A Prescriptional Study of Ojeoksan on Clinical Application (오적산(五積散)의 임상(臨床) 활용(活用)에 대한 방제학적(方劑學的) 고찰(考察))

  • Objectives : This study was intended to establish for medical treatment range of Ojeoksan and to help extending application of medical insurance through prescriptive research. Methods : We analyzed "Dongeuibogam", "Taepyeonghaeminhwajegukbang" and other books Result : 1. composition of Ojeoksan in "Dongeuibogam" is same as in "Gogeumeuigam" 2. Ojeoksan is composed of five kinds of basic prescripitions; Er Chen Tang, Ping Wei San, Ma Huang Tang, Si Wu Tang etc. and it used for five pathological factors; cold, energy, eating, phlegm, blood. 3. Property of Ojeoksan is warm so it can be used for any cold diseases. 4. Ojeoksan is used for 16 kinds of diseases including chest pain, abdominal pain etc. Conclusion : Ojeoksan can be used for cardiovascular disorders and Ojeoksan is possible to apply or to extend medical insurance coverage.

Studies on the Anti-inflammatory and Analgesic Activities of Ohjuksan (오적산(五積散)의 소염(消炎) 진통작용(鎭痛作用)에 관한 연구(硏究))

  • Ohjuksan has been used for the treatment of cold and pantalgia in traditional medicine. The anti-inflammatory activity of the aqueous extract from Ohjuksan (OJSE) was investigated utilizing acetic acid-induced edema and adjuvant arthritis in rats. The effects of this agent on acute toxicity and acetic acid-induced writhing syndrome in mice were also examined. OJSE did not showed acute toxicity at 2400mg/kg (p.o.) and 1200mg/kg(i.p.) body weight. It was also showed to have significant analgesic action on the writhing syndrome in mice induced by 0.7% acetic acid at 300,600mg/kg body weight. It showed anti-inflammatory activity in 5% acetic acid-induced edema and adjuvant arthritis with oral administration in rats and exhibited significant preventive effect on edema at 300 and 600mg/kg(p<0.01). In the method of adjuvant arthritis, orally administered for 19 days, it inhibited the hind paw edema in rats with 300 and 600 mg/kg body weight from 5 th day to 19th day. These results suggest that OJSE had analgesic and anti-inflammatory action.

Effects of Ojuck-san on Hyperlipidemia in Rats (오적산(五積散)이 흰쥐의 고지혈증에 미치는 영향(影響)에 대한 실험적(實驗的) 연구(硏究))

  • In order to study the effects of Ojuck-san on hyperlipidemia, we divided the rats into three groups(normal group, control group & sample group) and performed the experimental research. Hyperlipidemia rats were induced by oral for 14days. The sample group was administerd the extract of Ojuck-san for 14 days and control group was administerd equal dose of oral. And then we measured the amount of serum Total cholesterol, Triglyceride, LDL-cholesterol, HDL-cholesterol, Phospholipid, Cholinesterase and Cardiac risk factor. The results were as follows : 1. Ojuck-san showed decreasing effects on Total cholesterol, Triglyceride, LDL -cholesterol, and Phospholipid level in serum significantly(p<0.001). 2. Ojuck-san showed increasing effects on HDL-cholesterol level and Cholinesterase in serum significantly(p<0.05). 3. Ojuck-san showed decreasing effects on Cardiac risk factor in serum significantly.(p<0.001). According to the above results, Ojuck-san showed significant decreasing effects on hyperlipidemia, and it is considered that it is appropriate to apply for hyperlipidemia.

Brain Metal Level in Ohjeoksan-Treated Rats (오적산(五積散)을 투여한 흰쥐의 뇌중 금속농도변화에 관한 연구)

  • This study was to investigate the metal accumulation from Ohjeoksan-Decoction to rat brain(Sprague Dawley). 1. In Control group, each metal concentration was within 26.65mg/kg. and in each experimental groups, was within 28.39mg/kg. And there were no significant metal concentration between control and experimental groups. Exceptionally, Cr and Ni level of control group was lower than experimental groups significantly. (p<0.05) and Cr and Ni level of experimental I group was lower than other experimental groups too. (p<0.05). But, Pb level of control group was higher than experimental groups significantly (p<0.05) Pb level of experimental I group was higher than other experimental groups(p<0.01) 2. In non-hazardous, hazardous and total metal concentration, there was no increase tendency in brain according to the high dose of OD(Ohjeoksan-Decoction) intake. Reversely, in experimental groups, hazardous metal concentration was decrease by high OD intake. (P<0.05) 3. Correlation among each metal in brain was various in each groups Briefly under the intake of Ohjeoksan-Decoction, that if very busy herb prescription , this study was defined within safety in brain metal level by P.O. during 10 days. But, there should be a more research about Pb increase by high dose OD intake

Effects of Ojeok-san on the Maturation of Follicles and NGF Expression in Rats with Estradiol Valerate-induced Polycystic Ovaries (오적산(五積散)이 Estradiol Valerate로 유발된 다낭성 난소 모델에서 난포의 성숙 및 NGF발현에 미치는 영향)

  • Objectives: This study was designed to investigate the effect of Ojeok-san(OJS) on the estradiol valerate(EV)-induced polycystic ovaries(PCO) in rats. Methods: PCO was induced by single intramuscular injection with estradiol valerate(EV)(4mg) in female rats. Normal group(n=8) were injected with sesame oil and orally administrated distilled water for sixty days. PCO control group (n=8) were injected with EV and orally administrated distilled water for sixty days. OJS treated group(n=8) were injected with EV and orally administrated OJS for sixty days. Then, we measured weights of body, ovaries, adrenal glands and uterus of rats. The histomorphometrical changes of ovaries were also evaluated. And we observed the NGF and CRF expression by immunohistochemistry. Results: The results were as follows - The weights(mg) of ovaries in OJS treated group($57.4{\pm}9.4$) were significantly increased(p < 0.05) compared with PCO control group($42.3{\pm}8.5$). - The numbers of mature follicles in OJS treated group($9.3{\pm}2.5$) were significantly increased(p < 0.05) compared with PCO control group($6.1{\pm}2.1$). - The expressions of NGF-immunoreactive cells in the ovarian granulosa cells in OJS treated group were weaker than PCO control group. Conclusion: From the these results, we concluded that Ojeok-san(OJS) has inhibitory effect on the development of EV-induced polycystic ovary. And it's effect maybe related with decreased NGF activities in the ovary.

A Case Report of Diabetic Hyperlipidemia in a Patient with Cerebral Infarction Treated with Ojeok-san (뇌경색 환자의 당뇨병성 고지혈증에 대한 오적산가감방(五積散加減方) 호전 1례)

  • Hyperlipidemia is one of the major factors causing the atherosclerosis of coronary arteries and well-documented modifiable risk factors of stroke, especially of the ischemic type. For Insulin-Dependent Diabetes Mellitus sufferers, if blood sugar is appropriately maintained, lipid and lipoprotein are normal, but if blood sugar is inappropriately maintained or clinical Diabetic nephropathy induces metabolic disorder of lipid, then Total cholesterol, low density lipoprotein cholesterol, triglyceride and very low density lipoprotein cholesterol levels go up and high density lipoprotein levels go down. The purpose of this study is to evaluate the effect of treatment with Ojeok-san and to observe the changes in Fasting Blood Sugar(FBS), 2 Hours Postprandial Blood Sugar(PP2h), Haemoglobin A1c(HbA1c), Total Cholesterol(T-Chol) and Triglyceride(TG). After the treatment, Fasting Blood Sugar decreased from 149mg/dl to 89mg/dl. 2 Hours Postprandial Blood Sugar decreased from l85mg/dl to 110mg/dl. Haemoglobin A1c decreased from 6.3% to 5.7%. Total Cholesterol decreased from 268mg/dl to 217mg/dl. And Triglyceride decreased from 438mg/dl to 265mg/dl. These results support a role for oriental medical therapy in treating Diabetic Hyperlipidemia. Further case studies of herbal treatment of this ailment are needed.

Acute Toxicity Study on Fermented Ojeok-san(Wuji-san) Extract in Mice (발효 오적산 추출물의 급성독성 실험)

  • Purpose: This study was carried out to investigate the acute toxicity and safety of fermented Ojeok-san(Wuji-san) extract. Methods: To evaluate the acute toxity and safety, 0(control group), 1250, 2500 and 5000 mg/kg of fermented Ojeok-san(Wuji-san) extract were orally administered to 20 male and 20 female ICR mice. After single administration, we observed survival rates, general toxicity, changes of body weight, and autopsy. Results: Compared with the control group, we could not find any toxic alteration in all treated groups (1250, 2500 and 5000 mg/kg). Conclusions: $LD_{50}$ of fermented Ojeok-san(Wuji-san) extract might be over 5000 mg/kg and it is very safe to ICR mice.

Activation of Immune System & Antimetastatic Effects of Ojeok-san by Oral Administration (오적산(五積散) 경구투여에 의한 면역활성과 종양 전이 억제 효과)

  • Objectives: This study was designed to investigate intestinal immune system activation and antimetastatic effect of Ojeok-san on cancer cells by oral administration. Methods: Cell viability of Ojeok-san was tested with colon 26-M3.1 carcinoma cells and Peyer's patch cells in vitro. Antimetastatic experiments were conducted in vivo mouse model by using colon 26-M3.1 carcinoma cell. To observe immunomodulating effects of Ojeok-san on Peyer's patch cells, we measured interleukin (IL)-4, GM-CSF. In addition to observing effects of Ojeok-san on hematopoiesis, we measured proliferation of bone marrow cells mediated by Peyer's patch cells in vitro. IgA induction activated in serum and intestinal content was measured to observe the effect of orally administered Ojeok-san on mucosal immune system. After administering Ovalbumin (OVA) with Ojeok-san, Proliferation of Peyer's patch cell was measured to investigate gut immunostimulatory effect. Results: in vitro cytotoxicity analysis, the inhibitory concentration $(IC)_{50}$ of the colon 26-M3.1 carcinoma cell was $890{\mu}g/ml$. $IC_{50}$ of the Peyer's patch cells with LPS was $990{\mu}g/ml$. We found that orally administered Ojeok-san significantly inhibited tumor metastasis in vivo. In addition, the amounts of IL-4 and GM-CSF in the culture supernatant of Peyer's patch cells were significantly increased compared to the control group. The proliferation of bone marrow cell was significantly up-regulated with Ojeok-san. These results indicate that oral administration of Ojeok-san enhances the secretion of hematopoietic growth factors such as GM-CSF and IL-4 from Peyer's patch cells, and these cytokines also act on modulator of bone marrow cell proliferation. After orally administering Ovalbumin (OVA) with Ojeok-san, IgA induction and Proliferation of peyer's patch cell was up-regulated with Ojeok-san. These results means orally administered Ojeok-san activates intestinal immune system and has an inhibitory effect on tumor metastasis. Conclusions: Orally administered Ojeok-san appears to have considerable activity on the anti-metastasis by activation of immune system.
