Taxonomic status of Goodyera rosulacea (Orchidaceae): molecular evidence based on ITS and trnL sequences (로젯사철란(Goodyera rosulacea: Orchidaceae)의 분류학적 위치: ITS와 trnL 염기서열에 의한 분자적 증거)

  • Goodyera rosulacea, which is morphologically similar to G. repens, is described recently as a new species based on its distinct morphological characters such as rosette-formed leaves, short rhizome and habitat. To verify the taxonomic identity of G. rosulacea and its taxonomic relationship within Korean Goodyera taxa, sequences of the internal transcribed spacer (ITS) region of nuclear ribosomal DNA and the trnL region of cpDNA from 24 accessions including 1 outgroup accession were analyzed. Aligned sequences were analyzed using maximum parsimony and distance method, and the taxonomic identity and the taxonomic relationships among the related taxa were estimated by the existence of private marker gene and the phylogenetic tree of the aligned sequences. Molecular data indicate that G. rosulacea gas several private marker genes and shows monophyly in phylogenetic trees of both ITS and trnL sequences. the pairwise distance between G. rosulacea and the orher taxa of Korean Goodyera was 3.49-6.68% for ITS region and 5.05-9.53% for trnL region, indicating that G. rosulacea could be treated as an independent species. Therefore, our molecular data support the taxonomic of G. rosulacea as a distinct species of Korea. In phylogenetic trees, G. rosulacea formed same clade with G. repens, which has similar morphological characters with G. rosulacea, and showed the lowest pairwise distance with G. repens among Korean Goodyera taxa. These molecular data sugguested that G. rosulacea and G. repens are closely related taxa.

Effects of Root Diameter and Peeling Methods on Drying Time and Paeoniflorin Content of Paeonia Lactiflora Pallas (작약(芍藥)의 뿌리굵기와 각피(刻皮)정도가 건조시간 및 품질에 미치는 영향)

  • This experiment was carried out to gather basic information for improvement of peeling methods in peony (Paeonia lactiflora) roots which have been used for medicinal resources in Korea. Drying time required was shortened in thin root. Optimum drying time after machine peeling appeared to be 20 hrs in 5 to 10mm of root diameter, 39 hrs in 10 to 15mm, 48 hrs in 15 to 20mm, 56 hrs in more than 20mm, but those of unpeeled roots took 11 hrs in 5mm of root diameter and 4 hrs in exposed parent material. Paeoniflorin content in dried roots after peeling was lower than that of unpeeled root. Paeoniflorin content in root below 5mm in diameter was highest (6.15 %) and that was decreased in the increased root diameter up to 20mm, but it was slightly increased in root greater than 20mm in diameter.

Habitat Characteristics of Saussurea chabyoungsanica (자병취의 생육지 특성)

  • Saussurea chabyoungsanica was recorded in new species by Lim Hyoung Tak in 1997. In order to understand the entity of new species, we investigated the new distribution sites, vegetation structure and soil environmental factors. Additionally discovered distribution sites of S. chabyoungsanica were Manduckbong, Mt. Sukbyoung, Mt. Duckhang, and Sukgaejae, which were located on ridge of the Taebaek Mountains. Those sites are typical limestone zones of Korea and among them Sukgaejae belongs to lower great limestone area and Manduckbong, Mt. Sukbyoung and Mt. Duckhang belong to upper great limestone area. According to the result of phytosociological study, plant communities of S. chabyoungsanica were classified by 1 Community group, 4 Communities and 2 Subcommunities; Carex humilis var. nana-Saussurea chabyoungsanica Community group in wide sense Quercus mongolica Community group, Lespedeza maximowiczii-Saussurea chabyounsanica Community, Galium kinuta-Saussurea chabyoungsanica Community, Salvia chanryonica-Saussurea chabyoungsanica Community, Zabelia coreana-Saussurea chabyoungsanica Community. Environmental factor analysis of habitat showed that the distribution site of S. chabyoungsanica was ridge of North aspect and displayed pH $7{\sim}8$ typical of limestone. Soil moisture content was high, whereas organic matter content was low. Considering its high occurrence in sites of $1{\sim}10cm$ in soil depth, and of $30{\sim}45%$ in relative light intensity, major distribution sites were outcrops and boundaries between shrubland and forest. After community division by vegetation structure in habitat of S. chabyoungsanica, we investigated characters of soil environment by community. As a result of PCA analysis of soil sample by community, it was possible to divide community by characters of soil environmental factor. The cumulative value of contribution rate represented in second dimension space was 73% and the major factors for that value were soil texture, organic matter content and field capacity.

  • 지리산은 해발 1915m인 남한 본토 최고인 명산이며 국립공원으로 지정된 바도 있으나 아직 그 동물상에 대한 조사는 불충분한 바 있다. 1972년 8월 11일-14일의 4일간에 걸쳐 남궁 윤은 한라산 거미상과의 비교 검토를 위하여 지리산의 제 2 고봉인 반야봉과 제 3고봉인 노고단을 중심으로 한 지리산 서남면인 "겉지리산"의 일부를 조사하였다. 한편 백외는 1971년 7일 하순 화엄사, 노고단 및 피아골의 거미상에 대해 조사한 바 있다. 광대한 지리산의 전체적인 면모를 파악하기에는 미급한 조사였지만 대치적인 특징을 추찰한 수 있었기에 이에 보고코져 한다. 지리산 거미에 대하여는 백 갑용(1942-1972), 백운하외 (1970) 등에 의해 18과 76종 (3미확정종포함)이 보고된 바 있으나 채집기록에 불분명한 바가 있고, 그 환경이나 수직분포 등에 대하여는 언급된 바가 없었다. 금번 조사한 장소를 고도별로 보면 다음과 같다. 연곡사(300-400m), 화엄사(400-600m), 피아골(500-900m), 화엄사골(600-900m), 용수바위 (900-1300m), 코재 (900-1300m), 임걸령(1400-1500m), 노고단(1400-1550m), 반야봉(1550-1750m) (1) 이 조사보고에서는 종전의 보고에서 기록이 분명한 것을 종합하여 21과 135종 (미확정종 6종 포함)의 목록을 작성하였다. (2) 지리산 추가종 56종을 확인하였으며 그 중에서 한국 미기록종은 16종이다. (목록에 *표를 한 것) (3) 미확정종은 개체특징이 분명한 것만 수록했으며 이들 중 금후 연구로써 신종으로 기재됨 것도 있다. (4) 이번 조사의 결과로는 지리산의 거미상은 북방종 26종$(20.2\%)$ 남방종 8종$(6.2\%)$ 세계공통종이 3종$(2.3\%)$으로 북방온대성 $(71.3\%)$의 경향이 짙으다. (5) 고도분포는 반야봉 (1550-1750m)에서 18종$(7.3\%)$, 1400-1550m가 38종$(15.5\%)$, 900-1400m에서 39종$(15.8\%)$, 600-900m에서 72종$(29.2\%)$, 600m이하에서 79종$(32.1\%)$으로 고도가 높아짐에 따라 종수가 점감되고 1300m 이상에 서식하는 것은 44종$(32.6\%)$에 불과하였다. (6) 지리산의 우점종은 대륙접시거미 (Prolinyphia emphana)로 600m 이상부터 산정부에 걸쳐 널리 분포하며, 개체수에 있어서도 가장 많았다. 깡충거미과(Salticidae), 늑대거미과(Lycosidae)가 극히 적은 것도 특징적이라 하겠다.

Characteristics of the Outbreak Area of Diprion hani (Hymenoptera: Diprionidae) and a Report of its Torymid Parasitoid (잣나무솔잎벌, Diprion hani (Hymenoptera: Diprionidae), 대발생지 특성과 기생천적인 가시꼬리좀벌에 대한 보고)

  • Diprion hani Smith and Cho was first reported as a new species from South Korea in 2007. Major outbreaks occurred in September 2007. Outbreaks were restricted geographically to only three inland areas of central Korea where had previously been reforested with Korean white pine, Pinus koraiensis Seib. & Zucc. The outbreak areas were approximately 50 km in diameter. The sawfly populations decreased drastically in 2008. Environmental factors of the outbreak sites were investigated as a first step to characterize the sites and to understand the population dynamics of the sawfly. During the outbreak in 2007, the average temperature of the sites were $1.3^{\circ}C$ higher than the 30 years average, and the annual precipitation was 384 mm higher than the average from 1971 to 2000. In addition, Monodontomerus dentipes (Dalman), a well-known torymid parasitoid of many sawflies, is reported as a gregarious cocoon parasitoid of D. hani.

Effect of $GA_3$, Kinetin and Physical Treatment on the Seed Germination of Zanthoxylum piperitum A.P. DC. ($GA_3$, Kinetin 및 물리적 처리가 초피나무 종자의 발아에 미치는 영향)

  • This study was conducted to improve germination ratio of Zanthoxylum piperitum A.P. DC. seeds. Stratification for 60 days after scarification of seed with sand was germination percentage to 5.4% and $GA_3$, 50ppm for 24 hrs after scarification of seed with sand showed 8.9%. Soaking the seeds in $GA_3$, 50ppm for 24 hrs after 40 to $70^{\circ}C$ hot water treatment for 10 minutes showed low germination of 4.4%. Based on $H_2SO_4$, NaOH and $HNO_3$, treatments, germination percentage did not improve at all regardless of soaking time. The highest germination of 91.1% was observed when seed was soaked in $GA_3$ 100ppm for 48 hrs after stratification for 60 days at $4^{\circ}C$. Kinetin treatment at 50ppm for 24 hrs had the greatest germination percentage of 31.7% but it did not improve germination ratio compared to $GA_3$ treatment.

Effects of Removing Time of Flower Buds on Root Yield and Paeoniflorin Content in Paeonia lactiflora Pallas (작약(芍藥) 화뇌제거시기에 따른 근수량(根收量)과 Paeoniflorin함량(含量) 변화(變化))

  • This experiment was carried out to determine the proper time for flower bud removing to improve growth, yield and paeoniflorin content of root in peony(Paeonia lactiflora Pallas). The flower buds removement caused short stem length by $7{\sim}9cm$ compared with control, but number of stem and stem diameter were similar to that of control. The number of root, fresh root weight and paeoniflorin content in peony increased when their flower buds were removed at earlier stages. The root dry weight was highest of 1.647kg/10a, and increased by 27% compared with control when their flower buds were removed at early stage (bud diameter was less than 10mm). The root dry weight increased by 13% and 10%, respectively. when their flower buds were removed prior to flowering and at flowering stage. Paeoniflorin content in leaf was higher than that in stem, and was higher at early stage. Leaf and stem showed higher paeonif1orin content when their flower buds were removed.

  • The appropriate drying method in medicinal compounds and color of peony root was that predrying at briquet fire $(40{\sim}60^{\circ}C)$ for 6 hours or more, and then hot air drying at $40^{\circ}C$ during 60 hours. But this method needs too much time in drying. And the method that drying at $40^{\sim}C$ with hot air drying has bad result in color. In case Peony was seld by cutting product, before cutting, the Peony root was retted for 30 min. and sealed for 12 hrs. is good for drying time, Peoniflorin content and commodity.

A taxonomic study of the genus Hosta in Korea (한국산 비비추속(Hosta Tratt.) 식물의 분류학적 연구)

  • A taxonomic study of the genus Hosta (Lilliaceae) in Korea was conducted based on morphological characteristics. Hosta clausa varieties share several characteristics (rhizomes with underground stolons, smooth scapes, and purple anthers). Hosta clausa var. normalis has an open perianth, long pistils, and fertile fruit. H. clausa var. clausa can be distinguished from H. clausa var. normalis by its closed perianth and sterile fruit. H. clausa var. geumgangensis has sterile fruit, green petioles, short pistils, and short narrow perianth tubes. H. clausa var. ensata has very narrow, sword-shaped leaves. It can be included in H. clausa var. normalis, which has a large degree of variation in leaf and petiole shape. H. jonesii has smooth scapes, yellow anthers, and bell-shaped flowers. It can be distinguished from the related species H. tsushimensis found on the Tushima islands in Japan by its wide perianth tube. H. yingeri is distinguished by its funnel-shaped flowers, 3 long and 3 short stamens, and smooth scape; the related species H. laevigata has 6 equally-sized stamens, narrowly lanceolate leaves, and wavy leaf margins. Because H. laevigata is not found in its type locality Isl. Heugsando, its taxonomic position must be determined at a later time. H. minor shares several characteristics (scapes with ridges and yellow-colored anthers) with the related taxa H. venusta. However, H. venusta is changed to H. minor var. venusta because it has overlapping characteristics in plant size, number of flowers, leaf size, and leaf shape. Consequently, the genus Hosta in Korea is classified into 5 species, 3 varieties, and 6 forms.
