Effects of Butyrate on Colorectal Cancer (대장암에 대한 butyrate의 효과)

  • Due to the high incidence of cancer and cancer-related mortality in Korea, there is increased concern and psychological anxiety about this disease, leading to increasing numbers of cancer studies. Despite these, the trend of the cancer incidence rate has shown a significant increase. The detection of colorectal cancer, which has a high incidence rate, often tends to be delayed, causing a high mortality rate. Therefore, the prevention of colorectal cancer has become an important emergent issue. The cause of this cancer has not been confirmed. However, it may be attributable to westernized dietary patterns, which include consuming a high quantity of red meat. Consumption of dietary fiber promotes the production of butyrate short-chain fatty acids by enteric bacteria. In the treatment of cancer, anticancer medications have been shown to lead to the apoptosis of tumor cells, and a strong relationship between apoptosis mechanisms of tumor cells and cancer treatment has been confirmed. The results of many studies have confirmed that butyrate can directly promote the apoptosis of colorectal cancer cells. Therefore, increased consumption of dietary fiber, which promotes the production of butyrate shortchain fatty acids, can be expected to have an effect on the prevention and treatment of colorectal cancer.

Antioxidative and Anticancer Activities of Julbernardia globiflora Extract in Human Colon Adenocarcinoma HT29 Cells (Julbernardia globiflora 추출물의 항산화 활성 및 인체 대장암 세포 HT29에 대한 항암 활성 분석)

  • Julbernardia globiflora, a tropical African tree widespread in Miombo woodland, has been used in folk medicine for the treatment of depression and stomach problems. However, the antioxidative and anticancer activities of J. globiflora remain unclear. The objective of this study is to evaluate the antioxidative and anticancer effects of methanol extract of J. globiflora (MEJG) and the molecular mechanism of its anticancer activity in human colon carcinoma HT29 cells. MEJG exhibited significant antioxidative effect with an $IC_{50}$ (concentration at 50% inhibition) value of $1.23{\mu}g/ml$ measuring by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay and inhibited cell proliferation in a dose-dependent manner in HT29 cells. We found that MEJG induced apoptosis of HT29 cells with the increase of apoptotic cells and apoptotic bodies using Annexin V staining and 4,6-diamidino-2-phenylindole (DAPI) staining, respectively. The MEJG treatment showed the increase of Fas, a death receptor, and Bax, a pro-apoptotic protein, and the decrease of Bcl-2, an anti-apoptotic protein, resulting in the release of cytochrome c from the mitochondria into the cytosol and activation of caspase-3, -8 and -9. The apoptotic effects of MEJG were confirmed by cleavage of poly (ADP-ribose) polymerase (PARP). Collectively, these results suggest that MEJG may exert the anticancer effect in HT29 cells by inducing apoptosis via both the intrinsic and extrinsic pathways.

  • Scleroderma is a connective tissue disease of unknown etiology, but known as a kind of auto-immune disease. It is most common in women especially in $30{\sim}50$ years, and very rare in childhood. It can be classified into two main classes : localized scleroderma & systemic sclerosis. Localized type has better prognosis, and usually involves skin only, or in some cases, the muscle below, except inner organs. Systemic type involves skin, oral mucosa and major internal organs. Involving facial skin, we can see small and sharp nose, expressionless stare and narrow oral aperture. Usually they have Raynaud's phenomenon, and in progress, show mouth opening limitation and sclerosis of tongue and gingiva. It is called CREST syndrome showing calcinosis cutis, Raynaud's phenomenon, esophageal dysfunction, sclerodactyly, and telangioectasia. Treatment of scleroderma is systemic and localized steroid therapy, use of collagen-link inhibitor (D-penicillamine), immune depressor and etc. Mouth opening limitation can be improved by mouth stretching exercise. We report a 6 years old boy, diagnosed with localized scleroderma who had mouth opening limitation. We could get additional mouth opening, and have done successful restorative treatment of mandibular and maxillary 1st and 2nd deciduous molar under deep sedation with nitrous oxide and enflurane.

The Relationship Between Loss of Blood Group Antigen A in Cancer Tissue and Survival Time in the Antigen A Positive Non-Small Cell Lung Cancer (A 항원 양성 원발성 비소세포폐암 조직에서의 A 항원 소실과 생존기간과의 관계)

  • Background : The moot important prognostic factor in non-small cell lung cancer is the TNM stage. Even after complete resection in early non-small cell lung cancer, the five-year survival rate is still low. However, new prognostic factors, including molecular biologic factors, have recently been found to guide the treatment of patients with non-small cell lung cancer. We evaluated the prognostic value of the loss of blood-group antigen A in tumor tissue, which has been implicated as an important prognostic factor for overall survival and the timing of the disease progression. Methods : The loss of blood-group antigen A was assessed immunohistochemically in paraffin-embedded tumor samples from 26 patients with blood types A or AB, who had undergone curative surgery. Monoclonal antibody was used to detect the blood group antigen A expression. Results : Fifteen patients (58%) expressed antigen A in their tumor tissue, whereas 11 patients (42%) did not show antigen A. The median survival time of the blood A antigen positive group was 11 months, while the median survival time of the blood A antigen negative group was 18 months. The difference in survival between the two groups was not statistically significant. Conclusion : The loss of blood-group antigen A in tumor tissue was not found to be a significant prognostic factor in patients with non-small cell lung cancer. This study needs to be extended for further evaluation.

  • Leflunomide is a new disease modifying anti rheumatic drug (DMARD) for the treatment of active rheumatoid arthritis. Its mechanism of action differs from other DMARDs in that it inhibits the de novo pyrimidine synthesis by inhibiting dihydroorotate dehydrogenase and therefore prevents the proliferation of activated lymphocytes. As it has been prescribed worldwide, there is a great deal of much concerns regarding its potential adverse effects. Because leflunomide has an active metabolite with a long elimination half-life of approximately 2 weeks, serious adverse reactions may occur even after the leflunomide treatment has been stopped. The profile of serious reactions includes liver dysfunction, hematological disorders, severe skin reactions and respiratory dysfunction. Respiratory dysfunctions with leflunomide therapy are very rare and its incidence is lower than that of methotrexate therapy. However, there are reports in Japan showing that 5 patients died of interstitial pneumonitis and another 11 patients developed serious lung complications associated with leflunomide. This suggests the possibility of fatal respiratory toxicity of leflunomide. There are no reports of interstitial pneumonitis associated with leflunomide in Korea. We report a case of a 62-year old woman who developed interstitial pneumonitis, which might have been induced by leflunomide during the treatment of rheumatoid arthritis.

Antitumoral, Antioxidant and Antimicrobial Activities of Solvent Ftactions from Grifola umbllatus (저령추출물의 항암, 항산화 및 항균효과)

  • Grifola umbellatus was extracted using methanol, and the extract was further fractionated by water and ethyl acetate. Assay of each fraction with MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium-bromide] revealed significant cytotoxicity effect of the methanol extract of Grifola umbellatus against human gastric cancer cell but not normal human lymphocytes. The methanol extract showed the highest antioxidant activity as well. Antimicrobial activity of Grifola umbellants against Helicobacter pylori was higher in method extract than in other fractions. Grifola umbellatus had a significant inhibitory effect on Helicobacter pylori reducing both its growth and urease activity. These results show that the methanol extract of Grifola umbellatus possesses therapeutic potential on gastric diseases.

Preparation of Positively and Negatively Charged Carbon Nanotube-Collagen Hydrogels with pH Sensitive Characteristic (양전하와 음전하를 띄며 pH 감응성인 카본나노튜브-콜라젠 Hydrogel의 합성)

  • In this study, preparation of positively and negatively charged carbon nanotube (CNT)-collagen (CG) hydrogels with pH sensitive characteristic was reported. The positive and negative characteristics of the prepared hydrogels were created by introduction of positively functionalized CNT-NH2 and negatively functionalized CNT-COOH, respectively, into the collagen hydrogel. The surface charge of CNTs (CNT-NH2 and CNT-COOH), CG and CNTs/CG hydrogels was measured by Zetasizer. The swelling ratios of CNT-NH2/CG and CNT-COOH/CG hydrogels in aqueous solution were checked by measuring of weight changes of the hydrogels in the range of pH 2~10. In detail, the positively charged CNT-NH2/CG hydrogel swelled up to 5% at pH 4 in comparison to the weight at pH 7, while the negatively charged CNT-COOH/CG hydrogel swelled up to 10% at pH 10. The prepared CNT-NH2/CG and CNT-COOH/CG hydrogels will be very useful as pH sensitive oral drug-delivering systems for gastrointestine (pH ~2) and small intestine (pH ~9), respectively.

Combined Therapy with Carboplatin and Meloxicam for Oral Squamous Cell Carcinoma in a Dog (구강 편평 세포암 이환견에서 Carboplatin과 Meloxicam의 병용투여 증례)

  • A 16-year-old, spayed female, Maltese dog weighing 4.3 kg was presented with a 2-week history of yellowish nose-and oral-discharge, anorexia and lethargy. In physical examination, indefinite ulcerative mass about $3{\times}3$ cm in size in the left sublingual region, severe gingivoperiodontitis, halitosis, moderate dental calculus, fever and swelling of mandible soft tissue were noted. In radiographic view, periodontitis and bone lysis findings on the left rostral part of maxilla and mandible were observed. In complete blood count and blood chemistry test, thrombocytosis, increased levels of serum $NH_3$, AST and ALP were obtained. Urinalysis revealed hematuria and proteinuria. In histological examination, squamous cell carcinoma was diagnosed. No surgery was performed, but combined treatment with carboplatin and piroxicam was initiated. Five days after initiation of medication, because of insistent vomiting, piroxicam was substituted to meloxicam. Although the therapeutic efficacy of meloxicam could not be known; administration of meloxicam might be less potential to gastrointestinal side-effects than piroxicam. The dog totally received 3 times of carboplatin infusion. Five days after third infusion of carboplatin, the dog was hospitalized with severe lethargy, vomiting and bloody diarrhea. In examination, severe renal failure signs were noted, and the dog was euthanized.

An histochemical study of endocrine cells in the alimentary tract of the red-eared slider, Trachemys scripta elegans (붉은귀거북이 소화관 내분비세포에 관한 조직화학적 연구)

  • The regional distributions and relative frequencies of endocrine cells were studied histochemically (Grimelius and Masson-Hamperl (M-H) silver methods) in the alimentary tract of the red-eared slider, Trachemys scripta elegans. Samples were taken from the esophagus, fundus, pylorus, duodenum, jejunum, ileum and large intestine. Argyrophil (Grimelius-positive) cells and argentaffin (M-H-positive) cells were found in the whole alimentary tract in this study. Spherical to spindle and/or oval to round-shaped argyrophil or argentaffin cells were located in the gastric glands of the stomach regions, in the basal portion of the epithelium of intestinal tract or the esophagus with variable frequencies. Argentaffin cells were more numerously detected in the whole alimentary tract compared with those of argyrophil cells in this study. Argyrophil cells were observed in the whole alimentary tract including the esophagus and the most predominant region was the rectum with moderate frequency. The relative frequency of these cells was rare in the esophagus, fundus, duodenum, jejunum and ileum, respectively and a few frequency in the pylorus. Argentaffin cells were also observed in the whole alimentary tract including the esophagus and the most numerously demonstrated region was the rectum with numerous frequency. They were observed with a few frequencies in the remaining regions of the alimentary tract except for the rectum, respectively. However, to know the exact type of the argyrophil cells and argentaffin cells that were observed in this study, more developmental methods such as immunohistochemistry were needed.

Link between Periodontal Disease and Cancer: A Recent Research Trend (염증-치주 질환과 암에 관한 최근 연구 동향)

  • The multifaceted role of chronic inflammation in multistep carcinogenesis has been extensively investigated and well documented. Periodontal diseases are associated with multifactorial agents, including bacterial endotoxins and the generation of an inflammatory response, indicating that poor oral health is associated with a variety of systemic diseases. The association between poor oral health, chronic inflammation, smoking, and increased alcohol consumption as risk factors for tumorogenesis is well established. More recently, associations between oral health and tooth loss and gastric, lung, and pancreatic cancers have been explored, with some studies pointing to smoking and oral health as a common link with an increased risk for malignant disease. In addition, epidemiological studies consistently indicate increased risks of various cancers with periodontal disease or poor oral condition caused by oral bacteria, which may activate alcohol- and smoking-related carcinogens locally or act through chronic inflammation. Appropriate oral care is vital in preventing cancer, as well as many other diseases. Thus, research on the correlation between oral care and periodontal inflammation and cancer is required. This review highlights the association between oral health and the risk of certain malignancies, such as periodontal disease-associated chemoprevention of inflammation" in this sentence.
