Inhibitory Effects of Prunus mume Solvent Fractions on Human Colon Cancer Cells (매실 분획물에 의한 인체 대장암세포 억제 효과)

  • Prunus mume, also known as maesil, is a popular fruit consumed in East Asia (Korea, Japan, and China). It contains high amounts of organic acids, minerals, and polyphenols and has been used as a medication for fever, vomiting, and detoxification. In this study, the anti-proliferative and apoptotic effects of solvent fractions from maesil were evaluated using sulforhodamine B (SRB) assays, morphological evaluations, Hoechst 33258 staining, and western blotting. Addition of the maesil methanol fraction (MMF) and the maesil butanol fraction (MBF) significantly and dose-dependently decreased the cell viability of HT-29 human colon cancer cells. Colony-forming assays confirmed that the MMF and MBF treatments decreased colony numbers when compared with untreated control cells. Treatment of HT-29 cells with MMF and MBF caused a distortion of the cell morphology to a shrunken cell mass. Treatment with MMF and MBF also dose-dependently increased nuclear condensation and the formation of apoptotic bodies in HT-29 cells. Treatment with MMF and MBF significantly and dosedependently increased the expression of Bax (a pro-apoptotic protein), caspase-3, and poly ADP-ribose polymerase (PARP) and decreased the expression of Bcl-2 (an anti-apoptotic protein). MMF significantly and dose-dependently inhibited cell proliferation induced by bisphenol A, an environmental hormone. Therefore, MMF may have potential use as a functional food and as a possible therapeutic agent for the prevention of colon cancer.

Substantial Estimated Availability and NDF-Bound Trace Minerals (Fe, Zn, Cu, and Mn) Contents of Seven Different Seaweeds (7종 해조류 NDF에 결합된 미량무기질(Fe, Zn, Cu, Mn) 함량 및 추정 이용률)

  • The substantial trace mineral usabilities of seven seaweeds for nutrient evaluation have been studied. As the neutral detergent fiber (NDF) bound trace minerals have been known to be indigestible, these contents were excluded in the digestible trace mineral contents evaluation. Therefore, those four trace mineral contents in seven seaweeds and their NDF-bound contents were estimated. Consequently, the NDF-bound Zn, Cu and Mn contents, except Fe, in seven seaweeds were between 10 and 20% of total trace mineral contents, thus 80~90% of seaweeds trace minerals were observed to be substantially digestible. Although the average estimated availability of Fe in the seaweeds were relatively low, those Zn, Cu and Mn were high in relative to those total contents. Thus, even with consideration of their digestibility, seaweeds could be one of good sources of trace minerals for human nutrition because of their high availabilities of Zn, Cu and Mn.

  • For orthodontic treatment of growing patients and those which involve long treatment times, knowledge of growth and development are essential. Data from the general growth observation chart being used at present does not reflect the growth transition of modern times. This is because these data are out of date. The present study, therefore, aims to introduce the process of producing a growth curve and growth rate curve based on data which represents a more accurate description of the present situation. The data used were from the 5th nationwide surrey, SIZE KOREA 2004 study, carried out by the Technology and Standards Policy Division, Department of Technology and Standards Planning, Ministry of Commerce. Processing and rearranging produced data with variations divided into the three quartiles and the 1st, 5th, 95th and 99th percentiles were included to produce a growth observation chart according to sex, height and weight differences. In the same way, a growth rate curve based on the geometric mean value was produced. The resulting growth charts can be used as an index for growth and development, and used for better communication between the orthodontist, patients and parents.

Contamination and Risk Analysis of Heavy Metals in Korean Foods (국내식품의 중금속 오염과 위해성 분셕)

  • Foods constitute a large portion of heavy metal exposure toward general population and attract a deep concern with respect to assuring human health. This study summarized published data in Korea on the content, and dietary intake of heavy metals and assessed their risk potential in comparison with foreign data. An analysis for the yearly fluctuation of metal contents including arsenic, cadmium, mercury and lead by flood group (marine fishes, coastal shellfishes, freshwater fishes, cereal grains) exhibited a decreasing trend from the 1970s to the 1990s. When compared with domestic standards of heavy metals, their mean contents were below the limit and their maximum values seldom exceeded the limit. The data on the dietary intake of heavy metals by Koreans showed a decreasing trend from the 1980s to the 1990s. The average intakes offs and Hg were 6∼8% and those of Cd and Pb were 50∼80% of PTWI(provisional tolerable weekly intake), all of which were below the tolerance. As the extreme intakes of these metals may exceed the PTWI, a careful assessment for them may be necessary. Dietary intakes of Cd, Hg and Pb by Koreans lie in the mid-level among countries cited in the GEMS/Food monitoring data. As fishery foods are suspecious of contamination with Hg, Cd and As, and floods in general are with Pb, it is necessary to establish legal limits for these metals and monitor any progress of their contamination. Furthermore, overall assessment of exposure to heavy metals from all sources including floods, air, drinking water and occupation should be made in order to confirm the dietary risk factors and to assure the safety of food resources.

  • Cosmeceuticals products we not the products simply for moisturizing and protecting skin but the ones for encouraging to make improvements on wrinkles and helping to whiten, tan, and protect skin against ultraviolet light. In a broader sense, they refer to products with aiming to maintain healthy condition of skin such that skin troubles could be improved or aging of skin could be delayed. However, cosmeceuticals should not have any side effects because they are applied to the whole body for a long period of time differing from medical products which are used on specific areas for a short time. The number of such cosmeceuticals has increased from 500 in 2001 to 2300 in 2003. In order to develop and broaden the scope of cosmeceuticals, the concept of cosmetics needs to be changed from caring skin to delaying aging of the skin and reviving cells. For this purpose, high-valued materials should be developed through basic researches related to the biological function of skin, in vivo experiment and reaction of skin in response to various stimuli using biotechnologies and bioengineering methods. At the same time, it should be proceeded to develop new nano materials for overcoming skin barriers and transfer matericals for helping to absorb effective substances and maintaining stability.

A Study on the Awareness of medical and Health-Related Majors on Drinking Culture (의·보건 계열 대학생의 음주문화 인식에 관한 연구)

  • The purpose of this study was to examine the drinking awareness of medical and health-related majors in a bid to find out the impact of their drinking awareness on drinking. The subjects in this study were 153 students who were selected by rendem sampling from colleges located in two different cities in the province. Data collection was performed from may 1 to september 20, 2011 with total 153 college students. Out of them, 86 students majored in medical departments, and 67 students were in the health-related departments. After a self-administered survey was conducted, the following findings were given: A great deal of the students who accounted for 75.8 percent were ever disturbed in daily routine life by drinking over the past year. As for the influence of drinking awareness on the amount of drinking, the amount of drinking was under the significant influence of the following five items. The first was that alcohol was an energizer(p<.001), and the second was that drinking led to a sound sleep(p<.01). The third was that I drank too much with my close friends often(p<.05), and the fourth was that alcohol used as a hangover-chaser was conducive to a hangover(p<.05). The fifth was that alcohol had the largest impact on the liver(p<.05). All the variables made a 43.4% prediction of the amount of drinking. In conclusion, the medical and health-related majors who learned about the impact of drinking on the human body in a firsthand or secondhand manner had a tendency to drink a less amount of alcohol and do less binge drinking. It's advisable to take advantage of the knowledge of medical and health-related majors on the human body and drinking as resources of anti-drinking education.

Investigation of Microbial Communities in Sulculus diversicolor supertexta Through 16S rRNA Sequencing and Antibacterial Monitoring of Harmful Strains (16S rRNA 염기서열 분석을 통한 오분자기(Sulculus diversicolor supertexta)내 미생물 군집 조사 및 인체유해 질병세균에 대한 항균활성 모니터링)

  • This study investigated the muscles, intestines, and gonads of Sulculus diversicolor supertexta to examine the diversity of microbial communities within examples collected from the Jeju Coast. Using different media, initial pure isolation in MA, 1% BHIA, and 1% TSA indicated that the muscles, intestines, and gonads supported more communities, respectively. In analysis of relative similarity with 16s rRNA sequencing, 190 pure colonies were isolated, and further analysis with NBLAST identified 71 species, 39 genera, 25 families, and five phyla. Homogeny with the reference strain was 91-100%. Microbial communities in S. supertexta consisted of gamma and alpha Proteobacteria (48%), Actinobacteria (32.5%), Firmicutes (16.9%), Deinococcus-Thermus (1.3%), and Bacteroides (1.3%). In all tissue, Psychrobacter cibarius in Moraxellaceae was dominant. Alteromonadaceae, Enterobacteriaceae, Pasturellaceae, Moraxellaceae, Rhodobacteraceae, Geminicoccaceae, Dietziaceae, Intrasporangiaceae, Microbacteriaceae, Micrococcaceae, Micromonosporaceae, Streptomycetaceae, Aerococcaceae, Bacillaceae, Paenibacillaceae, Planococcaceae, and Staphylcoccaceae were commonly isolated across all tissues, and Flavobacteriaceae, Corynebacteriaceae, Yesiniaceae, Vibrionaceae, Hahellaceae, Pseudomonadaceae were also identified from the intestines. In microbial monitoring of four harmful bacteria, Streptomyces albus (96%) showed antibacterial activity against all four strains, and Agrococcus baldri (99%) and Psychrobacter nivimaris (99%) presented against E. Coli and E. aerogens. In addition, some strains with low homogeny were isolated and further experiments are therefore required, for example to refine the antimicrobial substances including new strain investigations. These additional experiments would aim to establish generic resources for the microbial communities in S. Supertexta and provide basic data for applied microbiological research.

Metabolizing analysis according to the sawdust media of Pleurotus ostreatus (산겨릅나무와 옻나무 톱밥배지에서 재배한 느타리버섯의 함유성분 분석)

  • The transitivity of Chemical constituents by Pleurotus ostreatus cultivated in different raw sawdusts, which are Acer tegmentosum MAX, Rhus verniciflura, was investigated. The HPLC chromatography patterns on the chemical constituents of P. ostreatus showed the similar chromatography patterns in the different raw sawdusts and control sawdust. The unknown chemical constituents of P. ostreatus cultivated in the mixed medium added 10 %, 20% raw sawdusts, respectively, were increased. But the significance results in the mixed medium added 50% raw sawdusts were not showed. The chromatography patterns of mycelia grown in media added the 80% MeOH extracts of A. tegmentosum and R. verniciflura showed the similar patterns in comparison with control mycelia. In the results, the secondary metabolites of functional media were not degrade and changed to other derivatives compounds by P. ostreatus.

In Vitro Antioxidant and Anticancer Potential of n-Hexane Extract from Ginseng Marc (인삼박 n-Hexane 추출물의 in vitro 항산화 및 항암 활성)

  • A lipid-soluble extract in ginseng marc was prepared by n-hexane extraction to evaluate its antioxidant and anticancer potential. A hexane extract of ginseng marc (HEGM) possessed a 2,2-diphenyl-1-picryl-hydrazyl free radical scavenging activity which was related to the amount of total phenolics. Also, HEGM showed a potent inhibitory activity on human non-small cell lung cancer (A549, $GI_{50}=34.0{\mu}g/mL$) and colon cancer (SNU-C4, $GI_{50}=45.2{\mu}g/mL$) cells proliferation in vitro in a concentration-dependent manner as did the hexane extract of ginseng with $GI_{50}$ values of $20.0{\mu}g/mL$ in A549 and $37.0{\mu}g/mL$ in SNU-C4. These results imply that HEGM can be utilized as an antioxidant and anticancer substance.
