Evaluation of Anti-inflammatory and Antioxidant Abilities of Complex Extracts Produced from Leonurus japonicus Houtt., Houttuynia cordata Thunberg, and Citrus unshiu Markovich (익모초, 어성초, 진피 복합 추출물의 항염증 및 항산화 평가)

  • This study aimed to confirm the potential as a material for food, medicine, and cosmetics by measuring the physiological activity of the complex extract(Leonurus japonicus Houtt., Houttuynia cordata Thunberg and Citrus unshiu Markovich). The complex extract was with hot water, The evaluation of cytotoxicity, antioxidant, anti-inflammatory, and MAPKs of the extract was performed by using ELISA and western blot. Total polyphenols and flavonoid contents of complex extracts were 126.16 mg/g and 105.84 mg/g, respectively. The scavenging activities of DPPH and ABTS radical significantly increased in a dose-dependent in complex extracts. Compared to the control group, the complex extracts treatments significantly reduced concentrations of nitric oxide and inflammatory cytokine (i.e., IL-1β, TNF-α, and IL-6). Furthermore, the complex extracts treatments significantly reduced protein expression levels of MAPKs (i.e., ERK, JNK, and p38). The results indicate that the complex extract can be used for oxidative damage and inflammation. Thus, the results of this study can be used as basic data for composite materials for food, medicine, and cosmetics.

  • This study investigated the anti-inflammatory, trypsin activity, and antioxidant effects of 11 kinds of plant extracts to discover materials for developing optimal mixtures that improve inflammation and help digestion. Ziziphus jujuba Mill. (ZJ), Leonurus japonicus Houtt. (LJ), Scutellaria baicalensis (SB), Platycodon grandiflorum, and Aster scaber extracts had excellent anti-inflammatory effects by reducing excessive nitric oxide (NO) and tumor necrosis factor-α content in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. The malt (MA), Pyrus pyrifolia Nakai (PP), Raphanus sativus L., Platycodon grandiflorum extracts among the 11 kinds of plant extracts had high trypsin activity. The antioxidant activity of the plant extracts was examined by the DPPH radical scavenging activity, and the SB, PE, JU, and MA extracts had high antioxidant activity. Therefore, PP, MA, ZJ, LJ, and SB were selected to develop optimal mixtures that improve inflammation and help digestion. The extract of plant mixture containing PP, MA, ZJ, LJ, and SB in the ratio 1:1:2:1:2 (w/w) significantly inhibited NO production than the extract of PP, MA, ZJ, LJ, and SB, respectively, in LPS-stimulated RAW 264.7 macrophages. The DPPH radical scavenging activity of the mixture extract was significantly higher than the extract of PP, MA, ZJ, and LJ, respectively.

  • The essential oil fraction of Leonurus sibiricus was analyzed by TLC and gas chromatography. By utilizing silica gel column, a ketone compound, m.w. 167, was isolated from the essential oil. The essential oil showed considerably the diuretic action, but the water extract exhibited weak action. This diuretic action of the water extract was potentiated by combined administration of essential oil. On the isolated rabbit's uterus the essential oil decreased spontanous movement and showed relaxation.

  • By the 505 chromotest which utilized Escherichia bolt PQ 37, Korean medicinal plants had been screened to Investigate the antimutagenic effect to aflatoxin B1(AFBl). Ikmocho(IMC, Leonurus sibiricus L.) was extracted with hot water. The extract was not found to be mutagenic in the Salmonella mutation test with or without metabolic activation, and the extract was showed to possess the antimutagenic properties towards AFB1-induced metation. The mutagenicity of AFB1 was inhibited by methanol soluble fracstion (IMC-MS) in dose-dependent. However, water-soluble fraction exhibited comutagenic activity. The greatest inhibitory effect of IMC-MS on AFB1 mutagenicity occurred when IMC-MS was first incubated, AFB1 followed by a second incubation with the cells and 59 mixture. Also lower inhibition was occurred when S9 mixtures were first incubated, with IMC-MS followed by a second incubation with AFBI. The results of the sequential incubation study support the probability that one mechanism of inhibition could involve the formation of chemical complex between IMC-MS and AFB1 rather than deactivation of S9 enzyme.

  • Ig Mo Cho (益母草) has been used as remedy for invigorating blood circulation to remove blood stasis and relieving edema by inducing diuresis. With regard to the botanical origin of Ig Mo Cho, it has been considered to be Leonurus spiecies of Labiatae, but there has never been pharmacognostical confirmation on it. To clarify the botanical origin of Ig Mo Cho from Korea, the anatomical characteristics of Leonurus japonicus and L. sibiricus were studied. As a result, the botanical origin of Ig Mo Cho from Korea was proved the herba of Leonurus japonicus.

  • For the quality control of Leonuri Herba, alkaloid compound, leonurine, was isolated from the MeOH extract of Leonurus sibiricus L. (Labiatae) and identified by the spectroscopic analysis. A quantitative analysis of leonurine using HPLC method showed that the average contents were $0.124{\pm}0.065%$ in 34 samples collected throughout the regions of Korea.

Anti-hypertensive Effects of Pini Folium and Leonuri Herba Extract on Spontaneously Hypertensive Rat (SHR) (SHR(Spontaneously Hypertensive Rat)를 이용한 송엽, 익모초 추출물의 항고협알 작용)

  • The aim of this study was to investigate a potential effect of treatment of spontaneously hypertensive rats (SHR) with Pini Folium or Leonuri Herba extracts. Male SHR were treated with extracts for 2 or 4 weeks starting at 13 weeks of age. We found that oral treatment with the methanol extract from Pini Folium or Leonuri Herba (1g/Kg/day) significantly decreased mean blood pressure to 85.4% and 78.6% respectively in compare with untreated control SHR. Treatment with the extract from Pini Folium for 4 weeks during hypertension development phase (starting 9 weeks of age) resulted in a partial prevention of hypertension development. There were no significant differences for body weight between untreated and Pini Folium treated SHR.

  • These studies were carried out to investigate effects of Leonurus sibiricus green juice on gastric secretion of rats and acute toxicity in mice ; $LD_50(mg/kg)$ of Leonurus sibiricus extract was 625mg/kg intraperitoneally in mice. The groups administered green juice and water extract of Leonurus sibiricus showed more excellent increasing effect in total acidity and peptic activity, and decreasing effect on gastric volume than control group. But any differences were not found between sample and control group in gastric pH. It is suggested that Leonurus sibiricus can he used as the bitter stomachics to promote the gastric digestion.

  • These studies were conducted to investigate the effects of Leonurus sibiricus extract (L.S.E.) on experimentally $CCl_4-induced$ liver damage and alloxan-induced diabetes in rabbits; Leonurus sibiricus juice (L.S.J.) showed the most rapid recuperation of liver function among samples compared to the control group in $CCl_4-intoxicated$ rabbits. SGPT activity stowed an apparant decreasing effect in 4 days, and the blood level of total cholesterol and total bilirubin in 6 days, and not in alkaline phosphatase level. L.S.J. exhibited more excellent hypoglycemic effect in alloxan-induced diabetes of rabbits, that is, significant to the control group after 2 days and adjacent to the normal level on 8th day. And SGPT activity and total cholesterol level were gradually decreased and showed markedly decreasing effect after 4 days.

  • Objectives:The present study purposed to evaluate the effects of Leonuri Herba and Leonuri Semen on hypercholesterolemia and provide data for the appropriate clinical application of Leonuri Herba and Leonuri Semen. Methods:We applied the water extracts of Leonuri Herba and Leonuri Semen to rats with hypercholesterolemia induced by high-cholesteroldiet, and examined their effects. Results:The levels of the total cholesterol and LDL-cholesterol were reduced by the uptake of Leonuri Herba to rats with hypercholesterolemia, but there was no significant effect on the level of TG(triglyceride), HDLc(HDL-cholesterol), FFA(free fatty acid), and Fc(free cholesterol), while the amount of AST and ALT was inhibited. The application of Leonuri Semen brought about the decrease of the total cholesterol and Fc, the increase of TG, HDLc, and FFA, and the significant decrease of the activation of AST. Leonuri Herba had only a weak effect on hypercholesterolemia, while Leonuri Semen had a strong effect on the reduction of the symptoms of hypercholesterolemia. Conclusion:Based on the fact that Leonuri Herba had only a weak effect on hypercholesterolemia, while Leonuri Semen had a strong effect on the reduction of the symptoms of hypercholesterolemia, it is concluded that Leonuri Semen is a better solution for hypercholesterolemia than Leonuri Herba.
