A study on the Determination of Trace Se and Bi in the Scalp Hair by Hydride Generation- Inductively Coupled Plasma Atomic Emission Spectrometry (수소화물발생 유도결합플라즈마 원자방출분광법에 의한 머리카락 시료 중 미량의 Se와 Bi의 분석에 관한 연구)

  • A method to determine the trace amount of Se and Bi in the scalp hair using the hydride generation inductively coupled plasma atomic emmission spectrometry was studied. The optimum operation conditions of ICP for hydride generation are 0.6~0.8L/min for the carrier gas flow rate, and 6mm above the induction coil for the observation height. Hydrochloric acid concentrations for the optimum hydride generation conditions were greater than 1.5M when 2.5% $NaBH_4$ and NaOH were used, and greater than 0.5M when 2.5% $NaBH_4$ and 0.1% NaOH were used. Severe interference effects are observed from transition metals such as Cu and Ni, and they could be circumvented by the coprecipitation with lanthanum hydroxide.

  • Acid effects on the ICP-AES signals are studied as the concentrations of nitric acid, hydrochloric acid, sulfuric acid, and 1:1 mixture of nitric acid and hydrochloric acid are changed. Almost all analyte signals are depressed. The extent of the depression due to the pressence of the acids became to be pecular when the acid concentration was over 1%. Among the acids used, the suppression due to sulfuric acid is most severe and unexpectable. The ratios of the analyte signal to Ar signal and the Mg II signal to Mg I signal are measured as the concentration of the acids changed. In this study, it is proved that the main reason of the signal reduction is the change in the nebulization efficiency, for example, droplet size distribution, viscosity and surface tension variation, not the alteration of plasma excitation characteristics. There was no relationship found between ionization potential and analyte signal reduction in ICP-MS.

  • Tellurium in simulated nuclear spent fuels (SIMFUEL) has been determined by hydride generation-inductively coupled plasma atomic emission spectrometry (HG-ICP-AES). Parameters such as concentrations of HCl and $NaBH_4$, flow rate of HCl and $NaBH_4$ were optimized and then the effects of U, Mo, Pd, Rh and Ru on the Te intensity were investigated. A thiourea as a masking agent was used to eliminate or minimize such interferences specially caused by palladium. Tellurium was measured by HG-ICP-AES and ICP-MS after separation of tellurium from SIMFUEL with cation exchange chromatography. The relative deviation between direct measurement and separation method was less than 6% based on the data by ICP-MS.

Ultrasound-Triggered Drug Release of Hydroxyapatite Coated Liposomes (하이드록시아파타이트 코팅 리포솜의 초음파에 의한 약물방출)

  • Liposomes, which can deliver payload at target site, have been studied as drug carrier. However, conventional liposomes have limitation for drug release at target site. Therefore, we developed hydroxyapatite (HA) coated ultrasound sensitive liposomes to increase drug release at target site and to enhance stability in blood stream. Control liposome was prepared using hydrogenated soy phosphatidylcholine (HSPC) and cholesterol, and then we assessed HA coating on the surface of control liposomes using calcium acetate, phosphoric acid, and 25% ammonium solution. Doxorubicin was used as a model drug. Size of HA coated liposomes was 120 nm and encapsulation efficiency of doxorubicin in liposomes was up to 95%. Size of HA coated liposomes are not changed in 30% serum solution, however, the control liposomes was 1.4 fold increased. After ultrasound triggered drug release from liposomes, intracellular efficiency of drug released from HA coated liposomes was 3 fold increased compared to control liposomes. In this study, we developed ultrasound sensitive liposomes to enhance drug release, which will be applied in controlled drug release at disease site.

  • The coating cracking on a substrate system was analyzed using a fracture mechanics approach. Multiple cracking in the bending configuration was analyzed using a variational mechanics approach to fracture mechanics of coatin $g_strate system. The strain energy release rate on bending geometry developed permits the prediction of crack growth in the coating layer on a substrate. Also, it can be used appropriately to the characterization of multiple cracking of coating. The obtained critical strain energy release rate (in-situ fracture toughness) will be a material property of coating and it will provide a better insight into coating cracking.ng.

  • 목적 : 정천탕(定喘湯, JCT)이 IgE/mast cell로 유포되는 천식(喘息)등의 알러지성 질환에 대한 효과를 실험적으로 검증하고자 하였다. 방법 : IgE의 혈중농도와(in vivo) 분비 량을 측정하였으며(in vitro), 알러지 관련 유전자의 발현을 RT-PCR로 검색하였고, JCT의 수용성 복합 추출물이 compound 48/80로 유발되는 mast cell line(IC-2)의 histamine의 방출에 미치는 직접적인 영향을 측정하였다(in vitro). 결과 : 정천탕은 ovalbumin으로 유발시킨 마우스의 혈중 IgE량과 IL-4와 CD-40로 유도시킨 spleen B 임파구의 IgE 생성량도 현저하게 감소시켰다. 또한 INF-r의 발현은 증가시킨 반면, IL-4와 IL-6의 발현과 mast cell의 histamine 방출은 억제하는 것으로 나타났다. 결론 : 정천탕의 항알러지 효과를 실험적으로 확인 할 수 있었으며 임상적으로는 천식등 알러지성 질환에 사용 할 수 있을 것으로 사료된다.

  • The 4-Hydroxynonenal (HNE) affects vascular dysfunctions probably through the interruption of the cellular redox balance. To better understand vascular abnormalities resulting from the accumulation of HNE, we delineated mechanism by which mitochondrial apoptosis occurs in the YPEN-1 endothelial cells. HNE treatment led to the loss of mitochondrial membrane potential (${\delta}{\Psi}_m$), resulting in the release of cytochrome c. Data showed decreased Bcl-2 and increased Bax protein levels in HNE-treated cells. NAC, a reactive oxygen species (ROS) scavenger, and penicillamine, the peroxynitrite scavenger, blocked HNE-mediated ROS generation, thereby thwarting the cytochrome c release and apoptosis. The treatment of the cells with zVAD-fmk, a broad range caspase inhibitor did not suppress HNE-induced apoptosis, suggesting that the apoptosis might be the possibility of caspase-independent process. Our findings delineate the underlying mechanism of the HNE induced endothelial apoptosis by triggering depolarization of mitochondria membrane potential that can lead to the deterioration of vasculature homeostasis and subsequent vascular dysfunction with aging.

  • Coronal mass ejection (CME) released due to solar flare explosion cause geomagnetic disturbance. The induced current by massive geomagnetic disturbance can cause damage to the transformer. The calculated geoelectric field is a major parameter of the geomagnetically induced current (GIC). The method applying a Fourier transform has a high accuracy but it needs all data measured for 24 hours. And the other method applying a integral equation can calculate in real time but it requires to check an accuracy. To reduce the gap between the calculated results of two methods, it adjusts the integration section. As a result, the correlation between two calculated geoelectric fields is high, and the event time and direction of the calculated current is the same as that of the measured current, and it's accuracy rate is above 92 percent.

  • Zaltoprofen, a propionic acid derivative non-steroidal anti-inflammatory drug, was known to have powerful inhibitory effects on acute, subacute and chronic inflammation. For initial release and sustained release, the microspheres were prepared using an emulsion-solvent evaporation method like an O/W emulsion method with varying the ratio of zaltoprofen-loaded polyoxalate (POX)/PLGA micropheres. The morphology of the microspheres was confirmed by scanning electron microscopy. The crystallinity of microspheres was analyzed by X-ray diffraction and differential scanning calorimeter. Fourier transform infrared spectroscopy was used to analyze the chemical structure of microspheres. The increased ratio of POX microspheres affected the initial drug release, and the sustained release of drug was influenced by ratio of PLGA microspheres. In this study, the initial release behavior of zaltoprofen can be controlled by the ratio of POX/PLGA microspheres.

  • Bismuth germanate crystals well known as scintillator were grown by Czochralski method. In order to understand a mechanism of radiation resistance in Eu-doped BGO, we measured radiation induced-absorption spectra, excitation spectra, emission spectra and luminescence lifetimes of BGO crystals. We found that the charge transfer state of $Eu^{3+}$ ion is to play a key role to enhance the radiation resistance in BGO crystal. The $^{5}D_{0}$ emission of $Eu^{3+}$ ions that is not suitable for the radiation detectors due to a long decay time was found to be increased with increasing europium concentration. In the BGO crystal doped with 0.1 mole%, the density of radiation induced color centers was reduced about twenty times and the light output of $^{5}D_{0}$ was negligible by comparing to that of BGO.
