Seed Production by Induction of Male Flowers on Female Plants of Hemp(Cannabis saliva L.) (대마 자주의 웅화 유도에 의한 자성종자 생산에 관한 연구)

  • This study was conducted to overcome the deliciousness by sex reversal in hemp plants. When female plants grown under long day length condition were transferred to short day length condition in 3th and 6th leaf stage, a few male flowers were formed and the gap of flowering days between sex reversed male and normal female flowers were around 10 days and 3 days, respectively. Although the flowering amounts were small, this short gaps of flowering days were enough for artificial pollination. When female plants grown under long day length condition were transferred to short day length condition just after treatment of silver nitrate in the vegetative stage, a great amount of male flower was formed and the gap of flowering days between sex reversed male and normal female flowers were only 1 to 3 days. The increased sex reversed male flowers and induced short gap of flowering days were adequate for natural wind pollination. When female seeds were cultivated, seed yield was 141 kg/10a, while it was 96 kg/10a when normal seeds cultivated.

Herbicidal Response and Control of Scirpus juncoides Roxb. Resistant to Sulfonylurea Herbicides (Sulfonyurea계 제초제 저항성 올챙이고랭이(Scirpus juncoides Roxb.)의 제초제 반응과 방제)

  • This study was conducted to investigate the herbicidal response and effective control strategy of sulfonylurea(SU)-resistant Scirpus juncoides Roxb. occurred in the paddy fields of Korea. A biotype of Scirpus juncoides Roxb. resistant to SU was identified in the paddy fields treated with SU herbicide-based mixtures for seven consecutive years. The apparent SU resistance observed in Scirpus juncoides Roxb. was completely confirmed in greenhouse tests. The susceptible biotype was almost controlled at the recommended dose of all the tested, but the resistant biotype was survived 20 to 30% even at 10 times higher dose of each the recommended dose of SU herbicides. The $GR_{50}$ values of 4 SU herbicides for the resistant biotype were 53 to 88 times higher than those for the susceptible biotype. The acetolactate synthase(ALS) isolated from the resistant biotype against bensulfuron-methyl and pyrazosulfuron-ethyl was less sensitive than that of the susceptible biotype. The $I_{50}$ values of the resistant biotype against bensulfuron-methyl and pyrazosulfuron-ethyl were 498 and 126 times higher than those for the susceptible biotype. A rapid diagnosis for identifying resistance of Scirpus juncoides Roxb. was possible within at least 3 days after SU herbicides. Three herbicides having different mode of action from SU herbicide, carfentrazone-ethyl, pyrazolate and simetryne exhibited excellent controlling effects on the resistant biotype of Scirpus juncoides Roxb. till 3.5 leaf stage. Among the SU-based herbicides, pyriminobac-methyl+pyrazosulfuroil-ethyl+carfentrazone-ethyl GR and azimsulfuron+carfentrazone-ethyl+pyriminobac-methyl GR were very effective to control resistant biotype of Scirpus juncoides Roxb. without rice injury. The resistant biotype which were not controlled with SU herbicise-based herbicides survived from the fields were effectively controlled by bentazone SL.

The Palliative Effect of Endobronchial Brachytherapy for Previously Irradiated Patients with Lung Cancer (이전에 외부방사선치료를 받은 폐암 환자에서 기관지내 근접치료의 고식적 효과)

  • Purpose: To evaluate the palliative effect of endobronchial brachytherapy (EBB) for patients with lung cancer that previously received external beam radiotherapy (EBRT). Materials and Methods: From July 1992 to May 2003, 29 patients with a recurrent or persistent lung cancer were treated with palliative EBB at our institute. EBB consisted of three fractions (once a week) of a dose of 5 Gy using the high dose-rate remote afterloader. Symptomatic improvement was assessed subjectively, and patients were divided into two groups according to whether symptoms were improved or not. Factors such as age, performance status, duration from EBRT to EBB and the location of the tumor were compared between the improved and unimproved groups of patients. Results: Overall symptomatic improvement was found in 27 out of 52 symptoms (52%). Improvement as to the type of symptoms was seen in 41 %, 50%, 82% and 33% of patients with cough, dyspnea, hemoptysis, and obstructive pneumonia respectively. The rate of improvement of hemoptysis was more than that of cough (p<0.05). The median time to symptom relapse was 5 months. The improved patient group (n=17, 59%) had a better performance status and longer duration from EBRT to EBB than the unimproved patient group (p<0.05). Lesions located in the distal trachea and/or main bronchus were found more frequently in the improved group of patients than in the unimproved group of patients, but the difference was not statistically significant (p=0.06). Fatal complications developed in two patients (7%), which were a hemoptysis and bronchopleural fistula respectively. Conclusion: Symptom improvement was found in 60% of patients after EBB and improvement was maintained for 5 months. Palliative EBB, even when EBRT was given previously, can be effective for a patient that has an endobronchial symptom, such as hemoptysis, and for a patient with good performance and a long duration from previous EBRT to EBB.

A Clinical Review of Mucoepidermoid Carcinoma of The Lung in Korea (점액상피암의 임상적 고찰)

  • Background: Mucoepidermoid carcinoma of the lung arises from submucosal gland of tracheobronchial tree. Histologically, the tumor is composed of mucin-secreting cells, squamous cells, and intermediated cells, which show no particular differentiating characteristics, in varying proportions. The tumor is divided into low grade and high grade depending on the proportion of cells, and the degree of the mitotic activity, cellular necrosis and nuclear pleomorphism. While favorable prognosis of low grade tumor, high grade tumor, which is very difficult to differentiate from adenosquamous carcinoma, has an aggressive clinical course. The tumor is rare, comprising 0.1 to 0.2% of primary lung cancers and 1 to 5% of bronchial adenomas. Method: A retrospective clinical study was done on 17 cases of mucoepidermoid carcinoma. The study investigated the clinical features, radiologic findings, bronchoscopic findings, histology and clinical courses. Results: Age ranged between second to seventh decade with a mean age of 42 years. Twelve out of 17 cases were male. Five out of 17 cases were smokers with a mean 11 pack-years. Common symptoms included dyspnea, cough, hemoptysis, and wheezing. Two out of 17 cases was asymptomatic. Atelectasis or mass was common radiologic finding. Plain chest radiography was normal in one patient whom the tumor was located in upper trachea. Bonchoscopy revealed exophytic mass in 12 cases and nodular infiltrations in 4 cases. One case having solitary pulmonary nodule in the right lower lung was normal on bronchoscopy. Histologically, ten out of 17 cases were low grade, and seven out of 17 cases were high grade. Among 10 patients with low grade tumor,9 patients were performed operation and have been alive without recurrence during a mean follow-up of 30 months. Two out of 7 patients with high grade tumor were performed pneumonectomy and have been alive during a follow-up of 3 and 8 months, respectively. Conclusion: Most of mucoepidermoid carcinoma is located at central airway and is presented symptoms by mucosal irirtation. Although atelectasis or mass is common radiologic finding. chest X -ray can be normal. The histologic grading and the extent of tumor are two most important factors for prognosis.

Plant Regeneration of Bupleurum spp. through Somatic Tissue Culture (자호(紫胡)의 체세포조직배양(體細胞組織培養)에 의한 식물체재분화(植物體再分化))

  • This study was conducted to determine the optimum conditions of inducing callus, proliferating callus, forming somatic embryos, and regenerating plantlets via somatic embryogenesis, for the purpose of producing artificial seeds and substantially developing plant factory technologies that can be employed to all seasons production of Bupleurum plants. Callus was efficiently induced from leaf tissues at three leaf stage in the MS medium supplemented with 2, 4-D 2mg /1 and thidiazuron(TDZ) 0.lmg /1. Callus induction from leaf tissues at maturity was mostly effective in the mixture of 2,4- D 2mg /1 and TDZ 1.0mg /1 while that from flower bud tissues was fairly good in the MS medium containing 2,4-D 1 or 2mg /1.Callus was formed in 15 to 20 days after culture initiation in the MS media supplemented with 2, 4- D 1-2mg /1 and TDZ 0.l-1.0mg /1. Such hormones as kinetin 3mg /1, GA 1mg /1, and the mixture of GA 1mg /1 and TDZ 1mg /1 effected markedly to proliferate the callus cells.The optimum temperature and light intensity for callus culture were found to be $25^{\circ}C$ and 3000 Lux, respectively. Direct plant regeneration from cultured callus was fairly made on hormone-free MS or half-strength MS medium. Somatic embryogenesis was most frequently observed in hormone-free media:60 somatic embryos per 20ml in MS medium and 28 somatic embryos per 20ml in half -strength MS medium. There were three stages-globular, heart, and torpedo-in development of somatic embryos, among which globular stage was more frequently observed in MS medium rather than in half-strength MS medium. Somatic embryos induced from suspension culture fairly differentiated a number of shoots and roots on hormone-free and half-strength MS solid medium.

Physiological and Proteome Responses of Korean F1 maize (Zea mays L.) Hybrids to Water-deficit Stress during Tassel Initiation (옥수수 영양생장기 한발 스트레스에 의한 광합성의 생리적 반응 및 프로테옴 변화 분석)

  • Severe droughts in spring have occurred frequently in Korea in recent years, exerting a critical impact on corn yield. Therefore, it is necessary to find physiological and/or molecular indicators of the response to drought stress in maize plants. In this study, we investigated the effects of water-deficit stress on two Korean elite F1 maize hybrids, Ilmichal and Gwangpyeongok, by withholding water for 10 days at tassel initiation. The water deficit drastically reduced the relative leaf water content, leaf number, leaf area, and stem length, leading to dry matter reduction. Moreover, it reduced the SPAD values and stomatal conductance of leaves in drought-stressed plants of both hybrids. Importantly, the number of leaves and SPAD value were non-destructive and easy to investigate in response to water-deficit stress, suggesting that they may be useful indicators for screening drought-tolerant genetic resources. We detected more than 100 spots that were differentially accumulated under drought stress. Of these spots, a total of 21 protein spots (≥1.5-fold) from drought-exposed maize leaves were successfully analyzed by MALDI-TOF-TOF mass spectrometry. Functional annotation using Gene Ontology analysis revealed that most of the identified proteins were involved in carbohydrate metabolism, stress response fatty acid catabolism, photosynthesis, energy metabolism, and transport. The protein expression levels were increased in both Ilmichal and Gwangpyeongok, except for triosephosphate isomerase, fructose-bisphosphate aldolase, and an uncharacterized protein. The lactoylglutathione lyase delta (3,5)-delta (2,4)-dienoyl-CoA isomerase was overexpressed in Gwangpyeongok only. The results obtained from this study suggest that the drought-specific genes may be useful as molecular markers for screening drought-tolerant maize genotypes.

Controlling Effect of Agricultural Organic Materials on Phytophthora Blight and Anthracnose in Red Pepper (고추 역병과 탄저병에 대한 친환경유기농자재의 방제 효과)

  • A total of 20 agricultural organic materials including inorganic compounds, plant oils and plant extracts were used in the study for assessing the control efficacy on pepper diseases. Among inorganic compounds, only copper hydroxide showed inhibitory effect on both Phytophthora capsici causing Phytophthora blight and Colletotrichum acutatum causing anthracnose. Phosphorous acid inhibited the growth of P. capsici on PDA, and Sulfur/quicklime had it on that of C. acutatum. Plant essential oil, rosemary oil, and rapeseed oil among plant oils and plant extract of Japanese apricot/ginkgo nut inhibited the mycelial growth of the two pathogens. In the screening using pepper plant seedlings, the control efficacy on Phytophthora blight in 6-leaf stage of seedling was superior to that in 4-leaf stage of seedling. A protective effect on Phytophthora blight was displayed by copper hydroxide, sulfur/quicklime, water soluble calcium, phosphorous acid, plant essential oil, and cloves extract. When C. acutatum was inoculated by the non-wound method, copper hydroxide and rapeseed oil showed excellent protective activities with control values of 91.3% and 82.6%, respectively. However, copper hydroxide did not show any activity, when C. acutatum was inoculated after wounding pepper fruits. All organic materials never showed the curative effect on Phytophthora blight and anthracnose in pepper seedling assay and fruit assay.

Herbicidal Efficacy of Newly Developed Several Herbicides in Rice (최근(最近) 개발(開發)된 수도용(水稻用) 제초제(除草劑)의 작용성(作用性)과 제초효과(除草效果))

  • Newly developed several herbicides were evaluated as paddy rice herbicide at the Yeongnam Crop Experiment in 1986. And also, the general situation of rice cultivation between Korea and Japan was compared. Twenty-nine herbicides of the total 59 herbicides were used as paddy rice field in Korea while these were 100 and 187, respectively, in Japan. Among paddy rice herbicides, butachlor was the most important herbicide in both countries. However, the degree of concentration to a particular herbicide was greater in Korea compared to Japan; consumption rate of single butachlor to the total herbicide were 66.5% for Korea and 11.9%r for Japan, respectively. Pyrazolate, pyrazoxyfen and quinclorac were the most promising hebicides in pressed-type rice nurserybed in terms of herbicidal efficacy and phytotoxic effect. For transplanted paddy rice field, single application of NC-311 or combining applications of NC-311 with butachlor or quinclorac gave excellent results at the weed community that was dominated by Echinochloa crus-galli, Aneilema japonica, Ludwigia prostrata, Scirpus hotarui, Cyperus serotinus, Potamogeton distinctus and Eleocharis kuroguwai. Particularly the above combining applications maintained their excellent herbicidal effect up to 3 leaf stage of E. crus-galli without arising herbicidal phytotoxicity. Pyrazolate and three sulfonyl urea herbicides (DPX-5384, NC-311 and CGA 142464) exhibited very high safety against rice. However, Japonica type rice cultivar was a little bit more sensitive than Indica/Japonica type rice cultivar. On the other hand, the effect of these herbicides against E. crus-galli was very strong. Herbicidal effect against E. crus-galli was enhanced through shoot absorption for sulfonyl urea herbicides and root abosorption for pyrazolate and quinclorac, respectively.

Physiological and Ecological Studies on the Low Temperature Damages of Rice (Oryza sativia L.) (수도의 저온장해에 관한 생리 생태학적 연구)

  • Experiments were conducted to investigate rice varietal response to low water and air temperatures at different growth stages from 1975 to 1980 in a phytotron in Suweon and in a cold water nursery in Chooncheon. Germination ability, seedling growth, sterility of laspikelets, panicle exertion, discoloration of leaves, and delay of heading of recently developed indica/japonica cross(I/J), japonica, and indica varieties at low air temperature or cold water were compared to those at normal temperature or natural conditions. The results are summarized as follows: 1. Practically acceptable germination rate of 70% was obtained in 10 days after initiation of germination test at 15\circ_C for japonica varieties, but 15 days for IxJ varieties. Varietal differences in germination ability at suboptimal temperature was greatest at 16\circ_C for 6 days. 2. Cold injury of rice seedlings was most severe at the 3.0-and 3.5-leaf stage and it was reduced as growth stage advanced. A significant positive correlation was observed between cold injury at 3-leaf stage and 6-leaf stage. 3. At day/night temperatures of 15/10\circ_C seedlings of both japonica and I/J varieties were dead in 42 days. At 20/15\circ_C japonica varieties produced tillers actively, but tillering of I/J varieties was retarded a little. At 25/15\circ_C, both japonica and I/J varieties produced tillers most actively. Increase in plant height was proportional to the increase in all varieties. 4. In I/J varieties the number of differentiated panicle rachis branches and spikelets was reduced at a day-night temperature of 20-15\circ_C compared to 25-20 or 30-25\circ_C, but not in japonica varieties although panicle exertion was retarded at 20-15\circ_C. The number of spikelets was not correlated with the number of primary rachis branches, but positively correlated with that of secondary rachis branches. 5. Heading of rice varieties treated with 15\circ_C air temperature at meiotic stage was delayed compared to that at tillering stage by 1-3 days and heading was delayed as duration of low temperature treatment increased. 6. At cold water treatment of 17\circ_C from tillering to heading stage, heading of japonica, I/J, and cold tolerant indica varieties was delayed 2-6, 3-9, and 4-5 days, respectively, Growth stage sensitive to delay of heading delay at water treatment were tillering stage, meiotic stage, and booting tage in that order, delay of heading was greater in indica corssed japonica(Suweon 264), japonica(Suweon 235), and cold tolerant indica(Lengkwang) varieties in that order. Delay of heading due to cold water treatment was positively correlated with culm length reduction and spikelet sterility. 7. Elongation of culms and exertion of panicles of rice varieties treated with low air temperature 17\circ_C. Culm length reduction rate of tall varieties was lower than that of short statured varieties at low temperature. Panicle exertion was most severaly retarded with low temperature treatment at heading stage. Generally, retardation of panicle exertion of 1/1 varieties was more severe than that of japonica varieties at low temperature. There was a positive correlation between panicle exertion and culm length at low temperature. 8. The number of panicles was increased with cold water treatment at tillering stage, but reduced at meiotic stage. As time of cold water treatment was conducted at earlier growth stage, culm length was shorter and panicle exertion poorer. 9. Sterility of all rice varieties was negligible at 17\circ_C for three days but 30.3-85.2% of strility was observed for nine-day treatment at 17\circ_C. Among the tested varieties, sterility of Suweon 264 and Milyang 42 was highest and that of Suweon 290 and Suweon 287 was lowest. The most sensitive growth stage to low temperature induced sterility was from 15 to 5 days before heading. There was positive correlation between sterility of rice plants treated with low temperature at meiotic and heading stage. 10. Percentage of spikelet sterility was greatest at cold water treatment at meiotic stage (auricle distance -15~-10cm) and it was higher in 1/1 (Suweon 264, Joseng tongil), japonica (Nongbaek, Towada), and cold tolerance indica(Lengkwang) varieties in the order. Level of cold water and position of young-ear affected on the sterility of varieties at meiotic stage; percentage of spikelet sterility of variety, Lengkwang, of which young-ear was located above the cold water level was high, but that of short statured variety, Suweon 264, of which young-ear was located in the cold water was lower. 11. Percentage of ripened grains was not reducted at 15\circ_C air temperature for three days at full heading stage in all varieties. However, at six-day low temperature treatment Suweon 287, Suweon 264 showed percentage of ripended grains lower than 60%, but at nine-day low temperature treatment all varieties showed percentage of ripened grains lower than 60%. Low temperature treatment of 17\circ_C from 10 days after heading for 20 days did not affect on the ripening of all varieties. 12. Uptake of nitrogen, phosphorous, potassium, calcium, and magnesium in whole plants was higher at average air temperature of 25\circ_C, but concentration of the elements was lower compared to those at 19\circ_C. However, both total uptake and concentration of manganese were higher at 19\circ_C compared to 25\circ_C. 13. Higher application of nitrogen, phosphorus, silicate, and compost increased yield of rice due to increased number of panicles and spike let fertility in cold water irrigated paddy.

Antagonistic Interaction between Quinclorac and Bensulfuron-methyl on Growth of the Rice Plants (Quinclorac과 Bensulfuron-methyl의 혼합처리(混合處理)에서 벼의 생장(生長)에 대한 제초제간(除草劑間) 길항작용(拮抗作用))

  • Field and pot expeiments were carried out to evaluate the interaction between quinclorac and bensulfuron-methyl on growth of the rice plants(Oryza sativa L. cv. Choocheongbyeo) at 20, 45, 65 days-old stages. Quinclorac and bensulfuron-methyl showed antagonistic interaction at both stages, which were detected by the Chisaka's method at isobles of 10% growth inhibition. The antagonism indices were -0.63 and -1.67 at 20 and 65 days-old seedling stages, respectively. Leaf-rolling of rice occurred when quinclorac was applied at 600g ai/ha or more at 20 days-old seedling stage, while it occured at the dose of 900g ai/ha at 65 days-old stage. Bensulfuron-methyl reduced plant height and dry weight as well as tiller production at both stages. Leaf-rolling of rice was reduced when mixture of quinclorac and bensulfuron-methyl was applied due to antagonism of the two herbicides. High temperatures increased the phytotoxicity of bensulfuron-methyl, while the phytotoxicity caused by quinclorac alone was not responsive to temperature. The antagonistic effect between quinclorac and bensulfuron-methyl increased at low temperature as tested by the Colby's method.
