This study was performed to evaluate the quality characteristics of Sulgi (Korean steamed rice cakes) prepared with fresh or frozen red onions. Sulgi was prepared with different levels (10, 20 or 30%) of fresh or frozen onions and stored for 4 days. The L value significantly decreased with the addition of fresh red onions and the a value significantly increased with the addition of either fresh or frozen red onions. The pH value was also increased with the addition of fresh red onions. In terms of texture, the hardness of Sulgi with either fresh or frozen red onions was reduced. The gumminess and brittleness gradually decreased with the addition of fresh or frozen red onions. Sulgi composed of 30% red onions showed the best sensory results in terms of color, flavor, texture, taste, and overall acceptance.
This study was carried out to investigate the microbiological contamination levels of Dutch coffee products marketed in Korea. The temperature conditions during distribution and storage were also considered in this experiment. Retailed Dutch coffee were purchased from regional cafes, that is, these were self-blended by the cafes, and the marketed products were purchased from department stores and from Internet sites. The 21 samples were blended in a coffee house and 9 were obtained from department stores or were delivered from internet sites. House blended Dutch coffee contained $35.2{\pm}15.8CFU/mL$ of general bacteria, and this increased to $78.4{\pm}29.7CFU/mL$ at room temperature or $51.2{\pm}32.1CFU/mL$ after refrigeration for 5 days. These almost reached the highest criteria level for the Korea Food Sanitation Law. After 10 days, the count increased to $98.5{\pm}58.4CFU/mL$ at room temperature and $86.7{\pm}44.2CFU/mL$ at refrigeration temperature. In the Dutch coffee for distribution, $39.6{\pm}20.1CFU/mL$ of general bacteria were detected, but these did not increase after 5 days or 10 days both for room temperature and under refrigeration. The Coliform group was not found in any kind of Dutch coffee, and Fungi was founded in 60% of the Dutch samples purchased in coffee houses, department stores, and shopping sites mall. On day 0 day, $2.6{\pm}1.7CFU/mL$ of fungi were detected in the coffee house Dutch, and it did not increase significantly during the storage period at room and in a cold temperature. $3.5{\pm}3.4CFU/mL$ of fungi were detected in the Dutch coffee for distribution, and it didn't increase during further storage under any temperature.
The purpose of this study was to evaluate the physical properties of hydrophilic polyvinyl siloxane impression materials as change of material's storage temperatures. Working time, strain-in-compression, elastic recovery and consistency were tested according to ISO Standard NO. 1563. The results are as followed. 1. Working time decreased in cold storage. 2. Strain-in-compression was different in storage temperatures. Material's strain-in compression in cold temperatures were higher than in room temperature and in incubator. 3. A coefficient of elastic recovery varied by storage temperatures. The rate in cold temperature was the lowest and in incubator was the highest. 4. Consistency of impression substance different in storage temperatures. The extent in cold temperature is the highest and in incubator was the lowest. Statistical analysis(SPSS 14.0k, p>0.05) showed that storage temperature affect to material's physical properties. We recognized that the physical properties of polyvinyl siloxane impression materials were changed according to storage temperature.
This experiment was carried to investigate the germination pattern in relation to temperatures and lights, and the emergence pattern in relation to seeding depths, lights and the alpha amylase activity of Youngia sonchifolia, Lactuca indica var. laciniata, Ixeris dentata var. albiflora and Ixeris polycephala. In Y. sonchifolia, the optimum germination temperature was $25^{\circ}C$, the optimum seeding depth to emerge was 0 mm and it could emerge in 0-5mm. In L. indica var. laciniata under cool storage, the optimum germination temperatures were $19^{\circ}C-28^{\circ}C$, the optimum seeding depth was 5mm and it could emerge in 0-20mm. In L. indica var. laciniata under room storage, the optimum germination temperature was $25^{\circ}C$ the optimum seeding depth was 5mm and it could emerge in 0-10mm. In I. dentata emerge was and 0mm and it could emerge in 0-5mm. In I. polycephla, the optimum temperatures were $16^{\circ}C-19^{\circ}C$, the optimum seeding depth to emerge was 0mm and it could emerge in 0-5mm. The alpha amylase activity was lower Y. sonchifolia, L. indica var. laciniata and I. dentata var. abiflora than barley cultivar Dongbor#1. And the increased pattern of alpha amylase activity was likely to it of germination rate.

Ginseng-whey beverages were prepared with rennet whey, ginseng extract, sweetener, honey and Japanese apricot, inoculated with different strains of lactic acid bacteria and unfermented partly. The samples were stored at 4$^{\circ}C$ or 30$\pm$1$^{\circ}C$ and the sensory evaluations were carried out at 1st, 3rd and 5th week. As a result of sensory test, unfermented ginseng-whey beverage (A) with sweetener and honey (storage at cold temp.) in overall eating quality obtained the best score (8.64~8.86) due to stronger sweetness and weaker sourness, bitterness, astringent taste and aftertaste. The fermented ginseng-whey beverage (C) which was stored at 4$^{\circ}C$ with inoculation of Lac. acidophilus and Lac. delbrueckii sub-sp. bulgaricus and the unfermented samples stored at room temperature with sweetener, honey and Japanese apricot received a good evaluation. But, the fermented beverages (E, F) stored at room temperature obtained the lowest score (2.92~3.58).

농산물의 품질 및 성분을 측정하는데 있어 기존의 화학적 분석 방식은 정밀도가 높으나 측정에 소요되는 시간과 비용이 많이 들어, 현장 적용하기에는 한계가 있다. 일반적으로 근적외선 분광 분석(Near Infra Red Spectroscopy, NIRS) 방법은 가공 과정에 따라 빠르게 변화되는 단백질 조성 및 수분함량 측정 등에 이용되고 있다. 분석에 소요 시간이 많이 걸리는 켈달법(Kjeldahi method)에 비해 NIR 분광 분석을 통한 보정으로 연속적인 모니터링이 가능하다. 본 연구에서 사용된 시료를 고정시키기 위한 프레임을 제작한 후 NIR센서와 광원인 LED의 각도를 고정시키고 측정 대상체인 사절된 감자 크기에 따라 시료를 고정시킬 수 있는 프레임을 반사면에 위치시켰다. 확산 반사법을 이용하여 프레임에 씨감자 시료를 고정 시킨 후 백색 LED를 이용하여 감자 표면에 빛을 반사시켜 3일 동안 12시간 마다 해당 시료들(열처리, 비누용액 침지, 생감자)의 스펙트럼을 측정하였다. 해당 시료들은 측정 기간 동안 저온상태($4^{\circ}C$)와 실온상태($20^{\circ}C$)에서 보관되었다. 실험 결과는 파장대 145nm에서 저온상태에서 보관된 생감자는 시간경과에 따른 흡광도의 결정 계수값($r^2$)은 0.98 이었다. 이는 감자가 저온에서 생감자의 상태 변화가 일어나고 있다는 것을 의미하고 파장대 145nm에서 시간에 따른 저온상태에서 보관한 감자의 상태 변화 예측이 가능함을 의미한다. 비누용액에 침지시킨 후 실온에 보관한 감자는 시간이 경과함에 따라 파장이 증가함에 따라 흡광도가 증가하였다. 이는 감자에 들어있는 Polyphenol Oxidase 함량 변화로 갈변 현상이 일어난 것을 알 수 있다. 또한 실온에서 보관한 생감자도 시간 경과에 따라 갈변 현상이 일어났지만 용액에 침지시킨 감자보다는 갈변 현상이 36시간 이후로 발견되었다. 열처리 후 실온에서 보관한 감자의 경우에는 갈변현상이 나타나지 않았다. 저온상태에서 보관한 감자시료들 모두 갈변형상이 나타나지 않았지만, 24시간이 지난 후 용액에 침지시킨 감자는 갈변 현상이 발생되었다. 생감자와 열처리한 감자는 시간 경과에 따른 갈변현상이 일어나지 않았으므로, 감자의 갈변현상은 감자의 표면 처리 방법에 국한되지 않고 온도에 영향을 더 많이 받는다는 것을 나타내고 있다. 본 연구는 향후 감자의 품질 및 성분 측정에서 간편하게 사용될 수 있는 감자의 품질 계측 기술에 기여할 것으로 판단된다.
The most important progress in diagnostic sciences is the increased sensitivity and specificity in diagnostic procedures due to the development of micromethodologies and increasing availability of immunological and molecular biological reagents. The technological advances led to consider the diagnostic use of saliva for an array of analytes and DNA source. The purpose of the present study was to compare DNA from saliva with those from blood and buccal swab, to evaluate diagnostic and forensic application of saliva, to investigate the changes of genomic DNA in saliva according to the storage temperature and period of saliva samples, and to evaluate the integrity of the DNA from saliva stored under various storage conditions by PCR analysis. Peripheral venous blood, unstimulated whole saliva, stimulated whole saliva, and buccal swab were obtained from healthy 10 subjects (mean age: $29.9{\pm}9.8$ years) and genomic DNA was extracted using commercial kit. For the study of effects of various storage conditions on genomic DNA from saliva, stimulated whole saliva were obtained from healthy 20 subjects (mean age: $32.3{\pm}6.6$ years). After making aliquots from fresh saliva, they were stored at room temperature, $4^{\circ}C$, $-20^{\circ}C$, and $-70^{\circ}C$. Saliva samples after lyophilization and dry-out procedure were stored at room temperature. After 1, 3, and 5 months, the same experiment was performed to investigate the changes in genomic DNA in saliva samples. In case of saliva aliquots stored at room temperature and dry-out samples, the results in 2 weeks were also included. Integrity of DNA from saliva stored under various storage conditions was also evaluated by PCR amplification analysis of $\beta$-globin gene fragments (989-bp). The results were as follows: 1. Concentration of genomic DNA extracted from saliva was lower than that from blood (p<0.05), but there were no significant differences among various types of saliva samples. Purities of genomic DNA extracted from stimulated whole saliva and lyophilized one were significantly higher than that from blood (p<0.05). Purity of genomic DNA extracted from buccal swab was lower than those from various types of saliva samples (p<0.05). 2. Concentration of genomic DNA from saliva stored at room temperature showed gradual reduction after 1 month, and decreased significantly in 3 and 5 months (p<0.05, p<0.01, respectively). Purities of DNA from saliva stored for 3 and 5 months showed significant differences with those of fresh saliva and stored saliva for 1 month (p<0.05). 3. In the case of saliva stored at $4^{\circ}C$ and $-20^{\circ}C$, there were no significant changes of concentration of genomic DNA in 3 months. Concentration of DNA decreased significantly in 5 months (p<0.05). 4. There were no significant differences of concentration of genomic DNA from saliva stored at $-70^{\circ}C$ and from lyophilized one according to storage period. Concentration of DNA showed decreasing tendency in 5 months. 5. Concentration of genomic DNA immediately extracted from saliva dried on Petri dish were 60% compared with that of fresh saliva. Concentration of DNA from saliva stored at room temperature after dry-out showed rapid reduction within 2 weeks (p<0.05). 6. Amplification of $\beta$-globin gene using PCR was successful in all lyophilized saliva stored for 5 months. At the time of 1 month, $\beta$-globin gene was successfully amplified in all saliva samples stored at $-20^{\circ}C$ and $-70^{\circ}C$, and in some saliva samples stored at $4^{\circ}C$. $\beta$-globin gene was failed to amplify in saliva stored at room temperature and dry-out saliva.

Purpose: For orthodontic bracket bonding, light curing resin cement is widely used because the process is convenient, and it can be polymerized at the desired time. This study compared the difference of bonding strength of orthodontic resin cement according to storage condition. Materials and Methods: After acid etching the bovine enamel surface with 37% phosphoric acid, 15 orthodontic brackets for mandible incisors were bonded with Ortho Connect and Orthomite LC according to following three conditions; 1) Immediate after 4℃ refrigeration for 3 months (IR), 2) One day room temperature after 4℃ refrigeration for 3 months (OR), 3) Room temperature for 3 months (RT). The shear bond strength was measured with a universal material tester and failure pattern of the specimen was observed. Two-way ANOVA and One-way ANOVA were used at the 95% significance level. Results: Ortho Connect that was applied immediately after refrigeration showed the maximum shear bond strength. Orthomite that was applied immediately after refrigeration showed the lowest shear bond strength, and the group stored at room temperature for three months showed the highest shear bond strength, and the difference between the two groups was significant. Conclusion: Ortho Connect can be used without worrying about bond strength even if it is used immediately after refrigeration, but Orthomite should be kept at room temperature sufficiently after refrigeration.
Background: Preanalytical factors can affect reliability of hormone assay results. Adrenocorticotropic hormone (ACTH) in blood is considered highly unstable because of proteolytic degradation, so storage of blood samples on ice until analysis is recommended. In clinical practice, however, this procedure may present logistical problems because most samples for ACTH measurement must be shipped from the place of sample collection to the laboratory. Therefore, we studied the impact of time and temperature before plasma separation and analysis on the results of ACTH assays. Methods: A total number of 22 patients were enrolled in this study. We obtained 2 blood samples. ACTH concentrations were 35~126 pg/mL. ACTH concentrations were measured by immunoradiometric assay (IRMA) using commercial kits (CIS Biointernational, Gif-sur-Yvette, France). Results: ACTH levels showed a significant difference between the samples of $22^{\circ}C$ EDTA and $4^{\circ}C$ EDTA. Measured ACTH concentrations significantly decreased with time before freezing at $-20^{\circ}C$. ACTH levels showed no significant difference between the groups of after storage for 24 hr without centrifugation at $22^{\circ}C$ and $4^{\circ}C$. Conclusion: We recommend that blood samples be obtained on pre-chilled EDTA collection tubes. The shortest possible time between sample collection and processing is always the best laboratory practice.
