We investigate the effect of enzyme pretreatment using protease, carbohydrase, and lipase on improvement of sludge treatment efficiency by measuring SCOD and TCOD. The enzyme-pretreatment increases SCOD of excess sludge. In addition, the amount of sludge reduction during digestion, in terms of SCOD and TCOD, are enhanced by enzyme-pretreatment. Among pretense, carbohydrase, and lipase, pretense showed the best enhancement of the sludge treatment efficiency. Sludge digestion followed by ozone and enzyme treatments showed more effective sludge treatment when compared with ozone treatment alone. Therefore, we expect that enzyme pretreatment can be used as a useful tool for enhancing the sludge treatment efficiency.

To assess the environmental risks of transgenic chili pepper with PepEST gene on non-target organisms before it exposes to the agro-ecosystem environments, we conducted the three sets of green peach aphids (Myzus persicas S.) life table experiment under laboratory conditions (Temp. $25^{\circ}C$, R.H. 50-70%, Photoperiod L16 : D8) in series during 2005-2006. We measured the net reproductive rate ($R_0)^*$, the intrinsic rate of increase ($r_m$), the mean generation time ($T_c$), $fecundity^*$, life span, and reproduction period between non-transgenic chili peppers and transgenic chili peppers, respectively. The life span of green peach aphids from three sets was 31, 27, 25 days, and the period of life span was similar to the general average length of green peach aphids, 25-29 days. Although the first reproduction of transgenic pepper was similar to the non transgenic pepper (P>0.05), the fecundity and the net reproductive rate ($R_o$) by using Jackknife method of transgenic pepper were lower than those of non transgenic pepper (P<0.05). Conclusively, we observed the adverse effect from our results but we should execute further experiments to confirm the results at the fields with the similar way.

In this study, antioxidative effects of the extracts of different species and flowering periods of Inula britannica were investigated. According to the free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity of the extracts, The I. britannica var. chinensis flower extract (500 ${\mu}g/mL$) was measured in a 79.89% free radical scavenging activity, but the flower extracts of similar species (I. britannica var. linariaefolia Regel, I. britannica var. ramosa, I. salicina var. asiatica) did not show any effect on the free radical scavenging activity. The effects of the free radical scavenging activity of I. britannica var. chinensis flower extracts were exhibited in the order of full bloom (93.68%), bud (43.28%), and fallen blossom (14.11%). Next, we established optimum condition of extract solvent, temperature, extraction time. The extract from ethanol at $60^{\circ}C$ showed the most free radical scavenging activity among other conditions and extraction time not relevant in free radical scavenging activity. The protective effects of the extract of I. britannica var. chinensis flower on the photohemolysis of human erythrocytes by using rose bengal were increased in a concentration-dependent manner (5 ~ 50 ${\mu}g/mL$). In particular, the extract in 50 ${\mu}g/mL$ concentration exhibited better protective activity (${\tau}_{50}$ = 116.1 min) than (+)-${\alpha}$-tocopherol (${\tau}_{50}$ = 73.44 min), which is a known lipophilic antioxidant. Principle component of I. britannica var. chinensis flower was identified as quercetin of flavonoids by high-performance liquid chromatography (HPLC). These results indicate that the extract of I. britannica var. chinensis flower can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging free radical and $^1O_2$, and protect cellular membranes against ROS. It is concluded that the antioxidative effects of the extract of I. britannica var. chinensis flower could be applicable to functional cosmetics.

Several tartaric acid-degrading bacteria were isolated from Korean grape wine pomace after enrichment culture at $30^{\circ}C$ for 10 days in liquid media containing tartaric acid Among them, strains KMBL 5777 and KMBL 5778 exhibited the highest level in the growth and tartaric acid degradability in a medium containing 0.2%(w/v) tartaric acid as a sole carbon source. They were identified as Acetobacter tropicalis based on their morphological and physiological characteristics as well as their 16S rDNA sequences. Blast search of the 16S rDNA sequences revealed that the isolated strains are closest to Acetobacter tropicalis. Homologies of the sequences of KMBL 5777 and KMBL 5778 were 96.0 and 98.9%, respectively with those of A. tropicalis LMG 1663. Both the two bacteria showed higher tartaric acid degradation at $25^{\circ}C$ that those at 20 and $30^{\circ}C$. They could degrade tartaric acid at a wide range of pH between 4.0 and 7.0 with the most rapid degradability at pH 7.0. However, when the bacteria were grown for 8 days, the same level of tartaric acid degradation was observed at pH 4.0, 5.0, 6.0 and 7.0, which was 90.0% of degradation of the acid.
Despite the fact that understanding customers satisfaction with transportation services is a subject of great importance, authors, so far, found no systematic researches referred to that issue. From this point, studying the satisfaction with subways services can be extremely useful. Empirical study of key factors in the satisfaction with subway services is the departure point, which holds as objectives, and we believe, will contribute to overall increasing in the number of subways services used and in the amount of public benefits derived from that usage. In order to achieve these goals: First, several items referred to some key factors in the satisfaction of subway usage were systemized. Second, a research of specific weights attached to those key factors by subway passengers was conducted. Knowledge of the satisfaction variables system can provide deep insights into ones perceptual experience when using a subway. The results were as follows: Various interrelated factors compose a passengers satisfaction with subway services. People do not just use subway passively; a number of key factors, like physical and personal services, exact timing, easiness to access etc. determine the passengers satisfaction with subway. In order to find out specific weights of these key factors multiple regression analysis was employed. Results showed that satisfaction with subway is determined by (in order of importance) easiness to access, quality of physical services, friendliness of working stuff and timing exactness. According to the findings, passengers do not use subway as a simple mean of transportation, rather they perceive it as a complex combination of environmental elements and overall satisfaction depends on these various factors. Therefore, to learn passengers satisfaction with subways services, passengers subway experience must be thoroughly studied and analyzed, and this is where papers value resides.
To find the preliminary environmental conditions for a short-time transport of living olive flounder, Paralichthys olivaceus, the stress response and physiological metabolic activity of the cultured fish to feed deprivation and slow temperature descending ($15.8^{\circ}C{\rightarrow}13.3^{\circ}C$) were monitored for 8 days. The monitored variables were the plasma alanine aminotransferase (ALT), aspartate aminotransferase (AST), glucose (GLU), total protein (TP), electrolytes ($Na^+$, $K^+$, $Cl^-$) and thyroid hormones ($TT_4$, $TT_3$, $FT_4$ and $FT_3$). In food deprivation experiment for 8 days, we did not find any statistical change of level in AST, ALT and electrolytes ($Na^+$, $K^+$, $Cl^-$), but found a significant decrease in TP and GLU. In thyroid hormones, the levels of four hormones in plasma were all showing a tendency to decrease. Especially, $FT_4$ and $TT_3$ were significantly decreased, indicating a withering of physiologic activity. In the temperature test, although no any significant change in AST, TP and electrolytes ($Na^+$, $K^+$, $Cl^-$), we observed a significant decrease of ALT and GLU following to temperature descending from $15.8^{\circ}C$ to $13.3^{\circ}C$ (P<0.05). In the levels of thyroid hormones, any significant change was not observed for experimental period. We conclude that the stress response and physiological activity of olive flounder were more influenced by feed deprivation than slow temperature descending at a transport of living fish, and plasma GLU appears to be sensitive factor to physiological metabolic activity, indicating that it could be used as a monitering mark or index for a health inspection of the fish.
In this study, three GRAS (generally recognized as safety) strain was isolated from Doenjang and Cheonggukjang and identified as a protease-producing microorganism, following the appearance of a clear zone around its colony when cultured on a medium containing skim milk. Based on an analysis of the nucleotide sequence of 16S ribosomal RNA, the strains wereas identified as Bacillus amyloliquefaciens and wereas therefore named Bacillus amyloliquefaciens CDD5, Bacillus amyloliquefaciens CPD4, and Bacillus amyloliquefaciens CGD3. Here, we analyzed the protease and ${\alpha}$-glucosidase inhibitory activities of the three B. amyloliquefaciens strains. Among the isolated strains, B. amyloliquefaciens CGD3 exhibited the highest protease activity (9.21 U/mL, 24 hr). The protease activities of B. amyloliquefaciens CDD5 and B. amyloliquefaciens CPD4 reached 1.14 U/mL and 8.02 U/mL, respectively, at 48 hr. The proteases from the three B. amyloliquefaciens strains showed the highest activities within a pH range of 8.0-9.0 at $50^{\circ}C$, and casein was found to be the preferred substrate on evaluating enzyme activity in the substrate specificity assay. The B. amyloliquefaciens strains exhibited maximal growth when the nutrient broth medium had an initial pH within the range of 5.0-10.0, 6-9% sodium chloride (NaCl), and 5% glucose. B. amyloliquefaciens CDD5 exhibited a low ${\alpha}$-glucosidase inhibition rate (5.32%), whereas B. amyloliquefaciens CPD4 and B. amyloliquefaciens CGD3 exhibited relatively higher inhibition rates of 96.89% and 97.55%, respectively.

In order to improve storability of subtropical yam produced in South Korea, the major pathogens found during the storage were isolated and identified of the pathogenicity, and rot inhibition effect was studied based on the curing treatment condition. Penicillium sclerotigenum and Penicillium polonicum were identified as major pathogens causing rot in subtropical yam during storage, and P. sclerotigenum had stronger pathogenicity. Only the cut surface which has been made during a harvest and has been made smooth before curing generated a normal callus layer. The cut surface of tuberous root was cured in 95% of relativity humidity for three days at $23^{\circ}C$, and cured at $28^{\circ}C$ and $33^{\circ}C$. The observation of callus layer showed that the $23^{\circ}C$ treatment group had similar color saturation between tuberous root and pellicle, while the groups treated above $28^{\circ}C$ showed clear distinction. The generation rate of callus 0.5mm or bigger was 93 percent at $28^{\circ}C$ treatment, 96% at $33^{\circ}C$ treatment, but was 52% at $23^{\circ}C$ treatment. The conventional curing treatment group that used wind or sunlight at room temperature created little callus layer. The infection rate of pathogens according to the relative humidity inside the storage room was low at 40% and 60% of humidity, and the curing treatment period did not make a difference. When the humidity inside the storage room was 80%, all treatment groups rapidly increased the fungal pathogens. The rotten rate of each treatment was studied after 180 days during which the storage temperature was maintained at $16^{\circ}C$ and relative humidity 60%. While the rotten rate of tuberous root that has been cut in conventional curing treatment based on solar and wind was 43%, the one cured at over $28^{\circ}C$ and created the callus layer was less than 18%. While even a healthy tuberous root showed 25% of rotten rate in the traditional treatment group, the one cured at over $28^{\circ}C$ was less than 10%. The weight loss was 1-6% lower in the forced treatment group than in the conventional treatment group.
Chrysanthemum is a cut flower species that normally lasts for 1 to 2 weeks, in some cases 3-4 weeks. This has been attributed to low ethylene production during senescence. Reduction in cut flower quality has been attributed to the formation of air embolisms that partially or completely blocks the water transport from the vase solution to the rest of the cut flower stem, increasing hydraulic resistance which may cause severe water stress, yellowing, wilting of leaf, and chlorophyll degradation. Standard type chrysanthemum can be harvested when buds were still tightly closed and then fully opened with the simple bud-opening solution. Standard type chrysanthemum can also be harvested when the minimum size of the inflorescence is about 5-6 cm bud which opened into the first flower full-sized flower. While spray varieties can be harvested when 2-4 most mature flowers have opened (40% opening). Cut flowers are sorted by stem length, weight, condition, and so on. Standard chrysanthemum is 80 cm length for standard type and 70cm for spray type. Pre-treatment with a STS, plant regulator such as GA, BA, 1-MCP, chrysal, germicide, and sucrose, significantly improved the vase life and quality of cut flowers. It is well established that vase solutions containing sugar can improve the vase life of cut chrysanthemum. Chrysanthemum is normally packed in standard horizontal fiberboard boxes. Chrysanthemum should normally be stored at $5{\sim}7^{\circ}C$. Precooling resulted in reduction in respiration, decomposition, and transpiration activities as well as decoloration retardation. There was significant difference between "wet" storage in 3 weeks and "dry" storage in 2 weeks. In separate pulsing solution trials, various germicides were tested, as well as PGRs to maintain the green color of leaves and turgidity. Prolonging vase life was attained with the application of optimal solution such as HQS, $AgNO_3$, GA, BA and sucrose. This also retarded senescence in leaves of cut flower stems. Fresh cut chrysanthemum can be transported using a refrigerated van with $5{\sim}7^{\circ}C$. Increasing consumption and usage of cut chrysanthemum of various cultivars would require efficient transport system, and effective information exchange among producer, wholesaler, and consumer.

The effects of purification using ${\alpha}-amlyase$ (Termamyl 120L) on physicochemical properties of ${\beta}-glucan$ from oat bran were studied. Four fractions were selected as fraction A ($55^{\circ}C$, 15%, pH 6), fraction B ($45^{\circ}C$, 15%, pH 6), fraction C ($50^{\circ}C$, 0%, pH 7), and fraction D ($50^{\circ}C$, 10%, pH 5) from the result of physiological test, and three consecutive subfractions were obtained by repeated ${\alpha}-amlyase$ treatments on the each fractions. The contents of ${\beta}-glucan$, protein, and ash after purification were in 81.4-88.2%, 4.1-6.3% and 2.6-6.2%, respectively. The apparent viscosities of purified ${\beta}-glucan$ aqueous solutions were similar to those of hydroxy methyl cellulose. Glucose was a major monosaccharide of ${\beta}-glucan$ extracts, and xylose and arabinose were also detected as minor constituents on TLC. The average molecular weight ranged $2.0{\times}10^6-5.1{\times}10^6$ and was decreased after purification. From the result of the differential scanning calorimetry, the melting point ranged $130-140^{\circ}C$ with purification step and thermal transition enthalpy was increased. The ratio of ${\beta}-(1{\rightarrow}3)\;to\;{\beta}-(1{\rightarrow}4) $ linkages were 1:2.22-1:2.52, and increased up to 1:5.50 after purification.
