Overexpression of Rice Chloroplast Small Heat Shock Protein Increases Thermotolerance in Transgenic Plants (벼 엽록체 small HSP의 과발현에 의한 형질전환 식물체의 내열성 증가)

  • To investigate the function of chloroplast small heat shock protein (HSP), transgenic tobacco plants (Nicotiana tabacum L, cv. SR-1) that constitutively overexpress the rice chloroplast small HSP (Oshsp26) were generated. Effects of constitutive expression of the Oshsp26 on thermotolerance were investigated with the chlorophyll fluorescence. After 5-min incubation of leaf discs at high temperatures, an increase in the Fo level, indication of separation of LHCII from PSII, was mitigated by constitutive expression of the chloroplast small HSP When tobacco plantlets grown in Petri dishes were incubated at $20^{\circ}C$/TEX> for 45 min and subsequently incubated at $20^{\circ}C$/TEX> leaf color of wild-type plant became gradually white and all plantlets were finally died. Under the conditions in which all the wild-type plants died, more than 80% of the transformants remained green and survived. It was also found that the levels of Oshsp26 protein accumulated in transgenic plants were correlated with the degree of thermotolerance. These results suggest that the chloroplast small HSP plays an important role in protecting photosynthetic machinery, as a results, increases thermotolerance of whole plant during heat stress.

Phylogenetic relationships of Korean campanulaceae based on chloroplast DNA sequences (엽록체 DNA 염기서열 분석을 이용한 한국산 초롱꽃과 (Campanulaceae)의 계통유연관계)

  • Phylogenetic studies were conducted to evaluate the taxonomic relationships among 28 taxa, including 2 outgroups of Korean Campanulaceae, using atpB, atpB-rbcL, atpF-H, matK, rbcL, rpl16, rpoC1 and trnL-F regions sequences in chloroplast DNA. The combined analyses of eight chloroplast DNA regions suggest that Codonopsis and Platycodon basally branches within the phylogenetic tree; Wahlenbergia distinguished an independent clade; Campanula forms a clade; Peracarpa and Asyneuma clade is a sister to the Adenophora-Hanabusaya clade; Hanabusaya is placed within the section Remotiflorae of Adenophora; Adenophora form a clade. Our present results support the generic level, although discordance remained at the infrageneric groups such as section and series based on morphological characteristics in the genus Adenophora.

Action Mechanism of S-23142 on the PPIX Biosynthesis System of Spinacia oleracea L. Chloroplast (엽록체내 Protoporphyrin IX(PPIX) 생합성계에서 S-23142의 작용기구)

  • The effects of S-23142{N-(4-chloro-2-fluoro-5-propargyloxyphenyl)-3, 4, 5, 6-tetrahydrophtalimide}, on protoporphyrin IX(PPIX) biosynthesis in Spinacia oleracea L, leaf in vivo and in vitro condition were investigated by reversed-phase HPLC with fluorescence detector. The stroma and the membrane fraction of spinach chloroplast were isolated by osmotic regulation. The conversion of ${\delta}$-aminolevulinic acid(ALA) to PPIX occured more in the stroma than in the membrane fraction. It suggested that the enzymes that catalyse PPIX biosynthesis from ALA were localized in the stroma. Also, the synthesized PPIX content from ALA was completely inhibited by $10^{-8}M$ of S-23412 or $10^{-7}M$ of acifluorfen in the stroma but not in the membrane fractions. Therefore, these results suggested that the target site of S-23142 and acifluorfen may exist in the stroma fraction of spinach chloroplast.

Effect of Antibiotics on Phospholipid Biosynthesis and Fatty Acid Composition in Chlorella ellipsoidea Chloroplasts (Chlorella ellipsoidea 엽록체의 인지질 생합성 및 지방산 조성에 미치는 항생제의 효과)

  • The effects of amphotericin B ($150\;\mu\textrm{g}/ml$) and cycloheximide ($10\;\mu\textrm{g}/ml$) on the biosynthesis of phospholipid and the composition of fatty acids in chloroplasts isolated from ChZorella were analyzed. The contents of the total lipid and phospholipid (PC, PE, PI) in treatment with antibiotics were lower compared with the control. In the whole cell system, the major fatty acids utilized for biosynthesis of phospholipid were palmitic acid (31.96%) and linoleic acid (16.96%) in control while those were palmitic acid (36.15%) and linolenic acid (16.71%) in treatment with amphotericin B. And in treatment with cycloheximide, palmitic acid (31.90%) and stearic acid (15.32%) were used in phospholipid formation. The major fatty acids in chloroplasts were analyzed as to be palmitic acid and linolenic acid in control (33.75%, 18.90%) and in treatment with amphotericin B (36.75%, 9.46%). However, it was shown that the major fatty acids in chloroplasts treated with cycloheximide were palmitic acid (28.01%) and oleic acid (19.27%).9.27%).

Effects of Ozone on Photosynithetic Activity in Chloroplast of Barley (Hordeum vulgare L.) (오존이 보리(Hordeum vulgare L.) 엽록체의 광합성 활성에 미치는 영향)

  • The effects of 0.5 and 1.0 ppm of ozone on the developing chloroplast of barley (Hordeum vulgare L) seedling during greening were investigated by PSI and II activities, chlorophyll fluorescence, and the contents of chlorophyll. Etiolated barley seedling was treated 0.5 and 1.0 ppm ozone for the first 4 h during greening. In 24 h greening experiment, the contents of chlorophyll were decreased by increasing ozone concentration from 0.5 ppm to 1.0 ppm. In 24 h greening experiment, Fo, Fv, Fm and qE were greatly decreased as the concentration of ozone was increased, but those were not considerably decreased in 48 h greening experiment. In another experiment, the developing barley seedling was treated with 0.5 and 1.0 ppm ozone for the last 4 h during greening period, which was 24 h or 48 h. In both experiments the PS II activity was decreased as the concentration of ozone was increased, but not in PS I activity. Fv, Fm and Fv/Fo were also decreased as the concentration of ozone was increased. qP and qR were strikingly decreased as the concentration of ozone was increased in both experiments.iments.

Intraspecific sequence variation of trnL/F intergenic region (cpDNA) in Sedum takesimense Nakai (Crassulaceae) and aspects of geographic distribution (섬기린초에서 엽록체 DNA 염기서열의 종내 변이와 지리적 분포 양상 연구)

  • Sequences of the trnL/F intergenic spacer of chloroplast DNA were used to investigate the intraspecific evoution and phylogeography of Sedum takesimense (Crassulaceae). The trnL/F intergeneric spacer sequences from 32 individuals of S. takesimense were either 291 bp (17 samples "without indel" in the following) or 297 bp (15samples "with indel 1") in length due to an indel of 6 bp. Two main cpDNA haplotypes were detected within S. takesimense. The haplotype with indel was found on Ulleung Island and without indel on Ulleung Island and Dok Island. This confirmed the existence of two cpDNA lineages with different geographical distributions. The cpDNA sequence analysis also suggested a putative long-distance dispersal event between Ulleung Island and Dok Island.

Preparation of Photosynthesis Nanofiber Composite Membrane by Using Chlorophyll and Polymer Nanofiber (식물 엽록소와 고분자 나노섬유를 이용한 광합성 나노섬유복합막의 제조)

  • In this study, chlorophylls were been extracted from common local plants, deposited on polypropylene (PP) substrate using various approaches, and the oxygen generation effect of the chlorophylls were investigated. The loading of chlorophylls on the substrates was achieved by dipping and spraying methods, where the spraying coating showed overall better results regarding oxygen generation from the combustion experiments in the closed vessel or in the isolated vacuum oven cell than those of dip coating. In addition, a composite substrate was prepared by nylon6/6 nanofiber on the PP substrate, and it exhibited an increase in the activation of chlorophylls. In the case of samples containing titanium dioxide ($TiO_2$), the reaching time of oxygen concentration from 16% to 21% and the combustion test using a candle for a sample with 50% chlorophylls showed similar results to those of a sample without $TiO_2$. As such, combining a spray coating and $TiO_2$ incorporation into gas separation membrane systems are expected to be useful to understand the fundamentals of material properties for their applications as oxygen generation membranes and air filtration systems.

Stabilization of Membrane Proteins by Benzyladenine during Wheat Leaf Senescence (노쇠중인 밀잎에서 Benzyladenine에 의한 막단백질의 안정화)

  • The effect of benzyladenine (BA) on lipid peroxidation and compositions of total insoluble proteins and chloroplast thylakoid protein from wheat primary leaves during senescence in the dark was studied. BA ($10^{-5}\;M$) treatment prevented conspicuously the loss of chlorophyll content and soluble and insoluble leaf protein contents in senescing wheat leaf segments during 4-day dark incubation. Under the BA treatment, especially, the level of insoluble protein was highly maintained than that of soluble protein. Also, the increase of malondialdehyde (MDA: the peroxidation product of membrane lipids) content was inhibited in the BA treated leaves. Three major polypeptide bands in quantity corresponding to 57, 26 and 12 KD molecular weight were clearly resolved with other minor bands by SDS-polyacrylamide gel electrophoresis (SDS-PAGE) in the insoluble protein fraction. The insoluble protein profiles of the control leaves showed a remarkable decrease in the intensity of the 57 and 12 KD band except for 26 KD band in the 72 h dark incubation. This loss during dark incubation was reduced by BA treatment. More than 20 polypeptides were resolved in the chloroplast thylakoid membrane fraction with the most prominent bands which are 59 and 57 KD ($\alpha\;and\;\beta$ subunit of coupling factor: CF) and 26 KD (apoprotein of LHCP). The changes in thylakoid protein profile during 72 h dark incubation showed the rapid degradation in control, but this degradation was prevented in quantity by BA treatment. The above results suggested that BA would inhibit the peroxidation of membrane lipids, thereby preventing the loss of membrane proteins which led to the maintenance of the membrane integrity including chloroplast thylakoid.

PCR-based markers to select plastid genotypes of Solanum acaule (Solanum acaule 색소체 유전자형 선발을 위한 특이적 분자마커 개발)

  • The tetraploid Solanum acaule is a wild potato species from Bolivia widely used for potato breeding because of its diverse attractive traits, including resistance to frost, late blight, potato virus X, potato virus Y, potato leafroll virus, potato spindle tuber viroid, and cyst nematode. However, the introgression of useful traits into cultivated potatoes via crossing has been limited by differences in endosperm balance number between species. Somatic fusion could be used to overcome sexual reproduction barriers and the development of molecular markers is essential to select proper fusion products. The chloroplast genome of S. acaule was sequenced using next-generation sequencing technology and specific markers for S. acaule were developed by comparing the obtained sequence with those of seven other Solanum species. The total length of the chloroplast genome is 155,570 bp, and 158 genes were annotated. Structure and gene content were very similar to other Solanum species and maximum likelihood phylogenetic analysis with 12 other species belonging to the Solanaceae family revealed that S. acaule is very closely related to other Solanum species. Sequence alignment with the chloroplast genome of seven other Solanum species revealed four InDels and 79 SNPs specific to S. acaule. Based on these InDel and SNP regions, one SCAR marker and one CAPS marker were developed to discriminate S. acaule from other Solanum species. These results will aid in exploring evolutionary aspects of Solanum species and accelerating potato breeding using S. acaule.
