Inhibition of Urease Activity of Helicobacter pylori by Mume Fructus

  • Several extracts of Chinese herbs are known to have antimicrobial efface on Helicobacter pylori. The inhibition action is believed to be anti-urease activity of the substance in the herbs. In this study ethanol (70%) extracts of Mume Fructus showed anti-urease activity far better than previously reported substance such as Caesalpinia sappan L. and Forsythiae suspensa VAHL.

Biological Activity Review of Mume Fructus and Processed Mume Fructus (오매(烏梅)와 증숙 오매(蒸熟 烏梅)의 생리활성 연구)

  • Objectives: Presently Mume Fructus (MF) undergoes fumigation, which produces benzo[a]pyrene. As a primary analysis with the aims to minimize the production of benzo[a]pyrene and to suggest standards for processing the MF, the steaming method was chosen among the various processing methods, and reviewed through a series of experiments.Methods: Methods:Pitted and un-pitted MF were steamed and processed into samples. After testing level of benzo[a]pyrene, the samples were analyzed for amount of polyphenol and flavonoids. Scavenging activities of the samples for the DPPH and ABTS radicals were tested. In order to measure anti-inflammatory effects of the samples, cell survival rate was investigated using CCK-8 Assay. Also, water extracts of dried and steamed MF were administered to the RAW 264.7 cells to compare expressions of NO, PGE2, IL-1β, and TNF-α. In addition, anti-diarrhea effects of the herbal medicine were tested on animal models with diarrhea induced by MgSO4 and Castor oil.Results: Regardless of pitting, processed MF contained no benzo[a]pyrene. Anti-oxidation effect increased in relation to the frequency of steaming process. However, extracts of dried and steamed MF suppressed different kinds of inflammation factors, and extract of dried MF showed superior anti-diarrhea effect than extract of steamed MF.Conclusions: It is suggested that steaming method of MF is recommended for processing the herbal medicine without the production of benzo[a]pyrene. But regarding that dried and steamed MF showed differences in their anti-oxidative, anti-inflammatory, and anti-diarrhea effects, it is recommended to perform further researches on different efficacies of MF according to their processing methods.

Effects of Some Herbal Extracts on Aflatoxin $B_1$ Production from Aspergillus parasiticus (생약추출물이 Aspergillus parasiticus Aflatoxin $B_1$ 생성에 미치는 영향)

  • The influences of the extracts from Cinnamomi Cortex, Eucommiae Cortex, Puerariae Radix, Lycii Fructus, Zizyphi Fructus, Schisandrae Fructus, Mume Fructus, Chaenomelis Fructus on mycelial growth and aflatoxin $B_1$ production from Aspergillus parasiticus were analyzed. The pH of the culture media were reduced to below pH 4 by all the herbal extracts after 3 days incubation. However, the pH of the culture media increased above pH 6 after 6 days incubation using the extracts from Cinnamomi Cortex, Eucommiae Cortex, Puerariae Radix and Lycii Fructus. The mycelial growth of A. parasiticus was increased over the amount of the control. Puerariae Radix produced the largest amount of mycelial growth and Chaenomelis Fructus produced the smallest amount of mycelial growth. The productions of aflatoxin $B_1$ from A. parasticus culture were increased by the extracts of Puerariae Radix and Zizyphi Fructus, while inhibited by the extracts of Cinnamomi Cortex, Eucommiae Cortex, Lycii Fructus, Schisandrae Fructus, Mume Fructus and Chaenomelis Fructus. In particular, the extracts of Cinnamomi Cortex, Lycii Fructus and Schisandrae Fructus almost inhibited the production of aflatoxin $B_1$. The production of the total protein from Cinnamomi Cortex, which produced much less aflatoxin $B_1$, and Puerariae Radix, which produced a great deal of aflatoxin $B_1$ from A parasticus were slightly higher than the production of the total protein of the control medium.

Antibacterial Activities of hot-water and ethyl alcohol Extracts of Medicinal Herbs on Fish Pathogenic Bacteria (천연 생약재 열수 및 알코올 추출물의 어병 세균에 대한 항균력)

  • Hundreds of medicinal herbs have been using for the purpose of diseases treatment and immune enhancement for human being and other animals including fishes. Among them, 49 species of medicinal herbs were selected and tested for antibacterial activities against 19 strains of fish pathogenic bacteria in different 4 species. The 49 medicinal herbs were extracted by water and ethyl alcohol. The extracts were freeze dried and some paper discs from the extracts were prepared for the evaluation of antibacterial activity. The tested pathogenic bacteria were 5 strains of Edwardsiella tarda, 5 strains of Vibrio sp., 4 strains of Lactococcus garvieae, 1 strain of Lactococcus raffinose, 1 strain of Streptococcus parauberius, and 3 strains of Streptococcus iniae. The Galla rhois (Obaeja), Gaeonnamu and Hwangleyon showed antibacterial activities on both gram negative and gram positive fish pathogenic bacteria. The Youkgae, Sangbaekpi, Bogolji and Gamcho showed very effective antibacterial activities on gram positive pathogens while Jiyu, Aeyoeb and Yeonkyo showed very effective on gram negative pathogens.

Antioxidant and Cell Activity Using Extracts of Mume Fructus (오매 추출물들의 항산화 및 세포 활성)

  • Mume fructus is the roasted fruits of Prunus mume and has been used as traditional chinese medicine. In this study, mume fructus extracts were prepared by three different methods including hot water extract (sample1), fermentation extract using Lactobacillus (Sample2-LP and sample-LA) and ethanol extract (sample3). Total polyphenol and flavonoid contents were improved by fermentation process, compared to water extract. Sample 3 showed the highest activity in DPPH radical scavenging. The cytotoxicity of the sample 2-LP was in the range of 83.3% cell viability at $300{\mu}g/m{\ell}$ concentration. Mume fructus extracts in a concentration-dependent manner inhibited melanogenesis and NO synthesis was inhibited. Specifically, the extracts fermented by L. plantarum (sample2-LP) showed higher anti-oxidant activity, anti-inflammatory and skin whitening activity than other extracts. It suggests that sample 2-LP could be potentially used as a resource of materials for functional cosmetics.

새로운 오메프라졸염의 약효검색에 관한 연구

  • 오메프라졸의 산 불안정성을 개선하기 위하여 신규 합성한 오메프라졸 cholestyramin resin 염 복합체와 오메프라졸 hydroxypropyl-$\beta$-cyclodextrin (HP-$\beta$-CD) 포접화합물의 약효를 검색하였다. 생체외 실험 (in vitro test)으로 H$^{+}$/K$^{+}$-ATPase 활성도 저해효과를 검토하였으며 생체내 실험 (in vivo test)으로 Shay 결찰법에 의한 위산분비 억제효과에 대한 실험을 실시하였다. 오메프라종 염 복합체와 오매프라졸 포접화합물은 1$\times$$10^{-5}$-1$\times$$10^{-3}$M농도 범위에서 용랴의존적으로 H$^{+}$K$^{+}$-ATPase 활성을 억제시켰으며 $IC_{50}$/치는 오메프라졸 결과와 유사하였다. 셍체네 실험에서는 오메프라졸 HP-$\beta$-CD 포접화합물이 오메프라졸과 그 resin염복합체보다 위액분비량, 펩신 활성도에 대한 $IC_{50}$/치가 낮았으며 이는 생체내에서 포접 화합물이 오메프라졸의 안정성을 증가시킴으로서 위산분비 억제효과를 증가시킨것으로 사료된다.

Effect of Butanol Extracts from Prunus mume on the Growth of Salmonella typhimurium (오매의 Butanol추출물이 Salmonella typhimurium의 생육에 미치는 영향)

  • The antimicrobial properties of extracts of Prunus mume were tested on Salmonella paratyphimurium and S. typhimurium. First, the Prumus mume was extracted with methanol at several temperatures, and then fractionation of the methanol extracts from Prunus mume was carried out by using petroleum ether, chloroform, ethyl acetate or butanol. Secondly, absorption column chromatoraphy (using a Diaion HP 20) was conducted to eliminate some water soluble materials that might inhibit the antimicrobial activity of some extracts. The antimicrobial activitry of each of the Prunus mume extracts was determined using a paper disc method against several food-borne pathogens, The growth inhibition curve was determine using butanol extracts of Prunus mume against Salmonella typhimurium. The extraction temperature did not have any significant effect on the yield of the extract or on the level of antimicrobial activity. The butanol extract of Prunus mume showed strong antimicrobial activity against Salmonella paratyphimurium and S. typhimurium; a 1,000 ppm of butanol extract of Prunus mume retared the growth of S. typhimurium up to 36 hours.   • Mume Fructus extract is used in folk medicine in Eastern Asian countries including Korea. However, its therapeutic effect on thrombosis is not known. Mume Fructus methanol extract (ME) dose-dependently inhibited ADP-induced platelet aggregation and also exhibited about 130% fibrinolytic activity compared to the natto. Oral administration of ME to mice significantly extended tail-bleeding time. ME prolonged both aPTT and PT in human citrated plasma also. These results suggest that the methanol extract from Mume Fructus have antithrombosis activity.

The Psychiatric and Central Nervous System Effects of Fructus Mume in Medical Classics (한의학 고문헌을 통한 오매의 정신의학과 중추신경계 관련 효능 연구)

  • Objectives : Fructus mume(F. mume) has been used as a medicine for thousands of years in East Asia and reported to have effect on cognitive deficits induced by chronic cerebral hypoperfusion. We investigated F. mume's effects on psychiatric and central nervous system in medical classics. Methods : 25 materia medica books and Donguibogam were searched to find psychiatric and central nervous system effects of F. mume. Two Korean Medicine doctors reviewed the effects from the clinical point of view. Results : 安心(relieve psychiatric discomfort), 令人得睡 治不眠(Treat insominia), 去煩悶(relieve chest discomfort) were psychiatric effects and 偏枯不仁(hypoesthesia accompanied with hemiplegia) was central nervous system effect of F. mume. Conclusions : Further studies will be needed to demonstrate F. mume's effects found in medical classics.

Effect of Ethyl Acetate Extract of the Dried Prunus mume on the Growth of Staphylococcus aureus (오매의 에틸아세테이트 추출물이 Staphylococcus aureus의 증식에 미치는 영향)

  • The dried Prunus mume, an alkaline food abundant in organic acids (citric acid, malic acid and tartaric acid), has been largely used in both folklore remedies and Chinese herbal medicine for a long time. This study was performed to investigate the antimicrobial activity of the dried Prunus mume. The fractionation of the methanol extracts from Prunus mume was conducted using petroleum ether, chloroform, ethyl acetate and butanol, respectively. The antimicrobial activity of the Prunus mume extracts was then determined against food-borne pathogens using a paper disc method. The ethyl acetate extract showed the strongest antimicrobial activity against the eigth food-born pathogens used in this present study. Diaion HP 20 column chromatography was performed to remove some sugars that might inhibit the antimicrobial activity of Prunus mume. The strongest antimicrobial activity of ethyl acetate fraction of Prunus mume was shown against Staphylococus aureus. The growth inhibition curve was determined using ethyl acetate extracts of Prunus mume against Staphylococus aureus, which showed the growth inhibition up to 72 hours at 1,000 ppm concentration.
