Purification and Characterization of ($Ca^{2+}$+$Mg^{2+}$)-ATPase of Sarcoplasmic Reticulum from Rat Skeletal Muscle (쥐 근소포체의 ($Ca^{2+}$+$Mg^{2+}$)-ATPase의 분리정제와 그 효소특성에 관하여)

  • The $(Ca^{2+}+Mg^{2+})$-ATPase has been purified homogeneously from sarcoplasmic reticulum of rat skeletal muscle by sucrose density gradient centrifugation. The purified enzyme has a molecular weight of 115,000 as judged by polyacrylamide gel electrophoresis in the presence of sodium dedecyl sulfate, and therefore has the same size of the enzyme in rabbit and chick skeletal muscle. $Ca^{2+}, Mg^{2+}, Fe^{2+}, Co^{2+}, and Mn^{2+}$ at 50 $\\muM$ show stimulatory effect on the ATP-ase, while $Zn^{2+}, Cu^{2+}, and Hg^{2+}$ inhibit it at the same concentration. The ATPase activity is insensitive to antimalarial drugs such as quinine and quinacrine, but is sensitive to inhibition by p-hydroxymecurie benzoate and phenylmethylsulfonylfluoride. The enzyme has optimum pH of 6 to 7 and Km value for ATP is estimated to be 98 $\\muM$. Thus, a number of biochemical properties of this enzyme appear to be different from those of the enzyme that have been isolated from rabbit skeletal muscle. The $(Ca^{2+}+Mg^{2+})$-ATPase appears to be selectively degraded in microsomal fraction. The activity of metalloendoprotease is evident in the microsomal preparation when assayed by radioactively labeled protein substrate, such as $^{3}H-casein and $^{125}I$-insulin. However, it is presently unclear whether the metalloendoprotease is responsible for the degradation of the $(Ca^{2+}+Mg^{2+})$-ATPase.

  • In 2007, 94% of organic wastewater sludge from industries located in Ulsan was disposed of by ocean dump. The ocean dump of organic sludge would be totally prohibited by the year of 2012. However, there is no alternative but incinerating the sludge from the industries located in Ulsan. Securing the technology for sludge treatment and on-land disposal is very important issue among the industries in the Ulsan Petrochemical Industry Complex. In this study, the material aspects of dried and carbonized sludge as a fuel were evaluated for petrochemical and sewage sludge from Ulsan. The dried and carbonized sludges from the factories producing terephthalic acid, BTX, propylene, chemical textile, etc. of which the low heat value exceeded 3,000 kcal/kg had high potential as a fuel according to the results of thermal characteristic analysis. However, the dried sludges with heat values lower than 2,100 kcal/kg and carbonized sludges, lower than 1,100 kcal/kg containing more Inorganic material from the industries producing pulp, paper, methylamine, amide, etc. had a little potential to be used as a fuel. In most cases, drying the sludge showed better results than carbonization in the aspect of thermal characteristics of sludge.

  • EHT and CNDO/2 calculations have been performed to determine conformations of acetamides and diacetamides, and of their protonated forms. Results show that: protonation is always favored on the cis position with respect to N due to greater attractive potential between $H^+$ and N; the trans-trans conformer of diacetamides gives the most preferred protonated form although the cis-trans conformer is the most stable one for the unprotonated diacetamides. Protonation on a carbonyl oxygen is predicted to increase both charge and orbital controlled $S_N$SN reactivities of the protonated carbonyl carbon due to increases in positive charge and AO coefficient of ${\pi}$-LUMO of the carbon atom. In the acid catalyzed hydrolysis of diacetamides therefore it appears highly probable that the rate determining attack by a water molecule occurs at the carbon of the protonated carbonyl group and the carbonyl carbon-nitrogen bond scission follows subsequently. This mechanism is consistent with that generally accepted for the hydrolysis of amides in dilute acid solution but disagrees with that proposed by Laureut et al., for acid hydrolysis of N-acetyl-lactams.

  • Lysyl Oxidase from fetal bovine aorta was purified to homogenity using extraction, Sephacryl S200HR chromatography, Hydropore AX ion-exchange high performance liquid column chromatography, Cibacron blue affinity chromatography, and Sephacryl S-300 HR chromatography in the presence of protease inhibitor. The purified enzyme was active toward lathyritic collagen as well as elastin and was sensitive to aminonitriles such as BAPN. Upon Sephacryl S-300 HR chromatography, the enzyme was eluted as a peak with a $K_{av}$ value of 0.45 (65% of $V_t$ ) and it eluted from high performance liquid ion-exchange column (Hydropore |AX) at single position (ionic strength, I = 0.1~0.15). Once purified, it showed one band upon SDS-PAGE. It migrated to a band the mobility of which corresponded to a Mr of 33,500 upon reduction while it migrated to a 24,500 Mr position under the non-reducing condition. In constrast to other reports, it is concluded that fetal bovine aorta contains only one type of lysy oxidase.

  • The smear media possible to sampling and radiation detection was prepared and evaluated for the surface contamination using indirect method. The films were made by impregnating Cerium Activated Yttrium Silicate (CAYS) in a polysulfone membrane. The membranes used solution as a dimethylformamide (DMF) and methylene chloride (MC), polysulfone as a polymer matrix and CAYS as a inorganic scintillator. The proximity membranes were prepared with single- and double-layered structure. The solidified methods were immersion to the nonsolvent bath such at water and ethanol and solvent evaporation. The measurement of the photon produced by interaction with radiation and inorganic scintillator used a photomultiflier tube (PMT), amplifier, and counter. In the comparison with the low background alpha/beta counter, the counter rate using inorganic scintillator proximity membrane for the $\^$14/C surface contamination was about 50%. Also. the $^3$H counting results revealed that the prepared membranes were efficient to monitor the surface contaminated with the low energy be-ray emitter nuclides.

  • When suspended solids are removed by natural sedimentation, it is necessary to use mainly huge equipments and waste great cost. This is a problem that must be solved certainly as soon as possible. In this study, suspended solids of yellow soil water were rapidly removed by controlling pH and amounts of magnetic powder, organic and inorganic flocculants. In the case of the suspended solids of 0.3% yellow soil water, the most excellent turbidity was achieved at pH between 7 and 7.5.

  • The surface of dense polysulfane films was modified through solvent treatment. The modification process consisted of dipping a film for one second in dimethylformamide and then immersing it Into a nonsolvent bath. After being solidified, the original transparent film transformed into an opaque white one, which is due to the light scattering on pores newly developed on the surface. The surface roughness entailing the pore formation was more explicit on a film coagulated by water as nonsolvent than on a film coagulated by isopropanol. The surface-modified films show the better pick-up efficiency than a conventional filter paper on the detaching of radioactive contaminants on the contaminated area. The pick-up efficiency of the film prepared by the water immersion process was superior to that of the film prepared in the isopropanol bath, which was consistent with the surface roughness result. The surface-modified films kept the dense inner structure, playing a major role preventing a possible secondary contamination during the pick-up process.

  • Reaction of monochloroacetic acid with zinc in presence of toluene, dimethylformamide, dimethyl sulfoxide, tetrahydrofuran, acetonitrile, and dioxane solvents showed the solvent effect in order of dimethylformamide, dioxane, dimethyl sulfoxide, toluene, acetonitrile, and tetrahydrofuran. The increasing reactivity of the reagents was observed in order of the polarity and hydrophilicity of the solvent. The same reaction of ethyl monochloroacetate in the same series of solvents showed also the solvent effect, but the difference was not large as compared to that of the acid. The reaction of the acid, ester, zinc, and carbonyl precursors such as benzaldehyde and 4-heptanone gave the Reformatsky reaction product in the case of the aldehyde, but the reactivity with 4-heptanone was slight. The yield of the product was varied by the method of addition of reagents. The best yield observed in case of the acid reaction was 38.5% of the hydroxy acid and 0.8g of the salt in presence of acetonitrile and in case of the ester reaction 19.3% of ethyl cinnamate and 21.6% of polymer. The variation of the reactivity of reagents due to the reaction temperature was observed in case of carbonyl reaction. The result was discussed in terms of the solvent effect and the procedures were described.

  • Toxic epidermal necrolysis (TEN) is a rare, acute and life-threatening cutaneous drug reaction. TEN is characterized by the sudden onset of extensive necrosis in the epidermis and frequent mucous membrane involvement. The pathogenesis has not yet been elucidated. In addition, no particular treatment for TEN has been established. We report a case of TEN in a 14-year-old-boy, which might have been caused by steroids with enalapril treatment for membranous nephropathy. He recovered after intravenous immunoglobulin therapy.

  • For the efficient recovering of collapsed sloped soil, using a soil binder that can support the soil strongly and help the growth of plants is very important. The soil binder should also have functions of recovering the soil ecologically as well as be environmental friendly materials. In this research, optimum values of the water content and permeability and direct shear strength were searched by adding the water absorbent and coagulant into the soil binder. The polyacrylamide (PAM) with various anionic strength, super absorbent polymer (SAP) and cellulose ether (CE) were used as a soil binder, water absorbent and coagulant, respectively. Effects of the soil binder on the characteristics of soil were observed by changing the mixing ratio of PAM, SAP and CE. Experimental results showed that the soil binder increased the direct shear strength tens of times and the water content around two times, whereas decreased the water permeability. Also, the addition of CE to increase the coagulation of SAP increased more of the direct shear strength and water content.
