Isolation and Characterization of Pectinase-Producing Bacillus sp. BS-214 (Pectinase를 생산하는 Bacillus sp. BS-214의 분리 및 특성)

  • A bacterial strain BS-214 producing extracellular pectinase was isolated from soil. The isolated bacterium was identified as a strain of Bacillus so. based on the morphological, biochemical, and physiological characteristics. Cell growth and pectinase activity of Bacillus sp. BS-214 were reached to a mixium in the culture condition of pH 8.5 at 4$0^{\circ}C$. Production of pectinase by the strain was the highest when polygalacturonic acid was added to culture medium as a carbon source, and its optimal concentration was 1%. Also, yeast extract was used as the best nitrogen source for the production of pectinase by the concentration of 0.25%. Decomposition of a constituent of Edzeworthia papyrifera by the strain was observed by scanning electron microscope.

  • This paper is about the development of automatic FPC(flexible printed circuit) punching instrument for the improvement of working condition and cost saving. FPC is used to detect the contact position of keyboard and button like a cellular phone. Depending on the quality of the printed ink and position of reference punching point to the FPC, the resistance and current are varied to the malfunctioning values. The size of reference punching point is 2mm and the above. Because the punching operation Is done manually, the accuracy of the punching degree is varied with operator's condition. To improve this manual punch ing operation to the FPC, automatic FPC punching system is introduced. Test algorithms and programs showed good results to the designed automatic punching system and led to the increasement of productivity and huge cost down to law material like FPC by avoiding bad quality.

  • In this paper, we have analyzed the BER (Bit Error Rate) performance of MC-CDMA systems under indoor wireless environment with class A impulsive noise suggested by Middleton. For the performance evaluation, we considered from strong to weak impulsive noise characteristics. And we have evaluated the degree of performance improvement by using turbo code as a compensation technique to impulsive noise. From the result of analysis, it is found powerful performance improvement scheme must be adopted in the system under impulsive noise environment, and when turbo code scheme is adopted, system BER performance is improved by the $10^{-2}$. As the impulsive noise characteristics approaches AWGN characteristics, the degree of performance improvement by adopting turbo code become larger.

Molecular Cloning and Phylogeny of the Human Endogenous Retrovirus HERV-W LTR Family in cDNA Library of Human Fetal Brain (인간 태아의 뇌로부터 만들어진 cDNA library에서 내생 레트로바이러스 HERV-W LTR의 클로닝 및 분자계통분류)

  • Long terminal repeats(LTRs) of the human endogenous retrovirus(HERV) heve been found to be coexpresed with genes located nearby. It has been suggested that the LTR elements have contributed to the genetic variation of human genome connected to various diseases. Recently, HERV-W family was identified in the cerebrospinal fluids and brains of individuals with schizophrenia. Using cHNA library derived from human fetal brain, we performed PCR amplification and identified seven new HERV-W LTR elements. Those LTR elements showed a high degree of sequence similarity(98∼99%) with HERV-W (AF072500). A phylogentic tree obtained by the neighbor-joining method revealed that seven new HERV-W LTR elements(FB-1, 2, 4, 8, 9, 10, 12) were closely related to the AX000960, AF072504, and AF072506 from Gen Bank database. Our data suggest that several copy numbers of the HERV-W LTR elements are expressed in human feta brain and may contribute to an understanding of biological function connected to neuropsychiatric diseases.

Characterization of Crude Oil Degradation by Klebsiella sp. KCL-2 Isolated from Sea Water (유류오염 지역에서 분리된 Klebsiella sp. KCL-2에 의한 원유분해 특성)

  • Several bacterial strains utilizing crude oil as their sole carbon and energy source were isolated from polluted marine by crude oil. One of the strains, named KCL-2 showed strong degradation activity for crude oil. This strain was identified as a Klebsiella sp. based on the morphological, biochemical, and physiological characteristics. The optimum cultural conditions were as follows; $27^{\circ}C$~$37^{\circ}C$ for temperature and 7.0 for initial pH. Additionally, the optimal concentration of sodium chloride was 3.0%, confirming indicating that this strain was derived from sea water.The strain KCL-2 could use several kinds of n-alkane hydrocarbones from octadecane to octacosane as a sole carbon source. The emulsifying activity by KCL-2 was the highest after 3 days of cultivation under the condition of 3.0% sodium chloride, pH 7.0 and 32$^{\circ}C$. This strain had several criptic plasmids.

Isolation and Characterization of Klebsiella pneumoniae WL-5 Capable of Decolorizing Triphenylmethane and Azo Dyes (트리페닐메탄계와 아조계 색소를 탈색할 수 있는 Klebsiella pneumoniae WL-5의 분리 및 특성)

  • A Klebsiella pneumoniae WL-5 with the capability of decolorizing several recalcitrant dyes was isolated from activated sludge of an effluent treatment plant of a textile and dyeing industry. This strain showed a higher dye decolorization under static condition and color removal was optimal at pH 6-8 and $30-35^{\circ}C$. More than 90% of its color of Congo Red were reduced within 12 hr at $200\;{\mu}M$ dye concentration. Malachite Green, Brilliant Green and Reactive Black-5 lost over 85% of their colors at $10\;{\mu}M$ dye concentration, but the percentage decolorization of Reactive Red-120, Reactive Orange-16, and Crystal Violet were about 46%, 25%, and 13%, respectively. Decolorizations of Congo Red and triphenylmethane dyes, such as Malachite Green, Brilliant Green, and Crystal Violet were mainly due to adsorption to cells, whereas azo dyes, such as Reactive Black-5, Reactive Red-120, and Reactive Orange-16 seemed to be removed by biodegradation through unknown enzymatic processes.

New Weight-reduction Design of the Fifth Wheel Coupler with a Trailer by Using Topology Optimization and Durability Tests (위상최적설계를 통한 트레일러 제5차륜 연결구조물의 경량화 및 내구성)

  • The fifth wheel coupler is a heavy automotive coupling structure which connects a tractor and a trailer used for heavy-duty trucks widely. It is subjected to various loads simultaneously such as rolling, pitching and yawing loads as well as coupling frictional and impact loadings. Most of existing couplers have been overdesigned and, therefore, it is necessary to reduce the dead weight to increase the fuel efficiency. The topology optimization was applied in order to find conceptual layout designs which could show major load paths and ribs locations, and then the size structural optimization was performed in order to determine the heights and thicknesses of coupler ribs with the predetermined various loading conditions for the development of a new slim coupler with a minimum weight and high enough strength and stiffness. As the results of the topology optimum design, an efficient new coupling structure for truck trailers was designed. The weight of the new fifth wheel coupler was reduced by 4.9 %, compared with the existing one, even though all strength requirements were satisfied. The fatigue test of the new coupler was performed with cyclic vertical loads (+78.4 to +235.2 kN) and horizontal loads (-91.2 to +91.2 kN) simultaneously at 1 Hz and the life of 2,000,000 cycles were achieved without failure.

  • We have been derived theoretically the SER's and CLP's of Wireless ATM (WATM) cells employing an OFDM/16QAM modulation scheme in wireless channel modeled as a frequency selective Rayleigh fading channel. The amount of the performance improvement of WATM- OFDM/16QAM systems adopting various coding techniques has been evaluated. In frequency selective Rayleigh fading channel, considering CLP : $10^{-3}$ as a criterion, it is observed that the performance improvement of about 14 dB is obtained in terms of $E_b/N_o$ by employing an OFDM scheme. It is also confirmed that convolutional coding technique gives better performance than the other coding techniques. Especially, when the convolutional codes are adopted to WATM-OFDM/16QAM systems, voice transmission services are sufficiently available with 5 dB of $E_b/N_o$.

  • In this paper we have evaluated the degree of performance improvement in Multi-Carrier CDMA system adopting CCI canceller against MUI under mobile channel coexisting MU which is a major interference degrading the performance of Multi-carrier CDMA system. As a result of analysis, BER performance was improved by adopting CCI canceller and it was found that the amount of performance improvement was largely increased as Eb/No became higher. As an example, in case that data service BER = 10$^{-5}$ must be achieved with $E_{b/}$ $N_{o}$ = 12 ㏈, 4 users could be supported without CCI canceller while 30 users could be supported with CCI canceller, Also, we found that performance improving effect of 5$\times$10$^{-3}$ in a point of view BER could be achieved by adopting CCI canceller with fixed $E_{b/}$ $N_{o}$. o/. o/.

Inhibition of DMBA-Induced Mouse Epidermal Carcinogenesis by Astaxanthin-Containing Egg Yolks (DMBA로 유발한 Mouse 피부암에 대한 Astaxanthin이 함유된 난황의 항암효과)

  • Anticarcinogenic activity of astaxanthin-containing egg yolks (designate AEY) was investigated for 7,12-dimethylbenz[a]anthracene (DMBA)-induced two stage mouse epidermal carcinogenesis. Female ICR mouse (6-7 weeks of age) were house in a humidity-and-temperature-controlled facility and subjected to feed and water ad libitum. AEY (10 mg/0.2 ml acetone) was painted on the back of mice 7 days, 3 days and 5 min before DMBA treatment (50 nmole/0.2 ml acetone). One week later after DMBA treatment, 6 ${\mu}g$ tetradecanoyl 12-phorbol 13-O-acetate (TPA) dissolved in 0.2 ml acetone was applied on the mouse twice weekly over a period of 22 weeks. No sample was given to control mice. Control egg yolk (CEY) and astaxanthin-containing oil (designate AO) from Phaffia rhodozyma were used as positive controls. Mouse treated with AEY exhibited 10 tumors per mouse whereas control mouse exhibited 15 tumors per mouse, the fact that 33% reduction of tumor per mouse by AEY treatment. Tumor incidence was also reduced to 15% by AEY treatment when compared to that of control group. Such effects were also seen in CEY and AO treatment groups, but leaser extent. AO gave reduction of food intake and body weights relative to those of AEY and CEY, indicating toxicity of AO. These results suggest that AEY exhibits anticarcinogenic activity for DMBA-induced mouse epidermal carcinogenesis.
