In this experiment, we report for the first time mass propagation by in vitro mircrotuberization of Codonopsis lanceolata. We first examined the effect of cytokinins on multiple shoot induction. 2.0 $mg{\cdot}L^{-1}$ of kinetin not only gave the highest rate of shoot induction (19.1%) but also the elongation of shoot (17.1 mm). Secondly, we investigated the effect of sugars on in vitro microtuberization from nodal segments. The diameter of tuberous roots was enlarged in the half-strength MS medium supplemented with 145.9 mM sucrose. Histological analysis revealed that the number of parenchymatous cell containing starch grains increased in the tuberous roots. In addition, unlike in non-tuberous root, vascular bundles were scattered inner cortex layer. Thirdly, in order to preserve and stimulate the germination, microtubers were stored at $4^{\circ}C$ refrigerator during 9 months and then transplanted to the artificial soils (vermiculrite : peatmoss = 1:1 v/v), resulting that the rates of survival and germination were 75% and 70%, respectively. These results indicated that mass propagation of C. lanceolata was achieved by in vitro microtuber formation, suggesting that this protocol might be applied for not only the propagation of elite clones but also conservation of C. lanceolata germplasm.
To study the conditions to enhance success of embryo transfer in the dog, 20 mixed-breed bitches were used for the experiment along with 4 male dogs for mating. The bitches were paired according to synchronism of natural estrus, or the counterpart as donor or recipient was treated with gonadotropin as FSH (follicular stimulating hormone) or PMSG (pregnant mare serum gonadotropin) for induction of estrus to be synchronized with estrus of the other bitch in natural estrus. Embryo recovery was performed in two ways for comparison, either by flushing each uterine horn after ovariohysterectomy or by flushing each horn in the state of non-ovariohysterectomy. In addition, the result of pregnancy according to the embryo stage and the repeatability of the experimental animals as donor or recipient were also investigated. FSH or PMSG was administered to the bitches which had passed over 4 months from last estrus, resulting in estrus-positive in 3 dogs of 6 FSH-treated dogs (50.0%), and in 5 dogs of 9 PMSG-treated dogs (55.6%), determined by proestrus signs and vaginal smear test. Estrus-positive bitches induced with gonadotropin were used as donor or recipient resulting in one embryo-recovered bitch as donor and one offspring-delivered bitch as recipient in 5 PMSG-treated dogs, whereas no result was obtained from 3 FSH-treated dogs. The rate of embryo recovery to be compared with number of corpus luteum was 68.2% in ovariohysterectomized dogs and 55.2% in non-ovariohysterectomized dogs, respectively. The number of dogs from which embryo was collected were 4 dogs of 6 ovariohysterectomized dogs (66.7%) and 6 dogs of 7 non-ovariohysterectomized dogs (85.7%), respectively. The result of parturition was obtained from one dog of 5 estrus-induced recipients, whereas no result was obtained from 3 natural-estrus recipients. The only dog which delivered a male puppy had been transferred 3 morulae and 2 blastocysts. Of 6 repeat-used bitches in canine embryo transfer, 3 dogs showed repeatability either as donor or recipient. These results indicated that inducing estrus of a dog with gonadotropin is feasible in canine embryo transfer to be synchronized with that of a natural-estrus dog, that embryo recovery is also possible in non-hysterectomized dogs, that the estrus-induced dog is also usable as recipient to result in parturition, and that repeat-use of a bitch as donor or(and) recipient is possible in canine embryo transfer.

The seed production of Ecklonia stolonifera Okamura was studied under laboratory conditions through the embryonic sporophyte stage and the field cultivation was conducted in eastern coast of Korea. The germination of zoospores occurred within 3 days and the growth of gametophytes was most rapid at $25^{\circ}C$ and $20\;{\mu}mol{\cdot}m^{-2}s^{-1}$. Sporophyte growth was highest at $20^{\circ}C$ and $20\;{\mu}mol{\cdot}m^{-2}s^{-1}$ and lowest at $25^{\circ}C$ and $80\;{\mu}mol{\cdot}m^{-2}s^{-1}$. In the nursery culture of E. stolonifera lasting for 2 weeks in January, the initial blade length of E. stolonifera (about $500\;{\mu}m$) grew to $526.3{\pm}176.0\;{\mu}m$ at water temperature of $12.05^{\circ}C$. The blade length and width reached their maxima in July, after which the ends of blade and stem began to degrade with the increase in water temperature. The degraded end of the blade started to regenerate in October, when water temperature began to decline. This species can be considered a potential candidate for aquaculture, increasing in the availability of raw material and aiding in recovery of seaweed bed.
Experiments were conducted to study the effect of auxins and cultural factors on rooting characteristics in offshoots cutting of Aloe vera. The result obtained were as follows;1. Short offshoots(15-20cm) with rhizome had more roots than that of the removed rhizome, while long offshoots(25-30cm) without rhizome had more roots and heavier root dry weight than short offshoots with rhizome. The highest rooting ability occured when IBA treated 500mg per liter in short offshoots with rhizome, whereas long offshoots without rhizome showed higher rooting ability at the level of 1,000mg/l of IBA and 200mg/I of NAA2. Among three kinds of anxin treated, both IBA and NAA showed better rooting ability than ethychlozate. Root number, root length and root dry weight were higher at 10 min. dipping treatment of IBA 500mg/I for short off-shoots without rhizome and 10 min. dipping treatment of IBA 1,000mg/l for long offshoots with rhizome. Rooting ability of long offshoots was higher than that of short offshoots.3. In addition to IBA 1,000mg/l, 7-day dryness under shading condition increased root number and dry weight of root in offshoots without rhizome. Control treatment lengthened root by increasing drying period under the shading condition.4. The plain soil with mamure had not only higher root dofferentiation but higher root growth of offshoots cutting than the artificial soil.5. Rooting ability in offshoots cutting had a tendency increasing with decrease in shading percentage to natural. Black color poly ethylene film mulching had better rooting ablility than transparency poly ethlene film mulching in offshoots cutting.
This experiment was carried out to investigate the fertilization and the size of mature female and male gametophytic parts of Pulsatilla koreana after artificial pollination. The size of pollen is $26.5{\mu}m$ at the time of anther dehiscence, that is, about $3{\sim}4$ days after pollination. Synergid nucleus, egg nucleus, and polar nucleus are 10.0, 15.0, and $32.5{\mu}m$ respectively at the time of completing egg apparatus formation, that is, about 2 days after pollination. Poller tubes germinate on stigma about 10 hours, passing lower part of style about 30 hours, penetrating into micropyle about 35 hours after pollination. Sperm nucleus penetrates into polar nucleus about 40 hours and egg cell about 48 hours after pollination. But, there seems to be different among the individuals. Multinuclei and multinucleoli are formed in egg cell, synergid, and polar nucleus about the time of fertilization. Proembryo is formed about 4 days, being changed to large globular form about $6{\sim}8$ days after pollination. Endosperm nuclei divide into free nuclei after fertilization and change to cotyledon in gymnosperm. There seems to be same phenomena in Pulsatilla koreana.

Metagenomics is a powerful tool to isolate novel biocatalyst and biomolecules directly from the environmental DNA libraries. Since the metagenomics approach bypasses cultivation of microorganisms, un-cultured microorganisms that are majority of exists can be the richest reservoir for natural products discovery. To discover novel herbicidal substances from soil metagenome, we established three easy, simple and effective high throughput screening methods such as cucumber cotyledon leaf disc assay, microalgae assay and seed germination assay. Employing the methods, we isolated two active single clones (9-G1 and 9-G12) expressing herbicidal activity which whitened leaf discs, inhibited growth of microalgae and inhibited root growth of germinated Arabidopsis seeds. Spraying butanol fraction of the isolated active clones' culture broth led to growth retardation or desiccation of Digitalia sanguinalis (L) Scop. in vivo. These results represent that the screening methods established in this study are useful to screen herbicidal substances from metagenome libraries. Further identifying molecular structure of the herbicidal active substances and analyzing gene clusters encoding synthesis systems for the active substances are in progress.

A new standard rose cultivar "Snow Day" was bred from the cross between pink standard cultivar "Saphia" and pink standard cultivar "Bridal Beauty" at the National Institute of Horticulture & Herbal Science. The cross was made in 2003 and "Snow Day" was finally selected in 2008 after evaluating characteristics for three years from 2006 to 2008. A white standard cultivar, "Snow Day" grows vigorously and resistant to powder mildew. The major characteristics of this cultivar are $152stems/m^2/year$ in yield, 81.8 cm in length of cut flower, 11.3 cm in flower diameter, 29.2 in petal number, and 13.7 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer's preference for this cultivar is relatively higher than that for the control cultivar, "Tineke".
A new standard white-pink flower rose cultivar 'Innocence' was bred from the cross between white standard cultivar 'Juvena' and green-white standard cultivar 'Green Success' at the Jeollanamdo Agricultural Research and Extension Services (JARES). The cross was made in 2001 and, 'Innocence' was finally selected in 2006 after investigating characteristics for three times from 2004 to 2006. 'Innocence' a white-pink standard cultivar has good flower shape with few prickles. The major characteristics of this cultivar were $118.7stems{\cdot}m^{-2}$ in year yield, 68.0 cm in length of cut flower, 10.9 cm in flower diameter, 34.4 in petal number, and 10.7 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer's preference of this cultivar was relatively higher than that of control cultivar, 'Rose Yumi'.

참삿갓사초는 전국에 분포하는 여러해살이풀로 사료용, 사방용 소재로 사용되며 조경용, 관상용 지피식물로 사용되기도 한다. 참삿갓사초의 대량증식을 위해 가장 효과적인 파종 시기를 추정하기 위해 생장상을 이용해 인공적인 온도조건을 주고 참삿갓사초 종자의 발아율을 조사하였다. 실험을 위해 온도를 항온조건(30, 25, 20, 15, $5^{\circ}C$)과 변온조건(15/30, 20/10, $25/15^{\circ}C$) 총 8가지 조건으로 설정하여 발아율을 매일 측정하였다. 광조건은 명/암을 매일 12시간씩 반복하였다. 항온조건에서 발아율은 $20^{\circ}C$가 $17{\pm}7%$로 가장 우수하였고, $25^{\circ}C$ ($12{\pm}2.8%$), $30^{\circ}C$ ($3{\pm}1.9%$) 순으로 나타났으며, $15^{\circ}C$, $5^{\circ}C$에서는 발아하지 않았다. 그러나 $20^{\circ}C$와 $25^{\circ}C$조건에서 통계적인 유의차는 발생되지 않았다. 한편, 변온조건에서의 발아율은 $15/30^{\circ}C$가 $91{\pm}3.8%$로 가장 우수하였고, $25/15^{\circ}C$ ($89{\pm}4.1%$), $20/10^{\circ}C$ ($69{\pm}3.4%$) 순으로 나타났다. 그러나 $15/30^{\circ}C$와 $25/15^{\circ}C$조건에서 통계적인 유의차는 발생되지 않았다. 따라서 참삿갓사초의 종자발아를 위해서는 변온이 필요하며, $25^{\circ}C$ 이상의 온도가 필요하다. 이러한 결과를 바탕으로 가장 효과적인 파종 시기를 추정할 때, 낮-밤 $10^{\circ}C$ 이상의 기온차가 있는 초여름~늦여름 사이가 가장 적절할 것으로 판단된다.

As a result of examining the germination rate between ginseng varieties, Jagyongjong varieties had the highest germination rate, and Yeonpung. had the lowest germination rate. In the ginseng seed germination rate experiment, the highest germination rate and growth condition were shown in artificial soil conditions of the ratio of Peatmoss 6.5: Pearlite 2: Masato 1.5. Good soil conditions require adequate soil moisture forces during the incubation period. The cultivation of ginseng medicinal crops requires optimal soil breathability, soil pH, and soil stabilization, which are important for root breathing. Microbial activity in the soil has a great influence on the growth of ginseng. The optimum pH of the soil for ginseng cultivation is 5.0-5.5 As a result of the experiment, the soil remained in an appropriate range after a month. In general, when the EC concentration value of the soil for ginseng cultivation is 0.2 mS/cm or more, growth deteriorates, and when the EC concentration value is 0.5 mS/cm or more, concentration obstacles such as root decay occur. As a result of the analysis, the higher the concentration value of EC, the more likely it is to interfere with ginseng growth.
