Environmental Factors Related to Mass Moralities of Young Red Seabream (Pagrus major) in the Artificial Seed Production (일본산 참돔, Pagrus major 종묘생산과정중 사육수 변화로 인한 대량폐사)

  • As seed production program is growing prosperously in various fishes in southern Korea, disease problems in larval and juvenile stages have emerged as a new research object. The following results were obtained from investigation about environmental factors related to mass mortalities of young Kinki red seabream, Pagrus major in process of artificial seedling production. Total length of red seabream larvae hatched was 2.93mm, and became 18.83~20.12mm at day 40. The first noticeable mortality of red seabream larvae (7.98~9.37mm) occurred in 25~30 day-old fish with the survival rates of 59.8~60.3%. Thereafter the mortality of larvae decreased, survival rate was 20.5~25.45% in day 40. After 20~30 days, the quality of pond water was bellow II class. During the experimental period COD, $PO_4$-P, $NO_2$-N, $NO_3$-N and $NH_$-N increased up to 3, 7, 34, 6 and 8 times, respectively, compared to initial ones. The number of viable bacteria in pond water and seabream larvae were $6.3{\times}10^6$~$2.3{\times}10^7$ cfu/ml, 4.3~$7.4{\times}10^6$ cfu/g in day 25, respectively. Among the isolated bacteria from the diseased red seabream in day 25, Vibrio spp. was considered to be the causative organism. External symptoms of this disease were floated, spined near the surface and inflated abdomen. When the isolated strain of the Vibrio was bathed to seabream larvae, $LD_50$ of seabream larvae was over $10^6$ cfu/ml of Vibrio spp.

  • Viral hemorrhagic septicemia (VHS) is one of the most serious viral disease of farmed rainbow trout and some marine fishes in Europe and North America. It has been reported in various marine fish species of Asian countries and induced cause mass mortality in Japanese flounder (Paralichthys olivaceus) culturing in Korea. The aims of this study were to monitor VHSV in wild marine fishes and to give critical information for controling the disease through prophylactic methods. Prevalence of the viral disease, geological distribution and reservoir of the virus were investigated using wild marine fishes captured in southern coast and east china sea for two years. (Reverse Transcriptase Polymerase Chain Reaction) RT-PCR results showed that VHSV were detected in 17 (10.6%) out of 160 fish. G gene sequences of viral strains isolated in this study were closely related to that of a reference strain, KVHS01-1, belonging to VHSV genotype Ⅰ. The results suggest that some of wild marine fishes are VHSV carriers and may spread the pathogen directly to fish farmed in coastal area.

  • Red sea bream iridovirus disease (RSIVD) cause massive economic losses in marine aquaculture industry in Korea. The causative agent of this disease (RSIV) infects a wide range of fish species. The aims of this study were to monitor RSIV in wild marine fishes and to give critical information for controling the disease through prophylactic methods. Prevalence of the viral disease, geological distribution and reservoir of the virus were investigated using wild marine fishes captured in southern coast and east china sea for two years. (Polymerase Chain Reaction) PCR results showed that RSIV were detected in 39 (24.3%) out of 160 fish. MCP gene sequences of viral strains isolated in this study were closely related to that of a reference strain, red seabream-K, belonging to Megalocytivirus subgroup Ⅲ. The results suggest that some of wild marine fishes are RSIV carriers and may spread the pathogen directly to fish farmed in coastal area.

Serological characteristics and antigenicities of Vibrio harveyi isolated from marine cultured fish (해산어류에서 분리된 Vibrio harveyi의 혈청학적 특성과 항원성)

  • Vibrio harveyi was a significant pathogenic agent and cause a high mortality of cultured fish and shrimp in the aquaculture industry. In this study, we have investigated biochemical, physiological, serological and immunological characteristics of V. harveyi isolated from marine cultured fish. The phenotypes of V. harveyi were differentiated with their own biochemical characteristics and colors of colony on the TCBS agar. V. harveyi were classified into more than four serogroups by agglutination test. Most isolates were classified into a group A which is categorized with the same band pattern generated by western blotting. Group A was characterized by a major protein, which is ranged from 26 and 34 kDa in size, and has a virulence to oliver flounder more than reference strain KCCM40866. Oliver flounders vaccined with FKC of V. harveyi C05011 were highly resistant to infection by other strains of group A.

  • The scenario of $CO_2$ disposal in the deep-sea are thought to be possible method to reduce atmospheric $CO_2$ concentrations. However, it is necessary to clarify the effects of elevated $CO_2$ concentrations on both marine organisms and marine ecosystems. In this paper the literatures on the biological impact of elevated $CO_2$ concentrations in seawater and recent studies on the effects of elevated $CO_2$ concentrations on marine animals are reviewed. Elevated $CO_2$ concentrations may affect the physiological functions of marine animals such as acid-base regulation, blood oxygen transport and respiratory system, and ultimately lead to the death of marine animals. Although the fish used in the early studies on $CO_2$ effects are temperate, shallow-water species, deep-sea species should be experimented for the future study on $CO_2$ sequestration in the deep ocean.

Inhibitory Effects of Candidate Probiotic Bacteria on the Growth of Fish Pathogenic Bacteria, Streptococcus sp. (Candidate Probiotic Bacteria의 어류병원성 Streptococcus sp. 성장에 대한 억제 효과)

  • For the treatments and protection of bacterial fish disease, many requirements are needed for aquatic probiotics so that they are effective in aquaculture animals but are also harmless to humans. In the present study, among 17 candidate probiotic bacteria (CPB) obtained from the edible part of the shellfish, Bacillus sp. CPB-St (CPB-St) were selected and in vitro evaluated for the possibility as a probiotic strain for the control of fish streptococcosis which frequently occurs in the olive flounder farms. CPB-St showed inhibitory effects on the growth of various fish pathogenic bacteria, Streptococcus sp., S. parauberis, S. iniae, Lactococcus garvieae and L. piscium by the double layer test ranging about 18~26 mm of clear zone. Inhibitory activity of CPB-St to Streptococcus sp. was observed 6 hours after and the growth of Streptococcus sp. was decreased to 8~55 folds in the co-culture of CPB-St with Streptococcus sp.. The safety of CPB-St to fish and survival of CPB-St in the intestine were assessed in the olive flounder, Paralichthys olivaceus. Fish mortality was not observed in artificial infection with CPB-St for 2 weeks. CPB-St was entirely excreted from the stomach and intestine 24 hours after oral injection. This results indicate that CPB-St has potential applications as a probiotic for the control of fish streptococcosis in aquaculture.

Studies on the mass mortality of the cultured grouper, Epinephelus septemfasciatus (양식(養殖) 능성어, Epinephelus septemfasciatus 대량(大量) 폐사(斃死)에 관(關)한 연구(硏究))

  • In the late summer of 1990 and 1991, mass mortality occured among cage-cultured grouper, Epinephelus septemfasciatus in south cost of Korea. The moribund fish didn't feed and became pale or dark chestnut colour and irregularly swimmed due to the loss of equilibrium, finally the diseased fish fell down side away on the bottom or the surface of cage showing the bent of body and died. The diseased fish showed the extensive hemorrahge in brain, the swelling of spleen and bile duct as the specific syptoms of internal organs. So the gill, skin and other organs of the diseased fish were examined for the presence of pathogenic parasites and bacteria. The parasitic Trichodina sp. were detected only from the gill lamella of the diseased fish, but these parasites seemed to be not a direct causative agents that induced the gross mortality of the cultured grouper. because these parasites were also observed in normal grouper, yellowtail, red seabream and rock bream co-cultured with the diseased grouper in same or near cages. In the viral examination, although isolation of the causative agent by the use of estabilshed cell Lines, RTG-2 and CHSE-214, was not succeed, the normal grouper inoculated intramuscularly with the filtered homogenate of the organs of the diseased fish showed the same external and internal signs with the naturally infected grouper. They died within a week. By using the naturally and the artificially infected fishes, electron microscopic observation revealed numerous hexagonal or polygonal particles in the cytoplasm of liver cells. Based on the these results, we suggest that the mass mortality of the cultured grouper would be occurred by the infection of a viral agent.

Quantitative analysis of the clinical signs in marine fish induced by Megalocytivirus infection (Megalocytivirus 감염 해산 어류에서 나타나는 임상증상의 정량적 변화 분석)

  • In quantitative studies of clinical signs, rock bream of adults and juveniles infected with Megalocytivirus IVS-1 isolated from rock bream (Oplegnathus fasciatus) in Korea showed average $4.49{\pm}1.13$ and $4.85{\pm}1.06$ of spleen index respectively. In challenge experiments, Megalocytivirus IVS-1 induced 100% cumulative mortality in both adult and juvenile rock bream. However we found 60% cumulative mortality in juvenile red sea bream (Pagrus major) even after 30 days of injection, which contradicted with the results of other laboratories. Interestingly, IVS-1 infected red sea bream of the same juvenile size with rock bream showed lower spleen index compared to that of rock bream. In real-time PCR, there was continuous increasing of the numbers of viral copies ($2.03{\times}10^7$ copies/mg) in the spleen of juvenile rock bream infected, which were different from those in adult rock bream showing plateau level after reaching to the peak level. Moreover, enlarged cell numbers in the infected spleen were also increased continuously in the juvenile but not in adult of rock bream, even decreased after reaching to peak level. Consequently, significant differences in clinical signs: cumulative mortality. spleen index and viral copy number were found between rock bream and red sea bream, but not between adult and juvenile rock bream. Certainly quantitative expression of clinical sign as in this study may be a way to compare the progression of megalocitiviral disease more accurately in different species or physiological conditions.

A Case Study of Mortality Caused by Viral Encephalopathy and Retinopathy (VER) in Cultured Sevenband Grouper, Epinephelus septemfasciatus during Winter (겨울철 양식 능성어의 바이러스성뇌망막증(VER) 감염사례)

  • In 2015, a nervous necrosis virus (NNV) was isolated from sevenband grouper, Epinephelus septemfasciatus, maintained in land-based aquaculture system at below $12^{\circ}C$ in winter. Mortality was up to 30% in brood fish, over 4 kg of body weight. Moribund fish showed clinical sings typical of viral encephalopathy and retinopathy (VER), also called viral nervous necrosis (VNN), such as uncoordinated, corkscrew-like swimming behavior, belly-up at rest, darkening of body, cloudy eyeball and hyperinflation of the swim bladder. Aetiology of the disease was confirmed by gross observation of clinical signs, histopathology and molecular diagnosis. Histological studies revealed severe vacuolation and necrosis in the brain. Molecular diagnosis by revere transcription-polymerase chain reaction (RT-PCR) specific to batanodavirus yielded a positive result. The nucleotide sequences of the PCR-amplified fragment were 99.48~100% similar to barfin flounder nervous necrosis virus (BFNNV) genotype and most closely aligned with Pacific cod betanodavirus (PCNNV). This is the first report of natural batanodavirus, NNV infection in sevenband grouper reared in low water temperature during winter (below $12^{\circ}C$) in Korea.

  • 현재 한국수자원공사에서는 시화호로 유입되는 유량을 저류하여 습지내 안정적인 유량을 공급함은 물론, 정화된 하천수를 방류하여 호소내 수질을 개선하고자 동화천 하류 약 840m 지점에 제수문을 설치하여 운영중에 있다. 그러나, 폭우시 상류농경지 침수우려에 따른 제수문 개방후 폐쇄시 제수문 담수어종이 상류로 올라가는 길이 차단되어 제수문 유출부에 어류가 폐사되는 현상이 반복되어 발생하고 있다. 따라서, 한국수자원공사에서는 동화천 제수문 하류부(약 36m)에 어도를 갖춘 수중보시설을 설치함으로써 제수문과 수중보사이에 Pond를 형성하여 어류의 자연사를 방지하고자 하였다. 이에 본 연구에서는 유량규모 및 수문개방 조건에 따른 보 설치 전 후 상황에 대한 1차원 수리특성분석을 실시함으로써, 보 설치로 인해 제수문 상류지역에 배수불량 상황 발생여부를 검토하고, 이를 배제하기 위한 제수문 개방조작 변경안을 제시하였다. 본 연구를 통해 제수문 하류단의 설치예정인 수중보가 제수문 상류지역의 배수상황에 미치는 영향은 제수문 상류 유입량이 저유량 구간인 $3.92cms{\sim}8.88cms$에서 발생하였으며, 이는 본 연구에서 제시한 개방수문 조작 변경안과 같이 간단한 수문조작을 통해 상류 수위를 저감시킬 수 있는 것으로 조사되었다.
