Sexing and Cell Cycle Induction Hanwoo Fetal Fibroblast Cells (한우 섬유아세포의 성 판별 및 세포주기 유도 분석)

  • For somatic cell nuclear transfer in Hanwoo, fetal fibroblast cell lines were established from 35, 50, 70 and 90-day fetuses of Korean native cattle. The sex of these fetal fibroblast cells were analyzed by PCR using Y-specific primers and confirmed that two cell lines were female and the other two cell lines were male. Karyotyping of these cell lines indicates that the chromosome numbers of fetal fibroblast cells were not affected by passage number and more than 80% of fetal fibroblast cells have normal chromosome number. To evaluate Go stage in cell cycle of fetal fibroblast cells, Western blotting was performed to detect the expression level of PCNA which is known to be expressed in all cell cycle stages except G$_{0}$ stage. Following serum starvation or confluent culture for 7 days, fetal fibroblast cells were effectively reached to G$_{0}$ stage. The cell cycle was resumed after culture of these Go stage-fetal fibroblast cells with normal medium. These results indicates that fetal fibroblast cells originated from Hanwoo were successfully isolated and culture system and induction of cell cycle of these cells were established for somatic cell nuclear transfer in Hanwoo.woo.

  • The idiogram of pine chromosomes was obtained from the length, the ratio of the long and short arm, and the position of the secondary constriction. The descending order of the long arm was found by analyzing the idiogram for 6 species of hard pines and 5 species of soft pines growing in Korea. The basic chromosome number of the genus Pinus was n=12, of which the ten chromosomes were the M-type showing similar S/L ratio, and the other two short chromosomes were the heterobrachial SM-type and the sub-median centric SM-type. The interspecific identification was able to made by comparing the number and the position of the secondary constriction, and the pattern of descending order of the long arm. The intraspecific variation was also able to be identified by comparing the long arms Descending order among the provenaces. Some differences were found in the chromosomal structures between the hard- and the soft-pines. However, the differences were not apparent as much as those in the morphological characteristics. The results might not be exactly reproducible because of the variable responses of chromosomes depending on concentration of the chemicals, the temperatures and time of the treatments, and the analytical errors during the preparateur preparation.

Taxonomic Study on the Genus Parasenecio (Compositae) of Korea by the Morphology and Somatic Chromosome Numbers (한국산 박쥐나물속(국화과)의 외부형태와 체세포 염색체수에 의한 분류학적 연구)

  • The morphology and somatic chromosome numbers of Korean Parasenecio were investigated to estimate their taxonomic values. Some morphological characters, such as the length of whole plants, the numbers of cauline leaves, the shapes of leaf and petioles, the numbers of involucral bracts, floret numbers per head, presence of trichomes on the style, ratio between wide part and narrow part of floret and the size of achene were found to be a good characters for delimiting each taxa. Based upon these characters, six taxa, Parasenecio auriculala var. auriculata, P. auriculata var. kamtschatica, P. hastata subsp. orientalis, P. koraiensis, P. firmus, P. pseudotaimingasa, were recognized about their distribution in Korea, but 2 taxa, P. adenostyloides, P. auriculata var. matsumurana, were needed to be studied their taxonomic position and distribution in Korea. The somatic chromosome numbers of six taxa, P. adenostyloides, P. auriculata, P. auriculata var. kamtschatica, P. hastata subsp. orientalis, P. firmus, P. pseudotaimingasa were 2n=60 and those of P. firmus and P. pseudotaimingasa were reported first in this study.

CYTOGENETIC ANALYSIS OF CHILDREN WITH AUTISM (자폐장애 환자의 세포유전학적 분석)

  • Twenty nine children with autism and thirty children with mental retardation were examined for association between autism and chromosomal disorders including fragile X. The peripheral blood was cultured in Medium 199 with methotrexate and without methorexate for 70 hours. Thirty metaphase cells in each case were karyotyped in all samples. Chromosomal abnormalities were found in 11 cases(37.9%) of autistic disorder and 10 cases (33.3%) of mental retardation, but in none of fragile(X)(q27.3) from all cases. Chromosomal abnormalities were present on group A, C, D and X in autistic disorder and on group A, B, C, D, E and X in mental retardation. No specific chromosomal region was found in both autistic disorder and mental retardation. Types of chromosomal disorders were only fragile and/or gap but no numerical abnormality was present in all cases. Number of cells which revealed fragile sites were 31 cells(3.6%) out of 870 cells in autistic disorder and 29 cells(3.2%) out of 900 cells in mental retardation Number of cells which revealed gaps were 43 cells(4.9%) out of 870 cells in autistic disorder and 35 cells(3.9%) out of 900 cells in mental retardation. Autistic disorder may not be directly correlated with fragile X but with nonspecific chromosomal breakages from these data.

  • Excised mature embryos of P. koraiensis were plated on a series of G.D. basic media supplemented with growth regulators as follow; $0.1mg/{\ell}$ NAA(M-1); $0.1mg/{\ell}$ NAA and 2,4-D (M-2); $0.1mg/{\ell}$ 2,4-D and BAP (M-3); $0.1mg/{\ell}$ 2,4-D and kinetin (M-4). Cytological study of the callus showed that thercentage of occurrance of diploid cells was observed 52% in M-3 and 36% in M-4, while that was revealed 29% in M-1 and 17% in M-1. The frequency of diploid cells was increased on the M-3 and M-4 media. The stable chromosome state is a crucial factor for organogenesis. Therefore, it can be inferred that the callus tissues cultured on media supplemented with both auxin and cytokinin have the greatest possibility of organogenesis.

  • Rhynchocypris oxycephalus and R. steindachneri show very similar karyotypes: 2n=50(EN=90), consisting of 12 metacentics, 28 submetacentrics and 10 acrocentrics with a gradual decrease in chromosome size, but with significant differences in nuclear DNA content of 2.64 and 2.52 pg/nucleus, respectively (P<0.05). Although the erythrocyte measurement and parameters of two species were similar, R. oxycephalus erythrocyte number was lower than that of R. steindachneri. Mode in karyological evolution within the genus Rhychocypris shows an increase of nuclear DNA without apparent changes in karyotype and erhthrocyte size.

  • A defective lambda transducing phase carrying acetyl CoA carboxylase gene (fabE) from Escherichia coli chromosome (72 min on the current linkage map) has been isolated. A restriction map of the chromosomal region from defective transducing phage was established by digestion with combination of the restriction enzymes. No cleavage site for the enzyme EcoRI was found in this region. Restriction fragments were cloned from defective transducing phage into high copy number plasmid vector pACYC184 to generate hybrid plasmids which were capable of complementation of fabE temperature sensitive mutation. We show here that the fabE gene is located on a 3.4 megadalton Bam HI-SalI fragment with a HindIII site, which lies within the 7.4 megadalton BglIIfragment, by complementation analysis.

Plant Regeneration Through Adventitious Bud Formation and Callus Induction from Scales of Lilium lancifolium Thunb. (참나리 (Lilium lancifolium Thunb.) 인편으로부터 부정아 발생과 캘러스 유도를 통한 식물체 재생)

  • This study was performed to investigate the effects of various media compositions in regeneration of Lilium lancifolium. The adventitious bud initiation from microscale was the best on MS medium supplemented with BAP 1.0 mg/L and NAA 0.1 mg/L after 4 weeks of culture. However, from bulbscales, adventitious bud initiation was the best in dark condition on MS medium supplemented with BAP 0.5 mg/L and NAA 0.1 mg/L. On the other hand, callus induction was found to be the best from the microscales incubated in complete dark condition for 8 weeks on MS medium supplemented with 2,4-D 1.0 mg/L and BAP 0.1 mg/L. The highest plantlet regeneration from callus was obtained after incubation in the light condition for 8 weeks on MS medium supplemented with NAA 0.5 mg/L and BAP 0.1 mg/L. Rooting of shoots was obtained easily on MS medium and the plantlets were transferred to soil pots after 8 weeks. The chromosome analysis of the root tip cells was revealed that the callus-derived plantlets had normal chromosome number, 2n=24. No variation was observed in the morphology of the plantlets.

A New Genetic Algorithm for Shortest Path Routing Problem (최단 경로 라우팅을 위한 새로운 유전자 알고리즘)

  • This paper presents a genetic algorithmic approach to shortest path (SP) routing problem. Variable-length chromosomes (strings) and their genes (parameters) have been used for encoding the problem. The crossover operation that exchanges partial chromosomes (partial-routes) at positionally independent crossing sites and the mutation operation maintain the genetic diversity of the population. The proposed algorithm can cure all the infeasible chromosomes with a simple repair function. Crossover and mutation together provide a search capability that results in improved quality of solution and enhanced rate of convergence. Computer simulations show that the proposed algorithm exhibits a much better quality of solution (route optimality) and a much higher rate of convergence than other algorithms. The results are relatively independent of problem types (network sizes and topologies) for almost all source-destination pairs.
