Effects of ${\gamma}-Irradiation$ on the Quality of Seasoned Cuttle during Storage (감마선 조사가 조미오징어의 저장 중 품질특성에 미치는 영향)

  • This study was conducted to investigate the changes in the qualify of gamma-irradiated seasoned cuttle during storage. Seasoned cuttle packed in PVC film (0.06 win) was stored at $15{\pm}1^{\circ}C$ for 6 months after treatment with doses of 0 to 7 kGy. Microbial populations of seasoned cuttle were $1.6{\times}10^{5}CFU/g$ in total aerobic bacteria, $10^{4}{\sim}10^{5}CFU/g$ in yeasts &molds, and negative in coliforms, which were effectively reduced by 3 kCy or higher up to the undetectable level(<20 CFU/g). The pH and moisture content of the samples were not changed with irradiation, but moisture was some decreased during storage. The instrumental color (especially Hunter b value), pigments (lipophilic &hydrophilic) and TBA value of the samples increased with storage time as well as irradiation dose more than 3 kGy. The influence of storage condition, however, were more significant. Irradiation did not induce any changes in volatile basic nitrogen (VBN) and trimethylamine (TMA) contents, thereby maintaining their contents lower than those of the non-irradiated samples during storage by reducing the microbial load.

Optimization of Conditions for the Preparation of W/O Emulsion Containing Eugenolchitosan (Eugenolchitosan 함유 유중수적형 유화 형성 조건 최적화)

  • Stabilities of W/O emulsions containing eugenolchitosan (EuCs) prepared from chitosan and eugenol were compared to determine the optimal conditions for the ratio of water (core phase) to corn oil (continuous phase), the concentration of EuCs, storage temperature, and the extent of homo-mixing. The optimal ratio of water to corn oil was 2:3 (w/w). The effects of EuC concentrations, and singular vs. binary system of emulsifiers on the storage stability of the emulsion were investigated with EuCs and polyoxyethylene sorbitan monolaurate. The emulsion was stable, showing more than 95% emulsion stability index (ESI) value, when the concentration of EuCs was more than 0.18% (w/v). ESI value of binary emulsifier system was almost equal to that of singular emulsifier system at the concentration of 0.18% (w/v). At this singular emulsifier system, the W/O emulsion formed by EuCs had ESI value of 100%. The optimal concentration of EuCs was determined as 0.18% (w/v). The highest stability of the emulsion was obtained from the homo-mixing at 11,000 rpm for 10 sec and the storage temperature ranging $25{\sim}65^{\circ}C$. EuCs produced from this study was mutagenecity-negative on Ames test and contained no heavy metal ions.

Quality and Thermoluminescence Properties of ${\gamma}$-Irradiated Boiled-Dried Anchovies during Storage (감마선 조사와 저장 기간에 따른 건멸치류의 품질 및 열발광 특성)

  • The effect of gamma irradiation was investigated on the microbiological and physicochemical qualities of boiled-dried anchovies (large- and smallsized) packed in PVC film (0.06 ㎜) during storage at 15$\pm$1$\^{C}$ for 6 months. On the other hand, thermoluminescence characteristics of minerals extracted from the sample were analyzed to evaluate its possibility in detecting the irradiated anchovies. The samples were contaminated by microbial levels of 104∼106 CFU/g in total aerobic bacterial counts and negative in coliforms, which were different depending on the sample sizes. Gamma irradiation at 3 kGy was effective for improving the hygienic quality of both samples for 6 months, keeping the microbial population less than 20 CFU/g. Besides it was not detrimental to the quality attributes, such as instrumental color parameters L, a, b, ΔE), browning, TBA value, volatile basic nitrogen, trimethylamine-N and sensory properties(appearance, color, odor, tape, overall acceptability). However, storage conditions (time, temperature, packing) showed a critical factor nfluencing the quality changes of dried anchovies. Thermoluminscence analysis was prove to distinguish irradiated from non-irradiated anchovies by comparing both temperature at which glow curves appear and the curve intensity, that was possible even after 6 months at - 20$\^{C}$.

Changes of Indicator Microorganisms and Pathogenic Bacteria in Spinach during Cook-Chill Process (시금치의 cook-chill 가공 중 오염지표균 및 병원성세균의 변화)

  • Spinach minimally processed using cook-chill and sous vide techniques was vacuum-packed in low gas permeable plastic film, pasteurized at $70^{\circ}C$ for 2 min, cooled rapidly at $3^{\circ}C$, and stored at 3 and $10^{\circ}C$. Contents of mesophilic bacteria, psychrophilic bacteria, anaerobic bacteria, spore-forming bacteria, total coliforms, yeast and molds, fecal Streptococcus, and Enterobacteriacea were measared to identify the degree of food contamination. Number of mesophilic bacteria, detected at $2.2{\times}10^8\;cfu/g$ in raw spinish, decreased to about $6.0{\times}10^3\;cfu/g$ after cook-chill process. During the storage at 3 or $10^{\circ}C$, levels of mesophilic, psychrophilic and anaerobic bacteria increased, whereas total coliforms, yeast and molds, fecal Streptococcus, and Enterobacteriacea were not detected. Twelve strains of Aeromonas hydphila, Escherichia coli O157:H7, Plesiomonas shigelloides, Pseudomonas aeruginosa, Salmonella spp., Shigella spp., Yersinia enterocolitica, Bacillus cereus, Campylococcus spp., Clostridium perfringens, Listeria monocytogenes, and Staphylococcus aureus were examined for detecting the presence of pathogenic bacteria in spinach. B. cereus and C. perfringens were isolated from raw, washed, and cook-chilled spinach, whereas A. hydrophila was isolated only from washed spinach. S. aureus was isolated from raw and washed spinach, but not from cook-chilled spinach. Other pathogenic organisms were not detected in raw, washed, and cook-chilled spinach.

The Cadmium Biosorption Mechanism in Gram Negative Bacteria, Serratia marcescens (Gram 음성 세균인 Serratia marcescens에 의한 카드뮴 흡착 기작)

  • Serratia marcescens, an enterobacterium of gram-negative bacteria, is characterized by resistance of the admium. Cadmium sensitive PM strain did not grow in the medium at cadmium concentration of 50 ppm. PA strain was induced to accommodate to cadmium by cultivating the mother strain (PC strain) in the medium with 50 ppm cadmium. As compared with PC and PM strains, PA strain revealed the excellent growth in cadmium media and accumulated four to five times higher cadmium concentration in cell than other strains. PA strain accumulated 23% of cadmium in cells when cultured in medium treated with 100 ppm cadmium and this cadmium was more accumulated in cytosol fractions than membrane fractions. Analysis by TEM indicated that cadmium was concentrated as a form of granule in cytosol. In protein patterns of cell after the treatment of cadmium, two inducible proteins (28 KDa and 64 KDa) and one reducible protein (45 KDa) were detected by SDS-PAGE. By Atomic Absorption Spectrophotometer, the amounts of cadmium attached to inducible proteins of 28 KDa and 64 KDa were 318.28 ㎍ and 325.37 ㎍ per gram of protein, respectively. It is assumed that these inducible proteins play an important role in the mechanism of cadmium accumulation in cells. A plasmid of 23Kbp was found in S. marcescens. The ability of resistance to cadmium in plasmid was confirmed by curing experiments.

  • As the automobile industry and technologies are developed, driver's tend to more concern about service matters than mechanical matters. For this reason, interests about recognition of human knowledge and emotion to make safe and convenient driving environment for driver are increasing more and more. recognition of human knowledge and emotion are emotion engineering technology which has been studied since the late 1980s to provide people with human-friendly services. Emotion engineering technology analyzes people's emotion through their faces, voices and gestures, so if we use this technology for automobile, we can supply drivels with various kinds of service for each driver's situation and help them drive safely. Furthermore, we can prevent accidents which are caused by careless driving or dozing off while driving by recognizing driver's gestures. the purpose of this paper is to develop a system which can recognize states of driver's emotion and attention for safe driving. First of all, we detect a signals of driver's emotion by using bio-motion signals, sleepiness and attention, and then we build several types of databases. by analyzing this databases, we find some special features about drivers' emotion, sleepiness and attention, and fuse the results through Multi-Modal method so that it is possible to develop the system.

Antioxidant and antimicrobial activities of ginger with aging and fermentation (발효숙성생강의 항산화 및 항균 활성)

  • Zingiber officinale Roscoe, commonly known as ginger, has long been used as a powerful health-promoting antioxidant that supports cellular health of the human body. The objective of this study was to compare the antioxidant and antimicrobial activities of the samples with aging and fermentation. Antioxidant activities of the samples were compared using total phenol, flavonoid contents, ABTS cation radical scavenging activity and DPPH radical scavenging activity. Antimicrobial activities were also examined using the paper disc method and minimum inhibitory concentration (MIC). Acidity of the fermented ginger (FG) with lactic acid bacteria showed a significantly higher value than that of the ginger (GG). The content of 6-gingerol, a bioactive component in ginger, decreased in all fermented gingers but 6-shogaol which is also one of the main valuable ingredients showed the increased content at ginger fermented with Streptococcus thermophilus and Lactobacillus acidphilus. Flavonoid contents of the FG and GG did not show significant differences. However, ABTS cation radical scavenging activity and DPPH radical scavenging activity were 10-30% increased in the samples with fermentation (p<0.05), respectively. The samples of the disc showed an inhibitory effect on growth of gram positive Staphylococcus aureus and Listeria monocytogenes. Zinger with fermentation showed higher antioxidant and antimicrobial activities. Thus, we conclude that aging and fermentation can be a helpful process to increase the functional effects of ginger.

Quality Characteristics of Mungbean Starch Gel Added with Salicornia herbacea L. Powder (함초 분말을 첨가한 청포묵의 품질 특성)

  • This study evaluated the quality characteristics of mungbean starch gel prepared with different amounts of Salicornia herbacea L. powder. In order to determine the optimal addition level of Salicornia herbacea L. powder, samples of mungbean starch with 0, 2, 4, 6 and 8% of Salicornia herbacea L. powder substitute were prepared, after which physicochemical, textural and sensory quality characteristics were measured. Upon increasing Salicornia herbacea L. powder levels in the formulation, moisture contents of samples decreased, except the 6% addition sample, and salinity levels increased. Total phenolic contents and DPPH radical scavenging activities also increased significantly upon addtion of Salicornia herbacea L. powder. According to the texture analysis, hardness and gumminess decreased with increasing amounts of added Salicornia herbacea L. powder. From the sensory tests, 4% Salicornia herbacea L. powder sample received the highest overall acceptability score with proper levels of flavor, saltiness and texture. As a result, to increase usage of Salicornia herbacea L., the optimal formulation consisted of 4% Salicornia herbacea L. powder substitute for mungbean starch.

Identification of Mycobacterium tuberculosis in Pleural Effusion by Polymerase Chain Reaction(PCR) (흉막 삼출액에서 중합효소 연쇄반응(PCR)을 이용한 M. tuberculosis의 검출)

  • Background: Since polymerase chain reaction(PCR) was devised by Saiki in 1985, it has been used extensively in various fields of molecular biology. Clinically, PCR is especially useful in situation when microbiological or serological diagnosis is limited by scanty amount of causative agents. Thus, PCR can provide rapid and sensitive way of detecting M. tuberculosis in tuberculosis pleurisy which is diagnosed in only about 60 % of cases by conventional method. Method: To evaluate the diagnostic usefulness of PCR in tuberculosis pleurisy, The results of PCR was compared with those of conventional method, including pleural biopsy. The pleural effusion fluid was collected from 7 proven patients, 7 clinically suspected patients and control group(7 patients with malignant effusion). We extracted DNA from pleural fluid by modified method of Eisennach method(1991). The amplification target for PCR was 123 base pair DNA, a part of IS6110. Result: 1) Sensitivity of PCR: We detected upto 50fg DNA. 2) In patients with pleural effusion of proven tuberculosis, the positive rate of PCR was 85.7%(6/7). In patients with pleural effusion of clinically suspected tuberculosis, the positive rate was 71.5%(5/7). In control group, positive rate was 0%(0/7). Conclusion: We concluded that PCR method could be a very rapid, sensitive and specific one for diagnosis of M tuberculosis in pleural effusion. Further studies should be followed for the development of easier method.

Formulation and Quality Characteristics of Noni Beverages Mixed with Red Ginseng, Rubus Coreanus and Pomegranate Extracts (홍삼, 복분자, 석류를 첨가한 노니 혼합음료의 개발과 품질특성)

  • The purpose of this study was to investigate the physicochemical and sensory characteristics of Noni beverages added with 10% of water(Noni 90), 1% of red ginseng(NR), 4% of rubus coreanus extract(NR), 5% of pomegranate extract(NP), and 4% of rubus coreanus extract and 5% of pomegranate extract(NPR). Calories, protein and total fiber contents were increased in NR, NP and NPR. Total sugar was significantly increased in NG and NPR. Fat, cholesterol and saturated fatty acid were not detected. Ca and Na contents were increased in NG while Fe content was increased in NG, NR, and NPR pH was significantly increased in NG and NP. Sugar degree was significantly increased in NP and NPR. L and b values were decreased in NG, NP, and NR and a value was increased in NG and NR General bacteria count was 0/mL, and Coliform, E. Coli and 0157:H7 were gram(-) in all beverages. In sensory evaluation, color was increased in NPR while taste and overall quality were increased in NG, NR and NP. Sour taste, sweet taste, and flavor were not different between Noni and the mixed beverages. Texture was significantly increased in NP and NPR. Therefore, mixed Noni beverages added with red ginseng, rubus coreanus extract, and pomegranate extract were better than Noni beverage.
