Carcinoma of the Tonsillar Region -Results of External Irradiation- (편도선 암의 방사선 치료)

  • Thirty-six patients with carcinoma of the tonsillar region treated with radical radiotherapy at Yonsei Cancer Center between Jan. 1971 and Dec. 1980 were retrospectively reviewed. The purpose of this study is to evaluate the clinical characteristics and the treatment results. Seventy-five per cent of patients were in advanced stage when they were first seen. Incidence of nodal involvement at the time of initial presentation was $64\%$. Bilateral nodal involvement was found in $19\%$ of cases. Three-year local control rates for T1, T2, T3, and T4 were $88\%,\;46\%,\;25\%,\;and\;11\%$, respectively. Advanced neck nodes were poorly controlled $(N1\;70\%,\;N2\;20\%,\;N3\;12\%)$. The prognosis is mainly affected by the status of the primary and neck nodes Overall three-year local control rate was $42\%$.

Effects of an Anti-cancer Drug, Tubastatin A, on the Growth and Development of Immature Oocytes in Mice (항암제 tubastatin A에 의한 생쥐 미성숙 난모세포의 성장과 발달에 미치는 효과)

  • In recent years, progress has been made in the search for the development of new anti-cancer agents by employing specific inhibitors of histone deacetylase (HDAC)-6 to block signal transduction pathways in cancer cells. This study examined the effects of tubastatin A (TubA), an HDAC-6 inhibitor, on the growth and development of immature oocytes in murine ovaries using RNA sequencing analysis. The results from a gene set enrichment analysis (GSEA) indicated that the expression of most of the gene sets involved in the cell cycle and control and progression of meiosis decreased in the TubA-treated group as compared with that in germinal vesicle (GV) stage oocytes. In addition, an ingenuity pathway analysis (IPA) suggested that TubA not only caused increased expression of p53 and pRB and decreased expression of CDK4/6 and cyclin D but also caused elevated expression of genes involved in the control of the DNA check point in G2/M stage oocytes. These results suggest that TubA may induce cell cycle arrest and apoptosis through the induction of changes in the expression of genes involved in signal transduction pathways associated with DNA damage and the cell cycle of immature oocytes in the ovary.

Can Hinokitiol Kill Cancer Cells? Alternative Therapeutic Anticancer Agent via Autophagy and Apoptosis (Hinokitiol에 의해 유도된 Autophagy 및 Apoptosis에 의한 대체 항암요법 연구)

  • Cancer is genetically, metabolically and infectiously induced life threatening disorder showing aggressive growing pattern with invasive tendency. In order to prevent this global menace from jeopardizing human life, enormous studies on carcinogenesis and treatment for chemotherapy resistance have been intensively researched. Hinokitiol (${\beta}$-thujaplicin) extracted from heart wood of cupressaceous is a well-known bioactive compound demonstrating anti-inflammation, anti-bacteria and anti-cancer effects on several cancer types via apoptosis and autophagy. This study proposed that hinokitiol activates transcription factor EB (TFEB) nuclear translocation for autophagy and lysosomal biogenesis regardless of nutrient condition in cancer cells. Mitophagy and ${\beta}$-catenin translocation into the nucleus under treatment of hinokitiol on non-small cell lung cancer (NSCLC) cells and HeLa cells were investigated. Hinokitiol exerted cytotoxicity on HeLa and HCC827 cells; moreover, artificially induced autophagy by overexpression of TFEB granted imperfect sustainability onto HeLa cells. Taken together, hinokitiol is the prominent autophagy inducer and activator of TFEB nuclear translocation. Alternative cancer therapy via autophagy is pros and cons since the autophagy in cancer cells is related to prevention and survival mechanism depending on nutrition. To avoid paradox of autophagy in cancer therapy, fine-tuned regulation and application of hinokitiol in due course for successful suppressing cancer cells are recommended.

Effect of LED LightIrradiation on the Mycelial Growth and Fruit Body Development of Hypsizygus Marmoreus (LED 광원이 느티만가닥버섯 균의 균사 생장과 자실체 생육에 미치는 영향)

  • A edible mushroom, Hypsizygus marmoreus is commercially cultivated. However, the researches of cultivation and physiological characteristics were not conducted in Korea. In this study, we conducted on artificial cultivation of H. marmoreus and elucidated the effect of light on the mycelial growth and fruit body development using LED light sources with different wavelength; blue (peak wave length 460nm), green(peak wave length 530nm), yellow(peak wave length 590nm), red(peak wave length 630nm), and white as positive control. Mycelial growth of H. marmoreus strains were inhibited about 30~40% in inhibition ratio under the illumination with blue, green, yellow LED light. However, red LED light was not inhibited. Elongation of stipe was effective under the long wave length such as yellow and red light. Abnormal fruit body was produced under the long wavelength and dark. However, development of pileus was effective under the short wavelength such as green and blue light. Also, as a result of cultivation with mixed light for high quality and harvest, many effective numbers and yields of fruiting bodies were obtained in the mixed treatment of blue and white light, and pileus developed well.

The impact of chlorothalonil on female gametophyte survival rate and relative growth rate of Undaria pinnatifida (Chlorothalonil이 미역(Undaria pinnatifida) 배우체의 생존 및 상대성장률에 미치는 영향)

  • Chlorothalonil is continuously introduced into the marine environment and has significant toxic effects on various marine organisms, however, research on its effect on seaweed is limited. Therefore, we analyzed the impact of chlorothalonil on the early life stages of major aquaculture species in Korea, Undaria pinnatifida. U. pinnatifida female gametophytes were exposed to different concentrations of chlorothalonil (0, 0.03, 0.05, 0.10, 0.20, and 0.40 mg L-1), and their survival rate and relative growth rate were analyzed. The no observed effect concentration (NOEC), lowest observed effect concentration (LOEC), and median lethal concentration (LC50) for female gametophyte survival were determined as 0.05, 0.10, and 0.141(0.121-0.166)mg L-1. NOEC, LOEC, and median effective concentration (EC50) for relative growth rate were 0.10, 0.20, and 0.124 (0.119-0.131) mg L-1. Therefore, female gametophytes of U. pinnatifida are expected to experience toxic effects at concentrations above 0.05-0.10 mg L-1 of chlorothalonil. These research findings are anticipated to serve as crucial reference data for evaluating the effects of chlorothalonil on the health of U. pinnatifida in the early life stages.

Shadows and Evil in Inferno of Divine Comedy (신곡의 지옥편에 나타난 그림자와 악)

  • This study is to illuminate the problem of shadows and evil appearing in contemporary events in the midst of a period of upheaval through Inferno of Divine Comedy. First, the concept of shadow and evil were briefly summarized in analytical psychology and discussed the importance of considering the concepts with the empirical aspect of relativity and ethics in the field of psychotherapy. The 14th century, the age of Dante, was the embryonic period of the Renaissance beyond the end of the Middle Ages. It was when Dante, who was writing Divine Comedy, had to take off his persona forcibly and live in exile. In a nutshell, it was a transition period for both the individual and the collective. The dark forest is a nigredo, darkness and chaos we face in this transition, but it can be a place of transformation and rebirth. The three beasts (leopard, lion, and she-wolf) encountered in the forest can be considered as the instinctual images that Dante ignored and alienated, which the medieval Christian world had suppressed and eliminated. Especially at the collective level, as destructive instincts, ferocious beasts roam throughout society when a crisis breaks dominant laws of values. The three beasts of Inferno appear as phenomena of shadows and evil. The aspect of leopard was explored Cerberus and Chiaco(pig) as a symbol of greed, and the lion, Farinata as the form of violent passion. The aspect of the she-wolf was examined as Geryon, a deceitful monster with a beast nature hidden behind goodness, the giants in the frozen lake of Cocytus, and Branca D'Oria, who betrayed and murdered the country and family. Inferno reveals the "state" of being trapped when one yields to the evil hidden within oneself and falls into prey.

Sarcoid-Like Reaction after Complete Remission of Malignancy: CT and 18F-FDG PET/CT Features for the Differential Diagnosis from Lymph Node Metastasis (악성종양의 완전관해 후 발생한 사르코이드증 유사 반응: 림프절 전이와의 감별진단에 유용한 CT와 18F-FDG PET/CT 소견)

  • Purpose To identify the imaging features indicative of sarcoid-like reactions in patients with intrathoracic lymphadenopathy after complete remission of malignancies. Materials and Methods This study enrolled five patients with histopathologically confirmed sarcoid-like reactions that developed after cancer remission. The clinical features and findings of CT and 18F-fluorodeoxyglucose (FDG) PET/CT were assessed. Results The underlying malignancies included breast, nasopharyngeal, colon, and endometrial cancer and lymphoma. The time intervals between complete remission of malignancy and the diagnosis of sarcoid-like reaction ranged from 6 to 78 months. CT findings of sarcoid-like reaction included bilateral hilar and mediastinal lymphadenopathies (n = 5), pulmonary nodules (1-15 mm) with peribronchovascular, fissural, or subpleural distribution, and interlobular interstitial thickening in the lungs (n = 4). 18F-FDG PET/CT revealed hypermetabolic uptake in the mediastinal and hilar lymph nodes and both lungs in the absence of extrathoracic uptake (n = 3). The sarcoid-like reactions resolved in all patients after corticosteroid treatment. Conclusion In patients with complete remission of malignancies, newly developed bilateral hilar and mediastinal lymphadenopathies with or without pulmonary nodules of perilymphatic distribution, in the absence of recurrence at the primary tumor site and extrathoracic metastasis, may suggest a sarcoid-like reaction. Such cases warrant histologic evaluation of the lymph nodes to prevent unnecessary systemic chemotherapy.

  • 최근의 생물학 연구를 위한 기기의 자동화 및 고속화는 생물학 관련 정보량의 급증을 가져오고 있다. 예를 들어, DNA chip에서 얻어지는 마이크로어레이(microarray)는 수천 종류의 유전자의 발현량을 동시에 측정한다. 이러한 기술들은 생물의 세포나 조직에서 일어나는 일련의 다양한 현상을 전체적으로 조망하는 관점에서 관찰할 수 있는 기회를 제공하고 있으며, 이를 통한 생명공학의 전반적인 발전이 기대되고 있다. 따라서 대량의 생물학 관련 정보의 분석이나 데이터 마이닝이 행해지고 있으며 이를 위한 대표적인 기법들로는 각종 클러스터링(clustering) 및 신경망 계열의 모델 등이 있다. 본 논문에서는 확률그래프모델의 하나인 베이지안망(Bayesian network)을 생물정보분석에 이용한다. 구체적으로 유전자 발현패턴과 약물의 활성패턴 및 암 종류 사이의 확률적 관계를 모델링한다. 이러한 모델은 NCI60 dataset(http://discover.nci.nih.gov)에서 베이지안망을 학습함으로써 구성된다. 분석의 대상이 되는 데이터가 sparse하기 때문에 발생하는 어려움들을 해결하기 위한 기법들이 제시되며 학습된 모델에 대한 검증은 이미 생물학적으로 확인되어 있는 사실과의 비교를 통해 이루어진다. 학습된 베이지안망 모델은 각각의 유전자 간, 혹은 유전자와 처리된 약물 간의 실제 생물학적 관계를 다수 표현하며, 이는 제시되는 방법이 생물학적으로 유의미한 가설을 데이터 분석을 통해 효율적으로 생성하는데 유용하게 활용될 수 있음을 보인다.

Treatment Results of Rhabdomyosarcomas of Head and Neck (두경부의 횡문근육종에서 다방면요법의 효과)

  • 최근 횡문근육종을 치료함에 있어서 수술적 요법, 방사선 치료, 강암 약물요법을 적절히 병행함으로써 치료에 따르는 후유증을 극소화하면서 괄목할만한 생존율 향상을 가져오게 되었다. 특히 두경부의 횡문근육종은 소아에서 다발하고 병소의 위치에 따라서 각각 다른 임상적 특성을 가지며 수술적 절제를 했을 때 기능 및 외견상 결손이 크므로 별도의 연구 대상이 되어 왔으며 수술적 방법보다는 방사선 치료 및 항암 약물요법이 강조되고 있다. 저자들은 1976년부터 1987년까지 두경부 횡문근육종으로 진단받고 연세 암센터에서 항암 약물요법 및 방사선 치료를 받은 22명의 환자를 대상으로 하여 발생 부위, 조직 병리, 병기별 분포 및 생존율, 치료 방법에 따른 생존율을 후향성으로 분석하였다. 22명의 환자중 10 세 미만이 12 예로 가장 많았고, 13명이 배아세포형이였으며, 임상병기는 병기 III 이 가장 많아서 14 예였다. 병소의 위치는 안와가 6명으로 가장 많았고, 뇌수막주변부가 7명 (상악동 3명, 중이도 2명, 비강 l명, 비인강 1명), 기타 9명 (경부 5명, 이하선 2명, check 2명) 이었다. 5년 무병 생존율은 안와 종양에서 가장 높아서 50% 였고, 안와외 두경부가 37.5%, 뇌수막주변부가 16.7% 로 가장 낮았으며, 낮은 병기(병기 III), 배아세포형, 항암 약물 요법 병용군에서 상대적으로 높은 생존율이 관찰되었다.

Lycorine induces apoptosis by enhancing protein degradation of survivin in human oral cancer cell lines (Lycorine의 사람 구강 암 세포주에서 survivin 단백질 분해 증진으로 세포자멸사 유도)

  • Lycorine, a natural alkaloid extracted from the Amaryllidaceae plant family, was reported to various physiological and pharmacological effects including anti-cancer activity. Nevertheless, there is no report of the anticancer effect of lycorine in oral cancer cells. The effects of lycorine on cell proliferation and apoptosis were examined through trypan blue exclusion assay, 4'-6-diamidino-2-phenylindole (DAPI) stain, Live/Dead assay, Western blot analysis and RT-PCR. Lycorine suppressed cell viability and induced apoptosis in MC3 and HSC-3 cell lines. Lycorine decreased survivin protein but did not affect its mRNA. It regulated survivin through accelerating protein degradation in a time-dependent manner although neither proteasome nor lysosome was not associated with lycorine-mediated protein degradation. Collectively, our results suggest that lycorine may be a potential therapeutic anti-cancer drug candidate for the treatment of human oral cancer.
