Evaluation of Potential for Antioxidant and Anti-inflammatory Material of Schisandra chinensis and Cudrania tricuspidata of Cultivated in Sunchang-gun (순창군 재배 오미자와 꾸지뽕의 항산화 및 항염증 소재 활용 가능성 평가)

  • 염증 (Inflammation)은 물리적인 상처나 세균감염이 되었을 때 손상된 조직을 재생하고 신체를 방어하기 위해 일어나는 선천성 면역반응으로 알려져 있다. 주로 선천면역을 담당하는 대식세포는 lipopolysaccharide, reactive oxygen species와 cytokine 등에 의해 활성화되어 tumor necrosis factor-α. interleukin-1β, interleukin-6 등 염증인자들의 생성에 관여한다. 특히, 산화질소는 superoxide 음이온과 쉽게 반응하고 peroxynitrite와 같은 독성이 강한 산화제를 생성하여 단백질 및 지질의 과산화를 유도하고 세포독성을 일으키는 것으로 보고되고 있다. 따라서 본 연구에서는 항산화 및 항염증 소재를 탐색하기 위해 오미자 (Schisandra chinensis), 꾸지뽕 (Cudrania tricuspidata)을 이용하여 항산화 활성을 평가하고자 하였으며, 세포주를 활용한 세포독성 및 항염증 활성을 확인하고자 하였다. 본 연구는 전북 순창군에서 재배된 오미자, 꾸지뽕을 열 건조(60℃) 통해 건조한 후 분말화하였다. 최적 추출 조건 선정을 위해 다양한 용매 (열수, 증류수, 주정 20, 40, 60%), 온도 (25, 40, 60, 80℃) 및 시간 (1, 2, 3, 4, 5 h) 조건에서 추출된 추출물의 총 폴리페놀 함량을 비교함으로써 최적 조건을 선정하였다. 오미자와 꾸지뽕의 DPPH 및 ABTS radical 소거 활성, 총 플라보노이드 함량을 확인하여 항산화능 및 기능성 성분 함량을 평가하였다. 또한 대식세포주인 Raw 264.7을 활용하여 MTT assay, 산화질소 생성 억제 활성을 확인하여 세포독성 및 항염증 활성을 평가하였다. 실험 결과, 오미자 및 꾸지뽕은 각각 주정 40%, 60℃ 그리고 증류수, 60℃에서 추출 시 가장 높은 총 폴리페놀 함량 (약 98.3 mg GAE/g 및 88.2 mg GAE/g)을 함유하며, DPPH 라디칼 소거 활성은 약 44.6% 및 24.4%, ABTS 라디칼 소거 활성은 약 30.3% 및 40.8%로 확인되었다. 총 플라보노이드 함량은 약 21.80 mg QE/g 및 35.68 mg QE/g으로 확인되었다. 또한 오미자 및 꾸지뽕 기능성 추출물을 100 ug/mL 처리 시 세포 독성이 나타나지 않는 것으로 확인되었으며, NO 생성량을 약 56.3% 및 21.7% 저감시켜 항염증 효능을 나타내는 것으로 확인되었다.

  • Safflower is natural red dye largely used for dyeing on protein and cellulose fabric. It contains safflower yellow and carthamin red. Safflower yellow is water-soluble dye, while carthamin red is soluble in alkaline condition. Therefore the former was extracted by cold water. Cartamon obtained by adding acidic solution to carthamin red shows the original hue of safflower. In this study, the condition of extraction with bean stem ash solution and dyeing behavior of carthamon in safflower were examined by using the traditional dyeing method. The relationship between the dye-uptake(K/S) of silk and ramie fabric and the various extractions and dyeing conditions was investigated.

Volatile components of Schizandra Chinensis Bullion (오미자(Schizandra Chinensis Bullion)의 휘발성 성분)

  • The essential oil of Schizandrae Fructus was extracted by simultanous distillation-extraction and steam distillation and analyzed by GC and GC-MS. The oil content of Schizandrae Frutus was 0.64% when extracted by steam distillation and 0.88% in case of simultanous distillation extraction and sensory analysis of the oil indicated woody, camphorous, spicy and sour characteristic notes. The experimental results confirmed the presence 47 volatile components, the major components were ${\gamma}-terpinene$, p-cymene, ${\alpha}-ylangne$ and ${\beta}-elemene$. The oil was fractioned into ten fractions and 4 fraction of them have a good aroma character. The application of the oil showed the utilization possibility as flavoring materials for Korea tea.

Antimicrobial Activity of Omija (Schizandra cheinensis) Extracts (오미자(Schizandra chjinensis) 추출물의 항균활성)

  • This research was conducted to investigate the antimicrobial activities of ethanol extracts from omija against the 12 icroorganism including bacteria, yeast and mold. The extracts inhibited the growth of bacteria, but not yeast or mold. Minimum bactericidal concentration (MBC) of B. subtilis and S. aureus was 1.6~3.2 mg/mL, and those of gram(-) bacteria, including E. coli, were 6.3~12.5 mg/mL. Growth of B. subtilis and S. aureus were retarded by adding 900 ppm and 300 ppm of ethanol extracts. Anthimicrobial activity of the ethanol extracts was not destroyed by heating. In comparison of endocarps extracts with the ethanol from the seed extracts, the ethanol extracts of endocarps showed the high antimicrobial activity.

A Quality Characteristic in Schisandra chinensis by Puff processing (팽화처리에 의한 오미자의 품질 특성)

  • Puff processing is one of the preparation and processing methods of herbal medicines. This study analyzed the quality characteristics in schisandra chinensis extracts by puff processing ($1kgf/cm^2$, $3kgf/cm^2$, $5kgf/cm^2$) through changes of the lignan contents and antioxidant activity. The lignan contents and antioxidant activity of puff processing schisandra chinensis extracts (PPSCE) increased higher than compared to a control. Also, as lignan contents and antioxidant activity of PPSCE was increased with the increasing pressure but it was reduced from $5kgf/cm^2$. The results suggest that puff processing was attributed to the quality increase schisandra chinensis.

Antioxidation activity of residue after omija (Schisandra chinensis) juice extract (오미자(Schisandra chinensis) 착즙박 추출물의 항산화 활성)

  • The physicochemical properties and schizandrin contents of various solvent ($H_2O$, 50% EtOH, 75% EtOH, 95% EtOH) extracts from residue after Omija juice was investigated using total polyphenol contents (TOC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity (RAS), anthocyanin contents (ANC), and schizandrin contents level (SCL). Total polyphenol contents, radical scavenging activity, and anthocyanin contents of 50% EtOH extract were the highest among all residue after Omija juice extracts, and was 16.70 mg/mL in the TOC and 86.16% in the DPPH-RAS. This meant that 50% EtOH extract from residue after Omija juice had more available antioxidant matters. As extraction time increases all extract treatments significantly reduced in the ANC contents (p <0.05). Amount of the SCL were observed higher value in 95% EtOH extract of residue after Omija juice.

Characteristics of Schizandra chinensis Baillon Orchard Soils Located in Jangsu-gun, Jeollabuk-do (전라북도 장수군 오미자 재배과원 토양 특성)

  • This study was conducted to investigate the physico-chemical properties of Schizandra chinensis Baillon orchard soils located in Jangsu-gun, Jeollabuk-do. Surface (0 to 10 cm) soils were collected from 200 experimental sites located at Jangsu-eup (53 site), Gyenam-myeon (31), Chunchun-myeon (73), Janggye-myeon (12), Bunyam-myeon (31). The soil texture was mostly loamy sand, and the mean values of degree of soil aggregate and soil porosity were 33.1 and 59.9%, respectively. The pH, EC, total-N, available-P, soil organic matter, and cation exchange capacity of the soils were $5.51{\pm}0.54$, $290{\pm}139{\mu}S\;cm^{-1}$, $946.3{\pm}65.5mg\;kg^{-1}$, $319.6{\pm}29.2mg\;kg^{-1}$, $29.0{\pm}13.9g\;kg^{-1}$ and $4.11{\pm}0.34cmol_c\;kg^{-1}$, respectively. The concentrations of Pb, Cd, Cu and Zn were $3.48{\pm}0.55$, $0.09{\pm}0.04$, $6.90{\pm}0.91$ and $97.7{\pm}42.2mg\;kg^{-1}$, respectively. The presented data can be utilized in better managing Schizandra chinensis Baillon orchard soils in the studied areas.

Twig Blight on Chinese Magnolia Vain Caused by Botryosphaeria dothidea in Korea (Botryosphaeria dothidea에 의한 오미자 줄기마름병)

  • The twig blight symptoms were observed in Chinese magnolia vine (Schisandra chinensis) at Mungyeong city, Gyeongbuk province, Korea in June 2015. The typical symptoms of infected plant were shriveled and wilted in leaves which led to blight resulted in death. Based on the morphological characteristics, the isolate was suspected as Botryosphaeria sp. Inoculation of isolated pathogen was performed to identify its pathogenicity according to Koch's postulates. Re-isolated fungi from the inoculated stem was showed same morphological characteristics with original pathogen. Phylogenetic analysis was performed using combined sequence of rDNA internal transcribed spacer region, EF1-${\alpha}$ and ${\beta}$-tubulin gene. The isolated pathogen was identified to the B. dothidea by phylogenetic analysis. This is the first report of twig blight on S. chinensis caused by B. dothidea in Korea.

Quality Characteristics of Omija (Schizandra chinensis) Extracts with Various Water Types (물의 종류에 따른 오미자 추출액의 이화학적 품질특성)

  • This is the basic study on the extraction characteristics of Omija (Schizandra chinensis) for juices of Korean traditional foods. Extraction is being performed as 25 times of the solvent ratio at room temperature and at $65^{\circ}C$ with deionized water, tap water and Cheong-song mineral water which have high contents of Ca and Mg. The overall good results of the extraction conditions are calculated from the values of L (Lightness), redness (a), yellowness (b), soluble solids content, acidity, pH levels and the DPPH radical scavenging activity by using the electron donating ability (EDA). The results are as follows: 12 hours of room temperature and 6 hours at $65^{\circ}C$ were good extraction times. The Cheong-song mineral water at $65^{\circ}C$ is available for rapid extractions when compared to deionized water and tap water. A longer extraction time shows lower values of acidity but increasing values of soluble solids content and DPPH radical scavenging activity when using the electron donating ability. The best sequences for the extractions are Cheong-song mineral water followed by tap water and distilled water after overall consideration.

Exfoliation of abalone, Haliotis discus hannai using edible plants (식용작물을 이용한 전복 박리)

  • Twenty nine edible plants were selected in order to study their exfoliation effect on abalone Haliotis discus hannai from its substrate. Exfoliation was observed in spearmint Mentha spicata, lemon balm Melissa officinalis, rose flower, onion Allium cepa, wasabi powder, mustard powder, omija Schizandra Chinensis, and hibiscus Hibiscus sabdariffa solutions. In particular, both the omija and hibiscus solutions exfoliated over 90% of the abalone. In field tests, 3% and 4% omija solutions exfoliated 93.6% (850/908) and 97.1% (810/834) of abalone, respectively, whereas 3% and 4% hibiscus solutions exfoliated 96.1% (780/812) and 97.4% (700/719), respectively. The detached abalone were recovered within 1 min 39 seconds. These results indicate that the omija and hibiscus solutions (3% and 4%) may be useful in the exfoliation of abalone.
