Uptake of Heavy Metal Ions by Water Dropwort (Oenanthe stolonifera DC.) and Identification of Its Heavy Metal-Binding Protein (미나리의 중금속 흡수량 측정 및 중금속 결합단백질의 동정)

  • Uptake of hen metal ions by water dropwort (Oenanthe stolonifera DC.) and its cadmium-binding protein were studied to probe for good method to remove heavy metal contaminants from environments. The plant was cultured in the culture medium (pH 7.0) containing the various concentrations of $Cd^{2+}$, $Cr^{3+}$ or $Pb^{2+}$, for 3 and 7 days. The residual heavy metals deposited in roots linearly increased as the metal ions concentration increased up to 17 ppm for $Cd^{2+}$, 20 ppm for $Cr^{3+}$ and 50 ppm for $Pb^{2+}$. Above these concentrations, the plant growth was inhibited and the uptake rates of the metal ions decreased. The heavy metals absorbed by the plant were mostly deposited in roots. In particular, the residual concentration of lead in roots was about four times higher than those of cadmium and chromium. When cultured in the medium containing 20 ppm of each metal ion, 80% of cadmium, 90% of cromium and 96% of lead were deposited in roots out of the total residual metal ions in the plant. These values correspond to 6.1 mg of cadmium, 5.2 mg of chromium and 23.6 mg of lead per one gram of roots tissue on a dry weight basis. A cadmium-binding protein was partially purified by extraction, gel filtration and DEAE-Cellulose chromatography from water dropworts that was grown in the medium containing 20 ppm $Cd^{2+}$. The purified protein was a single band on SDS- and non-denaturing- polyacrylamide gel electrophoresis. Its molecular mass was estimated to be ca. 5,000 dalton by gel filteration. Analysis of amino acid composition of the protein indicated that it had a typical amino acid composition of heavy metal-binding protein in that it contained 27% of acidic amino acids and 9.9% of cysteine. However, it is likely that the protein is a new plant metal-binding protein, since its amino acid composition is somewhat different from those of phytochelatins that have been known so far.

  • This study was conducted to reveal the effect of some meterological factors on the weight of cone and seed per cone in Korean white pine (Pinus koraiensis SIEB. et ZUCC.). The weight of cone and seed per cone for 7 years from 1992 to 1998 on 45(1992) year-old trees and some meterological factors for 9 years from 1990 to 1998 were surveyed in Hongcheon-Gun region, Gangweon-Do. Simple correlations and multiple regression between weight of cone and seed per cone and some meterological factors were analyzed. The results obtained from the above experiments were as follows: 1. Positive correlations were found between weight of cone and monthly mean temperature of February in flower bud differentiation year. number of annual hoarfrost days of the cone production year monthly mean temperature of may in the cone production year, as July respectively. There were negative correlations between weight of cone and monthly mean temperature of august in the flowering year, wind speed of April in the flower bud differentiation year, number of clear days of december in the flowering year, number of annual cloudy days of the flowering year, number of precipitation days of june in the flowering year, number of annual precipitation days of the flowering year, number of annual cloudy days of March in the cone production year. number of annual cloudy days from march to October in the flowering year as well as number of precipitation from march to october in the flowering year. 2. Positive correlation between weight of seed per cone and number of hours with sunshine duration of June in the flowering year, the percentage of sunshine duration of June in the flowering year, number of clear days of June in the flowering year. monthly mean temperature of may in the cone production year. as well as monthly mean temperature of July in the cone production year were found. Negative correlations were recognized between weight of seed per cone and monthly mean temperature of January in the flowering year, monthly mean temperature of august in the flowering year, wind speed of april in the flower bud differentiation year, number of annual cloudy days of the flowering year, number of precipitation days of June in the flowering year, number of annual cloudy days from March to October in the flowering year as well as number of precipitation from march to October in the flowering year.

Process Development of a Virally-Safe Acellular Bovine Amniotic Membrane for Biological Dressing (바이러스 안전성이 보증된 무세포 소 양막 생물창상피복재 제조 공정 개발)

  • A process for manufacturing virally-safe bovine amniotic membrane(BAM) has been developed for biological dressing. BAM was harvested from a healthy bovine placenta, and then the epithelium was removed. The remaining stromal layer was consecutively disinfected with 70% ethanol and 0.05% sodium hypochlorite. The stromal layer was incubated in a decellularization solution containing 0.25%(w/v) trypsin to remove the cellular components. The resulting acelluar BAM was lyophilized to preserve its biochemical and structural integrity. The BAM was packed and exposed to 25 kGy of gamma irradiation for sterilization purpose. Histological, electron microscopical, and biochemical observations showed that the acellualr BAM had intact structural integrity of three dimensional collagen fibers and contained several growth factors, accelerating wound healing, such as EGF (Epidermal growth factor), KGF (Keratinocyte growth factor), and FGF (Fibroblast growth factor). Bovine herpes virus (BHV), bovine viral diarrhoea virus (BVDV), bovine parainfluenza virus type 3 (BPIV-3), and bovine parvovirus (BPV) were chosen as the biological indicators for validation of viral safety of the acellular BAM. Samples from relevant stages of the production process were spiked with each virus and subjected to viral inactivation processes. Viruses were recovered from the samples and then titrated immediately. All the viruses tested were completely inactivated to undetectable levels within 1 h of 70% ethanol treatment. Enveloped viruses such as BHV, BVDV, and BPIV-3 were more effectively inactivated than BPV by 0.05% sodium hypochlorite treatment. BHV, BVDV, and BPIV-3 were completely inactivated to undetectable levels by 25 kGy of gamma irradiation. Also BPV was effectively inactivated by 25 kGy of gamma irradiation. The cumulative log reduction factors of BHV, BVDV, BPIV-3, and BPV were ${\geq}$13.30, ${\geq}$14.32, ${\geq}$15.22, and ${\geq}$7.57, respectively. These results indicate that the production process for acelluar BAM has a sufficient virus-reducing capacity to achieve a high margin of the virus safety.

Carrageenan Content and its Chemical Composition of Eucheuma cottonii from the Coast of Philippine (Philippine산(産) Eucheuma cottonii의 carrageenan 함량(含星)과 그 성상(性狀)에 대하여)

  • One species of Rhodophyceae namely Eucheuma cottonii from the coast of Philippine was analyzed with respect to the content of carrageenan and such chemical characteristics as the content of sulphate and 3,6-anhydro-D-galactose and its solubility in potassium chloride solution. In addition, the same chemical properties were tested in the fractions separated by the different concentrations of potassium chloride. In comparison of the results of carrageenan in Eucheuma cottonii samples from Philippine and Chondrus ocellatus samples from Korea, carrageenan content in Eucheuma cottonii was higher than that of Chondrus ocellatus. Both samples showed more than forty-five percent carrageenan content. The Eucheuma cottonii carrageenan showed a higher 3,6-anhydro-D-galactose than the Chondrus ocellatus carrageenan. The sulphate content was higher in Chondrus ocellatus than Eucheuma cottonii. In fractionation of carrageenan by the solubility methods using potassium chloride solution, the yield of Eucheuma cottonii was highest in fraction I, fraction III was next and fraction II was the lowest.

Behavioral Response of the Western Flower Thrips Frankliniella occidentalis (Thysanoptera: Thripidae) to Different Chrysanthemum Flower Colors (국화 화색별 꽃노랑총채벌레의 행동반응)

  • Frankliniella occidentalis is attracted to flowers and is a major pest of chrysanthemums. Even when some chrysanthemum plants are not flowering, the ones that have already flowered attract F. occidentalis. Therefore, we investigated the efficacy of chrysanthemum as a trap plant that attract F. occidentalis by using an olfactometer. The numbers of F. occidentalis collected from the flowers of pink, wihte and yellow standard chrysanthemums on a tray with wet paper during the flowering period were 18.4, 56.6, and 52.6 respectively; the numbers of F. occidentalis collected from leaves were 7.8, 16.6, and 15.4 respectively. the numbers of F. occidentalis collected from the buds of pink, white and yellow standard chrysanthemums were 15.2, 45.8, and 41.6 respectively; the numbers of F. occidentalis collected from the leaves were 2, 8.8 and 3.4 respectively. In the Y-tube olfactometer test, the number of F. occidentalis attracted to the 2-way arms of the Y-tube was not significantly different for the yellow, red, violet and white flowers. In the four-choice olfactometer test, when the same visual cues and odor cues were provided, the frequency of F. occidentalis was higher in the yellow (10.7) flowers than in the red (1.3), violet (3.7) and white (2.0) flowers. When visual cues with disturbed odor cues, F. occidentalis preferred yellow (10.0) color over red (3.3), violet (1.3) and white (3.0) colors. When the same visual and odor cues, except for yellow visual cues, were provided, F. occidentalis preferred white (8.3) color over red (4.7), violet (4.7) and yellow (2.0) colors. Therefore, F. occidentalis were attracted to buds before the flowering of chrysanthemum plants and attracted to yellow flowers after the flowering.

Prediction on Mix Proportion Factor and Strength of Concrete Using Neural Network (신경망을 이용한 콘크리트 배합요소 및 압축강도 추정)

  • An artificial neural network was applied to predict compressive strength, slump value and mix proportion of a concrete. Standard mixed tables were trained and estimated, and the results were compared with those of the experiments. To consider variabilities of material properties, the standard mixed fables from two companies of Ready Mixed Concrete were used. And they were trained with the neural network. In this paper, standard back propagation network was used. The mix proportion factors such as water cement ratio, sand aggregate ratio, unit water, unit cement, unit weight of sand, unit weight of crushed sand, unit coarse aggregate and air entraining admixture were used. For the arrangement on the approval of prediction of mix proportion factor, the standard compressive strength of $180kgf/cm^2{\sim}300kgf/cm^2$, and target slump value of 8 cm, 15 cm were used. For the arrangement on the approval of prediction of compressive strength and slump value, the standard compressive strength of $210kgf/cm^2{\sim}240kgf/cm^2$, and target slump value of 12 cm and 15 cm wore used because these ranges are most frequently used. In results, in the prediction of mix proportion factor, for all of the water cement ratio, sand aggregate ratio, unit water, unit cement, unit weight of sand, unit weight of crushed sand, unit coarse aggregate, air entraining admixture, the predicted values and the values of standard mixed tables were almost the same within the target error of 0.10 and 0.05, regardless of two companies. And in the prediction of compressive strength and slump value, the predicted values were converged well to the values of standard mixed fables within the target error of 0.10, 0.05, 0.001. Finally artificial neural network is successfully applied to the prediction of concrete mixture and compressive strength.

Comparative Sedimentology for the Lacustrine Deposits of the Upper Gyeongsang Supergroup in the Southeastern Gyeongsang Basin, Korea (경상분지 동남부의 상부 경상누층군에 발달한 호성퇴적층에 대한 비교퇴적학적 연구)

  • The lithofacies, biofacies, and paleosol development of the Jindong Formation, the Geoncheonri Formation, and the lacustrine deposits of Mt. Hwangryeong at Pusan, which occur in the southeastern part of the Gyeongsang Basin, were analyzed in comparative sedimentology and in stratigraphy. The common features of these lacustrine deposits are: 1) clastic deposits are prevailing, 2) deltaic deposits are not associated, 3) mudflat deposits are common, and 4) stromatolites are absent. The distinct differences among these deposits are: 1) in the Jindong Formation, the mudflat deposits are predominant, pedogenic calcretes are commonly present, and dinosaur tracks frequently occur, compared with other two lacustrine deposits, and 2) in the Geoncheonri Formation, invertebrate fossils are relatively common and storm deposits are not recognized, compared with other deposits, and 3) evaporite mineral casts and tuffaceous turbidite deposits are common in the Mt. Hwangryeong lacustrine deposits. In stratigraphy, the Geoncheonri Formation is correlated with the lower part of the Jindong Formation, and the Mt. Hwangryeong lacutsrine deposits are deemed to overlie the Jindong Formation. On the basis of comparative sedimentology and stratigraphic relationship among these lacustrine deposits, general paleoenvironements of the southeastern part of the Gyeongsang Basin from the late Hayang time to the early Yucheon time are interpreted as follows. During the late Hayang time, tectonic and volcanic activities were generally inacitive in the Gyeongsang Basin, and lacustrine environments expanded since the paleoclimatic condition became less arid compared with the middle Hayang time. In general, however, paleoclimate during the late Hayang time was still arid, and wetting and drying periods were alternated. The occasional occurrences of severe droughts were also characteristic of the late Hayang time. Mudflats existed in wide area in the southeastern part of the Gyeongsang Basin during the late Hayang time, and sedimentation rate was accordingly low. The sedimentation rate became relatively high during the latest Hayang time and the early Yucheon time since tectonic and volcanic activities had been active. Generally arid climate continued for the early Yucheon time, enough for evaporite minerals to precipate occasionally.

  • The members of the Korean Association of Pediatric Surgeons conducted a retrospective study of two hundred and twenty-two cases of intestinal atresia for the period from January 1, 2007 to December 31, 2009. Seventeen hospitals were involved. There were 76 duodenal, 65 jejunal, and 81 ileal atresias (3 colonic). The male to female ratio was 0.85:1 in DA and 1.34:1 in JIA. Ninety-four patients(43.3 %) were premature babies (DA 40.3 %, JA 64.6 %, IA 28.8 %), and 70 babies (32.0 %) had low birth weight (DA 38.7 %, JA 44.4 %, IA 16.0 %). Antenatal diagnosis was made in 153 cases (68.9 %). However, 27 infants (17.6 %) with antenatal diagnosis were transferred to the pediatric surgeon's hospitals after delivery. Maternal polyhydramnios was observed in 81 cases (36.59 %) and most frequent with proximal obstruction. In forty-four cases (19.8 %), only simple abdominal film was taken for diagnostic study. The associated malformations were more frequently observed in DA - 61.8 % in DA and 22.6 % in JIA. Meconium peritonitis, small bowel volvulus and intussusception were more frequently associated with ileal atresia. The overall mortality rate was 3.6 %. Seasonal Occurrences of the Apple Leaf Miner, Phyllonorycter ringoniella (Matsumura) and Its Parasites and Damaging Leaf Position (사과굴나방 및 그기생충류(寄生蟲類)의 발생소장(發生消長)과 가해엽위(加害葉位))

  • Apple leaf miner(ALM), Phyllonorycter ringoniella(Matsumura), occurs $4{\sim}5$ generations a year in Suweon; adult emergence peaks being in mid April, early June, early July, mid August, and mid September with the highest one in the 4th generation. Numbers of days required to complete the development(egg to adult emergence) were different with oviposited dates; 43days for late May, $32{\sim}37$ days for June to July, and 39days for early August. ALM larvae oviposited after late August did not emerge and went into diapause. Sites of the infested leaf on the shoot seemed to be somewhat different with the ALM generations; the preferred leaf sites being the 1st-6th leaf for the 1st, the 4th-9th for the 2nd, the 4th-18th for the 3rd or 4th, and the terminal leaves of the first growth shoot or the leaves of the secondary shoot for the 5th generation. Parasites of three families emerged from the mines of ALM in Suweon. The encyrtid (Holcothorax testaceipes Ratzelburg) occurs 4 generations and the eulophids 5 generations a year. A small number of the braconid(Apanteles sp.) occcrred only in September.

Host Preference of Ricania spp. (Hemiptera: Ricaniidae) at Different Developmental Stages (갈색날개매미충의 발육단계별 기주선호성)

  • We investigated the host preference of Ricania app. at different developmental stages to select the effective control methods. Ricania spp. at different developmental stages preferred different host plants. During the nymphal period, individuals moved to the bottom due to environmental factors such as wind and rain, rather than inhabiting trees, and fed on various plants ranging from 1 year-old herbaceous plants to shrubs. Especially, at Palbong Mt. in Gonju, Ricania spp. nymphs preferred Symplocos chinensis f. pilosa, Celastrus orbiculatus, Euonymus alatus, Kalopanax septemlobus, Aralia elata, Styrax japonicas, Rubus parvifolius, Ru. coreanus, Ru. crataegifolius, Quercus acutissima, Cornus officinalis, Lespedeza bicolor, Albizia julibrissin, L. maximowiczii, Rhododendron schlippenbachii and Rh. indicum. At Yongbong Mt. in Hongseong, Ricania spp. adults preferred herbaceous plants, such as Helianthus annuus, Solanum nigrum, and Oenothera biennis rather than trees during the pre-oviposition stage. Subsequently, as the oviposition season began, Ricania spp. adults were rarely found in H. annuus, S. nigrum, or O. biennis and appeared to migrate to the perennial trees and shrubs hosts to lay eggs. At Palbong Mt., the main laying hosts of Ricania spp. adults were Weigela subsessilis, Ailanthus altissima, Q. aliena, Boehmeria platanifolia, Diospyros lotus, Ziziphus jujubae var. inermis, St. japonicus, Prunus mume, P. serrulata var. spontanea, Spiraea prunifolia f. simpliciflora, Ru. crataegifolius, Salix koreensis, Ru. yedoense f. poukhanense, and Co. controversa. The degree of egg laying, number of egg masses and adult Ricania spp. were significantly higher in sunlit areas than in shaded areas, in addition, the lower the altitude, the more adults were present. This study contributes toward the development of traps that can attract and control Ricania spp. and reduce the density of Ricania spp. that disturb the ecosystem.
