NLRI or IALF on 7 days of age and reared during 14 days post-weaning. After then, all the piglets were fed nutritionally identical mash diets until 70 days of age. Compared with control group, body weight of piglets reared on automatic liquid feeders were lighter(P〈0.05) at 21 days of age [5.55kg (control) vs 4.97 (NLRI) and 4.98kg (IALF)], while heavier(P〈0.05) at 70 days of age(24.82kg vs 30.17 and 29.42kg). The results indicated that pigs reared on liquid feeding showed higher(P〈0.05) average daily gain than control pigs during the whole experimental period [346.7g (control) vs 425.8 (NLRI) and 416.1g (IALF)], while no difference was found in feed/gain (1.67 vs 1.78 and 1.84). There was no difference in growth performance and incidence of diarrhea between two automatic liquid feeders, NLRI and IALF. Compared with control group, intestinal villi tended to be shorter in liquid feeding group during the first week, but were recovered within two weeks. The results suggest that the automatic liquid feeder newly developed by National Livestock Research Institute, Korea can be successfully used for rearing young piglets weaned at very early age.

Effects of Extruded Corn in Nursery and Finishing Pigs (자돈 및 비육돈에 있어 옥수수의 가공 효과)

  • For the Exp. 1, a total of sixty pigs(15.95${\pm}$0.09kg average initial body weight) were used in a 28-d growth assay to determine the effects of extruded chinese corn on growth performance and nutrient digestibility in nursery pigs. Dietary treatments included 1) UCORN(U.S. corn-SBM based diet), 2) CCORN(Chinese corn-SBM based diet) and 3) ECCORN(Extruded Chinese corn-SBM based diet). Overall period, average daily gain of pigs fed ECCORN diet was higher than that of pigs fed CCORN diet(547 vs 522 g/d), however, there was not significant difference. On day 10 of the experiment, pigs fed UCORN and ECCORN diet had significantly increased in DM and DE digestibilities compared to pigs fed CCORN diet(P〈0.05). Also, on day 24 of the experiment, pigs fed UCORN and ECCORN diet had a significant increase in DM digestibility compared to pigs fed CCORN diet(P〈0.05). Pigs fed ECCORN diet had significantly increased DE digestibility compared to pigs fed CCORN diet(P〈0.05). For the Exp. 2, three cannulated barrows(54.09kg average initial body weight) were used to determine the apparent ileal digestibilities of amino acids and nutrient digestibility of extruded corn in finishing pigs. Dietary treatments were the same as in Exp. 1. Apparent ileal digestibility of cystine was greater for UCORN and ECCORN than for CCORN(P〈0.05). Apparent digestibility of DM at the total tract was greater for UCORN and ECCORN than for CCORN(P〈0.05). Pigs fed UCORN and ECCORN diet had a significant increase in apparent total tract digestibility of N compared to pigs fed CCORN diet(P〈0.05). In conclusion, the results obtained from these feeding trials suggest that the extruded corn for nursery pigs had affected growth performance and DM and DE digestibilities. In finishing pigs, extruded corn was an effective means to improve apparent total tract digestibilities of DM and N.

Effects of Mud Flat Bacteria Origin Protease Supplementation on Growth Performance, Amino Acid Digestibility, Blood Characteristics, Meat Quality, Fecal VFA and NH3-N Concentration in Finishing Pigs (갯벌 미생물 유래 단백질 분해 효소제의 급여가 비육돈의 생산성, 아미노산 소화율, 혈액성상, 육질특성 및 분내 휘발성 지방산과 NH3-N 함량에 미치는 영향)

  • This study was conducted to evaluate effects of mud flat bacteria origin protease supplementation on growth performance, amino acid digestibility, blood characteristics, meat quality, fecal VFA (volatile fatty acids) and NH3-N (ammonia nitrogen) concentration in finishing pigs. Eighty pigs (Landrace×Yorkshire×Duroc, 60.08±2.69 kg average initial body weight) were used during experimental period. Dietary treatments included 1) high nutrient density diet, 2) high nutrient density diet+0.1% protease, 3) low nutrient density diet and 4) low nutrient density diet+0.1% protease. For overall period, ADG was improved in treatment of high nutrient density diet added protease compared with treatment of low nutrient density diet without protease (P<0.05). DM and N digestibilities were improved in treatments of high nutrient density diet and low nutrient density diet added protease compared with treatment of low nutrient density diet without protease (P<0.05). Essential amino acid digestibility was improved in treatment of low nutrient density diet added protease compared with other treatments (P<0.05). Nonessential amino acid digestibility was improved in treatment of high nutrient density diet added protease compared with treatments of high and low nutrient density diet (P<0.05). BUN (blood urea nitrogen) concentration in blood was increased in treatment of high nutrient density diet added protease compared with treatment of low nutrient density diet without protease (P<0.05). L*value of M. longissimus dorsi muscle was increased in treatments of low nutrient density diet compared with treatments of high nutrient density diet (P<0.05). In conclusion, mud flat bacteria origin protease was effective for improving growth performance, amino acid digestibility and influencing BUN concentration and meat color in finishing pigs.

Association Between the Polymorphism in the Promoter Region of Porcine A-FABP Gene and Growth Traits in Duroc Pigs (돼지 Duroc 품종에서 A-FABP promoter의 다형성과 성장형질의 연관성)

  • A polymorphism was found in the promoter region of porcine adipocyte fatty acid binding protein gene(A-FABP) gene which plays a key role in the binding and transportation of free fatty acid in adipocyte and deposition of intramuscular fat. Mutation was detected a substitution(T406C) using SSCP analysis and subsequently confirmed by sequencing the fragment in Duroc pigs. This T-406C mutation might change the binding activity for transcription factor nuclear factor 1(NF1). In this population, this mutation was genotyped using HinfⅠRFLP, and found three kinds of genotypes(TT, TC, and CC) showing their frequencies of 42.3, 44.3, and 13.4%, respectively. We statistically analyzed the association between the A-FABP genotypes and growth traits and found that the body weights of the pigs containing 406C/(TC or CC) were heavier for the body weight at the age of 20 weeks than those containing genotype TT(P<0.05), but not for those at the age of 0, 3, and 10 weeks. Pigs containing genotype CC had also a higher value for the average daily gain and lower values for the date for 90kg of body weight and food conversion ratio than those of 406T/- genotype. In addition, without the significant difference of back fat thickness, there was a significant association between the existence of allele CC and lean meat and eye muscle area(P<0.05). As a result of this study, we suggest that the allele T406C in the promoter region of A-FABP gene play an important role in deposition of intramuscular fat and weight in the later growth period. This polymorphism will be an useful molecular marker for breeding of Duroc pigs.

Effects of Total Mixed Fermentations with Bean Curd Dregs on Growth Performance of Growing Hanwoo Heifers (비지박을 이용한 섬유질발효사료의 급여가 육성기 한우암소의 발육에 미치는 영향)

  • This study was conducted to determine the effects of total mixed fermentation (TMF) with bean curd dregs on body weight gain, feed intake and body size of growing Hanwoo heifers. Thirty Hanwoo heifers, 6 months of age and weighing $125.9{\pm}12.0$ kg, were randomly allocated to 3 experimental groups of 10 animals each for a 6-month feeding trial. The groups were T1 (rice straw offered ad libitum with restricted (1.7% BW/day) concentrate), T2 (TMF only offered ad libitum) and T3 (TMF and rice straw offered ad libitum with restricted (1.0% BW/day) concentrate). TMF feeds were composed of 60% bean curd dregs, 16.5% corn meal, 10.4% wheat bran, 5% rice straw, 7% rice hulls, 0.5% urea matter, 0.5% magnesia lime and 0.1% lactic acid bacteria. The chemical composition and pH value of TMF feeds were measured at 0, 24, 48 and 72 hr after storage. In TMF with bean curd dregs, there were no differences in concentrations of DM, EE, and CF according to storage periouds. The ADF and NDF at 72 hr of storage time showed a significant decrease (9.52 and 3.15 %, respectively) compared to 0h of storage time (p<0.05). The value of pH was gradually decreased during the storage periods. Body weight gain, feed intake and average daily gain were higher in the TMF groups (T2, T3) compared to the T1 group (p<0.05). Changes in body size were also higher in the TMF groups compared to the T1 group (p<0.05). Thus, the present results indicate that TMF with bean curd dregs may help improve growth performance in growing Hanwoo heifers.
