Immuno-modulatory and Antitumor Effect of Crude Polysaccharides Extracted from Sclerotium of Grifola umbellata (저령(Grifola umbellata)의 균핵에서 추출한 조다당류의 면역활성 및 항암 효과)

  • Neutral salt-soluble, hot water-soluble and methanol-soluble materials (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from sclerotium of Grifola umbellata. The Fr. NaCl and Fr. HW did not show any direct cytotoxicity against NIH3T3, Sarcoma 180 and MCF-7, but Fr. MeOH showed cytotoxicity against these cell lines at the concentration of $1,000\;{\mu}g/ml$. Intraperitoneal injection with Fr. NaCl showed antitumor effect with life prolongation of 66.7% and decrease the number of Sarcoma 180 cells of 54.2% in mice inoculated with Sarcoma 180. Fr. NaCl improved the immunopotentiation activity through alternative complement pathway and the alkaline phosphatase activity by $85.05{\sim}88.73%$ and 6 folds, respectively. The number of peritoneal exudate cells and the circulating leucocytes were increased by 1.7 and 3.6 folds in the Fr. NaCl treating group compared with the control group, respectively. The weight of immunoorgans such as liver, spleen and thymus were also gradually increased. The hematological analysis of the Fr. NaCl group was similar with that of the control group. The total polysaccharide and protein contents of Fr. NaCl were 98.25% and 1.44%, respectively. These results indicate that the antitumor activity of Fr. NaCl was exerted through immunopotentiation, but not through cytotoxicity against the tumor cells.

향류식 역삼투 농축공정을 이용한 NaCl 용액의 농축연구

  • 제염농축공정은 증류, 증발법을 일반적으로 사용하나, 에너지 절약차원에서 전기투석 (Electro dialysis)장치를 증발기 선단에 도입한 복합공정을 국내에서도 사용하고 있는 실정이다. 그러나, 전기투석장치는 전기적인 소모와 막의 재생, 교체처리비가 문제점이 되므로 역삼투 장치를 제염농축 공정 최선단에 도입한 복합공정을 이용할 경우 전기 투석 및 증발 복합공정에 비해 40%의 에너지 절감 효과를 기대할 수 있다. 이와 같은 장점에도 불구하고 역삼투 공정은 공급용액의 삼투압보다 큰 적용압력을 막표면에 가하여 물질분리를 수행하므로 농축공정에서 유발되는 배제액 농도의 상승은 삼투압의 증가를 일으켜 실적적용압력의 효과를 떨어뜨리게 되며 결과적으로 농축효과를 감소시키게 된다. 본 연구에서는 효과적인 염농축 공정을 위하여 막모듈 투과부에 고농도 삼투압 감소액(osmotic sink solution)을 향류식(막투과흐름을 맞받아치며 흐르는 방식)으로 유입시키는 향류식 역삼투 (counter-current reverse osmosis, CCRO) 나권형 모듈을 고안 제작하였으며, 제작된 모듈을 기존 역삼투 공정과 향류식 염삼투 공정에 적용하여 염농축도의 성능을 상호 비교하고 염농축에 관계되는 공급농도, 공급유량, 투과유량, 배제유량, 향류 유입유량, 압력구배, 삼투압차 및 농축단수 등의 인자들을 이용하여 두 공정에 대한 염농축 분리조작의 제반조건과 제작된 모듈 내의 농축관련 특성을 실험 및 수치적으로 비교, 고찰하였다.

Growth Inhibitory Effects of $Doenjang$, Prepared with Various Solar Salts, on Cancer Cells (다양한 천일염으로 제조한 된장의 암세포 성장 억제 효과)

  • The growth inhibition effect of $Doenjang$ that was prepared with various kinds of solar salt was investigated. $Doenjang$ was prepared using the bacterial koji and five kind of salt with 12% salt concentration (w/w): purified salt $Doenjang$, one-year aged solar salt $Doenjang$, four-year aged solar salt $Doenjang$, topan solar salt $Doenjang$, and boiled solar salt $Doenjang$. The $Doenjangs$ were fermented and aged for 18 months. The growth inhibition effects of the water extracts and the methanol extracts of the $Doenjangs$ were measured on AGS human gastric adenocarcinoma cells, HT-29 colon carcinoma cells, and BJ human foreskin normal cells using MTT assay. The water and methanol extracts of the $Doenjang$ samples showed growth inhibition effects on the cancer cells, in the following order of the samples with the strongest to the weakest effect: the four-year aged solar salt $Doenjang$, the topan solar salt $Doenjang$, the boiled solar salt $Doenjang$, the one-year aged solar salt $Doenjang$, and the purified salt $Doenjang$. The methanol extracts of the four-year aged solar salt Doenjang (AGS: 55% and HT-29: 48%) showed the strongest growth inhibition effect. In addition, decreased cancer cell numbers and morphological changes in the cancer cells (AGS and HT-29) were observed when the methanol extract of the four-year aged solar salt $Doenjang$ was treated. None of the $Doenjang$ extracts showed a growth inhibition effect on the BJ normal cells, though.

Lactic Acid Fermentation of Dioscorea batatas and Its Anti-Inflammatory Effects on TNBS-induced Colits Model (TNBS에 의해 유도된 마우스 대장염모델에서 유산균 발효 마의 항염효과)

  • To develop a health-aid preparation of Dioscorea batatas (DB), lactic acid fermentation was attempted using a mixed starter comprising of Lactobacillus acidophilus, Lactobacillus plantarum, Bifidobacterium longum. The anaerobic fermentation of a 10% DB flour suspension gave a uniform suspension of pH 3.65, containing $8{\times}10^6$ CFU/mL lactic acid bacteria. During the administration of the lactic acid fermented DB (FDB) and DB to trinitrobenzene sulfonic acid (TNBS)-induced colitis mouse model, histological lesions, morphological damage, and myeloperoxidase acitivity were significantly reduced at a dosage of 200 and 400 mg/kg/day. Dose-response (200 and 400 mg/kg/day) studies revealed that FDB pre-treatment of mice significantly ameliorated the appearance of diarrhoea and the disruption of colonic architecture. In FDB-pretreated mice, there was a significant reduction in the degree of both neutrophil infiltration (measured as decrease in myeloperoxidase activity) and weight loss rates. Theses findings suggest that FDB exerts beneficial effects in experimental colitis and may be useful in the treatment of inflammatory bowel disease.

Tenderness Improvement and Utilization of Low Quality Meat by High Temperature Aging (고온숙성에 의한 저급육의 연도개선과 그 이용)

  • The effect of high temperature aging on the meat tenderness improvement was studied, and also the effect of salt, pyrophosphate and succinic anhydride on binding characteristics of restructed beef were compared. At high temperature aging, shear force value decreased and myofibrillar fragmentation index increased as the aging progressed. From the electronic microscopic observation, the morphological change of myofibril appeared much faster when the meat was aged at high temperature. Added salt increased TBA values and rupture strength while reducing cooking loss. Increase in pyrophosphate decreased rooking loss and Increased rupture strength and TBA value. When salt and pyrophosphate were combined, the effects were somewhat additive. Added succinic anhydride increased cooking loss and hardness and decreased color rating, acceptability rating and adhesiveness, but cohessiveness was not significantly different from control group containing salt and pyrophosphate. The results suggest that high temperature aging have greater improving effect of meat tenderness of Korean native male cattle compared to low temperature aging and addition of succinic ahydride in combination with salt and pyrophosphate reduce binding ability of restructured beef.

Effect of Salt Type and Concentration on the Growth of Lactic Acid Bacteria Isolated from Kimchi (소금의 종류와 농도가 배추김치에서 분리한 유산균의 생육에 미치는 영향)

  • Tests show that the growth of lactic bacteria in kimchi varies according to the type and concentration of salt used. Weissella confusa, the early stage bacteria in kimchi fermentation, increased sharply after 5 hr of induction with 3% light salt and refined salt. However, the induction period lengthened to 12 hr with 3% sea salt and bamboo salt. Lactobacillus delbrueckii ss lactis and L. pentosus which grow in the middle stage of fermentation, and L. hamsteri, which grows at the end stage of fermentation, were found after 12 hr of induction with 3% salt of all kinds. When 5% light salt was added to the culture medium, the induction period of bacteria other than W. confusa lengthens to 12 hr. The trend is similar for sea salt, bamboo salt, and refined salt, with a higher NaCl concentration resulting in less growth. W. confusa showed salt tolerance, but L. hamsteri was affected by the type and concentration of salt. With 5% sea salt, bamboo salt, and refined salt, the growth of bacteria was inhibited by up to 24 hr.

Improvement Effect of the Eucommia ulmoides Extracts on CIA-induced Rheumatoid Arthritis Animal Models (두충 추출물의 류마티스관절염 동물모델에 대한 개선 효과)

  • The present study aimed to evaluate the effect of Eucommia ulmoides extracts on rheumatoid arthritis biomarker in a CIA-induced DBA/1 mice. For evaluation, Eucommia ulmoides extracts was administered orally at dose of 100 mg/kg/day for 4 weeks after production of an animal model of rheumatoid arthritis and we confirmed the treatments' effects based on serum biomarker, radiological, structural parameter analysis. Compared to the negative control group, the Eucommia ulmoides extracts treatments significantly reduced the serum level of inflammation and immunoglobulin markers (i.e., TNF-α, IgG, and hs-CRP), and significantly decreased the monocyte count of white blood cells. Furthermore, the Eucommia ulmoides extracts treatments effectively preserved the joint destruction, and little the joint deformation. Moreover, compared to the negative control group, the Eucommia ulmoides extracts treatments increased the bone volume, and significantly decreased bone inflammation. The results indicate that the Eucommia ulmoides extracts improved rhrumatoid arthritis symptoms. Thus, the Eucommia ulmoides extracts may be a novel therapeutic option for the management of rheumatoid arthritis.

Attachment of Bacillus subtilis to Al-Fe Bimetallic Oxide-coated Sand : Effect of Oxyanions (알루미늄.철 산화물 동시피복모래에서 Bacillus subtilis의 부착: 산화음이온의 영향)

  • This study investigated the influence of oxyanions (nitrate, carbonate, phosphate) on the attachment of bacteria (Bacillus subtilis) to Al-Fe bimetallic oxide-coated sand using column experiments. Results showed that bacterial attachment to the coated sand was independent of nitrate concentration. Bacterial mass recovery remained constant (10.9${\pm}$0.2%) with varying nitrate concentrations (0.1, 1, 10 mM). In case of carbonate, mass recovery increased from 25.6% to 39.0% with increasing carbonate concentration from 0.1 mM to 1 mM, and mass recovery also increased from 50.9% to 78.9% at the same concentration condition in case of phosphate. This phenomenon could be attributed to the hindrance effect of carbonate and phosphate to bacterial attachment to the coated sand. Meanwhile, with increasing carbonate/phosphate concentration from 1 mM to 10 mM, mass recovery decreased from 39.0% to 23.8% and from 78.9% to 52.6%, respectively. This phenomenon could be ascribed to the enhancement effect of free carbonate/phosphate ions present in solution phase due to increasing carbonate/phosphate concentration, which increase ionic strength and thus enhance bacterial attachment to the coated sand. In our experimental conditions, the effect of phosphate to bacterial attachment to the coated sand was the greatest among phosphate, carbonate, and nitrate.
