Isolation and Biochemical Properties of Pasteurella multocida from the pneumonic lungs of swine in Cheju (제주지방 돼지의 폐병변으로부터 Pasteurella multocida 분리 및 생화학적 특성)

  • The present study was conducted to investigate the incidence of Pasteurella multocida infection in cheju swine from March 1994 to December 1994 isolated organisms were identified by the biochemical properties, cellulose serological type and antibiotic susceptibilities. Pasteurella multocida was isolated from the Lungs of 96 pigs with pneumonia(51 %) among 188 slaughtered pigs. The majority of p multocida isolates were identical to those of the standard strains. On the classification of the capsular type of the isolated p multocida it consist of the 88 isolates of type A(91.6%) 2 isolates of type D (2%) and un classified 6 types(6.2%). The majority of the 96 isolates of p multocida highly susceptible to the antibiotics including ampicillin(Am), cephalotin(Ce), erythromycin(Em), gentamycin(Gm), kanamycin(Km), lincomycin(Lm), neomycin(Nm), penicillin(Pc), streptomycin(Sm), solfametoxazol/trimethoprim(Sxt) and tetracycline(Te)

모듈식 증기발생기를 사용한 일체형 원자로의 예비 개념설계

  • 일체형 원자로는 증기발생기 및 가압기를 압력용기 내에 설치한 것으로서 연결배관이 없기 때문에 배관의 파단에 의한 대형 냉각재 상실사고를 근원적으로 배제하고 전체계통을 단순화 시킬 수 있다. 증기발생기는 대부분 관류식으로서 일체식과 모듈식이 사용되고 있다. 본 연구에서는 모듈식 나선형 증기발생기를 사용한 일체형 신형경수로의 예비 개념설계를 수행하였다. 가압기는 원자로 내에 별도의 용기를 설치하는 내장형 자기가압기를 채택하였다. 제어봉 구동장치는 핵분열 반응열을 이용한 원자로 기동을 위하여 반응도를 미세하게 조정하는 것이 가능하고 지진하중과 같은 동하중의 영향을 최소화하기 위하여 원자로용기 외부로의 돌출부분을 최소화하는 설계개념을 채택하였다. 냉각재 펌프는 Canned Motor Pump를 원자로용기 상부에 수직으로 직접 부착하는 개념을 사용하였다.

A Reinterpretation of Sacheon Seonjin-ri Stone Monument's Chinese Characters of Empress Wu(則天文字) - On the Issue of the Introduction and Acceptance of Chinese Characters of Empress Wu to the Korean Peninsula - (사천 선진리비석 측천문자의 명문 재해석 - 측천문자의 한반도 전래 및 수용 문제에 대해 -)

  • Since the discovery of the inscriptions engraved with the "Chinese characters of Empress Wu(則天文字)" at 251 and 256-1, Seonjin-ri, Sacheon-si, Gyeongsangnam-do in 2003, researchers from the Gyeongnam Institute of Cultural Properties, Kwak Seung-Hoon, and Kim Chang-Gyeom have attempted to read and interpret the inscriptions. However, there are still different theories as to the origin of the "Chinese characters of Empress Wu" appearing in the inscriptions and the use of Idu(吏讀)-type notation. This study aims to clarify, this inscription was erected by the king of Silla in the late 7th and mid-8th centuries to commemorate his participation in the Sacheon and Jinju patrols and Buddhist activities, and it is revealed that the main character of the inscription is likely to be King Seongdeok who has visited southern countries. In addition, in the case of "上了言", which has been interpreted as a human name, it should be understood as an Idu-type notation or a 變體漢文 because of the use of the proposition 了, and it should be interpreted as a character that means the end of a specific act. Although it is not a religious text like 『Mugujeonggwangdaedaranigyeong (The Great Dharani Sutra)』, the reason why the "Chinese characters of Empress Wu(則天文字)" was used is that in the process of accepting the Huayan school, the royal family of Silla came into contact with the Buddhist scriptures of the period of the Empress Wu, which is closely related to the rise of the Huayan school in the Middle Ages, and it appears to have learned and used in this process.

  • It is important to calculate runoff, percolation and evapotranspiration in process hydrological cycle. Especially, a evaportranspiration in watershed has a very important effect on hydrological cycle. In the study, the watershed evapotranspiration was calculated by the water balance and a daily evaportanspiration coefficient(CE) was calculated by the modified Beken's formula.

Identification and Antimicrobial Susceptibility of Microorganisms Isolated from Bovine Mastitic Milk (젖소의 유방염 원인균 분리 및 약제 감수성 검사)

  • Microorganisms were isolated and identified from bovine 296 quarters which showed positive reaction by California Mastitis Test (CMT) in 40 farms of Jeju from September 1999 to June 2000. The organisms associated with the mastitis of bovine were 11 different bacterial species in this study. Which of them, Staphylococcus aureus was the most predominant species as 152 (51.4%) isolates. Other identified species included 49 (16.5%) coliform, 47 (15.8%) Streptococcus dysgalactiae, 15 (5.1%) Bacillus spp., 8 (2.7%) Staphylococcus epidermidis, 6 (2.1%) Streptococcus agalactiae, 5 (1.7%) Enterococcus faecalis, 5 (1.7%) Corynebacterium spp., 3 (1.0%) Streptococcus uberis, 1 (0.3%) Pseudomonas aeruginosa and 1 (0.3%) Pasteurella haemolytica. Almost of all the islolated beacterial species showed high sensitivity against kanamycin (98.6%), cephalothin (98.0%), streptomycin (94.9%), gentamicin (94.6%), ampicillin (92.2%) and polymyxin B (90.2%). On the contrary, they showed resistance against penicillin (47.0%), tetracycline (37.2%), cefazolin (26.0%), bacitracin (22.6%) and erythromycin (19.9%). Eighty-one isolates were not resistant to any antibiotics and 215 drug resistant isolates showed 89 different drug resistance patterns from single to nine multiple antibiotics resistance patterns.

  • The value of the cell enzyme-linked immunosorbent assay as a possible replacement for the indirect immunofluorescence antibody test for the estimation of antibodies against BD virus was assessed in four animal models. The serum antibody response was measured by both assay systems;the variability of both tests was less than one diluent step, and correlation of the two tests was assessed using regression analysis. The study showed that the all four animal models gave satisfactory correlation of CELISA and IFA. There, CELISA is acceptable for use in mouse, rabbit, chicken and rat models.

  • Enzyme-linked Immuno sorbent Assay (ELISA) for the serological diagnosis of Brucella abortus was developed and compared with plate agglutination test. Cell wall antigen was extracted from Brucella abortus 1119-3 by sonication and with a sodium deoxychlate solution. Optimum protein concentration of coating antigen were $0.4{\mu}g/100{\mu}{\ell}$ protein on each microtiter plate well. Horse radish peroxidase (HRP) labeled protein-G was used as a tracer of reacted antibodies. ELISA confirmed the agreeable results of 40 cases out of 43 cases by plate aggulutination test. ELISA diagnosed positive cases(10 out of 12) and negative cases (1 out of 12) with dubious sera by plate agglutination test. From this results ELISA could be used for the early diagnostic tools of Brucellosis in cattle.

  • In this experiment, we studied the sero-positive rate of akabane virus in cattle from Jeju-do and analyzed the seroepidemiological features. In an analysis of 1,051 samples, the positive rate for neutralizing antibody in sera collected in nine regions on Jeju-do was 56.7%. The rate varied with the region. The positive rate was 69.6% in Aewol, 63.1% in Jeju city, 54.4% in Anduck, 51.0% in Hallim, 69.8% in Jocheun, 47.6% in Pyosun, 40% in Daejeong, 30.0% in Harkyung, 71.6% in Namwon, 24.5% in Sungsan, 133.,3% in Seokypo and 44.5% in Gujwa, respectively The rate also depended on the age of the cattle. The positive rate was 67.2% in calves 0- to 12-month old, 48.3% in cattle 13- to 24-month old, 65.4% in cattle 25- to 36-month old, and 65.4% in cattle more than ,B7 months old. To isolate the virus from calves with malformations including arthrogryposis and hydranencephaly, cerebral homogenates were inoculated into Vero cells, which were determined for cytopathic effect (CPE). Vero cells with CPE were examined for Akabane virus using an electron microscope (EM) and indirect immunofluorescent antibody test (EM). Typical virus particles with a width of 90-130nm and specific immunofluorescence in the cytoplasm of infected cells were sought for identification.

  • The present study was conducted to investigate the incidence of equine herpesvirus 1(EHV-1) infection in Korean horses by cell EUSA and serum neutralization test A significant level of antibodies to EHV-1 was detected from 539(82.8%) by cell EUSA and from 524 horses(80.5%) by SNT indicating that EHV-1 infection in Korean horses are widely spreaded throughout the country.

  • The study was carried out to investigate the pathogenicity of EHV isolate to hamsters and mice and immunogenicity of experimentally produced. vaccine were evaluated in the horses. Hamsters infected. intranasally with $LC_1$ isolate showed symptoms of nasal discharge, conjunctivitis and body weight loss during the observation period of 12 days after infection, while only slight depression and body weight loss were noticed with mice infected with $LC_1$ indicating that hamsters are more susceptible to the virus. Antibody titer of mice and hamsters were gradually increased to highest level of 1:2560~10240, 1:640~1280, respectively, at 7~12 days post vaccination. Horses immunized against $LC_1$ killed vaccine reached to maximum antibody titer of 1:20480 around 4 weeks after 1st vaccination and declined after 12 weeks post vaccination. No significant antibody increase were detected after 2nd vaccination. Mean body temperature and mean total leukocyte counts remained within normal range and no adverse reaction were noticed after vaccination.
