Establishment of the Extraction Process by Evaluation of Proanthocyanidin Contents and Antioxidative Activities of Pine (Pinus densiflora) Needle Extracts. (적송잎 추출물의 proanthocyanidin 함량 분석 및 항산화 활성 검증을 통한 추출공정 확립)

  • We evaluated proanthocyanidin contents out of total polyphenolic compounds in two pine needle extracts prepared by combined extraction methods and compared their antioxidative activities. Amounts of total polyphenolic compounds of sub-supercritical hotwater precipitate (SSHP) and sub-supercritical hotwater supermatant (SSHS) were 225.4 and 116.6 mg/g, respectively. Amounts of proanthocyanidin of SSHP and SSHS were 9.9 and 3.6 mg/g, respectively. Thus, the ratio of proanthocyanidin contents over total polyphenolic compounds of SSHP and SSHS were 4.3 and 3.9%, respectively. Antioxidative activities ($IC_{50}$) of SSHP and SSHS measured by DPPH radical scavenging were 41.5 and $50.7\;{\mu}g/ml$, respectively. These results indicates that two combined extracts, SSHP and SSHS showed higher antioxidative activities than extract prepared by hot water or sub-supercritical. In conclusion, extraction procedures for SSHP and SSHS could be useful methods for preparing the effective components of functional food products.

Characteristics of marine algae extracts using subcritical water extract method (아임계 추출법을 이용한 해조류 추출물의 특성)

  • This study was performed in order to investigate the functional components of 5 kinds of marine algae. We have collected 5 samples of marine algae, such as the sea mustard (Undaria pinnatifida), sea tangle (Laminaria iaponice), sea weed fusiforme (Hizikia fusiforme), green laver (Entetomotpha), laver (Phophyratenera), which have been harvested in Jeollanam-do. In order to examine the functional effects, 5 kinds of marine algae were extracted with hot water ($80^{\circ}C$, 4 hr), ethanol and methanol (R.T., 4 hr), and subcritical water extract (SWE, 3 MPa, $90^{\circ}C$, $150^{\circ}C$, $210^{\circ}C$). A higher yield of extract was obtained through SWE method (3 MPa, $210^{\circ}C$) in all of the samples obtained. The highest total sugar content was 427.4 mg/g in green laver extracted with SWE (3 MPa, $210^{\circ}C$). The content of the SWE total phenolic compounds was higher than that of the water and solvent (methanol, ethanol) extracts. The anti-oxidative activities of the extracts from 5 kinds of marine algae were examined through the DPPH radical scavenging activity test. The SWE (3 MPa, $150^{\circ}C$ and $210^{\circ}C$) of the marine algae was the highest among all of the extracts. As per the results, the SWE of the marine algae contained more functional components and it had a higher antioxidant activity than those of the other extracts. The $IC_{50}$ value of tyrosinase in seaweed fusiforme and laver were higher than those of the other samples. These results strongly support the possible use of marine algae as functional materials.

Anti-inflammatory and Anti-oxidative Activities for the Subcritical Water Extract of Camellia japonica Flowers (동백 꽃 아임계 수 추출물의 항염 및 항산화 활성)

  • In this study, the anti-inflammatory and anti-oxidant efficacy of camellia subcritical water extracts (SWE, 135 ~ 180 ℃, 70 bar) was compared with 70% ethanol and hot water extracts. Among these extracts, the yield (57.9%) of the subcritical water extract, which was extracted under the condition of 180 ℃ and 70 bar was the highest, which increased the extraction yield by more than two times compared to the hot water extract (28.1%). The results of the nitric oxide (NO) production inhibition activity experiment using RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS) showed that subcritical water extracts had superior effects in inhibiting the production of NO without cytotoxicity than 70% ethanol and hot water extracts. In addition, DPPH and ABTS+ radical scavenging activity experiments showed that the radical scavenging activity of subcritical water extract was similar to that of 70% ethanol and hot water extract. Moreover, the content of gallic acid was determined by HPLC and the quantity was about 1.62 mg/g for the SWE (165 ℃, 70 bar), which was the highest among all of the extracts. Based on these results, it is concluded the SWE of C. japonica flowers could be potentially applicable as anti-inflammatory and anti-oxidative ingredients in cosmetic formulations.

  • Flavonoids in pine needles are known to be effective scavengers of free radical. Especially, proanthocyanidin, a kind of flavonoids possesses cardiovascular protection and antioxidative activities. Here, we evaluated proanthocyanidin contents in total polyphenolic compounds of pine needle extracts prepared by using hot water, ethanol, hexane or sub-supercritical $CO_2$. Analyses of total polyphenolic compounds and proanthocyanidin in each extracts indicated that hot water extract contained the highest concentrations, but sub-supercritical extract contained the lowest concentrations. On the other hand, evaluation of proanthocyanidin contents in total polyphenolic compounds in each extracts showed that sub-supercritical extract possessed the highest content, but hot water extract possessed the lowest content. These results indicate that extracts containing high concentrations of both total polyphenolic compounds and proanthocyanidin could be obtained by using hot water or ethanol extractions. Furthermore, extract containing high content of proanthocyanidin out of total polyphenolic compounds could be achieved by using sub-supercritical extraction. Measurement of antioxidative activities of extracts showed that hot water extract possessed the highest activity. In this study, we prepared extracts from pine needles by four different methods and evaluated the antioxidative compounds in extracts that could be used for effective components of functional food products.

Evaluation of the Basic Property Evaluation of Eco-powder, a Hydrothermal Synthesis Product for Improving Waste Vinyl Recycling Efficiency (농촌 폐비닐 활용률 제고를 위한 수열합성 생성물인 에코 파우더(Eco-powder)의 기초물성 평가)

  • This study aimed to improve utilization of the Class C vinyl waste generated in rural areas based on a preliminary investigation on the use of eco-powder, generated through pyrolysis, as a raw material for plastic. The efficiency of pre-processing treatments in controlling ash content of the generated eco-powder and its effect on the basic properties of manufactured plastic were evaluated. The basic properties included ash content of the compressed eco-powder at different levels of ash content, impact strength, flexural strength, and tensile strength. The experimental results confirmed that pre-processing improved the separation efficiency of soil particles and vinyl waste through physical impact. The eco-powder with ash content of less than or equal to 26% was found to satisfy the target performance during impact strength, flexural strength, and tensile strength evaluation. Thus, it was confirmed that the Class C vinyl waste, having low utilization and recovery rates, could be effectively utilized as a plastic raw material after optimum thermal treatment and physical processing using the eco-powder.

Antioxidative Activity of Pine (Pinus densiflora) Needle Extracts in Rats Fed High-Cholesterol Diet (고콜레스테롤 식이 섭취 흰쥐에 대한 적송잎 추출물의 항산화 효과)

  • This study was conducted to investigate the effects of pine (Pinus densiflora) needle extracts on hepatic antioxidative system in rats fed high-cholesterol diet. Eight-week old Sprague Dawley rats were divided into four groups; normal diet group (CON), 0.5% cholesterol diet group (0.5% CHOL), 0.5% cholesterol diet and pine needle extracts prepared by using hot water group (HOT water), and 0.5% cholesterol diet and pine needle extracts prepared by using sub-supercritical $CO_2$ group (Sub-supercritical). The serum ALP, AST, and ALT activities were increased by 0.5% cholesterol diet supplementation, but treatment with pine needle extracts showed decrease compared with 0.5% CHOL group. The SOD activity in liver at 0.5% cholesterol diet group was more decreased than control group while Sub-supercritical group had significantly higher SOD activity than 0.5% CHOL group. Moreover, groups with pine needle extracts supplementation had higher level of GPx in liver than 0.5% cholesterol diet group. These results suggest that pine needle extracts increased antioxidative activities.

The Effect of Pine (pinus densiflora) Needle Extracts on Blood Flow and Serum Lipid Improvement (적송잎 추출물의 혈행 및 지질개선 효과)

  • Pine needles have long been used as a traditional health-promoting medicinal food in Korea. To investigate the effects of pine (pinus densiflora) needle extracts on blood flow and serum lipid improvement were assessed in vivo. 8 week-old Sprague Dawley strain rats were divided into four groups of seven rats each; CON, 0.5% CHOL, HOT water and Sub-supercritical group. Serum total cholesterol and triglyceride contents were lower in the CON group than the 0.5% CHOL group. Three weeks of feeding hot water and sub-supercritical extract resulted in a decrease in serum triglyceride and total cholesterol level. The level of HDL-cholesterol in the 0.5% CHOL group was significantly (p<0.05) reduced compared to the CON group, but it had a tendency to increase with pine needle extract supplementation. Blood passage time of the pine needle extracts supplemented group was higher than the 0.5% CHOL group. Microscopic observation showed that whole blood passed smoothly through the micro channels in pine needle extracts supplemented groups. The platelet aggregation ability of the groups treated with pine needle extracts was less than that of the 0.5% CHOL group. All these results suggest that pine needle extracts might improve blood homeostasis mediated via antiplatelet activities.

Effect of Extraction Methods on Flavoring Compounds and Antioxidant Activity of Extracts from Cinnamon (Cinnamomum cassia Blume) (추출방법이 육계피 추출물의 향기 성분과 항산화 활성에 미치는 영향)

  • The interest in and development of healthy foods and nutraceuticals have increased because of the trend for a health-oriented society. Cinnamon is used as a food ingredient as well as a herbal medicine because of its functional properties. In this study, flavoring compounds and antioxidative activities of cinnamon extracts were investigated with different extraction solvents and extraction methods. The contents of flavoring compounds such as coumarin, cinnamic acid, cinnamaldehyde, and cinnamyl alcohol were investigated. The contents of courmarin, cinnamic acid, and cinnamylaldehyde in 70% ethanol extract were higher than those in hot water and subcritical water extracts. The contents of courmarin, cinnamic acid, and cinnamaldehyde in subcritical water extract were higher than those in hot water extract, whereas the content of cinnamyl alcohol was lower. DPPH scavenging activity increased with increasing concentration of the extracts, and the 70% ethanol extract showed the highest antioxidant activity. The ascorbic acid content of the 70% ethanol extract was largest in the antioxidative activity measurement by FRAP analysis. The ascorbic acid contents of the hot water and subcritical water extracts were similar.

Comparison on the Extraction Efficiency and Antioxidant Activity of Flavonoid from Citrus Peel by Different Extraction Methods (추출방법에 따른 감귤 과피 유래 Flavonoid의 추출효율 및 항산화 효과에 대한 비교)

  • The extraction of polyphenol and flavonoid from citrus peel was performed by the ethanol, sugar, hot water (80$^{\circ}C$), and subcritical water extraction methods. The maximum yields of total polyphenolic compounds (27.25${\pm}$1.33 mg QE/g DCP, QE and DCP indicate quercetin equivalent and dried citrus peel, respectively) and flavonoids (7.31${\pm}$0.41 mg QE/g DCP) were obtained by subcritical water extraction (SWE) with operating conditions of 190$^{\circ}C$, 1300 psi, and 10 min. The yields by SWE were over 7.2-, and 8.5-fold higher than those of total polyphenols (3.79${\pm}$0.73 mg QE/g DCP) and flavonoids (0.86${\pm}$0.27 mg QE/g DCP) obtained using the ethanol extraction, which showed the highest extraction efficiency among tested conventional methods, respectively. Antioxidant activities of extracts obtained by different methods showed no significant differences. However, the relative antioxidant yield per 1 g dried citrus peel by SWE (190$^{\circ}C$, 10 min) was over 9.5-fold higher than that by the ethanol extraction.

Effect of Subcritical Water for the Enhanced Extraction Efficiency of Polyphenols and Flavonoids from Black Rice Bran (흑미강으로부터 유용 폴리페놀 및 플라보노이드의 추출효율 증진을 위한 아임계수의 효과)

  • The extraction of polyphenol and flavonoid from black rice bran was performed by diverse extraction methods using the sugar solution, ethanol, hot water ($80^{\circ}C$), and by subcritical water extraction (SWE) method. By SWE under operating conditions of $190^{\circ}C$, 1,300 psi, and 10 min, the maximum yields of total polyphenolic compounds (35.06${\pm}$1.28 mg quercetin equivalent (QE)/g dried material and flavonoids (7.08${\pm}$0.31 mg QE/g dried material) could be obtained. These results were over 11.77- and 12.21-fold higher than those of the ethanol extraction, which showed the highest extraction efficiency among tested conventional methods in total polyphenols (2.98${\pm}$0.74 mg QE/g dried material) and flavonoids (0.58${\pm}$0.21 mg QE/g dried material), respectively. Though the highest antioxidant activity (87.14${\pm}$1.14%) was observed at the dried extract obtained from ethanol method, the relative antioxidant activity per 1 g dried black rice bran by SWE ($190^{\circ}C$, 10 min) was over 11.53-fold higher than that by the ethanol extraction.
