The Effects of gl, gh and wx Gene on the Grain Yield and Yield Components of Rice Plant (수도의 gl, gh 및 wx유전자가 수도수량 및 수량구성요소에 미치는 영향)

  • Isogenic lines for glabrous leaf (gl), hull color(gh) and glutinous endosperm (wx) were used to clarify the effects of the gl, gh and wx gene on the grain yield and yield components in rice plant. Eight genotypes: glgl ghgh wxwx, glgl ghgh WxWx, glgl GH-wxwx, glgl GH-WxWx, GL-ghgh wxwx, GL-ghgh WxWx, GL-GH-wxwx and GL-GH-WxWx were evalutated in the backcrossed $F_2$ generation. Significant effects were measured for gl genotypes on heading date, gh genotypes on grain fertility, and wxwx genotypes on number of panicles per hill and 1, 000 grain weight. The grain yield of straw hull and non glutinous lines were higher than gold hull and glutinous lines. The interaction of three genes, gl, gh and wx was significant for heading date, but not significant in other characters. The average value of isogenic lines which were combined with two recessive genes and one dominant gene were lower and the C.V. value was higher than the isogenic lines which were combined with two dominant genes and one recessive gene in all characters except plant height and number of grains per plant. The average value of isogenic line combined with three recessive genes was lower and the C.V. was higher than the isogenic line combined three dominant genes in all characters measured.

Construction of Genetic Linkage Map using Microsatellite and SNP Markers in Korean Native Chicken (Microsatellite와 SNP Marker를 이용한 한국재래닭의 유전적 연관지도 작성)

  • Chicken is one of the major livestock, especially for supplying proteins to human. The chicken genome size is approximately one-third compared with that of the human genome and regarded as a valuable model animal for genetics and development biology. In this study, we constructed the genetic linkage map for Korean native chicken (KNC) using 131 microsatellite (MS) and 8 single nucleotide polymorphism (SNP) markers. As a result, the total map length was calculated as 2729.4 cM and the average genetic distance between markers was 19.64 cM. The marker orders and genetic distances were well matched with the consensus linkage map except for the physical order of ADL0278 and MCW0351 in GGA8. In addition, the recombination rates in marcrochromosomes were 3.7 times higher than that of microchromosomes. The average numbers of alleles, expected heterozygosity (Hexp) and polymorphic information content (PIC) values were calculated as 5.5, 0.63 and 0.58, respectively. These results will give useful information for the understanding of genetic structure and QTL studies in KNC.

The Prevalence and Distribution of the P and G Genotypes of a Group A Rotavirus Detected in Acute Gastroenteritis Patients from Incheon (인천지역 급성 설사환자의 group A rotavirus 감염 실태 및 P와 G 유전자형 분포)

  • Rotavirus is the main cause of severe diarrhea in infants and young children of the world. However, the frequency of genetic alterations makes it hard to control the prophylaxis. Therefore, continuous monitoring of the rotavirus's genetic change is inevitable to prevent disease prevalence and is useful in inventing an efficient vaccine. From January 2005 to December 2010, we investigated 11,607 stool samples of acute gastroenteritis patients in the Incheon metropolitan area. About 13.18% (1,530 stool samples) of all samples had a positive reaction against rotavirus using an antigen capture enzyme-linked immunosorbent assay (ELISA). Then, the 160 stool samples were searched for subtypes of group A rotavirus by using a reverse transcription polymerase chain reaction (RT-PCR) and a nested multiplex RCR. In P sub-typing, P8 (56.3%) was an extremely prevalent genotype, followed by P6 (21.3%), and P1A (10.0%). G1 (39.4%) was most widespread in the G subtype, followed by G4 (25.0%) and G3 (18.8%). G1P8 (35.5%) was the most common G and P subtype combination, followed by G4P6 (19.3%) and G3P8 (13.1%). These results might be useful data for understanding the epidemiological status of group A-rotavirus dispersion in the Incheon metropolitan area.

Association of SNP Markers on Chromosomes 3 and 9 with Body Weight in Jeju Horses (제주마에서 3번 및 9번 염색체상의 단일염기변이와 생체중과의 관련성 연구)

  • This study was conducted to investigate the association of single nucleotide polymorphism (SNP) markers on equine chromosomes (ECA) 3 and 9 with body weight in Jeju horses. We used DNA samples and body weight data of 320 horses provided by the Livestock Promotion Agency, Jeju Special Self-Governing Province, and the Korean Racing Association, respectively. We genotyped all the experimental animals using nine SNP markers located on ECA 3 (BIEC2-808466, BIEC2-808543, BIEC2-808967, and BIEC2-809370) and ECA 9 (BIEC2-1105370, BIEC2-1105372, BIEC2-1105377, BIEC21105505, and BIEC2-1105840). These markers were selected due to their effects on body conformation traits in horses. The joint effect of the genotypes of the two SNP markers (BIEC2-808467 and BIEC2-1105377) regarding body weight were also evaluated. The estimated breeding value (EBV) of body weight was obtained as the dependent variable for association analyses using a linear mixed model. Significant associations were detected between SNP markers (BIEC2-808543, BIEC2-808967, BIEC2-809370, BIEC2-1105370, BIEC2-1105372, and BIEC2-1105377) and the body weight EBV. In addition, the joint genotype effect of the BIEC2-808467 and BIEC2-1105377 on the body weight EBV was significant. These results indicate that the SNP markers, which showed their significant effects on body conformation, can be used as genetic markers to improve the efficiency of the selective breeding program for the body weight traits in Jeju horses.

Breeding of "Wongyo 3111", Intermediate Strawberry Parent Line with High Sugar Content and Large-sized Fruit (고당도 대과성 특성을 지닌 중간모본용 딸기 "원교 3111호" 육성)

  • Strawberry is an out-crossed crop that is relatively sensitive to inbreeding, and as a result, most of breeding programmes in strawberry have been based on selection breeding where elite parents are selected from intercrossing. Also, pedigree breeding has been successfully employed in a number of instances to concentrate genes of interest. Therefore, in order to obtain inbred line having the genotype of excellent combining ability by selfing and incrossing to improve breeding efficiency of strawberry, elite varieties were self-pollinated. From the selfing line of "Benihoppe" cultivar, we selected "Wongyo 3111" having excellent horticultural traits such as plant vigor, sugar content and fruit hardness among these progenies. "Wongyo 3111" shows erect plant type, vigorous growth habit, early flower bud differentiation with 16~17 flowers per cluster. Fruits of "Wongyo 3111" are conical type having a bright red skin color, and 17.2 g in an average weight. Also that have a relatively more high sugar/acid ratio and firmness than check cultivars as sugar content of 11.3oBx, acidity of 0.50% and firmness of 21.7 g/$mm^2$. But although yield of "Wongyo 3111" is not significantly different from check cultivars, its marketable yield is remarkably lower than that of check cultivars because of poor fruit-setting under low temperature. Disease and pest reactions of "Wongyo 3111" are more sensitive to powery mildew, anthracnose and spotted spider mites than check cultivars. Therefore, "Wongyo 3111" is suitable for intermediate parent line better than as a cultivation cultivar.

The Trend and Achievements of Forest Genetics Research in Abroad (선진국(先進國)에 있어서의 임목육종연구(林木育種硏究)의 동향(動向))

  • The trend and achievements of forest genetics research in abroad were investigated through observation tours and reference work and following facts were found to be important aspects which should be adopted in the forest genetics research program in Korea. Because of world wide recognization on the urgency of taking a measure to reserve some areas of the representative forest type on the globe before the extingtion of such forest type as the results of continuous exploitations of the natural forests to meet the timber demand all over the world, it is urgently needed to take a measure to reserve certain areas of natural stand of Pinus koraiensis, Pinus parviflora, Pinus densiflora f. erectra, Abies koreana, Quercus sp., Populus sp., etc. as gene pool to be used for the future program of forest tree improvement. And the genetic studies of those natural forest of economic tree species are also to be performed. 1. Increase of the number of selected tree for breeding purpose. Because of the fact that the number of plus tree at present is too small to carry out selection program for tree improvement, particularly for the formation of source population for recurrent selection of parent trees of the 2nd generation seed orchard it is to be strongly emphasized to increase the number of plus tree by alleviating selection criteria in order to enlarge the population size of plus trees to make the selection program more efficient. 2. Progeny testing More stress should be placed on carrying out progeny testing of selected trees with open pollinated seeds. And particular efforts are to be made for conducting studies on adult/juvenile correlation of important traits with a view to enable to predict adult performances with some traits revealed in juvenile age thus to save time for progeny testing. 3. Genotype-environment interaction Studies on genotype and environment interaction should be conducted in order to elucidate whether the plus trees selected on the good site express their superiority on the poor site or not and how the environment affect the genotype. And the justification of present classification of seed distribution area should be examined. 4. Seed orchard of broad leaf tree species. Due to the difficulty of accurate comparison of growth rate of neighbouring trees of broad leaf tree species in natural stand, it is recommended that for the improvement of broad leaf trees a seedling seed orchard is to be made by roguing the progeny test plantation planted densely with control pollinated seedlings of selected trees. 5. Breeding for insect resistant varieties. In the light of the fact that the resistant characteristics against insect such as pine gall midge (Thiecodiplosis japonensis U. et I.) and pine bark beetle (Myelophilus pinipera L.) are highly correlated with the amount and quality of resin which are known as gene controlled characteristics, breeding for insect resistance should be carried out. 6. Breeding for timber properties. With the tree species for pulp wood in particular, emphasis should be placed upon breeding for high specific gravity of timber. 7. Introduction of Cryptomeria and Japanese Cypress In the light of the fact that the major clones of Cryptomeria are originated from Yoshino source and are being planted up to considerably north and high elevation in Japan, those species should be examined on their cold resistance in Korea by planting them in further northern part of the country.
