New Analytical Method for Separation and Identification of Heavy Metals (I) (중금속의 분리 및 검출을 위한 분석화학적 연구 (제 1 보). 새로운 분리방법의 개발)

  • In 10M chloride (4M HCl + 6M LiCl) solution, cobalt, but not nickel, formed complex anion (${CoCl_3}^-$), and this anion was extracted by a liquid anion exchanger with Amberlite LA-2. The ion exchange capacity was 2.175meq of cobalt complex per unit ml of Amberlite LA-2. Upon eluting the resin with 0.4M nitric acid, the cobalt complex was stripped and transfered into eluate quantitatively. By using this separation method in the chloride solution dissolved with 50mg of cobalt (II) and 500mg of nikel(II), recovery of cobalt were 99.6 percent.

A Study on the Separation of Neodymium from the Simulated Solution of $U_3Si/Al$ Spent Nuclear Fuel (모의 사용후분산핵연료($U_3Si/Al$) 용해용액으로부터 네오디뮴 분리에 관한 연구)

  • The separation of Nd from the simulated $U_3Si/Al$ spent fuel solution with sequential two-step anion exchange separation has been studied. To prepare the simulated $U_3Si/Al$ spent nuclear fuel, unirradiated $U_3Si/Al$ whose composition consists of small $U_3Si$ particle dispersed in an Al matrix with Al cladding was dissolved with a mixture of 4 M HCl and 10 M $HNO_3$ and 8 or 15 fission product elements were added to the dissolved solution. The trace amount of silica in the solutions was removed by evaporating to dryness with HF and the U was adsorbed on the first anion exchange resin. Neodymium can be purely isolated from the fission product elements with a methanol-nitric acid eluent using the second anion exchange resin. A large excess of Al didn't influence on the elution velocity of Nd, but reduced the eluted contents of Nd, Al, Eu, Gd, Sm and Sr, A large amount of Al was removed first from the column with 3 mL of loading solution (0.8 M $HNO_3$/99.8% MeOH) before Nd elution by the eluent [0.04 M $HNO_3$-99.8% MeOH(1:9)]. The recovery of Nd was more than 94%, regardless of Al contents. Taking the 9 to 13 mL fraction of eluate was effective to purely isolate Nd.

  • In this study, heterogeneous ion exchange membranes were prepared by casting method with various mixing ratios of PPO ion-selective solution and ion exchange resin. Then heterogeneous bipolar membranes were prepared by using this. The water content of heterogeneous cation and anion exchange membranes were 60~80% respectively, the ion exchange capacity was 2.81~3.26 meq/g, 2.31~2.74 meq/g and electrical resistances were $1.65{\sim}1.45{\Omega}{\cdot}cm^2$ and $1.55{\sim}1.05{\Omega}{\cdot}cm^2$. The tensile strength of heterogeneous bipolar membrane was lower than that of PPO resin before functionalization ($700Kg_f/cm^2$). The tensile strength of heterogeneous bipolar membrane with catalyst layer was lower than that of non-catalytic heterogeneous bipolar membrane. The water splitting voltage of the heterogeneous bipolar membrane with catalyst layer was low and stable at a minimum of 1.7~1.8 V, maximum 3.9~4.0 V, and the water splitting voltage of the non-catalytic heterogeneous bipolar membrane was constant at 3.8~4.0 V.

Study on the Elution Behaviors of Uranium and Vanadium in the Various Acids by Anion Exchange Chromatography (음이온 교환크로마토그래피법에 의한 여러가지 산에서 우라늄과 바나듐의 용리현상에 관한연구)

  • The species and equilibria of uranium and vanadium have been investigated in the various concentration of perchloric, hydrochloric and sulfuric acid by anion exchange chromatography. In the concentration range of $0.01\;{\sim}\;0.5M$ hydrochloric and $0.01\;{\sim}\;0.5M$ perchloric acid, uranium seems to be $UO_2^{2+}$species and in higher concentration than 0.5M hydrochloric acid $UO_2^{2+}$seems to form the chloride complex ion as $UO_2Cl^+$, $UO_2Cl_2$, $UO_2Cl_3^-$ and $UO_2Cl_4^{2-}$ according to the increase of the hydrochloric acid concentration. In the dilute(0.01N) sulfuric acid the adsorbability of uranium on anion exchange resin is strong and then decreases with increasing the sulfuric acid concentration. From this result we conclude that $UO_26{2+}$ formed the complex ion as $UO_2(SO_4)_2^{2-}$. In the perchloric acid of $0.01\;{\sim}\;0.5N$ concentration the existing equilibrium of vanadium and its constant calculated at $20^{\circ}C$ is $1.9{\times}108$ for $H_2V_{10}O_{28}^{4-}$ + $14H^+$ = $10VO_2^+ + 8H_2O$. The elution behaviors of vanadium in the hydrochloric and sulfuric acid are smiliar to those in perchloric acid.

  • 본 연구는 중금속에 의한 생체내 독성기전을 연구하고자 흰쥐간의 상층액에서 카드뮴과 잘 결합하는 HMWP들을 분리, 정재하고 나아가 생화학적 특성을 밝히고 이단백질이 카드뮴에 의해 생성되는 것인지 아니면 간의 기존 단백질인지를 밝히고자 함을 연구의 목적으로 하였다. CdCl %2 (3mg/kg body wt.)을 3일간 ip injection시킨 후 흰쥐의 간을 적출하고 균질화하여 원심분리한 crude extract를 직접 Sephacryl S-100에 흡착시켜 10mM phosphate buffer(pH 7.0)으로 용출시켰다. 용출된 fraction tube를 UV spectrometer로 흡광도를 측정하고 atomic absorption spectrophotometer로 카드뮴량을 측정하여 카드뮴량이 높은 분획을 모아서 ion exchange column chromatography(DEAE-Sepharose)에 흡착시켜 염농도 구배로 chromatography를 실시하고, 음이온 교환수지에서 흡착되지 않고 용출된 분획을 ultrafiltration으로 농축시킨 후 S-Sepharose에 흡착시켜 염농도 구배로 chromatography를 실시한 결과 두 종류의 카드뮴 결합 단백질(Cd-BP)를 분리, 정제하였다.

  • LOMI 제염제의 전기화학적 제조공정 중 전하운반체의 역할 및 V$^{3+}$ 이온의 침전현상을 방지하기 위하여 첨가되는 과량의 formic acid 는 제염제 제조시에는 중요한 역할을 하지만 제염 폐기물의 양을 증가시키는 요인이 되기도 한다. EPRI 공정에서는 formate : V 의 비율(FVR)이 10:1 인 조건에서 V$^{2+}$-formate 를 제조한 후 음이온 교환 수지를 이용하여 과량의 formate 를 제거하는 공정을 사용한다. 이러한 방법으로 줄일 수 있는 FVR의 한계가 약 2.5 : 1 이다. 본 연구에서는 제염제 중 formate 의 양을 더욱 줄일 수 있는 방법으로써 V$^{2+}$-formate 제조시 용액 중에 picolinic acid를 미리 첨가하는 영향을 조사하였다. V$^{2+}$ 이온의 농도가 0.1 M 일 때 FVR 를 1.5:1 까지도 줄일 수 있음을 확인하였다.

Identification of Proteins in Egg White Using Ion Exchange Cartridge and RP-HPLC (이온교환 카트리지와 RP-HPLC를 이용한 난백 단백질의 확인)

  • Approximately forty proteins in egg white have been widely studied for their functional properties. To develop a procedure of separation for pure and non-altered proteins from egg white, purification study was conducted to isolate lysozyme, ovotransferrin, and ovalbumin. Ion exchange cartridge can selectively separate proteins from egg white, and reversed-phase HPLC (RP-HPLC) could identify separated proteins. Proteins in egg white were purified by HI trap ion exchange cartridge SP and Q with buffers pH 8.0 and 5.2. C18 column (Phenomenex, USA) was used for RP-HPLC analysis and isocratic mobile phase was used with acetonitrile (ACN)/distilled water (DW)/trifluoroacetic acid (TFA) in the ratio of 50/50/0.1. Comparing the retention times of standards in RP-HPLC experiments showed that ovotransferrin, ovalbumin, and lysozyme in egg white were eluted successively in the RP-HPLC column after the pretreatment in SP and Q ion exchange cartridges.

Determination of trace actinide (Am, Pu, Th, U) using alpha spectrometry and neutron activation analysis (알파분광법과 중성자방사화분석법에 의한 극미량의 악티늄계원소 (Am, Pu, Th, U)분석연구)

  • Determination of actinides in the environmental sample requires separation of each element. This procedure is tedious and time consuming. And also, the detection limits of some nuclides using alpha spectrometry are rather higher. To overcome the lower detection limit and complicated separation procedure, a simple analytical technique for the determination of actinide isotopes in the environmental samples was developed and applied to IAEA and NIST reference sediment samples. For the separation of actinides from matrix, anion exchange resin and TRU-spec extraction chromatography resin were used and chemical yields were obtained using natural uranium, thorium, $^{242}Pu$ and $^{243}Am$ tracers. For overcoming the higher detection limits of U and Th in alpha spectrometry, neutron activation analysis was applied. Using combined method, the detection limit was increased about 10 times. The activity values of each isotope were consistent with the reference values reported by IAEA and NIST.

Screening of Adsorbent to Reduce Salt Concentration in the Plastic Film House Soil under Continuous Vegetable Cultivation (시설채소재배지의 토양특성과 흡착제 종류별 염류경감 효과)

  • Salt accumulation in the plastic film house soils under continuous cultivation condition causes problems such as salt damages to plants, nitrate accumulation in vegetables, groundwater contamination, etc. due to excess application of fertilizers. Objective of this research was to find an optimum adsorbent to reduce salt concentration in the soil solution of plastic film house soils, where crop injuries have been observed due to the salt accumulation. The soils were significantly high in available P $(1,431{\sim}6,516mg\;kg^{-1}),\;NO_3-N\;(117.60{\sim}395.73mg\;kg^{-1})$, exchangeable Ca $(4.06{\sim}11.07\;cmol_c\;kg^{-1})$ and Mg $(2.59{\sim}18.76\;cmol_c\;kg^{-1})$, as compared to those of the average upland soils in Korea. Soils were treated with each of adsorbent such as ion-exchange resin, zeolite, rice bran, etc. at 2% level and prepared into saturated-paste samples. After equilibrium, soil solution was vacuum-extracted from the soil and measured for changes of the pH, EC, and concentrations of $Ca^{2+},\;Mg^{2+},\;K^+,\;Na^+,\;{NH_4}^+,\;{PO_4}^{3-}\;and\;{NO_3}^-$. Rice bran effectively removed ${PO_4}^{3-}\;and\;{NO_3}^-$ in the soil solution up to 100%. Efficiency was decreased in the orders of rice bran > ion-exchange resin > zeolite. Removal efficiencies of zeolite and ion-exchange resin for $Ca^{2+}$ were ranged from 1 to 65% and from 7 to 61%, respectively. Ion-exchange resin was also effective for removing $Mg^{2+},\;K^+,\;Na^+,\;and\;{NH_4}^+$. Overall results demonstrated that rice bran and ion-exchange resin could be applicable for salt accumulated soil to remove the respective anion and cation.

Molybdenum Isotope Analysis of Standard Reference Materials (표준물질을 이용한 몰리브덴 동위원소 분석)

  • Mo isotope, one of highly redox-sensitive isotopes, has been shown to be useful tracers of geochemical processes. Many studies for Mo isotope have documented with the help of recently developed analysis tools, but it has not yet been documented in the Korea. In this study, we introduce two-stage column separation method of Mo using column tube (BioRad PolyPrep(R) column, 10 ml) and anion exchange resin (BioRad Resin AG(R) 1-X8, 200-400 mesh). Mo isotope ratios in the solid SRMs (BHVO-2, SDO-1, PACS-2) and liquid SRM (IAPSO) were measured on MC-ICP-MS (Multi-collector Inductively Coupled Plasma Mass Spectrometer) and then compared with reference Mo isotope ratios. Mo isotope ratios in our study overlap with reference Mo isotope ratios within analytical error.
