  • 연구목적 : 수심 깊이에 따른 수중운동이 하지근력에 미치는 영향을 알아보고자 하는데 목적이 있다. 연구방법 : 신경근골격계 질환이 없는 여자 10명을 대상으로 하였고, 무작위로 수성 0.7m 그룹과 1.4m 그룹으로 배치하였다. 수중운동은 1회 40분, 주 3회 6주 동안 적용하였다. Ultrasound MyLab25(Esaote, Italy)를 이용하여 하지근육의 우모각과 밀도를 실험전과 후에 측정하였다. 연구결과 : 골격근 구조의 역학적 특성을 나타내는 우모각 변화를 비교한 결과 0.7m 그룹의 오른쪽 넙다리곧은근과 왼쪽 넙다리곧은근, 앞정강근 그리고 장딴지근에서, 1.4m 그룹의 오른쪽 앞정강근과 장딴지근, 왼쪽 넙다리곧은근에서 통계학적으로 유의한 차이가 나타났으나(p<.05), 수심 깊이에 따른 유의한 차이는 없었다(p>.05). 또한, 질적 특성을 나타내는 근육 영상 밀도 변화를 비교한 결과 0.7m 그룹의 오른쪽 넙다리곧은근, 장딴지근과 왼쪽 넙다리곧은근, 앞정강근, 장딴지근에서 1.4m 그룹의 양쪽 넙다리곧은근, 앞정강근, 장딴지근에서 통계학적으로 유의한 차이가 나타났고(p<.05), 특히 1.4m 그룹에서 현저한 증가를 보였으나 수심 깊이에 따른 유의한 차이는 없었다(p>.05). 결론 : 본 연구에서는 수심 차이에 관계없이 수중운동 후 근밀도와 우모각에 영향을 미치는 것을 확인할 수 있었다.

Production of a Keratinolytic Protease by a Feather-Degrading Bacterium, Bacillus megaterium F7-1 (우모분해세균 Bacillus megaterium F7-1에 의한 Keratinolytic Protease의 생산)

  • Bacillus megaterium F7-1 producing keratinolytic protease was isolated from decayed chicken feather. The optimal culture conditions for the production of keratinolytic protease by B. megaterium F7-1 were investigated. The composition of optimal medium for the keratinolytic protease was 0.2% glucose, 0.8% skim milk, 0.05% NaCl, 0.01 % $(K_2HPO_4$, 0.02%, $(KH_2PO_4$ and 0.01 % $MgCl_2$. Especially, skim milk was found to be the most effective compound in keratinolytic protease production. The optimal temperature and initial pH were 6.5 and $25^{\circ}C$, respectively. The keratinolytic protease production under optimal condition reached a maximum of 269 U/ml after 5 days of cultivation. B. megaterium F7-1 degraded 98% of the feather used in the optimized medium within 6 days.

Characterization of Protease Produced by Elizabethkingia meningoseptica CS2-1 and Optimization of Cultural Conditions for Amino Acid Production (닭 우모 분해세균 Elizabethkingia meningoseptica CS2-1이 생산하는 단백질분해효소의 특성 및 아미노산 생산을 위한 배양조건)

  • A feather-degrading bacterium Elizabethkingia meningoseptica CS2-1 was isolated from compost in a chicken farm. Cultured on a basal medium containing 2% chicken feather, the bacterium showed 729.7 ${\mu}mol/mL$ of amino acid. Optimal culture conditions for feather degradation by E. meningoseptica CS2-1 were $25^{\circ}C$, pH 7.5, and 180 rpm. The optimal pH and temperature for protease activity were 8.0 and $40^{\circ}C$, respectively. The composition of an optimal medium for amino acid production was 0.05% NH4Cl, 0.05% NaCl, 0.03% $K_2HPO_4$, 0.03% $KH_2PO_4$, 0.01% $MgCl_2{\cdot}6H_2O$, 0.1% urea, and 2% chicken feather. Characteristics of amino acids extracted from the optimal medium under the optimal culture conditions of E. meningoseptica CS2-1 were analyzed. The total amino acid content of strain CS2-1 was 1063 ${\mu}mol/mL$, which was 46% higher compared to the basal condition (729.7 ${\mu}mol/mL$). The essential amino acid content in the total amino acid was 315.9 ${\mu}mol/mL$, which was 44% higher than that of the basal condition. Major amino acids were proline (14%), aspartic acid (12%), glutamic acid (11%), serine (10%), alanine (10%), glycine (9%), and tyrosine (7%) by strain CS2-1. These results suggest that strain CS2-1 can be used as a potential microbial resource for the production of amino acid using chicken feathers.

Nematicidal Effect of Root-Knot Nematode (Meloidogyne incognita) by Amino Acids Biochemical Agent Extracted from Chicken Feather (닭 우모로부터 추출한 아미노산 생화학제의 고구마뿌리혹선충 증식억제 효과)

  • This study examined the control effects of amino acid biochemical agent extracted from chicken feathers on Meloidogyne incognita for the purpose of developing an environmentally friendly nematicidal agent that can replace chemical control of root-knot nematodes (RKN). We investigated the lethal effects of J2 juveniles for 19 types of commercial amino acids. As a result, five kinds of amino acids (L-asparagine, L-aspartic acid, L-methionine, L-tyrosine, L-cysteine) showed mortality rate of more than 50% at a concentration of 50 mM. L-asparagine showed the highest mortality rate at 94%. We also investigated the lethal effect of J2 juveniles and suppressive effects of egg hatching by feather amino acids (FAA) biochemical agent. It showed that the mortality rate of J2 juveniles was more than 80% and suppression rate of egg hatching was 74% at 1/50 concentrations of FAA. As a result of conducting a tomato pot culture experiment for 60 days after treating 1/50 concentrations of FAA biochemical agent in rhizosphere soil, it showed that the control effects were 63% of juveniles density in the soil, 59% of egg mass and 61% of root gall index, respectively. Based on the above results, it is considered that the FAA biochemical agent extracted from chicken feathers can be used as an environmentally friendly nematicidal agent of RKN.

백혈병 마렉병을 중심으로 한 최근의 계병대책

  • 1. 마렉 감염계는 환경을 오염한다. 3. 감염원인 병독은 오랫동안 생존한다. 3. 따라서 감염성계는 오염환경에서 직접, 간접 간염한다. 이 병의 감염경로는 아직 확정되어 있지 않으나 주로 기도감염일 것으로 인정되고 있다. 4. 본 병의 감염은 병독 보유계의 우모근이 각화하여 탈각하는 상피세포중에 함유되는 병원성 병독이 야기 한다는 것은 확정적이다. 또 이 방법이 마렉병 감염을 반복하는 유일의 원인으로 생각하고 있다. 5. 본 병의 개란전파에 대해서는 거의 없는 것으로 알고 있으나 Sevoian은 전달의 가능성이 있다고 한다. 6. 임파종증일때와 마찬가지로 저항성품종과 감염성품종을 선택 도태를 함으로써 만들어져 있다. 이들은 마렉병에 대하여 저항성품종은 5$\%$의 발병성을 나타내고 감염성 품종은 90$\%$의 발병성을 나타내고 있다.

  • The development of protein utilization with keratin(cattle hair and human hair) have heen investigated. The include a) conditioning of hydrolysis of keratin, b) isolation of crude protein, c) pepsin HCl digestibility of isolated protein, d) the amino acid composition of isolated protein and amino acid concentrate. The results are obtained as follow ; 1. Human hair and cattle hair were softened and hydrolysed with hot dilute alkali(especially 1% NaOH) 2. Crude protein were isolated from hydrolysed keratin by means of different solubility and isoelectric point. 3. The protein isolated was of good quality on the point of color, protein content and pepsin digestibility. 4. The amino acid concentrate obtained by acids processing was of good quality on the point of color, taste and amino acid composition.

Isolation and Characterization of Feather Keratin-Degrading Bacteria and Plant Growth-Promoting Activity of Feather Hydrolysate (우모 케라틴 분해세균의 분리, 특성 및 우모 분해산물의 식물 생육촉진 효과)

  • This study was conducted to isolate and characterize a novel feather-degrading bacterium producing keratinase activity. A strain K9 was isolated from soil at poultry farm and identified as Xanthomonas sp. K9 by phenotypic characters and 16S rRNA gene analysis. The cultural conditions for the keratinase production were 0.3% fructose, 0.1% gelatin, 0.04% $K_2HPO_4$, 0.06% $KH_2PO_4$, 0.05% NaCl and 0.01% $FeSO_4$ with an initial pH 8.0 at $30^{\circ}C$ and 200 rpm. In an optimized medium containing 0.1% chicken feather, production yield of keratinase was approximately 8-fold higher than the yield in basal medium. The strain K9 effectively degraded chicken feather meal (67%) and duck feather (54%), whereas human nail and human hair showed relatively low degradation rates (13-22%). Total free amino acid concentration in the cell-free supernatant was about 25.799 mg/l. Feather hydrolysate produced by the strain K9 stimulated growth of red pepper, indicating Xanthomonas sp. K9 could be not only used to increase the nutritional value of chicken feather but also a potential candidate for the development of natural fertilizer applicable to crop plant soil.

Effects of Heat Treatment of Three Animal by-products on Ruminal Degradation Characteristics and Intestinal Availability of Crude Protein (동물성 부산물 사료 세 종류에 대한 열처리가 조단백질의 반추위내 분해특성 및 하부장기내 이용성에 미치는 영향)

  • In order to investigate the effects of heat treatment of three animal by-products(feather meal, tallow meal, viscera meal) on in situ ruminal degradation characteristics and gastrointestinal availability of dietary crude protein(CP), three ruminally and duodenally cannulated dry Holstein cows were employed. Cows were fed a diet containing 60% concentrate and 40% orchard grass hay, and had free access to water and mineral block. Experimental feeds were processed for 4 hr at 149$^{\circ}C$ in a forced-air oven, and were passed through a 1-mm screen. Degradation kinetics of feed protein in the rumen were fitted to an exponential type model, and intestinal availability was estimated by the mobile nylon bag technique. Effective CP degradabilities in the rumen for feather meal, tallow meal and viscera meal were 30.2%, 75.0% and 56.4% at 5% passage rate per hour(k=0.05), respectively. In addition, heat treatment increased effective ruminal CP degradability on feather meal and viscera meal treatments, whereas decreased in tallow meal treatment(P$<$0.05). Gastrointestinal CP disappearances of feather meal, tallow meal and viscera meal were 56.2%, 18.6%, and 37.9%, respectively. In addition, heat treatment decreased the gastrointestinal CP disappearance on feather meal and viscera meal treatment, but increased in tallow meal treatment(P$<$0.05). Intestinal availability of rumen undegradable protein(A-UDP) was 80.4% for feather meal, 83.8% for tallow meal and 86.9% for viscera meal. In addition, heat treatment increased A-UDP on feather meal and tallow meal treatment, 94.0% and 91.3%, respectively, but decreased on viscera meal treatment, 76.5%(P$<$0.05).

The Effects of Dietary Supplementation of Feather Meal Digests on the Performances and Muscular Taurine Contents in Broiler Chickens (육계 사료내 우모 Digest 첨가시 생산성과 계육내 Taurine 함량에 미치는 영향)

A total of 100 broiler chickens were assigned to five dietary treatments; Control, PM diet(FM), NaOH treated FM diet(NaOH-FM), HN03 treated FM diet(HN03 - FM) and 0.5% synthetic taurine supplemented diet(Taurine). Treated diets were supplemented with FM or FM digests at the level of 5% to the control diet Treated diets were fed during the last 3wks of 6wks feeding. During the finisher period, weight gain of chicks fed FM treatments tended to decrease in general. Feed intakes of FM and Taurine were significantly lower than the control but were not significantly different from NaOH - FM or HN03- PM. Feed conversion rate(feed intake/gain) of NaOH-FM was significantly higher than those of FM and HNOJ - FM but not different from those of the control and Taurine. Taurine content of the control was highest in heart muscle(1482 J1.g/g) followed by leg muscle(778f.lg/g) and breast muscle(79 J1.g/g). Taurine contents of leg and breast muscle were significantly(P< 0.01) affected by treatments but that of heart muscle was not. Taurine content of Taurine treatments was highest in both leg and breast muscle. Taurine content of leg muscle was increased 170"10 by Taurine supplementation, 123% by FM supplementation, 122% by NaOH - FM supplementation and 63% by HNOJ- FM supplementation compared with control. Taurine content of breast muscle of Taurine treatment was 246% higher than the control, but other treatments were not different from the control. In conclusion, supplementation of feather meal at 5% level of broiler diet can enrich taurine content of leg muscle. Chemical treatments of FM were not effective in improving taurine enrichment of broiler meat.

Cultural Conditions for Pretense Production by a feather-Degrading Bacterium, Bacillus megaterium F7-1 (우모분해세균 Bacillus megaterium F7-1에 의한 단백질 분해효소 생산에 영향을 미치는 배양조건)

  • The effects of inorganic salts and feather concentrations on pretense production by Bacillus megaterium F7-1 were investigated. Pretense production was dependent on the presence of phosphates in the medium. Supplementation of medium with calcium ion slightly increased protease production. The highest protease production was obtained at $1.4\%$ feather. The optimal medium contained $2.0\%$ glucose, $0.8\%$ skim milk, $0.06\%\;K_{2}HPO_{4}\%,\;0.04\%\;KH_{2}PO{4},\;0.06\%\;NaCl,\;0.03\%\;MgCl_{2}\cdot6H_{2}O,\;0.002\%\;CaCl_{2}\cdot2H_{2}O,\;and\;1.4\%$ whole feather. By using this optimized medium, increased production of the protease was achieved compared with the cases of using basal medium.
