Radio-sensitivity of Dark-striped Field Mice, Apodemus agrarius, as a Biological Dosimeter in Radio-ecological Monitoring System (환경 방사선 생물학적 감시 지표로서 야생 등줄쥐의 방사선 감수성)

  • This study examined the possibility of using dark-striped field mice as a biological indicator for the environmental radio-surveillance. For this study, dark-striped field mice were caught from five areas of Kyonggi, Kyongsang, Chungchong and Cholla provinces. The external morphological characteristics and isoenzymic types of dark-striped field mice were studied after they were captured. Among the external morphological characteristics, the dark-brown coat, dark back stripe, head-to-tail length, tail length, and ear length matched the taxonomical characteristics of dark-striped field mice. The analyses on L-lactate dehydrogenase, aspartate aminotransferase, and malate dehydrogenese revealed that one species of dark-striped field mice, called Apodemus agrarius, was inhabitated throughout a wide range of Korea. On the other hand, the frequency of micronuclei in peripheral polychromatic erythrocytes to survived mice after irradiation also analyzed. The LD50/30 of A. cgrarius and ICR mice were approximately 5 Gy and 7.9Gy, respectively. The results of the study reveal that wild A. asrarius have a high potential as a biological monitoring system to determine the impact of radiation in areas such as those within the vicinity of nuclear power plants.

  • Effects of Various Chemical Substrates on Heat Resistance of Isolated EnterobTo pursue the various properties of Enterobacter sp. which may give an outbreak of food-borne disease to man, isolated from the meats and the small intestines of wild mice, and to offer the significant data in the fields of food hygiene and public health this study was carreied, 5 strains of Enterobacter sp. were isolated from the above samples. 5 strains isolated from sample sources were 1 strain (E. aerogenes) from the beeves, 2 strains (E. aerogenes, E intermedium) from the pork and 2 strains (E. aerogenes, E. cloacae) from the small intstines of wild mice. Of total isolatd 89 strains, including non-Enterobacter sp., these numbers of 5 isolates were showed as 5.6% of dection rate. On heating at temperature of $55^{\circ}C$ for 30min., these 5 stratins were generally more resisted in the 0.1M phosphate buffer when such chemical substrates as 0.1M glycine, 5% mannitol or 0.5% sorbitol was added to it whereas they were appeared weaker resistances in 0.1M phosphate buffer when 1% sodium citrate, 1% casein, 10% sodium chloride or 0.1M systein was infused into it.cter sp.

  • In this paper, we applied bayesian hierarchical model to analyze the field mice example introduced by Demster et al.(1981). For this example, we use Gibbs sampler method to provide the posterior mean and compared it with LSE(Least Square Estimator) and MLR(Maximum Likelihood estimator with Random effect) via the EM algorithm.

Morphological Characteristics and Habitat Types of Rattus norvegicus and R. tanezumi Collected in Jeju Island (제주도에서 채집된 Rattus norvegicus와 R. tanezumi의 서식지 유형과 형태적 특성)

  • This study was investigated the morphological characters, secondary sexual dimorphism, and habitat patterns of Korean Rattus animals collected on Jeju Island from April 2005 to October 2012. Two wild rat species, namely R. norvegicus and R. tanezumi were identified on Jeju Island based on morphological characteristics and molecular data; however, R. rattus, which had recorded formerly, was not found in this study. Individuals of R. norvegicus were captured from urban, rural, and natural habitats, while those of R. tanezumi were specially found in animal farms and the surrounding areas. Comparing of morphological characters of two species, R. norvegicus had a shorter tail and ears than R. tanezumi (p<0.05), and the ratios of tail length and ear length to those of head-body length showed significantly differences between two species (p<0.05). The body weights (BW) of urban populations of R. norvegicus were significantly heavier than those of rural populations (p<0.05). No secondary sexual dimorphism was found in R. norvegicus, but females of R. tanezumi showed heavier BW than those of males (p<0.05). These findings suggest that it is necessary to revise the records for the existence of R. tanezumi and to confirm the animal fauna and elucidate the distribution and ecological characteristics from further studies using extensive sampling and detailed investigations on Jeju Island and also on the Korean Peninsula.

Agronomic Characteristics and Aromatic Compositions of Korean Wild Codonopsis lanceolata Collections Cultivated in Field (한국 야생더덕 수집종의 노지 재배시 생육 특성과 향기성분 조성)

  • The native ecological environment and aromatic constituents of Korean wild Codonopsis lanceolata and one Japanese strain were investigated to find Codonopsis lanceolata strains showing high aromatics, and to know regional differences among these strains. The results were as follows : There were no remarkable differences among the Korean wild C. lanceolata strains in ecological environments. Recovery yield of essential oils was highest in Togyusan strain with 0.009%. Difference in protein band patterns among these strains was not recognized, and peroxidase and esterase pattern changes were appeared in different collected regions at the leaf and root tissues. Major aromatic constituents were 11 kinds of aliphatic alcohols such as trans-2-hexenal, 1-hexanol, cis-3-hexanol, and trans-2-hexanol. And Togyusan strain, Sobaeksan strain, and Kayasan strain have the highest aliphatic alcohols of plant essential oils. In particular, BHT(butylated hydroxytoluene), one of the antioxidants, was detected in Chirisan strains.

  • We investigated the cytogenetic characteristics of male black-striped field mouse (Apodemus agrarium) in Korea. Chromosome slides were obtained from blood cell cultures which were synchronized with thymidine blocking or not. In the chromosome slide which synchronization with thymidine blocking was employed on, the GTG(G bands by trypsin using Giemsa)-bands of high resolution were observed. The male black-striped field mouse has 48 chromosomes composed 46 autosomes and XY sex chromosomes. The centromeric regions of autosomes were positive to GTG-banding. According to this investigation, thymidine blocking in cell culture process was useful to get lengthened chromosomes. It may be necessary to employ RBG-banding technique to investigate complementary band patterns between R- and G-banding in black-striped field mouse.

Effects of Hambag Mushroom (Grifola frondosa)-Powder on Hyperglycemia and Hyperlipemia in STZ and High Fat Diet-induced Diabetic Rats (STZ 및 고지방식이에 의해 유도된 흰쥐의 고혈당 및 고지혈에 대한 함박잎새버섯분말의 효능)

  • Hambag mushroom is edible mushroom, and one of new strains in Grifola frondosa. It was developed from Korean wild Grifola frondosa by National Institute Science & Technology. In this study, we investigated the effects of the powder extracted from fruit-body of hambag mushroom on hyperglycemia, hyperlipemia and pancreatic cells in rats with streptozotocin (STZ) and high fat diet-induced diabetes. The diabetic rats were fed with hambag mushroom-powder (1 and 2% of diet weight) for 6 weeks. And then the levels of blood glucose and lipid and the pancreatic tissue were analyzed and compared. The diabetic rats fed with hambag mushroom-powder lost less body weight than those fed with the powder-free normal diet. flood glucose levels in the diabetic rats fed with hambag mushroom-powder at all concentrations was lower by 50% than in the diabetic rats fed with normal diet. The levels of total cholesterol, triglyceride and LDL-cholesterol in the diabetic rats fed with hambag mushroom-powder were increased compared to those in the diabetic rats fed with normal diet. There were the blood levels of cholesterol, triglyceride and LDL-cholesterol by hambag mushroom-powder concentrations in a dose-dependent response. The decreased damage of pancreatic tissue in the group treated with hambag mushroom-powder could be suggested possibly as one of mechanisms for the effects of hambag mushroom-powder on hyperglycemia and hyperlipemia.

Organ Weights and Splenocytic Apoptosis in γ-irradiated Korean Dark-Striped Field Mice, Apodemus Agrarius Coreae (방사선 조사된 국내 야생 등줄쥐 (Apodemus agrarius coreae) 장기무게 및 비장세포 세포고사)

  • The present investigation was planned to estimate potential possibility of striped field mice, Apodemus agrarius coreae (A. a. coreae), as a biological dosimeter in radio-environmental ecology. We bred captured wild A. a. coreae at laboratory and classified taxonomically based on external, cranial and tooth characters. Organ weights and splenocytic apoptosis were observed in order to establish a basic data on radiation biology of A. a. coreae (male, 40 weeks old). The biological effects was observed at 24hrs following irradiation (doses : 0, 0.5, 1, 2 Gy, dose rate : $0.8Gymin^{-1}$, $^{137}Cs$). Only thymus weights was significantly decreased. Splenocytic apoptosis was increased after irradiation. But splenocytic apoptosis was decreased in 0.5 Gy ${\gamma}$-irradiated mice compared to those of 0, 1, 2 Gy (P < 0.05). These data suggested that events in thymus and spleen of Korean dark-striped field mice, A. a. coreae THOMAS, could be a potential radio-biological indicator in human environments.
