  • 최근 도시의 발달은 하상공간에 대한 이용도를 높이는 방향으로 개발이 진행되어가는 추세이며, 하상도로 및 하상주차장의 이용은 이제 도시 내에서 이용 가능한 마지막 여유 공간으로 인식될 정도로 그 의존도가 높아져가고 있다. 그러나 하상공간의 활용도가 높아져갈수록 도시홍수의 발생으로 인한 대피문제가 발생하게 되고 돌발홍수로 인하여 하상도로의 차단 혹은 하상 주차장에 주차된 차량의 소거가 늦어지는 경우 고스란히 피해를 보게 되는 등 그 부작용도 계속 증가되고 있다. 도시홍수의 특성을 살펴보면 국지성 돌발 강우에 의한 유량의 급격한 증가와 짧은 유하시간, 작은 유역면적 등에 의하여 주요 예보지점까지의 도달시간이 매우 짧아 수문학적 홍수예측 모형을 이용하여 홍수예측 업무를 수행하는데 선행시간을 충분히 확보할 수 없다는 단점을 지니고 있다. 이에 따라 본 연구에서는 기존의 하천시스템에 대한 설계 등을 목적으로 하여 모형의 적용을 통한 시뮬레이션 기법을 적용하고 이를 통하여 홍수 예경보를 발령하기에는 선행시간의 확보(대피시간의 확보)라는 측면에서 상당한 어려움을 지닐 수 있으므로 시시각각으로 측정되는 실시간 수위측정 자료 및 실시간 강우자료를 이용하여 모형의 수행과정을 생략하고 하천의 수위변동을 직접 예측하고 대피할 수 있는 시나리오 기반의 수문모형을 개발하였다. Analyzing Vomit of Platalea minor (Black-faced Spoonbill) to Identify Food Components using Next-Generation Sequencing and Microscopy (차세대염기서열 및 현미경 분석을 통한 저어새의 토사물 내 먹이생물 분석)

  • We sampled vomit of black-faced spoonbills(Platalea minor) during the brood-rearing season (from June 2011 to June 2014) at the Namdong reservoir in Incheon and analyzed the food components in the vomit using microscopy and next-generation sequencing (NGS). Microscopic observations primarily helped in identifying osteichthyes (bony fishes), crustaceans, and polychaetes. In particular, species belonging to the families Mugilidae and Gobiidae among the fish, and Macrophthalmus japonicas among the crustaceans, were observed at high frequency. Results of NGS analysis revealed the predominant presence of bony fish (42.58% of total reads) and crustaceans (40.75% of total reads), whereas others, such as polychaetes (12.66%), insects (0.24%), and unidentified species (3.78%), occurred in lower proportions. At the species level, results of NGS analysis revealed that Macrophthalmus abbreviates and Macrobrachium sp. among the crustaceans, and Acanthogobius hasta, Tridentiger obscurus, and Pterogobius zacalles among the bony fish, made up a high proportion of the total reads. These food species are frequently found at tidal flats in the Songdo and Sihwa lakes, emphasizing the importance of these areas as potential feeding sites of the black-faced spoonbill. Feed composition of the black-faced spoonbill, as evaluated by analyzing its vomit, differed when the evaluations were done by microscopic observation or by NGS analysis. Evaluation by microscopic observation is difficult and not error free, owing to the degradation of the samples to be analyzed; however, NGS analysis is more accurate, because it makes use of genetic information. Therefore, accurately analyzing food components from morphologically indistinguishable samples is possible by using genetic analysis.

Immunomodulatory activities of crude polysaccharide fraction separated from Perilla frutescens Britton var. acuta Kudo (자소엽(Perilla frutescens Britton var. acuta Kudo) 조다당류 추출물의 면역활성 효과)

  • This aim of this study was to examine the immunomodulatory activities of crude polysaccharides from Perilla frutescens Britton var. acuta Kudo (PCP) in mouse bone marrow-derived dendritic cells (BMDC) and splenocytes. The immunomodulatory activity was determined by cell viability, nitric oxide (NO) production, cell surface marker expression (CD 80/86 and MHC class I/II), and cytokine production in BMDC, and cell viability, and cytokine production in splenocytes. Cell proliferation and cytokine production (tumor necrosis factor; TNF-α, interleukin (IL)-6, IL-1β, and IL-12) tested in BMDC were significantly increased by PCP treatment. Additionally, the cell surface markers (CD 80/86, MHC class I/II) were highly increased by PCP treatment. For cytokine production in splenocytes, PCP treatment significantly increased the production of Th 1 cytokines [IL-2 and interferon (IFN)-γ], but not Th 2 cytokines (IL-4). Therefore, PCP can induce immune cell activation and is a potential candidate for the development of nutraceuticals to boost the immune system.

Cisplatin Suppresses Proliferation of Ovarian Cancer Cells through Inhibition Akt and Modulation MAPK Pathways (Cisplatin의 난소암 세포 증식 억제에 관한 신호 전달 기전)

  • Cisplatin (CDDP) is a chemotherapy agent used for patients with ovarian cancers. CDDP activates multiple signaling pathways, which causes various cellular reactions according to the type of cancer cells. Therefore, it is difficult to clearly conclude its signaling pathways. The purpose of this study is to determine the role of the signal protein of Akt/ERK1/2 and MAPK by CDDP-induced apoptosis in ovarian cancer cells (SKOV3). As a result, the number of apoptosis increased according to the TUNEL assay, and flow cytometric analysis confirmed that the percentage of sub-G1 early apoptosis was 8.73% higher than the control. The PARP and caspase-3 activity that appeared in the process of apoptosis was increased and the Bcl-2 expression was decreased. It was verified that the Akt and ERK1/2 activity was decreased, and p38 and JNK activity increased in a time dependent fashion. In conclusion, these results demonstrate that cisplatin inhibits the proliferation of ovarian cancer cells by inhibiting Akt activity and induces apoptosis by modulating the MAPK signaling pathway. However, a decrease in the ERK1/2 activity by CDDP was the opposite result to the result shown from the HeLa cells. For this reason, further research on signaling pathways is necessary. These results are expected to be useful for ovarian cancer treatment strategies targeting the MAPK pathway.

Changes of Lactulose Content during Heat Treatment of Milk (우유의 열처리 및 저장 조건에 따른 Lactulose의 함량 변화)

  • The aim of this work to determine the formation of lactulose during heat treatment process as a contribution to the estabilishment of limits of chemical indicators for different types of heat processed milk and analyze of lactulose for the reconstituted milk added samples. The HTST(75\^C/15s) and UHT(130\^C/2∼3s) treatment realized with a pilot plant and heat-treated samples were stroed at 4, 10, 30\^C for 4 weeks. Changes in lactulose was evaluated at 7 days intervals. The other heat treatment was sealed in glass tube and heated at 75\^C for 10 to 120s and heated at 130\^C for 2 to 60s in a thermostatically controlled constant temperature bath of glycerol. The reconstituted milk was made with full fat dry milk that reconstituted with deionized water to 10% total solid, and was added to milk at 10, 20, 30% respectively. The samples processed with a HTST pilot plant showed that lactulose was contained at 1.47∼1.52mg/10()ml and 8.19 ∼8.32mg/100ml for UHT-treated samples. Changes in the lactulose content of heat-treated samples during storage at 4 and 10\^C for 4 weeks caused a slight increase, however a noticeable increase was observed at 30\^C for 4 week. The glass tube samples showed that high correlations between relative increase in content of lactulose and increasing processing times(75\^C : r = 0.986, 130\^C : r = 0.987, respectively). Added with reconstituted milk would cause a increase of the lactulose content linear with increasing addition amount(r = 0.982). This results observed for lactulose in commercial milk samples would applied to the detection of chemical changes during heat treatment and illegal use of reconstituted milk.

Anisomycin, an Inhibitor of Protein Synthesis, Overcomes TRAIL Resistance in Human Hepatocarcinoma Cells via Caspases Activation and Bid Downregulation (Caspase 활성 및 Bid의 발현 저하를 통한 단백질 생성 억제제인 anisomycin의 인체간암세포에서 TRAIL 매개 apoptosis 유발의 활성화)

  • Anisomycin, also known as flagecidin, is an antibiotic produced by Streptomyces griseolus that inhibits protein synthesis by binding to the ribosomal 28S subunit. The tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a protein that induces apoptotic cell death. TRAIL primarily causes apoptosis in tumor cells by binding to death receptors. Many human cancer cell lines are refractory to TRAIL-induced cell death. In this study, we investigated whether anisomycin could enhance TRAIL-mediated apoptosis in TRAIL-resistant human hepatocarcinoma Hep3B cells. Treatment with anisomycin and TRAIL alone did not reduce cell viability in Hep3B cells. However, in the presence of TRAIL, the anisomycin concentration dependently reduced the cell viability. Our results indicate that anisomycin sensitizes Hep3B cells to TRAIL-mediated apoptosis and that this occurs, at least partly, via caspase activation. Interestingly, Bid knockdown by small interfering RNA significantly reduced the induction of apoptosis in combination with anisomycin and TRAIL, indicating that anisomycin effectively acts to lower the threshold at which TRAIL-mediated truncated Bid triggers the mitochondrial-mediated apoptosis program in Hep3B cells. Therefore, the use of TRAIL in combination with anisomycin might provide an effective therapeutic strategy for the safe treatment of some TRAIL-resistant cancer cells.

Growth Inhibitory Effect of Extracts of Propolis on Epithelial Ovarian Cancer Cells (상피성 난소암 세포에서 프로폴리스 추출물의 세포 증식 저해 효과)

  • Propolis is a natural product collected from plants by honey bees product used extensively in traditional medicine for its antioxidant, anti-inflammatory, immunomodulatory and anti-cancer effects. Propolis exhibits a broad spectrum of biological activities because it is a complex mixture of natural substances. Ovarian cancer is the second most common newly diagnosed cancer from all cancers among women in Korea and the leading cause of death from gynecological malignancies. While most ovarian cancer patients initially respond to surgical debulking and chemotherapy, patients later succumb to the disease. Thus, there is an urgent need to test novel therapeutic agents to counteract the high mortality rate associated with ovarian cancer. In this study, we investigated the anti-cancer properties and the active mechanism of Australian propolis in human epithelial ovarian cancer A2780 cells. Our data revealed that propolis showed a cytotoxic activity in a dose-dependent manner. Flow cytometric analysis for cell cycle arrest and apoptosis using propidium iodide staning and annexin V-FITC indicated that propolis could induce cycle arrest in the G0/G1 phase and apoptosis in a dose-dependent manner on human epithelial ovarian cancer cells. These results suggest that the Australian propolis is potential alternative agent on ovarian cancer prevention and treatment.

Studies on the Rheological Properties of Sugar Derivative Sweeteners (대체감미료 당유도체의 유변성에 관한 연구)

  • The rheological properties and food functionality of the novel sugar derivatives, fructo-oligosaccharide, high maltose syrup(HMS), maltitol and sorbitol were examined and compared to those of sucrose. All samples tested showed Newtonian fluid property at the concentration range of 10% to the original concentrated products containing 6981 solid. HMS showed the highest viscosity. The viscosity increased(r=0.8038) as the average molecular weight of sugar derivatives were increased. The viscosity increased exponentially as the concentration increased, and sugar alcohols had lower value of the exponent compared to HMS and fructo-oligosaccharide. The viscosity of sugar derivatives solutions decreased by the increasing temperature following the Arrhenius equation. The flow activation energies of sorbitol and HMS were higer than that of sucrose. Substitution of sucrose with fructo-oligosaccharide in apple jam processing did not change the textural characteristics, but in redbean jelly(yanggaeng) it reduced the hardness, adhesiveness, springiness and cohesiveness. When sucrose was 100% replaced by HMS, the texture of apple jam and redbean jelly was not changed, but by mixing sucrose and HMS 1 : 1 ratio, the hardness decreased substantially The sugar alcohols reduced the hardness, adhesiveness, springiness of apple jam and redbean jelly significantly. Addition of fructo-oligosaccharide and HMS to sucrose did not influence the solidifying rate of candy, but sorbitol, even at 10% addition, retarded the candy moulding.

Neuroprotective Effect of Rice with Phellinus linteus Mycelium in HT22 Cells (상황버섯균사체 쌀의 HT22 신경세포 보호 효과)

  • In this study, the protective effect of rice with Phellinus linteus mycelium (PLMR) against hydrogen peroxide-induced oxidative stress was assessed in a mouse hippocampal neuronal HT22 cell line through (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) salt (MTS) assay and western blot. MTS assay using HT22 cells showed that PLMR extract did not affect viability at a concentration range from 1 mg/mL to 5 mg/mL. However, at concentrations over 10 mg/mL, PLMR extract resulted in increased cell death. Cell viability of HT22 was significantly reduced by H2O2 treatment, and reduction of cell viability was efficiently restored by treatment with PLMR extract in a dose-dependent manner from 0.1 to 1 mg/mL. Cells treated with H2O2 showed increased expression of Bax, a pro-apoptotic protein, which was down-regulated by treatment with PLMR extract. On the other hand, cells treated with H2O2 resulted in reduced expression of Bcl-2, an anti-apoptotic protein, which was restored by treatment with PLMR extract. In addition, treatment with PLMR extract reduced expression of cleaved caspase 3 and PARP, which were up-regulated by H2O2 treatment. The results may suggest that treatment with PLMR extract would suppress H2O2-induced apoptosis of HT22 cells.

중서의결합치료소아백혈병사로(中西醫結合治療小兒白血病思路) -중서의 결합으로 소아백혈병 치료에 대한 접근-

  • 백혈병은 조혈계통의 악성 증식성 질환으로서 현재까지 주로 화학약물요법에 의존하고 있는 실정이다. 화학약물요법은 백혈병 세포를 소멸시킬 수 있지만 또한 인체에 여러 가지 독성 반응 및 부작용을 일으키기도 한다. 따라서 중서의결합으로 백혈병 치료에 접근하는 것은 중요한 의미를 갖는다. 중의학에서 소아 백혈병을 치료하는 경우 청열해독법(淸熱解毒法), 부정보허법(扶正補虛法), 활혈화어법(活血化瘀法)을 주로 사용한다. 청열해독법은 백혈병의 조기치료에 주로 활용되는데, 인체의 저항력을 증강시켜 화학약물요법을 실시하는 동안 흔히 나타날 수 있는 감염증상을 예방하는 효과를 얻을 수 있다. 부정보허법(扶正補虛法)은 주로 화학약물요법의 유도 완화기 및 치료효과의 유지를 위하여 활용되는데, 이는 인체의 면역력을 향상시켜 화학약물요법이 인체에 미치는 손상을 경감시킬 수 있다. 활혈화어법(活血化瘀法)은 미세순환을 개선시키는 작용을 하며 골수의 조혈기능을 촉진하고 면역기능을 조절하며 또한 일부 활혈화어제(活血化瘀劑)는 백혈병 세포에 직접적인 억제효과를 보인다. 소아백혈병에 대하여 화학약물요법을 진행하는 동안 중약을 같이 병행하는 경우 다음과 같은 과정으로 나누어 실시할 수 있다. 1. 유도완화치료(화학요법)단계: 이와 같은 치료과정은 대개 화학약물요법으로 인한 극심한 독성반응이나 주작용을 나타내게 되는데, 이 과정에서 중약치료를 병행하면 신속하게 증상을 개선시킬 수 있다. 만약 구토나 설사와 같은 소화계 부작용이 나타나면 화위강역법(和胃降逆法)을 활용하고, 감염 증상이 나타나면 부정(扶正)과 거사법(祛邪法)을 병행할 수 있다. 화학약물요법을 진행한 후 신체가 극도로 허약해지고 골수의 기능이 심하게 억제되는 경우는 주로 부정(扶正)시키는 중약을 사용하면서 익기양혈제(益氣養血劑)를 곁들이고 보조적으로 단삼(丹蔘), 당귀(當歸), 천궁(川芎), 계혈등(鷄血藤) 등과 같은 활혈화어제(活血化瘀劑)를 사용하여 골수의 조혈기능을 회복시킨다. 2. 치료효과의 유지단계: 본 과정에서는 중약치료에 있어서 부정(扶正)과 거사법(祛邪法)을 병행한다. 화학약물요법을 실시하는 동시에 거사제(祛邪劑)를 중용(重用)함으로써 화학약물요법의 효과를 강화시킨다. 화학약물요법이 끝난 뒤 부정(扶正)시키는 약물을 중용(重用)하여 인체의 면역기능을 증강시키고 백혈병세포를 억제시킨다. 3. 치료효과의 유지 및 강화단계: 치료효과의 유지단계에서는 변증논치(辨證論治)의 원칙에 입각하여 항암효과가 있는 중약을 활용할 수 있는데, 예를 들어 백화사설초(白花蛇舌草), 산자고(山慈?), 청대(靑黛), 용규(龍葵) 등을 사용할 수 있고, 육신환(六神丸)을 장기적으로 복용하여도 된다. 소아백혈병 치료에 있어서 중서의결합의 치료법을 활용하는 경우 다음과 같은 내용에 주안점을 둘 수 있다. 화학약물요법을 진행하는 과정에서 중약을 병행하여 투여하는 경우 사진합삼(四診合參)을 근간으로 종합적인 분석을 통하여 병인(病因)을 살피어 치료에 임하도록 한다. 약물의 선택과 처방의 구성은 반드시 변증논치(辨證論治)의 원칙하에 이루어져야 한다. 화학약물요법과 중약치료를 병행하는 과정에서 변증(辨證)과 변병(辨病)이 서로 결합되고 부정(扶正)과 거사법(祛邪法)을 병행하여 활용한다. 정체관(整體觀)에서 출발하여 환자를 관찰하는 동시에 특징적인 증후(證候)에 대한 변증논치(辨證論治)도 중요하며, 또한 백혈병 환자의 유형(類型)이나 임상 혈액검사 소견, 골수의 양상, 화학요법의 진행단계 및 환자의 연령과 체질 등을 충분히 가만하여 종합적인 분석을 토대로 치료법을 선택하여야 중약요법과 화학약물요법의 협동적인 효과를 증폭시키고 백혈병치료에 새로운 전기를 마련할 수 있을 것이다.
