Emulsification of O/W Emulsion Using Non-ionic Mixed Surfactant: Optimization Using CCD-RSM (비이온성 혼합계면활성제를 이용한 O/W 유화액의 제조 : CCD-RSM을 이용한 최적화)

  • A mixing ratio of the oil in water (O/W) emulsion of palm oil and the non-ionic surfactant (Tween-Span type) possessing different hydrophile-lipophilie balance (HLB) values was evaluated in this work. An optimum condition was determined through analysis of main and interaction effects of each quantitative factor using central composite design model-response surface methodology (CCD-RSM). Quantitative factors used by CCD-RSM were an emulsification time, emulsification speed, HLB value and amount of surfactant. On the other hand, the reaction parameters were the viscosity and mean droplet size of O/W emersion. Optimized conditions obtained from CCD-RSM were the emulsification time of 12.7 min, emulsification speed of 5,551 rpm, HLB value of 8.0 and amount of surfactant of 5.7 wt.%. Ideal experimental results under the optimized experimental condition were the viscosity of 1,551 cP and mean droplet size of 432 nm which satisfy the targeted values. The average error value from our actual experiment for verifying the conclusions was below to 2.5%. Therefore, a high favorable level could be obtained when the CCD-RSM was applied to the optimized palm oil to water emulsification.

Adhesion control of Campylobacter jejuni in chicken skin using emulsifiers (유화제를 이용한 계육 표면에서 Campylobacter jejuni의 부착 제어)

  • To prevent contamination by Campylobacter jejuni during chicken carcass processing, the effect of emulsifiers on C. jejuni inoculated on chicken skin was investigated using confocal laser scanning microscopy. Among the 8 emulsifiers (SWA-10D, L-7D, M-7D, S-1670, L-1695, P-1670, polysorbate 20, polysorbate 80) tested for antimicrobial activity by the paper disk method, 4 emulsifiers (L-7D, L-1695, polysorbate 20, polysorbate 80) were screened further. Emulsifier L-1695 showed the largest clear zone at a concentration of 200 mg/mL. The 4 emulsifiers subjected to primary screening were screened for heat and pH stability. In the contact surface test, emulsifier L-1695 showed the lowest log CFU/㎠ value on both stainless steel and ceramic surfaces. When emulsifier L-1695 was applied via general and electrostatic spray methods, the number of C. jejuni entrapped inside chicken skin follicles was significantly reduced in both methods. In conclusion, the emulsifier L-1695 could be employed as a microbial detachment agent in the chicken carcass processing industry.

Evaluation of Coconut Oil-based Emulsion Stability Using Tween-Span Type Nonionic Mixed Surfactant (Tween-Span계 비이온성 혼합계면활성제를 이용한 Coconut Oil 원료 유화액의 유화안정성 평가)

  • In this study, the influence factors on the stability of the O/W (oil in water) emulsions prepared with coconut oil and the nonionic mixed surfactant (Tween 80-Span 80) were evaluated. The concentration and HLB value of the nonionic mixed surfactant, and the degree of agitation were used as manufacture factors. The stability of prepared O/W emulsions were measured with the mean droplet size, zeta-potential, emulsion stability index (ESI), and thermal instability index (TII). The mean droplet size of the prepared O/W emulsions was from 100 to 200 nm. As the concentration of mixed surfactant and the homogenization speed increased, the droplet sizes decreased, while the zeta-potential values increased. The effect of HLB values increased in the order of 6.0, 10.0 and 8.0, and at the HLB value of 8 the smallest mean droplet size as 120 nm was obtained whereas the largest value of the zeta-potential between 10 and 60 mV. From the results of ESI and TII, the stability of prepared O/W emulsions increased in order of 6.0, 10.0 and 8.0 of HLB values, and ESI and TII values were above 80% and below 20% respectively at HLB value of 8.0.

Relationship between Emulsion Stability Index and HLB Value of Emulsifier in the Analysis of W/O Emulsion Stability (W/O형 유화계의 유화안정성 분석에 있어서의 유화안정지수와 HLB값과의 관계 규명)

  • The stability of W/O emulsions (milk fat : water=4 : 1, w/w) containing various emulsifiers was compared to determine the effect of different chemical types of emulsifiers in relation to the change of HLB value caused by emulsifier type and the influence of single vs. binary emulsifier systems. These variables were compared at emulsifier HLB values of 0.5~16.7 and at emulsifier concentrations of 1.0~3.0%(w/w). Eleven emulsifiers used as 11 different single mixtures and 16 different binary mixtures were evaluated in W/O type emulsion systems containing 20.0%(w/w) of water in milk fat. This W/O emulsion was stable (more than 90.0 of ESI value) in the range of low value of emulsifier HLB (less than 4.7 of HLB value). All the ESI values of binary emulsifier systems were higher than those of single emulsifier systems. But, the influence pattern of emulsifier HLB on this emulsion stability in single emulsifier systems was very similar to the trend in binary emulsifier systems.

  • 동식물성 기름과 메탄올의 전이에스테르화 반응에 의해 생산되는 바이오디젤은 환경친화성과 지속가능성이 인정됨에 따라 그 생산량이 급격히 증가하고 있어 대두유, 유채유, 팜유 등의 원료유 부족과 가격 상승, 수급 불안정 등의 문제가 대두되고 있다. 이를 해결하기 위한 방안으로 유리지방산 함량이 높은 저가유지 자원(폐식용유, 폐돈지, 폐우지, soapstock, trapped grease)과 새로운 오일 작물을 이용한 생산 기술 연구가 활발히 진행되고 있다. 본 연구에서는 비활용 해외 열대작물 씨앗에서 착유한 식물성 오일을 정제하여 바이오디젤 원료유를 생산하는 과정에서 발생하는 폐기물(폐유, 폐수)의 경제적 처리 방안으로 유화유 제조 원료(벙커C유, 물)와 유화유 제조 첨가제(무기계, 유기계)로 활용 가능성을 검토하였다. 열대작물 오일의 물성 분석 결과 고형물, 수분, 인지질(phospholipid), 유리지방산(free fatty acid) 함량이 기존 원료유보다 매우 높게 나타났다. 인지질은 바이오디젤 제조 반응후 에스테르와 글리세린의 층분리를 방해하고 유리지방산은 염기촉매와 결합하여 지방산염을 생성해 생산 수율을 감소시킨다. 고형물과 수분 역시 촉매반응에 악영향을 가지나 여과와 감압증발에 의해 쉽게 제거가 가능하다. 유리지방산은 산촉매 에스테르화 반응에 의해 제거가 가능하다. 인지질은 탈검(degumming) 과정을 통해 제거하며 탈검은 수용성 탈검, 산 탈검, 세정 공정으로 구성된다. 착유한 원료유의 고형물을 제거 후 물과 수세하여 수용성 인지질을 수화하여 층 분리해 제거하고 상층의 오일은 추가적인 산 탈검을 수행한다. 그 뒤 세정을 통해 사용된 탈검제인 산과 추가적으로 수화된 인지질을 제거하게 된다. 이러한 3단계의 탈검 과정에서 하층으로 오일과 물이 폐기물로서 배출되며 본 연구에서는 배출 폐기물을 다시 층분리하여 오일층과 물 층으로 구분하여 유화유 제조에 사용되는 벙커C유, 물, 그리고 기존 유기계 및 무기계 유화제의 대체 가능성을 조사하였다. 유화 연료유는 기름과 물을 균일한 분산상으로 혼합한 연료유로 연소시 오일계 성분의 미연분을 감소시켜 연료 효율 제고와 배출가스 성상을 개선하기 위해 개발되어 왔다. 본 발표에서는 다양한 종류의 상용 첨가제 및 바이오디젤 원료유 생산 폐기물을 활용해 유화 연료유를 제조하였으며 각 유화유의 장시간의 상(phase) 안정성을 비교하였다. 바이오 폐기물 중에는 천연 계면활성제(surfactant)인 인지질이 다량 함유되어 있어 기존의 무기계 및 유기계 유화제보다 상 안정성이 우수하게 나타났으며 바이오디젤 원료유 생산 공정의 폐기물인 폐유과 폐수의 활용이 가능한 것으로 나타났다.

Improvement in functional properties of conger eel skin gelatin by succinylation (Succinylation에 의한 붕장어껍질 젤라틴의 기능성 개선)

  • To effectively utilize fish skin gelatin as a material for quality improvement in surimi gel from fish with a red muscle, conger eel skin gelatin was modified with succinic anhydride, and funtional properties such as emulsifying activity and emulsifying stability were determined. The degree of chemical modification incresed up to 0.3 g of succinic anhydride/g of gelatin, above this adding ratio a nearly constant value was reached. The maximum amount of modification was about 90%. The emulsifying activity and emulsifying stability of gelatin gradually increased up to 89.8% of succinylation extent, little changed above of succinylation extent. The other functional properties as solubility, water holding capacity, foam expansion and foam stability were improved following succinylation with 0.3 g of succinic anhydride/g of gelatin. Amino acid composition of succinylated gelatin was similar to that of unmodified gelatin. Heavy metal contents such as cadmium, lead, copper and zinc of succinylated gelatin were lower than those of unmodified gelatin.

  • The emulsion stabilizing properties of soy protein isolate (SPI) were investigated in terms of the protein and oil concentration. Particularly, the dependence of emulsion stability on the oil particle size and viscosity of emulsion was studied in conjunction with the adsorption pattern of protein onto the water/oil interface during emulsification. The data showed that increasing protein concentration decreased the oil particle size and increased the emulsion viscosity, resulting in the enhanced emulsion stability. In contrast, increasing oil concentration increased both the oil praticle size and the emulsion viscosity, and thus emulsion stability varied depending on which factor predominated the overall emulsion system.

Preparation of Cosmeceuticals Containing Scutellaria baicalensis Extracts: Optimization of Emulsion Stability and Antibacterial Property (황금추출물이 함유된 Cosmeceuticals의 제조: 유화안정성 및 항균특성 최적화)

  • To optimize the emulsion stability and antibacterial activity against Escherichia coli (E. coli) of cosmeceuticals using Scutellaria baicalensis extracts and olive wax as natural emulsifiers, we conducted a study. The independent variables were the amounts of Scutellaria baicalensis extracts and olive wax added. The response variables included the emulsion stability index (ESI) of the cosmeceuticals product and the inhibition diameter against E. coli. Through central composite design-response surface methodology (CCD-RSM), we obtained a statistically significant and reliable regression equation within a 95% confidence interval. By optimizing multiple responses, we determined that the optimal emulsification conditions that satisfied both ESI and E. coli inhibition diameter were 3.7 wt% of Scutellaria baicalensis extracts and 2.7 wt% of olive wax. The predicted ESI and E. coli inhibition diameter were 97.9% and 9.7 mm, respectively. When actual experiments were conducted under the optimal conditions, the measured ESI and E. coli inhibition diameter were 95.0% and 9.4 mm, respectively, with an average error rate of 3.2 ± 0.4%.
