Functionalization of Electrospun Nano/Micro-fibrous Scaffolds Using Gamma-ray Irradiation (감마선 조사법을 이용한 전기방사 나노/마이크로 섬유 지지체의 표면 기능화)

  • In tissue engineering application, a fibrous structure of scaffolds has been issued as an alternative system to regulate cell survival and tissue regeneration, and electrospinning technique has been popularly used to generate fibrous meshes or sheets mimicking the structure of native extracellular matrix (ECM). However, recent strategy in the scaffold development is expanded to provide the structural property as well as a biological property of native ECM, a variety of surface modification techniques have been used to introduce biological property. In this study, we developed biomimetic poly(L-lactide-co-${\varepsilon}$-caprolactone) (PLCL) nano- and micro-fibrous scaffolds as a unique platform with structural and biological properties with native ECM using electrospinning method and gamma-ray irradiation. Surface morphology of the scaffolds was observed by scanning electron microscopy, and alteration of surface property was evaluated with toluidine blue O staining, water contact angle measurement and ATR-FTIR analysis.

Physiological Responses of Rice by Acute and Chronic Gamma Irradiation (방사선 완·급조사에 따른 벼의 생리적 영향 평가)

  • We investigated the physiological responses by acute and chronic gamma-irradiation in rice. The rice, Oryza sativa L. cv Dongan, plants were irradiated with 100 and 400 Gy of gamma-rays for acute and chronic irradiation, and their morphological, chlorophyll content, MDA, proline and activities of antioxidant enzymes were examined. The plant height of chronic irradiation samples were decrease with increase of a does than the control, but the number of tiller and dry weight of shoot were increased 100 Gy. Carotenoid and chrolorphyll content were decreased of all irradiated plants than non-irradiated plants. But, MDA and proline content were increased in 400 Gy both acute and chronic gamma irradiation. And the activities of antioxidant enzymes were different as gamma-irradiation patterns.

Effect of Gamma Irradiation on the Splenocyte Proliferation and Cytokine Production of Chaga Mushroom Hot Water Extract (방사선 조사된 차가버섯 열수 추출물의 비장세포 증식능 및 사이토카인 발현에 미치는 영향)

  • This study investigated the effect of gamma irradiation on immune enhancing activity of Chaga mushroom extract (CME). CME was prepared by hot water extraction at $70^{\circ}C$ for 4 hours and lyophilized. Lyophilized CME powder was dissolved with deionized water at $10mg\;ml^{-1}$ and then irradiated at the doses of 10, 30 and 50 kGy by cobalt 60 gamma irradiator. The gamma-irradiated and non-irradiated CME were treated into the splenocyte separated from mouse. Cell proliferation and cytokine production of the immune cells were increased by gamma-irradiated CME and these increases were more prominent when CME was irradiated at higher doses. Therefore, it is considered that gamma irradiation can be an effective method for improvement of the immunomodulating activity Chaga mushroom extract.

Synthesis of 18F Labeled Clotrimazole Derivatives as a Potential PET Imaging Agent (18F을 표지 암 영상용 클로트리마졸 유도체의 합성)

  • Clotrimazole [1-{(2-chlorophenyl)-diphenylmethyl}-1H-imidazole, CLT] has been reported to inhibit the proliferation of vascular endothelial and act as an in vitro anti-VEGF drug. It is also shown to inhibit angiogenesis in an animal model. The radioisotope labeled clotrimazole derivative can be utilized to monitor the physiologic processes of cancer. In this study, we synthesized [$^{18}F$]fluoride labeled clotrimazole derivatives as a new tumor imaging agent for PET. The references were prepared by a refluxing with clotrimazole and an excess of fluoroalkyltosylate in acetonitrile for 36 h and clotrimazole reacted with ditosylalkane to give precursors. [$^{18}F$]Fluoride labeled reaction was performed with precursor in Kryptofix[2.2.2]/$K_2CO_3$ for 10 min at $80^{\circ}C$. The radiolabeling mixture was passed through a silica Sep-Pak cartridge to remove $^{18}F^-$. The [$^{18}F$]F-clotrimazole derivatives were synthesized with a 20~25% yield. In the radiofluorination step, we used acetonitrile and DMSO as a solvent and observed a higher yield at the acetonitrile (25%) reaction compared with the DMSO reaction (5%).

Effect of Gamma-Irradiation on the Cell Proliferating and Interleukin-2 Producing Activity of Mouse Splenocytes of Staphylococcal Enterotoxin B (감마선 조사가 Staphylococcal Enterotoxin B의 비장세포 증식률 및 Interleukin-2 분비능에 미치는 영향)

  • The purpose of this study was to investigate the cell proliferating and interleukin-2 producing activity of staphylococcal enterotoxin B by gamma-irradiation. Staphylococcal enterotoxin B was gamma-irradiated with the various doses of 0, 2, 20 and 50 kGy. SDS-PAGE analysis showed that gamma-irradiation caused the sharp decrease of the content of staphylococcal enterotoxin B and the effect was irradiating dose-dependent. Non-irradiated staphylococcal enterotoxin B increased the cell proliferation of splenocytes isolated from female Balb/c mouse, whereas 2 kGy-irradiated toxin significantly decreased the activity. 20 and 50 kGy-irradiated staphylococcal enterotoxin B was no effect. A similar effect on the interleukin-2 production of mouse splenocytes was observed with non-irradiated and irradiated staphylococcal enterotoxin B. It was considered to be due to the decrease of the antigenicity of staphylococcal enterotoxin B by gamma-irradiation. Therefore, these results suggest that gamma-irradiation can be effective for the decrease of the antigenicity of staphylococcal enterotoxin B as superantigen.

Radiation Preparation and Characterization of Antimicrobial Hydrogels (방사선 이용 항균성 천연추출물 함유 하이드로겔 제조 및 특성 연구)

  • Taraxacum platycarpum (TP) has been used for years without restriction on usual dose for its non-toxic nature and nonexistence of the side effects. To develop antimicrobial hydrogel, poly (vinylalcohol) (PVA) hydrogels containing the aqueous extracted TP as an antimicrobial agent were prepared by using gamma-rays irradiation. The antimicrobial activities of the TP hydrogels were tested against Staphylococcus aureus and Staphylococcus epidermidis by disc diffusion method. The inhibition zones (IZ) of the TP extracts and TP hydrogels against S. aureus were 16 mm and 20 mm and against S. epidermidis was 10 mm and 13 mm, respectively. In conclusion, the TP hydrogel that has an excellent antimicrobial activity was proved to be a valuable material for functional skin care.

Isolation of Cesium and Radiation Resistance Bacteria for Bioremediation (생물정화를 위한 세슘 및 방사선 저항성 세균의 분리)

  • The global problem of handling radioactive materials is facing limitations. Eco-friendly bioremediation methods using microorganisms are being studied. This study was conducted to screen cesium-resistant microbial strains. M1 strain was selected from the soil sample by enriched culture in R2A medium containing 100 mM CsCl. In liquid medium containing above 40 mM of CsCl, the growth of M1 was inhibited in a concentration-dependent manner. Otherwise, M1 can survive up to 80mM CsCl in solid medium although the growth rate was slow and colony size was small. M1 strain was genetically identified as a strain of the genus Acinetobacter through 16S rRNA sequencing, and radiation resistance (D10 value) of M1 was found to be 0.307 kGy. These results showed that M1 strain is highly resistant to cesium and can grow in radiation environment. It was considered that M1 strain is useful in the field of biological decontamination of cesium.

  • 한국 원자력연구소(소장 윤용구)는 73년도 구구개편으로 정부조직으로부터 분리하여 비영리 법인으로 새 출발한지 제 3차년도로 접어들게 되었다. 이 연구소는 운영의 자율성과 안정성을 갖추고 연구에 전념하고 있다. 1975년의 슬로건을 「획기적인 실천의 해」로 내건 동연구소의 기본방침과 사업계획을 살펴본다.
