Effects of Alternative Medicine Extract on Bone Mineral Density, Bone Strength and Biochemical Markers of Bone Metabolism in Ovariectomized Rats (난소적출 수술을 받은 흰쥐에서의 골밀도, 골강도 및 골대사의 생화학적 지표에 대한 한방요법의 효과)

  • To prevent and treat the osteoporosis, diverse therapies have been applied, which is still need to solve negative side effects. We investigated the effects of the extract from alternative medicine(AM) on the progress of bone loss in ovariectomized-rats fed with Ca-deficient diet for 7 or 14 weeks. Ovariectomy(OVX) concomitant with Ca-deficiency caused bone loss evidently decreased in bone mineral density and bone strength of femoral epiphysis and vertebrae, which were ameliorated with administration of AM extract. Also, the effect of AM extract on the biochemical markers were measured. The increased serum alkaline phosphatase caused by OVX and Ca-deficiency were observed, which were not affected by administration of AM extract. Administration of AM extract may have preventive effect on the elevated serum acid phosphatase concentrations caused by OVX and Ca-deficiency at 14 wks, implicating that AM extract possibly acts toward reducing born resorption, even though the results were not statistically significant. Serum osteocalcin and urinary deoxypyridinoline, the markers of bone turn over, were not changed by estrogen deficiency or AM extract. We concluded that the AM extract treatment had potently preventive effects on the decreased bone density and bone strength induced by OVX and Ca-deficiency. The changes of biochemical markers related to the effect of AM extract were not manifested but it still suggest that AM extract may inhibit the bone resorption derived from OVX and Ca-deficiency.

Association between vitamin D deficiency and anemia among Korean adolescent girls and young women (여자 청소년 및 젊은 여성의 비타민 D 결핍과 빈혈과의 연관성 분석)

  • Purpose: Although vitamin D deficiency is common among Korean adolescent girls and young women, few studies have explored the potential health effects of vitamin D deficiency in this vulnerable population. This study examined the association between vitamin D deficiency and anemia in Korean adolescent girls and young women. Methods: The data from the Korea National Health and Nutrition Examination Survey 2008 ~ 2014 were used. A total of 3,643 girls and adult women aged 12 to 29 who provided all the information (including serum 25-hydroxy vitamin D, hemoglobin, and/or serum ferritin) needed for the analysis were included in the analysis. Demographic, lifestyle, and health data were obtained through survey questionnaires. Anemia and iron deficiency anemia were defined according to the World Health Organization cut-offs. Multivariable logistic regression, and restricted cubic spline regression were used in the analysis. Results: In fully adjusted logistic regression models, the vitamin D deficiency was significantly associated with higher prevalences of anemia (odds ratio (OR): 1.61, 95% confidence interval (CI): 1.04 ~ 2.49) and iron deficiency anemia (OR: 1.43, 95% CI: 1.01 ~ 2.03). In a cubic spline regression model, we observed a dose-response relationship between serum 25(OH)D concentration and anemia, and this linear relationship was also clearly observed between serum 25(OH)D concentration and iron deficiency anemia. Conclusion: Vitamin D deficiency may be associated with a higher prevalence of iron deficiency anemia and anemia in adolescent girls and young women. Alternatively, vitamin D deficiency may be a concurrent event for patients with anemia, which we cannot distinguish in this cross-sectional study. Further studies are needed to verify the causality in this population of low vitamin D levels.

  • 식이 중 지방함량이 성장기 뇌의 미토콘드리아 지방산 조성에 미치는 영향을 살펴보기 위해 옥수수기름을 각각 10, 2, 0.5% 포함한 3종류의 식이를 임신한 Sprague Dawley종의 흰쥐에게 임신말기부터 제공했다. 수유기간 중의 영향을 살펴보기 위해 대조군(10%옥수수기름 섭취)의 일부에 분만 후 0.5% 결핍식이를 섭취시켰고, 또 이유후부터 0.5% 결핍식이군의 일부에 10% 정상식이를 주어 회복효과를 관찰하고자 하였다. 전 실험기간 동안 체중과 뇌 무게, 단백질 함량을 측정하였고 뇌에서의 미토콘드리아를 분리하여 지방산 조성을 관찰하였다. 출생후 수유기간 동안에는 대조군에 비해 지방결핍식이군에서 체중과 뇌의 무게가 다소 높았으나 이유 후 부터 점점 감소하여 유의적인 차이를 볼 수 없었고 특히 0.5% 식이군의 체중은 생후 6주와 7주에 유의적인 감소를 보였다. 그러나 체중에 대한 뇌무게와 뇌의 단백질 함량에 있어에는 대조군과 결핍군 사이에 유의적인 차이를 볼 수 없었다. 따라서 지방결핍이 뇌 성장에 미치는 영향은 다른 영양소에 비해 크지 않음을 알 수 있었다. 뇌 미토콘드리아의 지방산 조성은 나이와 식이에 따라 변하였다. 나이를 먹음에 따라 단쇄 포화 지방산은 감소하고 다 불포화 지방산의 함량은 증가하였다. 또한 결핍식이군과 대조군 사이에 뇌 미토콘드리아 지방산조성을 살펴 본 결과 지방 결핍의 좋은 지표가 되고 불포화도, triene 과 tetraene의 비, 그리고 ${\omega}-6$ 계열과 ${\omega}-9$ 계열의 비에 있어서 큰 차이를 볼 수 있었다. 이들로 부터 성장초기에 지방함량을 조절한 식이에 의해 뇌 미토콘드리아 지질 조성이 크게 변하고 이에따라 미토콘드리아의 기능도 유의적인 영향을 받고 있음을 알 수 있었다.

  • Transient symptomatic zinc deficiency (TSZD) is clinically similar to acrodermatitis enteropathica. The patients are experienced dermatitis on periorifice and extremities, alopecia, diarrhea, growth retardation, anemia, delayed wound healing and infection. They showed a characteristically rapid response to zinc supplement without recurrence. We report 3 cases of TSZD infants, 2 premature infants with inadequate zinc supply and 1 mature infant with chronic diarrhea.

철분강화 유제품 제조 및 생이용성 평가연구

  • 국민의 영양상태가 크게 향상되어왔고 일부에서는 영양과잉과 운동부족으로 인한 비만에 대한 우려가 고조되고 있지만 최근에도 유아나 성장기 어린이들의 철분부족상태에 대한 문제점이 제기되고 있다. 외국에서는 철분 필요량이 높은 성장기 어린이나 유아들에게 영양학적 품질을 개선한 철분강화제품을 공급하기 위하여 이에 대한 연구가 오래 전부터 진행되어왔으나 여러 실험자에 의하여 행해진 실험조건, 사용된 철분염의 종류가 달라서 정확한 비교평가를 하기에는 미흡하다고 사료된다. 여러 가지 식품에서 특히 우유는 영양학적으로 완전식품이라고 하지만 철분은 거의 함유하고 있지 않아서 효과적으로 철분을 강화할 수 있는 기술은 국민영양개선 효과면에서 매우 유용하다고 사료된다. 본 연구에서는 식품첨가물로서 사용되고 있는 철분염 11종을 우유에 100 ppm수준으로 첨가하고 4$^{\circ}C$에서 저장하면서 pH, 지방산패도, 색도변화를 평가하여 우유에 사용하기 적합한 철분염을 1차적으로 선정하였다. 품질평가 결과를 종합하여 분석할 때 ferric citrate, ferric ammonium citrate, ferrous lactate가 철분강화우유 제조에 적합한 것으로 평가하였다. 또한 지금까지 철분강화식품에 일반적으로 자주 사용하고 있는 ferrous sulfate는 우유에 적합하지 않은 것으로 나타났으나 비교치로 사용하기 위하여 다음 실험에 사용하였다. 이상에서 screening된 철분염을 첨가한 우유를 HTST, LTLT으로 각각 살균하고 저장하면서 품질을 평가한 결과 HTST법이 LTLT법보다 PH, 지방산패도, 색도변화에서 나쁜 것으로 평가되었고, 철분염 종류에 따른 차이는 LTLT, HTST법에서 ferrous lactate가 우수한 것으로 나타났다. 본 연구에서 사용된 철분염 가운데에서는 ferrous lactate, ferric citrate가 유제품에 사용하기에 적합한 것으로 나타났다. 생이용성을 평가하기 위하여 우유에서 low molecular weight components(ILC)를 분리하고 철분과 복합체를 형성시킨 다음, 철분 결핍된 쥐의 소장에서 loop을 형성시켜 ILC-철분 복합체를 injection하여 철분 흡수도를 조사하였다. Ferrous lactate 100ppm에서 약 25.6%흡수되었고 ferric citrate 100ppm은 24.7%, ferrous sulfate는 19.7%흡수되었다. ILC를 첨가하지 않은 100ppm 철분염 용액은 ferrous sulfate를 제외하고는 흡수도가 감소되었다. 철분 결핍된 쥐에게 gavage 방법에 의하여 철분강화우유를 투여하였을 때 철분 25ppm 시료에서는 ferrous sulfate가 12.5%로 가장 높았고 ferrous lactate는 8.1%, ferric citrate는 6.5% 흡수되었다. 철분 100ppm수준에서는 흡수율이 낮아져 ferrous sulfate는 25ppm 시료보다 절반이하 수준이었다. Ferric citrate는 차이가 거의 없었으며 ferrous lactate는 70%수준이었다. 이상의 결과에서 철분강화우유에 사용하기 적합한 철분염은 ferrous lactate, ferric citrate였는데 특히 ferrous lactate는 제품의 이화학적 품질, 생이용성 측면 모두에서 가장 좋은 것으로 나타났다.

Physicochemical Characteristics and Antioxidant Activity of Kanjang made from Soybean Cultivars Lacking Lipoxygenase and Kunitz Trypsin Inhibitor Protein (Lipoxygenase와 Kunitz Trypsin Inhibitor 단백질 결핍콩으로 제조한 간장의 이화학적 특성 및 항산화 활성)

  • In order to evaluate suitability by processing for non-GM soybean cultivars such as Gaechuck#2, Jinyangkong and CJ#1 lacking lipoxygenase (LOX) and kunitz trypsin inhibitor (KTI) protein, physicochemical characteristics and antioxidant activity of Kanjang made from above soybean were compared to Kanjang made from a conventional cultivar (Taekwangkong). Proximate compositions of soybeans were similar for the 4 kinds cultivars. Total phenol and flavonoid contents were significantly higher in cultivars lacking LOX and KTI protein than the Taekwangkong. In Kanjang, contents of total and reducing sugar were higher in Taekwangkong Kanjang than Kanjang from made cultivars lacking LOX and KTI protein. Contents of total and amino type nitrogen were the highest in the Jinyangkong Kanjang. Mineral contents were higher in the Jinyangkong and CJ#1 Kanjangs, amino acid contents were higher in the Kanjang made from 3 cultivars lacking LOX and KTI protein than the Taekwangkong. Taste of the Jinyangkong Kanjang with higher sweety and savory was also found to be superior to that of others in overall acceptability evaluation. Total phenol and flavonoid contents in Kanjang were significantly higher in the Kanjang made from cultivars lacking LOX and KTI protein than the Taekwangkong. Radical scavenging activity of Kanjang was increased in the total phenol contents dependent on. Reducing power by ferric-reducing antioxidant potential (FRAP) was significantly higher the Kanjang made from Gaechuck#2 and CJ#1 than the Taekwangkong. $Fe^{2+}$ chelating activity was higher in Taekwangkong Kanjang than the other cultivars, but its activity was similar to Jinyangkong Kanjang. Therefore, higher nutritional composition, total phenol and flavonoids contents and antioxidant activity in the Kanjang made from soybean cultivars lacking LOX and KTI protein might be provide better benefit for manufacture of another their products.

Effect of n-3 Fatty Acid Deficiency on Fatty Acid Composition in Brain, Retina and Liver Using a Novel Artificial Rearing System (인공 사육 동물 모델 시스템을 이용한 n-3 지방산 결핍이 쥐의 뇌, 망막, 간의 지방산 조성에 미치는 영향)

  • Docosahexaenoic acid (22:6n-3, DHA) is highly enriched in membrane of brain and retina, and plays an important role in maintaining an optimal function of the central nervous system. We investigated the effect of n-3 fatty acid deficiency on rat brain, retina and liver fatty acyl composition at two different ages (3 wks and 15 wks) under DHA deficient condition. Rat pups born to dams fed a diet with $3.1\%$ of total fatty acids as $\alpha-linolenic$ acid (LNA) were fed using an artificial rearing system either an n-3 deficient (n-3 Def) or n-3 adequate (n-3 Adq) diet. Both diets contained $17.1\%$ linoleic acid (LA) but the n-3 Adq diet also contained $3.1\%$ LNA. Rats consuming the n-3 Def diet showed a lower brain $(50\%\;in\;13\;wks\;and\;70\%\;in\;15\;wks,\;p<0.05)$ and retinal $(50\%\;in\;13\;wks\;and\;63\%\;in\;15\;wks,\;p<0.05)$ DHA than those on the n-3 Adq diet, which was largely compensated for by an increase in docosapentaenoic acid (22:5n-6, DPAn-6). In the liver of the n-3 Def group, the percentage of DHA decreased by $97\%$ at 3 wks of age with an apparent increase in DPAn-6 relative to the n-3 Adq group (p<0.05), while there was a $65\%$ lower liver DHA in n-3 Def group at 15 wks of age than the n-3 Adq group (p<0.05). Liver arachidonic acid (20:4n-6, AA) was increased at 3 wks of age but decreased at 15 wks of age in the n-3 Def group compared with n-3 Adq group (p<0.05). In conclusion, the replacement of DHA by DPAn-6 in brain and retina fatty acid composition may be related to the suboptimal function in spatial learning, memory and visual acuity. This artificial rearing method presents a first generation model for n-3 deficiency that is similar to the case of human nutrition that commonly employed two generation model.

The Relationship between Helicobacter pylori Infection and Iron-Deficiency: Seroprevalence Study in 937 Pubescent Children (Helicobacter pylori 감염과 철 결핍의 관계: 937명의 사춘기 환아의 혈청 유병률 연구)

  • Purpose: The purpose of this study was to investigate the relationship between Helicobacter pylori (H. pylori) infection and iron-deficiency anemia in pubescent children, susceptible to iron deficiency due to the high iron requirements for growth. Methods: Hemoglobin, serum iron, total iron-binding capacity, serum ferritin, and serum IgG antibodies to H. pylori were measured in 937 children (475 boys and 462 girls). Their ages ranged from 10 to 18 years. The prevalences of H. pylori infection were compared between groups, based on the presence or absence of anemia, hypoferritinemia, iron deficiency, and iron-deficiency anemia. The levels of hemoglobin, serum iron, total iron-binding capacity, transferrin saturation, and serum ferritin were obtained according to the presence or absence of H. pylori infection. Results: The prevalences of anemia, iron deficiency, iron-deficiency anemia, and H. pylori infection were 8.1%, 9.1%, 3.1%, and 20.8%, respectively. The H. pylori-positive rates in anemia, hypoferritinemia, and iron-deficiency group were 34.2%, 29.5%, and 35.3%, respectively, compared to 19.6% in the non-anemia group, 19.2% in the non-hypoferritinemia group, and 19.4% in the non-iron deficiency group. The H. pylori-positive rate in the iron-deficiency anemia group was 44.8% in comparison with 20.0% in the non-iron-deficiency anemia group. Hemoglobin and iron levels did not show any significant differences between the H. pylori-positive and -negative groups, whereas the serum ferritin level decreased significantly in the H. pylori-infected group. Conclusion: H. pylori infection is thought to be associated with iron deficiency in pubescent children.

Comparative Analysis of the Effect of Physical Activity and Stress Experience on the Vitamin D Deficiency according to Occupations: results from KNHANES dataset for 2008~2013 (신체활동수준과 스트레스수준에 따른 Vitamin-D 결핍률에 관한 직군별 비교분석연구: 2008~2013년 KNHANES 데이터 분석을 중심으로)

  • The purpose of the study is to investigate the occupational difference of Vitamin D deficiency according to the level of physical activity and the level of stress. For this purpose, We performed empirical approaches and adopted KNHANES 2008~2013 dataset, to which were applied such methods as crosstabulation analysis, logistic regression and SOBEL test. As a result, we found that Vitamin D deficiency was higher in the group of people with indoor-working-job than in the group of people with out-door working job. Besides, Vitamin D deficiency tends to increase in the people with lower physical activity and higher stress experience. As a result, those employees highly related with in-door activities must be ensured that they need to be exposed to sunlight on a regular basis. Moreover, improving the physical activity levels of employees could be one of appropriate solutions to alleviate Vitamin D deficiency problems. Besides, lowering stress levels in workplace needs to be seriously considered in order not to drop the physical activity levels of workers.
