Studies on the Isolation of Auxotrophic Mutants of Bacillus subtilis and Escherichia coli (Adenine 요구변이주(要求變異株)의 분리(分離)에 관(關)한 연구(硏究))

  • In order to obtain amino acids and nucleic acid derivatives from adenine auxotrophic mutants of Bacillus subtilis and Escherichia coli, vitamin, nucleic acid analogue, streptomycin as well as ultraviolet light were adopted for the production of adenine auxotrophic mutants and the results showed efficient production of desired mutants. 1. Ultraviolet ray $(2530\;{\AA}\;2080\;erg/mm^2)$ irradiation to Bacillus subtilis and E. coli at a distance of 30 cm for 80-90 sec. and for 15-20 sec. respetively induced four and eight strains of auxotrophic mutants. 2. Treatment of aminopterine$(200\;{\mu}g/ml)$ inhibited the growth of Bacillus sultilis significantly but a subsequent irradiation of ultraviolet light at the above mentioned conditions induced six times as much mutants as compared to the irradiation alone. In case of E. coli a similar tendency was observed with treatment of streptomycin$(200\;{\mu}g/ml)$ with doubled induction rate of adenine auxotrophic mutants as compared to the irradiation alone.

  • Five varieties of sweetpotatoes recommended in Korea were investigated with respect to the storing capacity, resin content and the possibility of developing sweetpotato chips as a new processed food item. The results are summarized as follows: 1) Two varieties, Suwon No. 147 and Chun-Mi were more resistant to chilling injury and soft-rot decay than other varieties. 2) The contents of resinous and polyphenolic substances were quite different depending upon the variety. 3) Sweetpotato chips of different color were made from different varieties and rapeseed oil was found to be the best as frying oil. 4) Best conditions to prepare sweetpotato chips with fresh color and proper texture were to dip slices of 1-2 mm thickness in 0.25% sodium bisulfite solution at $40^{\circ}C$ for 30-40 minutes and to subject to deep frying in an oil bath at $150-160^{\circ}C$ for 2.5 to 3.5 minutes. 5) Polyethylene-cellophane film as packing material of sweetpotato chips was the film in the moisture proof and film-impact tests.

  • 1. In order to elucidate the differences between varieties of Korean soybean, forty-nine subjects were examined for their chemical constituents. Their proportions varied considerably with the differences in variety. The moisture varied from 10.93% to 12.57%(aver. 11.95%), protein; 33.79% to 47.00%(aver. 39.8%) crude fat; from 16.32% to 24.79%(aver. 19.2%), crude fiber; from 3.03% to 5.57%(aver. 3.98%), ash; from 3.95% to 5.23%(aver. 4.6%) and nitrogen free-extracts (earbohydrates); from 16.58% to 26,81% (aver. 20.57%) Chemical composition of soybean was affected by the varieties as well as their localities of growth. 2. Of the total protein, 33.26% based on the soybean meal was soluble protein and 1.94% insoluble protein in average.

  • As a part of the survey on the chemical compositions of Korean Sweet potatoes, the changes of chemical compositions of Suwon No. 118 and Suwon No. 147, which are representative varieties of sweet potatoes in Korea, were studied during storage. The result of the study showed the followings. 1. The moisture content of the both varieties were fairy uniform with higher value of Suwon No. 147 than another. 2. The starch content and crude starch yield in the stored root tuber were shown to be considerable uniform except decrease at the earlier part of storage and in January. 3. Reducing sugar content of them showed increase at earlier part, then almost uniform except gradual decrease in later part of the storage. 4. The content of total soluble sugar were gradually increased at the earlier part, then became almost uniform at the later part of the storage. 5. The sucrose content showed gradual increase except at the earlier part of the storage in which both varieties showed different feature in decrease of the content. 6. The content of crude protein and soluble protein were almost uniform throughout the storage.

  • In order to fractionate the rice protein employing paper electrophoresis, 9 subjects of Korean rice and one Indica type, Pin Galw 50 were examined, the results were as follows. 1. Polished rice protein was separated into albumin, globulin, prolamine, and oryzenin. The amount of these fractions was determined by Kjeldahl method showing respectively 0.26%, 0.65%, 0.41%, and 5.01% in average. Albumin was extracted with deionized water, globulin with 10% NaCl, prolamine with 70% ethanol, and oryzenin with 0.05N-NaOH. 2. Albumin was extracted with deionized water and dialyzed by a cellophan tube. The supernatant was submitted to paper electrophoresis using phosphate buffer (pH 7.6, ${\mu}$ 0.18). Albumin was identified as monocomponent in all of 10 varieties under study. Globulin was extracted and dialyzed to remove the albumin. The precipitates were resolved in 10% saline solution and examined by paper electrophoresis. The globulin consists of two components in phosphate buffer(pH 7.6, ${\mu}$ 0.18) Any subject, regardless the origin, appears to contain globulin I and globulin II. Prolamine was extracted with 70% ethanol, dialayzed against deionized water, resolved with ethanol, and analyzed by Paper electrophoresis. It was proved as one component in the 70% alcoholic buffer(pH 9.0, ${\mu}$=0.0095). On the contrary, paper electrophoresis with oryzenin demonstrated two or three components in Sorensen's buffer(pH 13.0, ${\mu}$ 0.11). Yookoo 132, Dungpan 5, Kwansan, and Jaekun contain oryzenin I, oryzenin II, and oryzenin III. On the other hand, Paldal, Jinheung, Sukwang, Eunbangzu, Damakum, and Pin Galw 56 contain only oryzenin II, and oryzenin III. On the basis of these analyses a discussion of the differences between the protein fractions of 10 varieties was presented. 3. Globulin I varied from 0.22% to 0.46% (aver. 0.35%) in the amount, globulin II from 0.21 to 0.44%(aver. 0.32%), oryzenin I from 0.17% to 0.44%(aver. 0.3%), oryzenin II from 1.59% to 2.88%(aver. 2.23%), and oryzenin III from 2.02% to 3.57%(aver. 2.66%).

Effects of the time, and the rate of potassium fertilization at nursery stage on the rooting activity in paddy rice plant (수도(水稻)의 발근(發根)에 미치는 묘대가리시용(苗垈加里施用)의 영향(影響))

  • In order to learn the effect of timing and rate of potash fertilization in the rice plant nursery on the root growth and the vegetative growth of later stage a pot experiment was carried out employing 'Jaekun' a rice variety. Potassium Chloride was applied at the rate of $0{\sim}100g\;K_{2}O/3.3m^2$ right before, and 30 days after seeding. Forty-day old seedlings were transplanted three times successively at the one week intervals with the roots cut each time. Each subject was observed in rooting and other useful traits. A part of seedlings after the third root scission were cultured intact thereafter and the yield characters examined. The results were shown as: 1. The rate of sound seedlings was high in the case that a small amount of potassium was applied as the basic placement while the application of the fertilizer over $75g/3.3m^2$ (as $K_{2}O$) yielded a far less crops. 2. The plant height, number of tillers, number of leaves, and grass weight increased as the amount of potassium was elevated. However, the application of $50g\;K_{2}O/3.3m^2$ in the basic placement and that of $75g\;K_{2}O/3.3m^2$ were the peaks over which the above mentioned characters were weakened. 3. As the amount of potassium was increased the increment in number of rooting was notable, especially when the scission of roots was repeated within the limit of $50{\sim}75g\;K_{2}O/3.3m^2$. 4. In the maximum root newly shooted length indicated was about the same tendency although no statistical significance was observed. 5. The plant height, number of tillers, and variation of weight between grass and root indicated a similar tendency as the number of root shoot. 6. The differences in number of ears, grain yield, and weight of straw between the treatments were not so great but showed somewhat similar trend as in the growth of transplanted plants. In the treatment-100g $K_{2}O/3.3m^2$ the yield (ears, grain and straw) decreased as in the non-fertilized.

  • A survey of the free amino acids and free sugars in the juice of fourteen citrus varieties was made by means of paper chromatography and the pectin content in the rind was detected The results of the survey are summarized as follows. 1) Sixteen aminoacids detecteu in fourteen varieties under the condition of this study were distributed with. different frequency as follow. Proline, gamma-amino-butyric acid, alanine, valine, serine, glutamic acid, aspartic acid (in 14 varieties), lysine(12), leucine threonine(11), isoleucine(10), arginine(9), glycine(6) ${\beta}-alanine$(4) asparagine(3) unknown(2). 2) Proline, gamma-amino-butyric acid were mostly abundant in all varieties and alanine was next in the amount. 3) The varieties in the decreasing order of number of amino acids container were C. grandis madow C. leiocarpa (14 acids), C. sulcata, C. hassaku. native citron, Fortunella japonica(13) C. grandis heiko, C. Tamurana iyo, C. nobilis(12) C. reticulata C. junos(11) C. natsudaidai(10) C. miyakawa unshiu, C. ohali unshiu(9). 4) The varieties which appear to contain all essential amino acids(6 acids)detected were C. grandis madow C. grandis heiko, C. sulcata, C. Tamurana iyo, C. hassaku and native citron, and C. natsudaidai, C. unshiu were the least (1-3 acids). 5) Glucose fructose, sucrose and maltose were detected in all varieties. 6) The pectin content in the rind ranged from 8.64% F.W.(C. junos) up to 17.0% for C, grandis madow and the mean was $11.63{\pm}2.69%$.

  • A pot experiment with $P^{32}$ was carried out to investigate the soil phosphorus availability to four leguminous forage crops and three graminaceous, of black volcanic ash soil and red one. Soil phosphorus was extracted with 6 different extractants and also fractionated in Fe, Al and Ca phosphorus. The results were: 1) Soil phosphorus availability was in decreasing order of Italian rye grass${\gg}$ soybean> cassia> corn> weeping love grass${\gg}$ Korean lespedeza> Red clover and they might be grouped into three levels by A-value, over 1000, 200-500 and below 40 $p_{2}O_{5}\;kg/ha$. 2) The amount of various available phosphorus and phosoborus fraction in the black soil was higher than that in the red soil. No difference in phosphorus availabiliy to forage crops was shown between two soils. Therefore an extractant able to draw out similar amount of phosphorus from two soil will be suitable for determining the phosphorus availability index. 3) Two extractants, one extracting 20 ppm as maximum and the other extracting 100 ppm as minimum will be recommendable for determining the availability of phosphorus; the former for red clover and Lespedeza and the latter for others. Truog method may be good for the former but no appropriate method for the latter was found in the methods used. 4) T/R ratios of legumes were negatively correlated at 5% level with % phosphorus from fertilizer (% pdF). Legumes showed below 50 of % pdF over 5 of T/R ratio and over 80 of % pdF below 5 of T/R.

Comparison of the Content of Shizandrin, Gomisin A and Gomisin N in Schisandra Fruit by Water Extraction Condition (오미자의 물 추출조건에 따른 shizandrin, gomisin A, gomisin N의 함량 비교)

  • This study compared the content change of physiological active substances according to the extraction condition when we generally drink the extractive of Schisandra fruit with using water. Physiological active substances of the Schisandra fruit, schizandrin, gomisin A and gomisin N were analysed with HPLC. The results of extracting 1 dose of the Schisandra fruit tea, which is some as 1 g of Schisandra fruit, in 100 mL of water by differing the extraction type, extraction temperature and extraction time were of the followings. The total content of schizandrin, gomisin A and gomisin N of powder type in $26^{\circ}C$ leaching was the highest (p < 0.05) as the leaching content of 24 hours (5.54 mg/g) showed difference with different times and the contents of raw type Schisandra fruit were less than 0.1 mg/g in all of extraction time. The contents of powder type Schisandra fruit in $60^{\circ}C$ heating extraction was over 4.50 mg/g except for the 5 minute (2.94 mg/g), and the contents of raw Schisandra fruit were less than 0.3 mg/g. In case of $100^{\circ}C$ heating extraction, the contents of powder type Schisandra fruit showed over 5.10 mg/g in every time zone, and the raw type Schisandra fruit was less than 1.0 mg/g.

  • Acrylamide gel electrophoretic pattern of 13 wild soybean (Glycine ussuriensis) was compared with that of G. gracilis and G. max var. Gwanggyo. Average Protein content (50%) of wild soybean was greater than that of C. gracilis (46%) and Gwanggyo (45%). Grain weight of wild soybean was one third of G. gracilis and one ninth of Gwanggyo. Electrophoresis of wild soybean protein showed total 16 different bands and three of which (Rm 0.09, 0.59 and 0.84) were specific and did not appeared in 86 var. of G. max which showed four specific bands (Rm 0.35, 0.45, 0.50 and 0.77) of total 17 bands. G. glacilis had all bands of Gwanggyo and two bands (Rm 0.53 and 0.59), one of which (Rm 0.59) was specific for wild soybean indicating that G. gracilis is middle type. Of 16 protein bands the third band (32%), the first band (28%) and the 5th band (13%) were main bands. Electrophoretic pattern could be sorted qualitatively into 4 groups, semiquantitatively into 6 groups and 2 or 4 groups depending on reference pattern by correlation or pattern similarity method. All sorting methods separated a wild soybean from Sogri mountain into a group and except that there were no similarity among methods but correlation methods seems more reasonable. Protein content was no relation with electrophoretic pattern but positively correlated with percent contribution of first band at 5% level suggesting that the first band may have a important role for protein synthesis.
