Production of Cellulolytic Enzymes by Aspergillus niger on Solid and Submerged State Fermentation (액체와 고체 발효 조건에서 Aspergillus niger의 셀루로오스계 효소 생산)

  • Microbial production of cellulolytic enzymes by Aspergillus niger in solid state fermentation (SSF) and submerged state fermentation (SF) in laboratory scale was compared. Czapek Dox liquid broth amended with cellulose (0.5%) was used for cultivation in SF, whereas rice bran was used as a solid support, moistened with cellulose, amended Czapek Dox broth for growth in SSF. The production of Carboxymethyl cellulase, Filter paperase and ${\beta}$-Glucosidase was monitored at regular intervals. The peak production of the enzymes occurred within 3 days of incubation in SSF as against $\geq$ 7 days in SF. SSF gave higher yields of enzymes in comparison to SF. Maximum titres of 0.40, 0.62 and 0.013 U/ml in respect of FPase, CMCase and ${\beta}$-glucosidase in SSF were recovered as against 0.13, 0.06 and 0.0013 U/ml in SF respectively, at their respective peak time intervals. Hence, SSF appeared to be a better choice for production of cellulolytic enzymes by Aspergillus niger.

  • In this study, we obtained fundamental data about Korean raw lacquer's physical and chemical properties to produce high quality lacquer. The tested raw lacquers were obtained from Won-ju in Korea, Shanxishang, Guizhoushang, Anhuishang in China. The drying time of refining lacquers, tensile strengths of dried films and uniformity of films are measured. The refined lacquers were prepared by experimentally scaled refining equipment. Films of lacquer were applied on glasses by film applicator. This films were tested by universal strength test machine. The films were pictured by scanning electron microscopy and confocal microscopy to define the uniformity of them. The refining method were not different among three different kind of raw lacquers which were different their collecting time and places. But the viscosity of them were quite different. When black refined lacquer is made with iron powder, the adding time of iron powder is critical to control the viscosity of it. The refining times, viscosity and tensile strength of refined lacquers were not depended the method of refining condition but the place of collecting of raw lacquer.

  • $\alpha$-Cyclodextrin glucanotransferase excreted from a newly isolated Geobacillus thermosacchalytycus was purified through the ultrafiltraion, hydrophobic Sepharose CD-4B affinity chromatography, and gel filtration on Sephadex G-75, respectively. The molecular weight of the purified CGTase was 69 kDa and its N-terminal amino acid sequence was determined to be Asn-Leu-Asn-Lys-Val-Asn-Phe-Val-Ser-Asp-Val-Val-Val-Gln-Ile. The optimum pH and temperature were pH 6.0 and$ 60^{\circ}C$, respectively, and stably at the pH range of 6.0-8.0 and $60^{\circ}C$ in the presence of $Ca^{++}$. The excreted CGTase from the thermophilic G. thermosacchalytycus was $\alpha$-type showing a high coupling activity for the transglycosylation on various glucosides. The coupling reaction was carried out according to the random ternary complex mechanism.m.

  • The production of heterologous protein using GAL promoter in conventional S. cerevisiae has several problems to s이ve for c commercialization. In this research, S. cerevisiae mutant(reg1-501, gaI1), which cannot use galactose and has alleviated g glucose repression level, is used as host for optimizing induction of GAL promoter. In this experiment, the effects of specific g growth rate on specific recombinant protein expression rate were tested in both cases and optimum fed batch fermentation m method was obtained in both cases. Through these experiments, optimum condition of recombinant protein production by G GAL promoter using S. cerevisiae mutant (reg1-501, gal1) were found.

  • Green fluorescent protein (GFP) is an attractive reporter for bioprocess monitoring. A fluorescence-based method was developed to quantify GFP levels in transgenic plants and protein extracts. In this study, GFP was produced and secreted from suspension cells derived from transgenic rice. The RAmy3E promoter placed before the GFP gene controlled by sugars such as sucrose. The effects of sucrose concentration on the secretion of GFP and total protein into the medium were investigated in batch suspension culture. It was possible, therefore, to induce the expression of the GFP by removing sucrose from the cultured media or by allowing the rice suspension cells to deplete sucrose catabolically. The dry cell weight (7.06 g/L) and GFP level were detected as highest at 12%, 3% sucrose after 20 day culture, respectively. However secreted GFP fluorescence at the other sucrose concentrations (6%, 12%, 18% and 24%) were a little amount in media.

  • This paper investigates the differentials in productivity and growth performance between small and medium size and large size firms in Korean manufacturing industries. According to the empirical analysis based on the Mining and Manufacturing Survey of Statistics Korea, this paper finds the following facts. First, the level of contribution of capital deepening on the growth rate of output(value added) is relatively greater in the small and medium size firm group, while the level of contribution of total factor productivity on the growth rate is relatively greater in the large size firm group. Second, the productivity polarization among the firms has increased relatively greater in the large size firm group over the past decade.

복제한우 생산효율과 산자의 성장능력

  • 본 연구는 한우 복제수정란의 이식에 의해 고능력 복제한우를 생산할 수 있는 효율과 산자의 성장능력을 검정하기 위해 수행되었다. 고능력 한우의 귀세포를 이용하여 체세포복제수정란을 생산하였고 그 신선수정란을 대리모에 이식한 결과는 다음과 같다. 2000년 (n=35)과 2001년 (n=121)에 이식한 복제소의 분만율은 각각 5.71%, 8.26%로 나타났다. 복제송아지의 임신기간은 평균 287일 (279~295일)이었으며 같은 기간내 인공수정 우군의 평균 임신기간인 287일 (255~293)과 동일하였다. 복제송아지의 분만시 사산율은 6.67%였으며, 인공수정시의 94.44%와 비슷하였다. 그러나 분만후 복제송아지의 생존율은 36.67%로서 인공수정의 100% 보다 매우 낮았다. 그리고 태어난 후 200일 이상 생존한 송아지의 생시 평균체중은 28.25kg (AI 23.67kg)이었고, 복제송아지의 경우 생시체중이 표준체중보다 적거나 (<20kg) 또는 거대체중 (>35kg)의 경우는 분만후 대부분 사망하였다. 상기 결과에 의해 현재의 복제기술에 의한 한우 생산효율은 아주 낮고 분만 후 거대체중등 비정상인 경우가 많아 생존율이 낮음을 알 수 있었다. 따라서 이에 대한 원인구명에 관한 기초연구의 확대가 요망된다.

  • A bacterium producing the neutral pretense was isolated from soil, and was identified as Bacillus sp. DS-1 by 16S rRNA sequence comparison and biochemical determinations. The production of protease from Bacillus sp. DS-1 was increased 20% and 30% by the additions of 1% glucose and 1% yeast extract, respectively. The optimum pH and temperature for the protease activity were pH 7.0 and 55$^{\circ}C$. Bacillus sp. DS-1 produced a metalloprotease as a major protease in culture medium, since the pretense activity in culture supernatant was inhibited by the presence of 1 mM EDTA significantly.

  • The physiochemical properties of extracellular lipid produced by an oleaginous yeast, Rhodotorula glutinis K-501 were examined. From the analytical experiments, it was suggested that the extracellular lipid produced is glycolipidic compound. Critical micelle concentration and minimum surface tension of the extracellular lipid in aqueous solution were 89mg/L and 31dyne/cm, respectively. Surface tension was also constant throughout wide range of pH. The emulsifying abilities and dispersing power of the extracellular lipid were much greater than those of commercial surfactants such as Tween 80 and Triton X-100 by factors of 2-3 and 1.3, respectively.

공기업민영화(公企業民營化)의 정책효과분석(政策效果分析)에 관한 연구(硏究)

  • 본(本) 연구(硏究) 1960년대말 이후 1980년대초 기간중에 민영화(民營化)된 공기업(公企業)에 대하여 민영화(民營化)가 개별기업(個別企業)의 생산효율성(生産效率性)에 미친 효과를 회귀분석모형(回歸分析模型)에 의하여 추정하고, 추정결과를 토대로 향후 정부의 바람직한 민영화정책의 방향을 제시하고자 한다. 본(本) 연구(硏究)의 분석결과(分析結果), 공기업민영화(公企業民營化)는 대한통운, 대한항공, 대한재보험, 그리고 대한석유공사의 경우 개별기업의 생산효율성(生産效率性) 증진(增進)에 긍정적인 효과를 미쳤으나, 5대(大) 시중은행(市中銀行)의 경우 공기업민영화(公企業民營化)가 기업(企業)의 생산효율성(生産效率性) 제고에 미친 효과가 미미하여 통계적 신뢰성이 낮게 나타나거나 부정적(否定的)인 효과(效果)가 추정(推定)되었다. 이러한 분석결과(分析結果)에 근거할 때, 기업민영화(公企業民營化)가 항상 기업(企業)의 생산효율성(生産效率性) 증가(增加)에 기여할 것이라는 일반적인 믿음은 현실적안 근거가 부족하며, 일정한 조건이 충족되어야 그러한 믿음이 현실화될 수 있음을 알 수 있다. 민영화(民營化)가 바람직한 정책효과(政策效果)를 나타내기 위해서는 공기업민영화시(公企業民營化時) 기업소유권(企業所有權)의 민간이전뿐만 아니라 실질적인 경영권(經營權)의 이전이 수반되어야 하며, 민영화(民營化)와 더불어 정부(政府)의 각종 규제(規制)와 간섭(干涉)이 배제되어야 한다.
