Analysis of the Contents and Physiological Activities of Calystegia japonica Leaf Extracts (메꽃(Calystegia japonica) 잎 추출물의 함유성분과 생리활성에 관한 연구)

  • This study examined the physiological activities as well as the total polyphenol and flavonoid content of water extract (WE), ethanol extract (EE) and hot water extract of Calystegia japonica leaves under high pressure (HWE). The xanthine oxidase inhibitory rate of EE was the highest with a value of 98.89% at a concentration of 1.0 mg/mL, whereas the rate of WE and HWE was over 90% at a concentration of 0.3 mg/mL. The superoxide dismutase (SOD)-like activity of HWE was the highest at 18.88%. The nitrite scavenging abilities were 64.59-66.46% at conditions of pH 1.2 and 1.0 mg/mL, and 52.78-55.89% at pH 3.0. The electron donating ability of EE was the highest with a value of 84.80% at a concentration of 0.1 mg/mL. All extracts showed the highest degree of electron donating at the concentration of 0.1 mg/ mL, and this effect decreased as the extract concentration increased. The EE had the highest content of total polyphenol compound (173.89 mg/g) and flavonoid compounds (40.68 mg/g).

Antioxidant Activities of Solvent Extracts from Pomegranate Endocarp (석류 내피 용매별 추출물의 항산화 활성)

  • This study was designed to investigate antioxidant activities of pomegranate endocarp extracts. Pomegranate endocarp extract contains the highest antioxidant function compared to pomegranate extracts from other parts. Pomegranate endocarp extract was fractionated with hexane, dichloromethane, ethyl acetate, butanol extract, and water, followed by evaluation for antioxidant activity. During this experiment, various antioxidant tests such as nitrite scavenging activity, reducing power, superoxide anion scavenging activity, $ABTS^+$ scavenging activity, SOD like-activity, and DPPH radical scavenging activity were conducted on the $CH_2Cl_2$, EtOAc, BuOH, $H_2O$ fractions of pomegranate endocarp extract. Results showed that the ethyl acetate fraction contained the highest DPPH radical scavenging activity among the samples. For SOD like-activity, the dichloromethane fraction had the highest antioxidant activity. For superoxide anion scavenging activity, the ethyl acetate fraction had the highest antioxidant activity. In the $ABTS^+$ scavenging activity test, the ethyl acetate fraction $IC_{50}$ was 39.26 ${\mu}g$/mL, whereas that of the butanol fraction was 40.95 ${\mu}g$/mL. In testing reduction power at 0.1 mg/mL, the O.D. of the ethyl acetate fraction was highest at 1.404 and showed higher activity than ascorbic acid at 1.332. The results of the nitrite scavenging activity test were very similar to those of the SOD like-activity test. These results suggest that pomegranate endocarp extract may have value as a natural antioxidant.

Antioxidative activity of some natural products which have been orientally used as ophthalmic drugs (한방에서 안질환에 사용되어 온 천연물들의 항산화 작용)

  • Antioxidative activities of methanol extracts of 7 natural products which have been orientally used or studied as ophthalmic drugs were investigated. All of 7 extracts assayed - Rubus coreanus, Dendrobium monile, Eriocaulon sieboldianum, Ligustrum japonicum, Cassia tora, Cicadae Periostacum and Polygonatum odoratum var pluriflorum had some electron-donating ability to OPPH radicals, in which R. coreanus and D. monile showed strong activity as similar as ascorbic acid or BHT. At the concentration of $200{\mu}g/m{\ell}$, methanol extract of L. japonicum showed superoxide dismutase-like activity the same as 0.1 mM ascorbic acid and methanol extract of R. coreanus inhibited effectively the production of ABTS radical produced by $H_2O_2$. Furthermore, methanol extract of C. tora showed the highest nitrite scavenging ability. The inherent antioxidative activities of these natural products appear to be responsible for its ophthalmic effects.

Extractive Optimization of Functional Components for Processing of Onion Health Promotion Drink (양파 음료 제조를 위한 기능성 성분 추출 최적화)

  • Onion, licorice, abgelia root, Chinese date, pine needle, and mulberry leaf were used to extract functional components far onion complex drink. No differences were observed between water extraction at room temperature and methanol extraction in electron-donating ability (EDA), thiosulfinate content (TSC), and ascorbic acid content (AAC), whereas water extraction resulted in higher nitrite-scavenging ability at pH 1.2 and 3.0, and lower superoxide dismutase-like ability than methanol extraction. Level of water extracts prepared by hard-boiling for 3 hr at above $100^{\circ}C$ was higher in all functional abilities except TSC than those prepared at room temperature. Optimal conditions for water extraction and storage were $100-120^{\circ}C$ for 6 hr and low-temperature storage, respectively.

  • This study was performed to investigate the effect of Portulaca oleracea extract on the antioxidant and antimicrobial activities against Helicobacter pylori. The each solvent extracts prepared from Portulaca oleracea were investigated by measuring total phenolic compounds, electron donating ability, superoxide dismutase-like ability and thiobarbituric acid reactive substances The herb extractor extract yielded the highest content of total phenolic compounds(72.2 mg%). The electron donating abilities(EDA) of ethyl acetate and methanol extracts showed high antioxidant activity. The superoxide dismutase (SOD)-like abilities of ethyl acetate and petroleum ether extracts also showed some activity. The antioxidant activity of thiobarbituric acid reactive substances was not significant. The petroleum ether extract of Portulaca oleracea showed the highest antimicrobial activity at 10,000 ppm concentration.

  • This study was designed to test the effect of dandelion (Taraxacum officinale) extracts on the intestinal microorganisms of streptozotocin-induced diabetic rats. Male Wistar rats were divided into normal, diabetic control and dandelion extract groups. The extracts were prepared by water, ethylacetate and ether from leaf and root, respectively. Diabetes was induced by injecting streptozotocin (55 mg/kg BW, i.p.) in citrate buffer. The extract was supplemented in 11.45g of raw dandelion / kg diet for 50 days. The growth of Lactobacillus was more enhanced in dandelion leaf-water and leaf-ethylacetate extract group than that of the diabetic control group, whereas the growth of E. coli decreased. Results indicate that the dandelion extracts would be effective to improve the intestinal microorganisms.

Effect of Ethanol Extract of Quercus mongolica Leaf as Natural Food Preservative (신갈나무 잎 에탄올 추출물의 식품보존제 효과)

  • This study was investigated to determine the antimicrobial effect of the ethanol extract of Quercus mongolica leaf on microbial growth. The ethanol extract at the concentration of $250\;\mu\textrm{g}/mL\;and\;500\;\mu\textrm{g}/mL$ inhibited the growth of gram positive and gram negative food-borne disease bacteria for 40 hours in tryptic soy broth, respectively. Antimicrobial activity of the ethanol extract from Quercus mongolica leaf was not affected by pH and heat treatment. The comparision between ethanol extract and commercially available preservatives on antimicrobial activity in food system was conducted. When the 0.1% ethanol extract of Quercus mongolica leaf was added to pine needle drink and carrot juice, antimicrobial activity was similar to those of containing 0.05% benzoic acid and 0.5% grapefruit seed extract. Also addition of 2~3% ethanol extract to the soybean paste inhibited the microbial growth up to 7 week, comparable to the inhibition of 2% ethanol. Thus, this results indicate that the ethanol extract of Quercus mongolica leaf may be useful as natural antimicrobial agents.

Antioxidant and Antihemolytic Activity of Ethanol Extracts of Rubus coreanus Miquel (복분자 에탄올 추출물의 항산화 및 항용혈 효능)

  • To develop a natural antioxidant and anti-hemolytic agents, we investigated the effects of ethanol extracts of Rubus coreanus Miquel (crude extracts). Crude extract was extracted with ethanol. Antioxidant activity of crude extracts was evaluated by employing two different assays, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity and super oxide dismutase(SOD)-like activity. Also, anti-hemolytic activity of crude extract was determined using $H_2O_2$-induced hemolysis in normal rat red blood cells (RBC) or plasma. The extracts obtained from crude extract dose-dependently increased the scavenging activity on DPPH-induced radicals and SOD-like activity. RBC oxidative hemolysis induced by $H_2O_2$ was significantly suppressed by the extracts of Rubus coreanus Miquel in a dose-dependent manner. These results suggest that crude extracts may have value as the potential antioxidant and anti-hemolytic medicinal plant.

  • This study was designed to analyze the election donating ability(EDA), superoxide dismutase(SOD)-like activity, nitrite scavenging ability and xanthine oxidase for food nutritional evaluation and excavation of functional materials in fruits of Elaeagnus multiflora THUNB. The electron donating ability of ethanol extracts in the 1.0 mg/ml concentration of extract was higher in extract of unripe fruits than others. The superoxide dismutase (SOD)-like activity was increased according to increasing of the extract concentration. The SOD-like activity was highest in the extract of unripe fruits among the extracts. The nitrite scavenging ability of unripe fruits ethanol extracts in 1.0 mg/ml of extraction solution in pH 1.2 was higher than extract of ripe and Ovenipe fruits. The nitrite scavenging ability was decreased according to increasing of pH. Also, inhibition of xanthine oxidase activity were higher in extracts of unripe fruits than those in extract of ripe and over-ripe fruits.

  • The studies on the carotenoids in the viscera of abalone (Haliotis discus hannai) have been-carried out. The pigments were extracted with acetone-methanol mixture (4 : 1) from the viscera of abalones which were caught around the coastal water of Korea from March to August. The individual carotenoid in the extracts was separated and purified by the silica gel TLC, $Mg(OH)_2$ impregnated paper chromatography and $Mg_2(OH)_2CO_3$ TLC. The isolated eleven carotenoids were investigated and identified by epoxide test, partition test, reduction with sodium borohydride, alkaline hydrolysis, co-chromatography and comparative test with reference carotenoids and electronic and IR absorption spectrophotometry. ${\alpha}$-Carotene, ${\beta}$-carotene. lutein, zeaxanthin, siponaxanthin, siponein, fucoxanthin, loroxan-thin-like and fucoxanthinol-like have been identified among the eleven carotenoids isolated. It has been found that fucoxanthin, on alkaline treatment, was transformed to the product of which chromophore was the same one as fucochrome and semifucoxanthol. Among the identified nein carotenoids siphonaxanthin, siponein, fucoxanthin, loroxanthin-like and fucoxanthinol-like have not been reported previously to be contained in the shellfish.
