The Anti-bacterial Effects and Epidermal Permeability Barrier Function of Red Onion Juice Produced in Jeon-Nam province in Korea (전남 함평산 자색양파 추출물의 항균효과와 피부장벽 보호효과)

  • Objective : In this study, we assessed the anti-bacterial effects and epidermal permeability barrier function of red onion juice comparing to yellow onion juice and $Houttuynia$ $cordata$ extract $in$ $vitro$. Methods : 3types of red and yellow onion juice were prepared as antibacterial agent candidates with Houttuynia cordata hot water extract using 4 different bacterial strains ($Escherichia$ $coil$, $Salmonella$ $enterica$ $subsp.$ $enterica$, $Staphylococcus$ $epidermidis$, $Staphylococcus$ $aureus$ $subsp$) by colony counting method. The expression of filaggrin, a marker of keratinocyte differentiation, and serine palmitoyl transferase (SPT), a marker of the formation of the stratum corneum lipid barrier, in human HaCat keratinocytes were analyzed using HaCaT cell line. The expression of COX-2 and AP-1 which is a factor of COX-2 transcription were also analyzed by western blotting method. Results : There was detectable anti-bacterial effects on $Staphylococcus$ $epidermidis$, $Staphylococcus$ $aureus$ $subsp$ among 1%, 5%, 10% extracts of yellow and red onion.(81%-100%) The bacteriocidal effects were not shown on $Escherichia$ $coil$, $Salmonella$ $enterica$ $subsp.$ $enterica$ among $Houttuynia$ $cordata$, yellow onion and red onion extracts. The in vitro results showed the concentration-dependent effects on the expression of both filaggrin and SPT in HaCat cells among 0.01%, 0.05%, 0.1%, 0.5% extracts in Houttuynia cordata and red onion, reflecting the notion that $Houttuynia$ $cordata$ and red onion can induce epidermal keratinocyte differentiation and improve the recovery of skin barrier functions. The concentration-dependent effects also have been shown on the expression of both COX-2 and AP-1 among 0.01%, 0.05%, 0.1%, 0.5% extracts in $Houttuynia$ $cordata$ and red onion, while slight effect in yellow onion. Conclusion : Red onion juice could be a potential candidate enhanser for the skin care and cosmetology.

Antimelanogenic Effect of Ligularia fischeri, Solidago virga-aurea, Aruncus dioicus Extracts from Ullung Island in Murine Melanoma Cells (마우스 악성흑색종세포에서 울릉도 곰취, 미역취, 삼나물 추출물의 멜라닌 생성 억제 효과)

  • The objective of the present study was to evaluate the skin whitening effect of the extracts of 3 herbs, Ligularia fischeri, Solidago virga-aurea and Aruncus dioicus, which were collected from Ullung island. Tyrosinase inhibition activities were 33% in pre-fermented extracts and 45% in post-fermented ones. When tyrosinase activities in B16F10 murine melanoma cells were tested, activities in pre- and post-fermented extracts were 41 and 56.5%, respectively. Thus, the post-fermented extracts might have greater skin whitening effects. The protein expression of MITF, TRP-1, TRP-2, and tyrosinase, which are all skin-whitening related transcription factors, showed that both pre- and post-fermented herbs inhibited protein biosynthesis in B16F10 melanoma cells. Post-fermented herb extracts especially showed a greater decrease of protein expressions. The expression of MITF, a regulatory transcription factor, was also decreased by both extracts but was greater in the post-fermented ones. From the results, it can be concluded that the 3 herb extracts from Ullung island may inhibit melanin biosynthesis by the suppression of MITF activity in a signaling pathway. Results indicate that the post-fermented herbs tested in the present study had skin whitening activities and can be used as functional ingredients for food and cosmetic compositions.

  • 본 논문은 변압기 이상진단을 위한 유중가스분석법의 진단신뢰도 향상을 위해 기존의 추출율이 높은 토플러 추출방법에 대한 결과와 Headspace 분석법에 의한 결과를 각각 비교하고, Headspace 추출법에 의한 결과의 정확도 향상을 위해 여러가지 인자들을 고려하였다. 향후 최적 조건을 수립하여 수은에 의한 인체 유해성이 없는 Headspace 분석법으로의 전환을 모색하고자 한다.

Forsythiae Fructus Induces VEGF Production via p38 MAPK Activation in Human Keratinocytes (각질형성세포에서 p38 MAPK 활성을 통한 연교의 VEGF 생성 효과)

  • Cutaneous microvasculature plays a critical role in age-associated skin changes. A considerable reduction of number and size of vessels has been observed in the upper dermis of elderly skin. Forsythiae fructus (FF), the dried fruit of plant Forsythia suspensa (F. suspensa), has been traditionally used as an herbal medicine to treat inflammatory diseases and bacterial diseases. However, its regulatory effect on angiogenic responses has not been elucidated in skin. Therefore, we analyzed secretory profiles upon treatment of FF extract using array designed to detect angiogenesis-associated mediators in human keratinocytes. Because keratinocyte-derived VEGF (vascular endothelial growth factor) has been regarded as a potent factor for new microvasculature under the epidermis, we further investigated the effect of FF extract on VEGF production. We observed that the VEGF expression of mRNA and protein level was increased by about 2 folds in a dose-dependent manner after FF extract treatment. In signaling experiments, FF extract induced rapid p38 MAPK activation within 5 min, and the activation was totally abrogated by pretreatment with a p38 MAPK specific inhibitor. The FF-induced VEGF upregulation was also significantly attenuated by a p38 MAPK inhibition. Taken together, FF extract induces VEGF production via p38 MAPK activation in human epidermal keratinocytes. These novel findings suggest that FF is useful as a potential agent with pro-angiogenic activity and may help to improve age-dependent reduction of the microvasculature in aged skin or to heal skin wound.

Cytoprotective Effect of Petasites japonicus Extract on Cadmium-induced Cytotoxicity in HaCaT cell (Cadmium으로부터 손상을 유도한 HaCaT 세포에서 머위(Petasites japonicus) 추출물의 세포보호효과)

  • In this study, we investigated the effect of Petasites japonicus extract on the cytotoxicity and cytoprotective effects against cadmium for cosmetics use. We measured the protein expression of apoptosis regulatory factor (Bcl-2 and procaspase-3) after treatment of Petasites japonicus extract in the cadmium-induced keratinocyte. As a result, high cell viabilities above 98% were observed in the all treated concentrations except at $200{\mu}g/mL$ of Petasites japonicus extract in keratinocytes with cadmium-induced damages. In keratinocytes with cadmium-induced damages, Bcl-2 and procaspase-3 protein expression increased in the experimental group treated with Petasites japonicus extract. Also HaCaT cells resulted in cleavage of PARP protein at 12 h post-cadmium exposure. Western blot analysis and relative density of the bands suggested that pretreatment of cells with Petasites japonicus extract inhibited cadmium-mediated cleavage of PARP. These results suggest that Petasites japonicus extract can be used as the cosmetic ingredients for cytoprotective effect.

Inhibitory Effects of Allium sacculiferum Max. Methanol Extracts on ROS Production and Lipid Accumulation during Differentiation of 3T3-L1 Cells (참산부추(Allium sacculiferum Max.) 메탄올 추출물의 지방세포 내 ROS 생성 및 지질 축적 억제 효능)

  • Allium sacculiferum Max. (ASM) is a perennial plant of the Liliaceae family and grows over the entire regions of Korea. Obesity is a serious health problem worldwide and has currently become a prevalent chronic disease. Adipocytes produced by preadipocyte differentiation during adipogenesis and adipocytes combined with abnormal accumulation cause obesity. Recently, intracellular reactive oxygen species (ROS) were shown to accelerate lipid accumulation in 3T3-L1 cells. In this study, we investigated the effects of ASM methanol extracts on ROS production and lipid accumulation in 3T3-L1 adipocytes. Our results indicate that the 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity of ASM methanol extracts increased in a dose-dependent manner. ASM methanol extracts suppressed ROS production and lipid accumulation during adipogenesis. In addition, ASM methanol extracts inhibited the mRNA expression of both pro-oxidant enzymes such as glucose-6-phosphate dehydrogenase as well as the transcription factors, including sterol regulatory element-binding proteins 1c, peroxisome proliferator-activated receptor ${\gamma}$, and CCAAT/enhancer-binding protein ${\alpha}$. Our results suggest that ASM methanol extracts inhibit ROS production and lipid accumulation by controlling ROS regulatory genes and adipogenic transcription factors. Thus, ASM has potent natural antioxidant, anti-adipogenic properties and have potential in the development of a potent anti-obesity agent.

Effects of dietary Gelidium elegans extract on fat metabolism in preadipocyte cell and mice fed a high-fat diet (개별인정원료 우뭇가사리추출물의 체지방 감소에 관한 기능성 고찰)

  • We investigated the anti-obesity effect of Gelidium elegans extract (GE) on 3T3-L1 preadipocytes and a high-fat-diet (HFD)-induced mouse model. The results of the present study demonstrated that GE prevents weight gain induced by a high-fat diet (HFD) by modulating the adenosine monophosphate-activated protein kinase (AMPK)-PR domain-containing 16 (PRDM16)-uncoupling protein-1 (UCP-1) pathway in a mice model. Moreover, in vitro results show that GE suppressed adipocyte differentiation by modulating adipogenic regulators, stimulated lipolysis by activating ATGL, and inhibited adipogenesis by downregulating various enzymes associated with triglyceride synthesis. GE was also found to upregulate AMPK phosphorylation as well as the expression of UCP1 and PRDM16 proteins, leading to measurable changes in the beige-like phenotype differentiation of 3T3-L1 cells. Taken together, these findings suggest the role of GE as a functional food ingredient extracted from Gelidium elegans to increase energy expenditure and anti-obesity efficacy.

  • Smilax sieboldii is one of the Smilax species. A number of Smilax plants have multiple physiologically-active components and anti-inflammatory/anti-oxidant effects. Antiobesity effects induced by Smilax sieboldii have not been reported. In this study, we investigated the effects and molecular mechanisms of anti-obesity activity of 70% ethanol Smilax sieboldii extract (SSE). The anti-obesity effect of SSE was determined using 3T3-L1 adipocytes. We confirmed that SSE was not cytotoxic to murine 3T3-L1 preadipocytes, we evaluated SSE dose-dependently decreased the accumulation of lipids via an Oil Red O assay and triglyceride assay. These anti-obesity activities of SSE were mediated by the inhibition of adipogenesis-related marker genes (peroxisome proliferator activated receptor-γ, CCAAT-enhancer-binding protein α, and SREBP1c) and lipogenesis-related marker genes (fatty acid synthase and aP2). These results suggest that SSE has the potential to exert anti-obesity and anti-hyperlipidemia effects by regulating adipogenic transcription factors and inhibiting the expression of adipogenic markers.

  • Since the industrialization spread out, the metropolitan areas of Seoul have been urbanized rapidly in Korea, without concern for the quality of further development and sufficient environmental management. Due to this fact, it has become increasingly more apparent that natural hazards, such as floods and landslides, occur frequently after the summer's heavy rains, and because of that, the scale of damage is getting larger. The purpose of this study is first to analyze the relationship between development and floods in the sub-basins of the study area. In addition to this, I would like to compare the influences of geomorphological characteristics upon the floods occurring in both the whole study area and the developed area in sub-basins.

Inhibitory Effect of the Ethanol Extract of Rudbeckia laciniata var. hortensis Bailey on Adipocyte Differentiation in 3T3-L1 Cells (겹삼잎국화 에탄올 추출물의 지방세포 분화 억제 효과)

  • Rudbeckia laciniata var. hortensis Bailey is used in home remedy for colic and gastritis in South Korea. Although Rudbeckia laciniata var. hortensis Bailey is used extensively for home remedies, no single study on its efficacy exists. In this study, we investigated the anti-obesity effects of Rudbeckia laciniata var. hortensis Bailey. The anti-obesity effect of a 70% ethanol extract from Rudbeckia laciniata var. hortensis Bailey on the differentiation of 3T3-L1 pre-adipocytes to adipocytes was investigated with an Oil Red O assay, western blot analysis, and mRNA analysis. Compared to the control (only treated with DM), the 70% ethanol extract of Rudbeckia laciniata var. hortensis Bailey significantly inhibited adipocyte differentiation and intracellular triglyceride (TG) levels at a concentration of $100{\mu}g/ml$. To determine how the TG content was reduced, we measured the level of protein and mRNA expression of obesityrelated agents, such as peroxisome proliferators-activated receptor ${\gamma}$ ($PPAR{\gamma}$), CCAAT/enhancer- binding protein ${\alpha}$ ($C/EBP{\alpha}$), AMP-activated protein kinase (AMPK) phosphorylation, LPL, and FAS. As a result, the 70% ethanol extract of Rudbeckia laciniata var. hortensis Bailey significantly increased the expression of AMPK and decreased the expression of genes related to adipogenesis and fat storage, such as $PPAR{\gamma}$, $C/EBP{\alpha}$, LPL, and FAS.
