The Variation of Leaf Form of Rare Endemic Berchemia berchemiaefolia Populations (희귀수종(稀貴樹種) 망개나무 자생집단(自生集團)의 엽형변이(葉型變異))

  • 7 populations of rare endemic Berchemia berchemiaefolia were analysed using multivariate analysis for 9 characteristics of leaf morphology. The aim of this study was to examine geographic variation to support a genetic resource conservation plan of B. berchemiaefolia in Korea. In the morphological characters, nine characters of leaf were 10.25 cm (blade length), 4.10 cm (maximum width), 2.52 (blade length/maximum width), 3.22 cm (upper 1/3 width), 3.42 cm (lower 1/3 width), 0.95 (upper 1/3 width/lower 1/3 width), 1.24 cm (petiole length), 8.91 (blade length/petiole length), 8.16ea (vein number), respectively. Nested anova showed that were statistically significant differences among populations as well as among individuals within populations in all 9 quantitative characters. In 7 of 9 characters, variance components among individuals within populations were higher than those among populations. Cluster analysis using complete linkage method showed two groups (Chungbuk and Gyeongbuk districts) to Euclidean distance 1.2. Among principal components, primary 3 principal components appeared to be major variables because of the loading contribution of 87.3%. The first contribution was blade length, blade length/maximum width and blade length/petiole length; the second one was maximum width, upper 1/3 width and lower 1/3 width; the third one was petiole length, respectively.

First Report of Apricot pseudo-chlorotic leaf spot virus Infecting Peach Trees in South Korea (복숭아나무에서 검출된 Apricot pseudo-chlorotic leaf spot virus의 국내 첫 보고)

  • In May 2016, 24 peach samples showing abnormal and virus like symptoms were collected in one of major peach producing area, Yeongcheon-si, Gyeongsangbuk-do, Korea. We performed RT-PCR diagnosis for confirmation of viral infection. The diagnostic targets are 17 species of viruses and viroids that quarantine and high risk pathogens when it occur. As a results, seven species of viruses and viroids, including an unreported (Apricot pseudo-chlorotic leaf spot virus, APCLSV) and a quarantine (Peach latent mosaic viroid, PLMVd) species in Korea, were detected. For the sequence analysis of unreported virus, APCLSV, the sequence of coat protein gene were amplified and cloned. The sequence showed 97% nucleotide identity with other APCLSV isolates and compared with other seven species of reported Trichoviruses. This virus was classified as APCLSV based on the sequence and phylogenetic analysis. This isolate was named Yeongcheon. As patterns of APCLSV occurrence, all samples that APCLSV detected were co-infected with Apple chlorotic leaf spot virus (ACLSV). As properties of ACLSV, APCLSV has high possibility of wide spread disease in fruit tree farms in Korea. Therefore, it is necessary to do related researches, such as infection route and influence of disease in commercial orchards.

The Variation of Needle Characteristics of Picea jezoensis(Siebold & Zucc.) Carriere Populations in Korea (한국산 가문비나무 자생집단의 침엽특성 변이)

  • The aim of this study was to examine geographic variation to support a genetic resource conservation plan of rare endemic Picea jezoensis (Siebold & Zucc.) Carriere. Three populations of P. jezoensis were analysed using multivariate analysis for 7 characteristics of needle morphology. In the morphological characters, seven characters of needle were 15.11 mm (needle length), 1.43 mm (needle width), 0.42 mm (needle thickness), 10.9 (length / width of needle), 3.6 (width / thickness of needle), 0.47 mm (Distance between resin duct and vascular), 17.4 ea. (number of stomata row), respectively. Nested analysis showed that were statistically significant differences among populations as well as among individuals within populations in 6 quantitative characters. In 5 of 7 characters, variance components among populations were higher than those among individuals within populations. Cluster analysis using complete linkage method showed two groups (Mt. Gyebang populations was genetically distinct from the other two populations) to Euclidean distance 0.4. Among principal components, primary 2 principal components appeared to be major variables because of the loading contribution of 100%.

Molecular Epidemiological Characteristics of Vibrio Parahaemolyticus as Recently wilde-spreaded in Korea (최근 한국에서 유행하는 장염비브리오균의 분자 역학적 특성)

  • The purpose of this study is to inquire into molecular epidemiological characteristics of Vibrio parahaemolyticus. For this study, 120 strains(120 strains of Vibrio parahaemolyticus sampled from diarrhea patients) were examined and analyzed for biochemical characteristics, TDH (thermostable direct hemolysin) antibiotics sensitivity and detection of toxR, gyrE, tdh, and tds gents. G-S PCR (Group Specific Polymerase), PFGE (Pulsed-field Gel Electrophoriesis) methods were performed on the materials from patients were results. 1 Vibrio parahaemolyticus didn't grow in 0% density of NaCl, but the fact was found that those grew in 8% density of NaCl. 2. O:K serotypes of Vibrio parahaemolyticus was turned up in domestic patients was 17 types. Among those O3:K6 was the most, it was 68.3%. 3. In 18 kinds of antibiotic tests resistant against Ampicillin, Ticacillin was comparatively high. the case of resistant against Ampicillin, Ticacillin, Vancomycin at the multiple resistant was 52.5%. 4. Toxin gene tdh had only 109 strains among 120 ones isolated from patients held the genes of 199bp size, and 11 strains was negativity 5. In the test of Kanagawa toxic productivity, 107 strains among strains isolated from patients appeared to be positivity reaction 6. The strain that held trh toxin was only 3, and those among test strains had the genes of 250bp size and that had tdh, trh genes at a time were 3 strains, and TDH toxic productivity of those were 16 times, and it was weak. 7. Group Specific-PCR appeared to be useful in the confirmation of O3:K6 serotype interrelations. 8. Three strains which showed difference of 7 DNA sequence even in the same serotype were detected by the result of analyzing the regular gene, toxRS DNA sequence. These strains are differ from general strains which carry infection easily. 9. These mutual dose epidemiological relations were classified into smaller-parts through PFGE method. As a result of such classify, 3 findings were found. V. parahaemolyticus sampled from diarrhea patients were classified into 3 types. And third, the result obtained through PFGE method can be used as a useful tool in a point of molecular-epidemiological view.

The Genetic Analysis Study of Ancient Human Bones Excavated at Janggi-dong site, Gimpo (김포 장기동 유적 출토 인골의 유전자 분석 연구)

  • Most human bones of Joseon Dynasty period are so good condition that we can do research in physical anthropology, genetics and chemistry with them. In this study, we analyzed DNA typing using 6 human bones of Joseon dynasty period excavated at Janggi-dong, Gimpo. The DNA typing was mitochondrial DNA haplotype, Y-chromosome haplotype and sex determination. Prior to DNA analysis, we distinguished histological index of 6 human bones. As the result of mitochondrial DNA analysis, most of bones were confirmed as haplogroup G, R11, M7, A5, etc. As the result of sex determination, 4 human bones were female and 2 human bones were male. The male haplogroup was confirmed as haplogroup O by the single nucleotide polymorphism analysis of Y chromosome. For extensive ancient human bone analysis, researchers need to apply a histological index to select ancient human bones and explain a relationship among ancient human bones with various analyses of mitochondrial and nuclear DNA.

The Variation of Germination, Growth and Leaf Form of Open-Pollinated Progenies of Cornus kousa Buerger ex Miquel in Korea (산딸나무(Cornus kousa) 풍매차대(風媒次代)의 발아(發芽), 생장(生長)및 엽형(葉型) 변이(變異))

  • A nursery trial comprising 109 progenies of 5 populations of Cornus kousa was analysed using multivariate analysis of 12 quantitative traits (growth and leaf form). The aim of this study was to examine geographic variation of the species based on the single, existing nursery trial to support a genetic resource conservation plan of Cornus kousa in Korea. Nested Anova showed that there were statistically significant differences among populations as well as among families within populations in all 12 quantitative traits. In 10 of 12 traits, variance components among families within populations were higher than those among populations. Cluster analysis using complete linkage method showed three groups to Euclidean distance 0.8. Among principal components, primary 2 principal components appeared to be major variables because of the loading contribution of 91.9%. The first contribution component was maximum width, vein number, blade length/petiole length and upper 1/3 width lower 1/3 width; the second one was height, diameter at root collar, blade length, upper 1/3 width, petiole length and petiole length/vein number, respectively. But all characters showed no significant difference with the pattern of geographic distribution.

Analysis for the Major Traits and Genetic Similarity of Native Ginseng (Panax Ginseng C.A. Meyer) Collections in Korea. (인삼(Panax ginsneg C.A. Meyer) 수집종의 주요 특성 및 유연관계 분석)

  • In this study, the major agronomic traits were investigated and RAPD technique was applied for the analysis of the genetic relations between the native ginsengs collected from Poonggi and Geumsan provinces in Korea. The main morphological traits were measured for a total of 54 collections of native ginseng from two areas based on UPOV standard. A total of 58 collections consisting of twenty-one native ginsengs collections from Poonggi area, twenty-nine collections from Geumsan area and four varieties of P. quinquefolium, P. japonicum, Chunpoong and Hwangsuk as controls were analyzed and clustered by RAPD. The results indicated that 01-9, 01-35 and 01-44 collections from Poonggi area were grouped into Geumsan area, while 332001, 332002 and 332003 collections from Geumsan area were grouped into Poonggi area. On comparison to the similarity of Poonggi collections (73-95%), the Geumsan collections showed 65-86% similarity in the population. Thus, the cluster should be applied according to the number of stem, number of leaves per stem and leaflet shape on the regionally native ginseng collections. The fourteen primers such as OPA02, OPA07, OPC08, OPD11, OPD20 and so on, will be used to select the native ginseng in the future studies.

Comparison of B and Q biotype distribution, insecticidal mortality, and TYLCV viruliferous rate between Korean and Chinese local populations of Bemisia tabaci (한국과 중국 채집 담배가루이 지역계통의 B, Q biotype 분포 및 살충제 약제반응, 토마토황화잎말림바이러스 보독율 비교)

  • International trade is one of the primary ways that non-native species spread worldwide. Korea and China are geographically close and have a large mutual trade volume. To investigate the population movement of the invasive whitefly(Bemisia tabaci Gennadius) between the two countries, we compared the biotype distribution, insecticidal response, and the TYLCV(tomato yellow leaf curl virus) viruliferous rate of local populations collected in 2019. Based on the mitochondrial DNA COI sequences of B. tabaci, only the Q biotype was found in all populations in Korea, whereas the B biotype (14.3%) and Q biotype (85.7%) were found in China. In the haplotype composition of the B. tabaci Q biotype, only the Q1 group[Q1H1(79.8%) and Q1H2(20.2%)] was observed in China, but the Q1 group [Q1H1(1.7%) and Q1H2(97.5%)] and the Q2 group(only one individual) were found in Korea. The Korean populations showed high mortality(more than 80%) from 15 commercial insecticides, but the Chinese populations showed significantly low mortality from eight insecticides. No TYLCV infections were observed in the Korean populations while the average TYLCV viruliferous rate was 21.4% in the Chinese populations. Taken together, the results suggest that the population structures of B. tabaci in the two countries are different and may have different immigration histories.

Development of Multiplex Microsatellite Marker Set for Identification of Korean Potato Cultivars (국내 감자 품종 판별을 위한 다중 초위성체 마커 세트 개발)

  • To analyze the genetic relationships among Korean potato cultivars and to develop cultivar identification method using DNA markers, we carried out genotyping using simple sequence repeats (SSR) analysis and developed multiplex-SSR set. Initially, we designed 92 SSR primer combinations reported previously and applied them to twenty four Korean potato cultivars. Among the 92 SSR markers, we selected 14 SSR markers based on polymorphism information contents (PIC) values. PIC values of the selected 14 markers ranged from 0.48 to 0.89 with an average of 0.76. PIC value of PSSR-29 was the lowest with 0.48 and PSSR-191 was the highest with 0.89. UPGMA clustering analysis based on genetic distances using 14 SSR markers classified 21 potato cultivars into 2 clusters. Cluster I and II included 16 and 5 cultivars, respectively. And 3 cultivars were not classified into major cluster group I and II. These 14 SSR markers generated a total of 121 alleles and the average number of alleles per SSR marker was 10.8 with a range from 3 to 34. Among the selected markers, we combined three SSR markers, PSSR-17, PSSR-24 and PSSR-24, as a multiplex-SSR set. This multiplex-SSR set used in the study can distinguish all the cultivars with one time PCR and PAGE (Polyacrylamide gel electrophoresis) analysis and PIC value of multiplex-SSR set was 0.95.

Environmental characteristics on habitats of Viola diamantiaca Nakai and its RAPD analysis (금강제비꽃(Viola diamantiaca Nakai) 자생지의 환경특성과 RAPD 분석)

  • This study investigated the environmental factors and conducted a RAPD analysis for a better understanding of the environmental characteristics and regional genetic variation in samples from 18 different areas of Viola diamantiaca. The habitats are mostly located on the slopes of mountains facing north at an altitude ranging from 614 m to 1,462 m above sea level with angles of inclination ranging from 3 degrees to 30 degrees. A total of 268 vascular plant taxa are identified in 35 quadrates of 18 habitats. The importance value of V. diamantiaca is 11.58%, and four highly ranked species, Sasa borealis (5.61%), Meehania urticifolia (5.21%), Ainsliaea acerifolia (3.62%), Pseudostellaria palibiniana (3.60%) are considered to have an affinity with V. diamantiaca in their habitats. The degree of their average species diversity is 1.36, while this metric for their evenness and dominance are 0.89 and 0.07, respectively. The average field capacity of the soil is 25.99%, with organic matter at 17.47%, and the pH is 5.19. The soil texture was confirmed as sandy loam of eleven and loam of seven. The result of the RAPD analysis, among 78 bands amplified with a primer, 64 (84.6%) showed polymorphism. Eighteen populations could be classified into five groups with similarity coefficient values ranging from 0.53 to 0.86. The Mt. Jiri population, which is geographically segregated, shows basal branching within the 18 populations. Five populations, including two in the southern district in Gangwon-do and three in Chungcheongbuk-do, form a distinct clade. Four populations in the central district of Gangwon-do and Mt. Bohyeon in the Gyeongsangbuk-do clade form a sister to the clade containing two populations in Gyeonggi-do and five populations of the northern district in Gangwon-do. The Mt. Gariwang population is placed between the southern district and the central district in the Gangwon-do clades.
