Effect of extraction conditions on mineral components and physical properties in cinnamon extracts (계피의 추출조건이 추출액의 무기성분과 물리적성질에 미치는 영향)

  • The dried cinnamon was extracted with water and 70% ethanol and studied for the several extraction conditions of temperature, time, solvent addition ratio and number of extraction on mineral components and physical properties of the extracts. Mineral components significantly increased as the extraction temperature increased. More than 8 hours of extraction showed a little improving while solvent ratio (V/W) up to 60 times (V/W) significantly increased the mineral components, particularly Cu, Zn, Fe, Na in water extracts and Fe, Mn, K in 70% ethanol extracts. Absorbance at 285 mm decreased as the temperature raised up to the range of $60{\sim}80^{\circ}C$. The Hunter a-value was much affected with extraction conditions and L-value and transmittance were changed similarily while pH was little changed.

Analysis of Vicamine Using High Performance Liquid Chromatography and Antioxidant Activity of Vincaminor Extract (High performance liquid chromatography를 이용한 빈카민 분석 및 빈카마이너의 항산화능 측정)

  • Vincamine, one of the major indole alkaloids in vincaminor (Vinca minor L.) is commonly used for treating cerebrovascular diseases. The antioxidant activity of vincaminor extracts and vincamine were measured by 1.1-diphenyl-2-picrylhydrazyl (DPPH) and lipid malonaldehyde (MA) assay. Vincaminor leaves were pulverized and extracted with various solvents such as water, methanol, and ethanol. The antioxidant activities of the extracts varied in accordance with solvents and assays. In DPPH assay, the water extract showed the highest antioxidant activity. In lipid MA assay, However, the ethanol extract inhibited MA formation from cod liver oil by 82% at the level of 5,000 ${\mu}g/mL$. Vincamine in the extract was analyzed by high-performance liquid chromatogram and the concentration of vincamine was 0.419$\pm$0.005 ${\mu}g/mL$.

  • The efficiency of mtCOI amplication after DNA extraction of benthic harpacticoid Tigriopus japonicus s.l. was tested under different conditions depending on fixative (99% Ethanol, or 4% Formalin) and additional chemicals (Ludox or Rose Bengal). Each experimental group by the fixative was subdivided into four groups, respectively: 1) Control (fixative only), 2) processed with Ludox HS40, 3) processed with Rose Bengal, and 4) processed with both Ludox HS40 and Rose Bengal. For the 99% ethanol-fixed sample, overall success rate of amplification by PCR was 96% or above, while for the 4% formalin-fixed one, success rate was much lower than those of ethanol-fixed: 1) Control: 27%, 2) Ludox HS40: 3%, 3) Rose Bengal: 7%, and 4) Ludox HS40 and Rose Bengal: 3%. As a result present study verify that 99% ethanol is a proper fixative for DNA extraction in meiofauna organisms.

Conditions for Pigment Extraction from Bokbunja (Rubus coreanus Miquel) Byproducts (복분자 부산물(박) 색소의 추출조건 모니터링)

  • With the aim of using Bokbunja byproducts as natural pigments, characteristics of extracts were investigated after application of different extraction conditions. Total optical density (TOD) of pigment extracts was greatest (9.51 units) when 80% (v/v) ethanol was used as solvent. The polyphenol content was greatest when 40% (v/v) ethanol was used and tended to decrease with higher concentrations of ethanol. The TOD values were increased when citric or lactic acid was added to extracting solvents. Polyphenols were effectively extracted by solvent containing ascorbic acid. As the level of byproducts increased with different extraction procedures, TOD and polyphenol content of byproducts tended to decrease. More than 90% of anthocyanin pigments and polyphenols could be obtained using two extractions with 40% (v/v) ethanol.

  • 중국 장홍화는 해발 1500m~3000m에 자생하는 고원식물이다. 장홍화에 함유되어 있는 크로신은 암세포를 파괴시키어 종양세포를 줄이는 작용을 하며, 카로티노이드 성분이 항산화 활성과 면역을 증진시키어 항암효과를 가져다 준다. 또한 다량의 칼륨을 함유하고 있어서 나트륨배출에 효과적이어서 혈압을 떨어뜨리는 작용을 한다. 중국 홍경천은 극한의 생육환경으로 고산병 예방, 피로회복, 작업능력향상, 운동능력향상과 세균의 저항력향상, 진정작용과 강심작용, 조혈작용, 항산화작용등이 신경전달물질의 활동을 증진시키어서 스트레스 해소와 치매 및 인지력을 높여준다고 알려져 있다. 본 실험에서는 홍경천과 장홍화 혼합추출액의 총 폴리페놀 함량과 항산화 물질 활성을 조사했던 바, 홍경천과 장홍화 혼합추출액에서 양호한 반응을 보였다. 또한 에탄올 추출물과 메탄올 추출물 처리구에서도 매우 양호한 항산화 반응을 나타내었다.

Mass Balance on the Pulping Extracts of Maple Hardwood using High Performance Liquid Chromatography (HPLC 분석기를 이용한 펄프용 단풍나무의 펄핑 추출액에 관한 물질수지)

  • At the University of Maine, a hemicellulose pre-extraction technology is now being investigated to improve pulp yields, reduce the organic and inorganic load for liquor recovery, and create a feed stream for the generation of new biomaterials. It is important to understand the composition of unextracted wood, extracted wood, and pulping extracts in the design of an economically viable pilot-scale ethanol plant. For analysis of wood pulp composition, the total analytical mass balance closure was 100.6, 100.3, and 81.6% for unextracted chips, extracted chips, and pulping extracts from HPLC-H column analysis. Meanwhile, the total analytical mass balance from the HPLC-P column was 97.8, 86.3, and 80.7%, respectively. This slight variability between H- and P-column results for analytical mass balance may be within the experimental error of the measurement. The data generated by this analysis are important to further design work in commercializing this process.

  • 감자는 전세계적으로 섭취되는 주요 작물로서 우리나라에서는 감자의 생즙을 관절염 및 통증을 억제하는 민간요법으로 사용되어 왔다. 본 연구에서는 감자 에탄을 추출물의 항산화 작용을 검토하였다. 감자 에탄올 추출물(0.05~5 mg/mL) 은 1,1-diphenyl-2-picrylhydrazyl radical에 대하여 강한 free radical scavenger 활성을 나타내었다. 또한 감자 추출물(0.5 mg/mL)은 쥐간 균질액 에서 생성 된 TBARS의 양을 유의적으로 억제하였으며 (p<0.05) Fe/sup 2+//ascorbate induction system에서 감자 추출물(0.5~2 mg/mL)은 DNPH 반응으로 측정된 carbonyl 형성을 억제하였다. 항산화 활성과 마찬가지로 감자 추출물의 환원력은 0.5 mg/mL 농도에서 가장 높았고 이는 α-tocopherol의 경우와 비슷하였다. 이 결과는 감자 추출물이 항산화력을 통해 생체에서 작용할 수 있는 가능성을 제시한다.

Effects of Water and Ethanol Extracts from Various Plants on the Germination and Growth of Lettuce Seeds (식물체로부터 추출한 물질이 상추 종자의 발아 및 생장에 미치는 영향)

  • This study was conducted to detect the allelopathic effect of water and ethanol extracts from 4 plant species (Lactuca sativa L., Ambrosia elatior L., Oenothenra odorata Jasp., Ginkgo biloba L.) on germination and growth of lettuce seeds. A little bit higher inhibitory effect was obtained in ethanol extracts rather than water extracts. Allelopathic effects varied in the source of extracts and concentrations of 4 plant species and the highest allelopathic effect appeared in 10% solution, regardless of plant species.

Antioxidative Activity of Ethanol Extraction Fraction from the Korean Red Tail Ginseng (홍미삼 에탄올 추출분획의 항산화 활성)

  • The purpose of this study was to investigate the extraction method and antioxidative activities of phenolic compounds from Korean red tail ginseng. Antioxidative activities of red tail ginseng were evaluated with its ability to donate hydrogen to DPPH, and to inhibit the oxidation of linoleic acid and LDL induced by $H_2O_2$ and $FeCl_2$, respectivly by measuring the MDA formation. Total phenolic compounds expressed as % caffeic acid were 0.80%, 0.12%, 0.06%, 0.03%, 0.01% when red tail ginseng was consecutively extracted with 60% ethanol for 5 times, most of the phenolic compounds was recovered in the extract obtained after 3 times of extraction. The extraction efficacy of 60% ethanol was superior to that of water in extraction phenolic compounds, and the efficacy did not change after evaporating the extract followed by dissolving with water. 60% ethanol extract of red tail ginseng had weak ability to donate hydrogen to DPPH. MDA determination showed the antioxidative effect with inhibition ratio of 72.23% on linoleic acid oxidation by addition of red tail ginseng extract at the concentration of 1,500 ppm. 22.52% of LDL oxidation was inhibited by addition of 250 ppm.

Protective Effect of Selected Amino Acids and Food Extracts on Ethanol Toxicity Decrement in Rat Liver (일부 아미노산과 식품 추출물의 에탄올 간독성에 대한 보호효과)

  • An rat liver enzyme test was carried out in order to investigate preventing effect of selested amino acids and some food extracts on ethanol induced liver toxicity in vitro. Solutions of aspartic acid, arginine, glutamic acid were prepared and treated on ethanol treated rat liver preparation. Protective effect of amino acids on lipid peroxidation was determined. Same experiments were conducted using aqueous extracts of Dried soybean sprout, Dried Alaskan pollack and Ganoderma lucidum. The TBA value indicating the lipid peroxidation decreased significantly (p<0.05) by addition of aspartate, glutamate and arginine, repectively at concentrations of $6.25{\sim}50\;{\mu}g/mL$. Similar results were observed by adding the aqueous extracts of Soybean sprout, dried Alaskan pollack and Ganoderma lucidum. The aqueous extracts added after ethanol treatment presemted more effect than added before the treatment.
