The Effect of Soil Conditioners on the Rutin Biosynthesis and the Yield of Buckwheat(Fagopyrum esculentum moench) (메밀의 rutin 생합성과 수량에 대한 토양개량제의 효과)

  • This experiment was conducted to investigate the effects of soil conditioners, such as lime, borax, poultry waste sawdust manure (P.W.S.M) and mixed oil cakes (M.O.C), on the rutin biosynthesis and the yield of buckwheat. The content of phosphorus ($P_2O_5$) in buckwheat plants before flowering was higher at before flowering than that of flowering time at all plots. The content of nitrogen (N), potassium ($K_2O$), calcium (CaO) and magnesium (MgO) was lower at before flowering than those of flowering time at all plots. The contents of rutin in buckwheat plant at flowering time was higher than that at before flowering content of rutin in leaf was higher than that in stem. The contents of rutin in buckwheat plant was the highest at the plot treated with borax (B) compared to other plots. Correlation between rutin and amino acid was positive in buckwheat leaf, while it was negative in buckwheat stem. The yield of buckwheat was 21 percent higher at the plot treated with borax than the plot applied with the other three ingredients.

  • This study aimed to analyze germination characteristics of dredged soil collected in Gunsan. This study develops the ecological environment materials for useable green soil that can be used in the stabilization of the dredged slope. In order to analyze the germination characteristics of dredged soil, specimens treated with Bio ameliorant (0, 6%), AC (Amino acid) ameliorant (0, 6%), Sawdust (0, 6%) and loess (0, 50%) were made. Based on the test results, it was found that dredged soils of Gunsan Saemangeum were classified as SM, and poor arboreal growth conditions were necessary to be treated with ameliorants (Sawdust, Bio ameliorant, etc.).

퇴비에서 분리한 우모(牛毛) 분해균 Bacillus pumilis RS7에 의한 우모분해산물이 식물성장에 미치는 영향

  • 전 세계적으로 방대하게 배출되는 도축 부산물인 우모는 높은 영양적 가치를 가짐에도 불구하고 현재 사용하는 물리화학적 처리법의 여러 단점으로 효율적으로 이용되지 못하고 있는 실정이다. 그 결과 2차 오염을 발생시키지 않는 경제적 방법인 생물학적 처리에 대한 연구의 필요성이 커지고 있다. 본 연구에서는 그러한 방법 중 대표적인 방법으로 미생물이 생산하는 keratinase를 이용한 생분해에 대해 연구를 하기 위해 경남 일대 퇴비화 볏짚에서 keratinolytic protease 생성능이 우수한 균주인 RS7을 분리하였고 생화학적 동정법과 16S rDNA를 이용한 동정결과 B. pumilus로 동정되었다. 본 실험에서 분리된 B. pumilus는 기존에 활발히 연구되어 있지 않는 균주로써 native feather 분해도가 기존에 알려진 Bacillus 속들이 3일 정도에서 분해 완료되는 것과는 달리 36시간 ${\sim}$48시간 내에 깃대까지 완전히 분해하였다. 또한 분리 균주의 경제적인 분해능을 토대로 분해산물이 식물 생장에 주는 영향과 아미노산 함유량을 검토한 결과 기존의 화학적 분해에 의한 분해산물보다 아미노산 함유량이 4배가량 많았으며 식물 생장에 있어서도 생장율과 개화시점으로 미루어볼 때 비료로써의 역할을 수행할 수 있었다.

Isolation of Protease Producing Microorganisms (단백질 분해효소 생산 균주 분리)

  • Protease producing microorganisms were isolated from many kinds of food waste and fermented foods, which contains high amount and variable kinds of degraded substances. Several microorganisms were identified by 16S rRNA full sequencing analysis methods. The activity of protease was analyzed and identified in variable conditions for the application. For industrial use for biowaste treatment some proteases were isolated, identified and selected from microbial cells. And the tests were carried for the further use. The protein degrading activity at low temperature is useful for the treatment of organic waste, which contains much proteins. By the protein degradation process the organic waste can be utilized in variable fields, for example from feedstuff supplement to fertilizer for agriculture. Bacterial cells with protease activity at low temperature were isolated and identified. The optimal conditions for microbial cultivation and protease production were studied.

  • A pot experiment was carried out using Seomjin-byeo and Samgang-byeo to investigate the effects of nitrogen and sulfur on the growth, yields, and sulfur containing amino acid content of rice. The yields and number of tillers at 50 days after transplanting were increased with the increase of the amounts of nitrogen and sulfur, but N/S ratio was decreased in both varieties. There were a positive correlations between yield and contents of nitrogen and sulfur in straw and brown rice, but a negative correlation between N/S ratio and yields at harvesting stage. Crude protein contents increased with the increase of amounts of nitrogen and sulfur. contents of amino acid containing sulfur in brown rice were positively correlated with the sulfur and nitrogen contents in rice straw.

Effects of Concentrated Pig Slurry Separated from Membrane Filter and Several Environment-Friendly Agro-Materials Mixtures on the Growth and Yield of Lettuce (Lactuca sativa L.) in Hydroponics (막분리 돈분농축액비와 몇가지 친환경농자재의 혼합액이 수경재배에서 상추의 생육과 수량에 미치는 영향)

  • This experiment was conducted to investigate the effects of concentrated pig slurry separated from membrane filter and by environment-friendly agro-materials mixtures on growth of lettuce in hydroponics. The swine waste treatment system having a ultra filtration and a reverse osmosis process was designed in this study. Filtration of pig slurry was necessary to prevent the hose clogging in hydroponics. Primary separation using ultra filter was followed by concentration by RO (Reverse Osmosis). The concentrated pig slurry (CS) was mixed by five different environment-friendly agro-materials mixtures. The chemical nutrient solution was the solution of National Horticulture Research Station for the growth of lettuce. The concentration of nutrient solution in hydroponics was adjusted a range of 1.5 mS/cm in EC. The concentrated pig slurry was low in phosphorus(P), suspended solid and heavy matal, but rich in potassium (K). The concentrated slurry was lowest in the growth characteristics of leaf lettuce. And also SPAD value in leaf was reduced in plot treated with concentrated slurry. But the growth of lettuce in the mixtures plot (CS+BM+AA, CS+BM+AA+SW) in hydroponics was significantly high compared to concentrated slurry. The fresh yield of lettuce was 78, 84% that of nutrient solution as 131.9, 142.2g in plot of CS+BM+AA and CS+BM+AA+SW, respectively. Our studies have shown that it is possible to produce organic culture using concentrated slurry and environment-friendly agro-materials mixture, although growth is slower than when using a conventional inorganic hydroponic solution.

Effect of Split-Application of Slow-Release Fertilizer on Yield and Quality of 2nd Harvested Tea Leaves (완효성 비료 분시방법에 따른 두물차의 수량 및 품질)

  • A field experiment was conducted to evaluate effect by split-application of slow-release fertilizer on the tea plant. The yield of the 2nd harvested tea leaves was not different to the slow-release fertilizer of two time split manuring had been doing Sep. or Mar. compared with the traditional manuring had been doing four time split manuring, but that of the slow-release fertilizer to one time split manuring in Sep. had decreased $12.5{\pm}1.5%$. In case of the 2nd harvested leave, the contents of chemical components related to quality such as total nitrogen, total amino acid were somewhat higher in the slow-release fertilizer (two time split manuring) than in the traditional manuring, but those of tannin, and caffeine were low, and those of chlorophyll, vitamin C, free sugar and theanine were not different to out of treatments. In scoring test, appearance and quality of green tea were more excellence in the two time split manuring compared with one time split manuring of slow-release fertilizer and with the traditional manuring (four time split manuring). Therefore, I thought that use of slow-release fertilizer be increased yield and quality of tea leaves, and improved efficiency nature of nitrogen, phosphate and potassium out of soil fertilizer components.

  • The soil management practices before transplanting the ginseng plant were studied with two organic matter sources such as a traditional organic matter (wild grass) and commercial organic fertilizer (byproducts of amino acid fermantation) during the late spring to late autumn. During the soil management practices, the soil received 40kg N/10a from five different combination treatments with above two organic matter sources, a wild grass and a commercial organic fertilizer. After the application of the treatments, the soil were ploughed regularly at the interval of 20 days and the changes of physicochemical properties during the soil management practices were investigated. The next year after soil management practices, ginseng plants were transplanted to each treatment, growth and the content of some organic components of ginseng plant were measured for comparision of the different treatments. 1. The decrease in bulk density observed during the first 40 days of management was considered to be the effect of the improved physical conditions caused by ploughing, The decrease in bulk density observed after 40 days of management was considered to be the effect of organic matter. Similar results were observed in particle density, however porosity increased with time. 2. Soil pH tended to decrease during the first 40 days of management, after which period the pH increased and was stabilized. However, CEC increased with organic matter treatment and the exchangeable $NH^+_4-N$ and $NO^-_3-N$ increased in 20 and 40 days after the management practices, respectively, and after that period it became steady. 3. The decomposition rate of treated organic matter was measured by the incubation test in laboratory conditions. The rate of decomposition was rapid during the first 20 days of management, after which period it showed slight changes. 4. The weight of ginseng root significantly increased in the treatment of 10kg N/10a organic fertilizer and 30kg N/10a wild grass. 5. The saponin content of ginseng root was highest in the 40kg N/10a wild grass treatment. The addition of organic fertilizer at the rate of more than 20kg N/10a caused the decrease in the saponin content.

Effect of Agro-chemical Alternatives on the Yield and Fruit Quality of Apple (대체농업자재가 사과의 수량 및 품질에 미치는 영향)

  • This study was tried to examine the efficacy of apples that had grown with agro-chemical alternatives in an apple orchard. The chlorophyll content of apple leaves was a little higher in the plot of Charcoal powder, Chitosan, and Peat moss treatment among the other agro-chemical alternatives. On the contrary, the sugar content of apple fruits was higher in the plot of Amino acids, and Green ion calcium treatment, but there was quite a difference among them. Vitamin C content of apple fruits was high at the cell division period. It became lower at the hypertrophic period and then got higher again at the harvest. Vitamin C content was the highest in the chemical fertilizer plot as well. In an apple orchard where agro-chemical alternatives were used only, the commercial grade of ripened apples had declined by 25%, compared to those in customary cultivation. The commercial grade of ripened apples with agricultural chemicals using fertilizers and pesticides was over 90% with over 250g of its weight in the plot of Charcoal powder, Amino acids, and Vitamin C treatment. Therefore. we guessed the environment-friendly method of cultivation in an apple orchard shall be established by reducing the usage of agricultural chemicals gradually and increasing the agro-chemical alternatives at the same time.

Composition and Nitrogen Distribution of Peat and Organic Matter from Paddy Soil in Korea (한국산(韓國産) 이탄(泥炭)과 답토양(畓土壤)의 유기물(有機物) 조성(組成)과 질소(窒素)의 형태별(形態別) 분포상(分布相))

  • In a series of studies on the chemical compositions and properties of organic matter from peats, volcanic ash soils and paddy rice soils, a comparative study on the composition and the fractional distribution of nitrogenous organic matters were undertaken for the elucidation on their characteristics. The results obtained are summerized as follows. 1. Gimpo and Peongtaeg peats showed higher content of organic matter, more than 60%, than Gimje and Yeongdong peats, and there is a great difference between the two groups in total-N, CEC and ash content. 2. The content of Lignin, Hollocellulose and Hemicellulose in organic matter was 12-25%, 15-31% and 7-14% respectively, and Gimpo peats were lots of Lignin content compared to others. 3. Sixty seven to eighty eight percent of the total-N in the soil was hydrolysable by 6 N-HCl and they were decreased in the order of volcanic ash soils (86.4%), mineral paddy soils (77.2%) and peats (72.3%). 4. The fractionations of acid hydrolysable organic-N were ${\alpha}$-amino acid-N(25-45%), unidentified-N (12-50%), amino sugar-N (1-7%) and ammonium-N (12-25%). 5. Almost same pattern in the amino acid composition of the acid hydrolysates of peat and soil organic matter was showed in the thin-layer chromatogram.
