  • 간의 약물대사는 흡수된 외인성물질의 배설을 위한 중추적인 역할을 수행하면 이 반응은 일상대사와 이상대사효소로 구성된 약물대사효소계에 의해 매개된다. 약물대사효소의 발현과 활성은 외인성물질의 노출에 의해 유도되거나 억제되며 이 결과는 약물상호작용을 발생시키는 주요한 원인이다. 또한 당뇨, 비만, 영양실조, 음주, 염증반응 등의 병적인 생리상태는 간에서 약물대사효소의 발현과 활성을 조절하는 것으로 보고되고 있다. 이러한 변동은 치료약물 또는 환경오염물질에 대한 인체의 반응성에 영향을 미치며 결과적으로 예측하지 못한 부작용이나 독성을 발생시킬 수 있다. 본 논문에서는 당뇨병에서 약물대사효소의 발현변화를 정리하였다.

Effects of Trialkyltin in vitro on the Microsomal Monooxygenase System of Digestive Gland in the Clam, Coelomactra antiquata (유기주석화합물이 명주조개 (Coelomactra antiquata)의 약물대사효소계에 미치는 영향)

  • This study was carried out to measure the in vitro interaction of trialkyltin with the microsomal monooxygenase (MFO) system of the clam, Coelomactra antiquata. Cytochrome P450 (CYP) level and 7-ethonvesorufin-O-deethylase (EROD) activity were invetigated in the microsome isolated from digestive gland of the clam (C. antiquata) exposed to tributyltin chloride (TBTC), bis-tributyltinoxide (TBTO) and triphenyltin chloride (TPTC). The specific contents of CYP in clam microsome exposedto 0.4 mM TBTC, TBTO and TPTC for 20 minutes were decreased 52, 72 and $40\%$, respectively, compared to control group. The EROD activities also were inhibited by exposure to TBTO ($92\%$) and TPTC ($85\%$) except for TBTC, The level of CYP and the EROD actintles were decreased according to the OTC exposure concentrations. The toxic effects on the level of CYP and the EROD activities were in order of TPTC>TBTC>TBTO in this study. The measurement of CYP level and EROD activity could be applied as a biomarker for environmental study.

Responses of MFO System in Surf Clam, Pseudocardium sachalinensis, Injected with Sea-Nine 211 Antifoulant (Tin-free 방오제인 Sea-Nine 211에 노출된 북방대합에서 MFO 효소계의 반응)

  • Many alternative biocidal additives were applied to antifouling paint to replace TBT, and Sea-Nine 211 is one of alternating organic booster compounds used in antifouling paint. In this study, extent of Sea-Nine 211 toxicity on marine benthic bivalve is evaluated. Sea-Nine 211 was injected to surf clam, Pseudocardium sachalinensis, that inhabitate northern part of Gangwon Province, Korea. Survival rate of the clam and xenobiotics metabolizing enzyme activities in digestive gland were measured during 4 day-exposure period. The results were compared with those of TBT exposed clam. There were no mortality of clam in the solvent (DMSO) control group and the three Sea-Nine 211 exposure groups (5, 25, 50 mg kg$^{-1}$ body weight), while the clam exposed to 1, 2 and 5 mg kg$^{-1}$ TBT chloride (TBTC) demonstrated 70, 30 and 0% survival rate, respectively. The Sea-Nine 211 exposure group showed a tendency of cytochrome P450 (CYP) induction according to the exposure duration, on the other hand, CYP content was decreased in the TBT exposure group. NADPH cytochrome P450 reductase activity slightly increase according to the exposure duration in the Sea-Nine 211 exposure group, while TBTC inhibit its activity as CYP content. Moreover, there was no significant change of NADH cytochrome b5 reductate activity in the clam epxosed to Sea-Nine 211. In the TBTC exposure group, its activity increased in early exposure period and then significantly decreased the rest of exposure period. All the results indicate that Sea-Nine 211 demonstrated a tendency to induce CYP level, while TBTC inhibits the CYP level, NADPH cytochrome P450 reductase and NADH cytochrome b5 reductase activities.

  • Induction of cytochrome P45O (CYP) and 7-etholqresorufin-O-deethylase (EROD) in the microsome exposed to 3-methylcholan-throne (MC), $\beta$-naphthoflavone (BNF) and phenobarbital-Na (PB) was investigated, Microsome was isolated from digestive gland of clam (Coelomactra antiquata) and then exposed to each chemical in concentration range of 0.1 to 1.0 mM for 7 hours. The CYP content and EROD activity in the microsome exposed to each chemical significantly increased compared to the control group. The overall CYP and EROD induction potency was in order of MC>BNF>PB. The induction response of EROD was two times higher than that of CYP level in the microsome exposed to MC, but the induction response of EROD was slightly higher than that of CYP level in BNF and PB exposure groups.

  • Meliae toosendan Fructus is the fruit of Melia toosendan $S_{IEB}$. et $Z_{UCC}$. (Meliaceae), which is written in oriental terminology as clearing heat and drying dampness, and also explained using liver, stomach and small intestine for channels entered. Among the five fractions prepared from methanol whole extractive of the herb, the chloroform fraction which suggests the presence of triterpenoid, flavonoid and alkaloid stimulated the activities of drug metabolizing enzymes and bile secretion and lowered the serum transaminase activities of liver damaged by carbon tetrachloride.

Responses in Hepatic Xenobiotic Metabolizing and Antioxidant Enzymes in Javelin Goby Acanthogobius hasta Collected at Shihwa Lake (시화호에서 채집한 풀망둑 Acanthogobius hasta의 간장 약물대사효소계 및 항산화계의 반응)

  • The aim of this study was to assess the responses of mixed function oxygenase (MFO) and antioxidative systems of feral Javelin goby, Acanthogobius hasta, caught in two sites of different pollution level in Shihwa lake, which has been a highly polluted lake by organic pollutants from nearby industrial complexes and sites. Enzymes analyzed in phase I of MFO system are cytochrome P450 (CYP), NADPH-cytochrome P450 reductase (P450R), NADH-cytochrome b5 reductase (b5R), and ethoxyresorufin deethylase (EROD). Phase II enzyme of glutathione S-transferase (GST) in MFO system was also investigated. Moreover, oxidative-enzyme system including catalase (CAT), glutathione reductase (GR) and total-glutathione peroxidase (GPX) activities and glutathione concentration in both of oxidized (GSSG) and reduced form (GSH) were determined. P450R, b5R, and GST activities of fish are relatively high in the polluted area, whereas hepatic EROD activity levels of fish in polluted area were lower than those of unpolluted area. CYP concentrations are not different between areas. These results indicated that feral Acanthogobius hasta were adaptive to highly polluted environment and exposed to oxidative stress in Shihwa lake.

Effect of Soybean Supplementation on Murine Drug-metabolizing Enzymes and Benzo(a)pyrene-induced Lung Cancer Develpoment (콩보충식이가 생쥐의 해독효소계 및 Benzo(a)pyrene에 의해서 유도된 폐암발생에 미치는 영향)

  • Soybean has drawn much attention mainly due to its chemopreventive action as well as antiestrogenic effect. Although suppression of breast and prostate cancers were believed to be exerted via antiestrogenic or antiandrogenic activity of genistein, its mechanism of prevention against other cancers has not been clearly demonstrated. We proposed that prevention by soybean from other cancers than sex hormone -related cancers was achieved via modulation of drug-metabolizing enzymes. Addition of acid hydrolysate of 80% methanol extract of soyflour to diet caused a significant induction of quinone reductase, an anticarcinogenic marker enzyme and one of drug-metabolizing enzymes, in mouse lung while it suppressed arylhydrocarbon hydroxylase, involved in bioactivation of procarcinogens, in kidney and small intestine. It is likely that active components exist in a conjugated form and released by acid hydrolysis to be able to affect drug-metabolizing enzyme and exert chemopreventive activity. Benzo(a)pyrene-induced tumor development in mouse lung was greatly reduced by soybean extract supplementation, which is consistent with the extract's capability to modulate favorably arylhydrocarbon hydroxylase and quinone reductase towards chemoprevention.
