Anti-climacterium Effects of Gagamguibiondam-tang in Ovariectomized Rats (난소적출로 유발된 랫트 갱년기 장애에 대한 가감귀비온담탕의 생리활성 효과 평가)

  • Purpose: The object of this study was to observe the anti-climacterium activity of Gagamguibiondam-tang (GGOT) on ovariectomized (OVX) rats, a well-documented rodent models resembles with women postmenopausal climacterium symptoms, as including cardiovascular diseases, obesity, hyperlipidemia, osteoporosis, organ steatosis and mental disorders. Methods: In this study, anti-climacteric effects were evaluated separated into three categories; 1) anti-obese, 2) anti-uterine atrophy and 3) anti-osteoporotic effects. Five groups were used (8 rats in each group); sham control, OVX control, GGOT 500, 250 and 125 mg/kg administered groups. Twenty-eight days after bilateral OVX surgery, GGOT were orally administered, once a day for 84 days, and then the changes on the body weight and gain during experimental periods, serum estradiol levels, abdominal fat pad and uterus weights with histopathology of abdominal fat pads (total thickness and mean adipocyte diameters) and uterus (total, epithelial and mucosal thickness, percentages of uterine gland regions) for anti-obese and estrogenic effects. In addition, femur, tibia and fourth or fifth lumbar vertebrae (L4 or L5) wet, dry and ash weights, mineral density (BMD), bone strength (failure load), serum osteocalcin and bone specific alkaline phosphatase (bALP) contents, histological and histomorphometrical analyses - bone mass and structure with bone resorption, were monitored for anti-osteoporosis activity. Results: As a result of OVX, noticeable increases of body weight and gains, food and water consumption, weights of abdominal fat pad deposited in dorsal abdominal cavity, serum osteocalcin levels were demonstrated in this experiment with decrease of uterus, femur, tibia and L5 weights, serum bALP and estradiol levels. In addition, marked hypertrophic changes of adipocytes located in deposited abdominal fat pads, uterine disused atrophic changes, decreases of bone mass and structures of femur, tibia and L4 were also observed in OVX control rats with dramatic increases of bone resorption markers, the Ocn and OS/BS at histopathological and histomorphometrical analysis in this study as compared with sham-operated control rats, suggesting the estrogen-deficient climacterium symptoms - obese and osteoporosis were induced by OVX, respectively. However, these estrogen-deficient climacterium symptoms induced by bilateral OVX in rats were significantly inhibited by 84 days of continuous oral treatment of GGOT 500, 250 and 125 mg/kg, respectively. Especially, GGOT 500, 250 and 125 mg/kg showed clear dose-dependent inhibitory activities on the OVX-induced climacterium signs. Conclusion: The results suggest that oral administration of GGOT 500, 250 and 125 mg/kg has clear dose-dependent favorable anti-climacterium effects - estrogenic, anti-obese and anti-osteoporotic activities in OVX rats in this experiment.

Effects of Occupation-Based Bilateral Upper Extremity Training for Chronic Stroke: Pilot Study (뇌졸중 환자의 작업기반 양측성 상지훈련 효과: 예비연구)

  • Objective : The purpose of this study is to observe effects of Occupation-Based bilateral UE training for chronic stroke. Methods : 4 patients with hemiplegic stroke were divided into two groups: experimental group and control group. From May to August 2016, we conducted a occupation - based bilateral UE training and a traditional bilateral UE training for 5 weeks, 3 times a week, and 60 minutes per session for each subject. EMG, accelerometer, ARAT, Y-BAT, SIS, and COPM were performed before and after the intervention. Results : The experimental group showed a large changes before and after intervention in ECR, DA and all items except for hand function and memory of SIS, compared with the control group. Also, the experimental group showed a large change before and after the intervention compared to the control group in the accelerometer measuring the amount used of bilateral hand and grasp, pinch, and gross movement items of ARAT and COPM, Y-BIT. Conclusions : Occupation-based bilateral upper extremity training was effective in the recovery of upper limb function and social participation through neurological changes. Future research will be required development studies to ensure a high content validity of the Occupation-based bilateral upper extremity training protocol.

  • Parkinson's disease (PD) is the most common movement disorder and the second most common neurodegenerative disease after Alzheimer's disease. Approximately 5~10% of PD patients are familial PD cases. Leucine-rich repeat kinase 2 (LRRK2) has been known to be a causal gene of PD when it is mutated. LRRK2 contains the functional kinase and GTPase domains as well as leucine-rich repeat (LRR) and WD40 domains that are known to play critical roles for protein-protein interaction, suggesting that LRRK2-interacting proteins are important regulators for PD pathogenesis. In an effort to identify proteins interacting with LRRK2, we carried out co-immunoprecipitation of LRRK2 antibody using extracts of NIH3T3 cells that express LRRK2 at a relatively high level. The mass spectrometry analysis of a precipitated band revealed that the co-precipitated band was methionyl-tRNA synthetase (MRS), an ancient enzyme that transfers methionin to its cognate tRNA. The interaction of MRS with LRRK2 was confirmed again by co-immunoprecipitation of endogenous proteins and GST pull-down assays. However, LRRK2 did not phosphorylate recombinant MRS protein in in vitro kinase assays, and over-expression of LRRK2 or MRS did not affect the stability of its partner protein. Our data indicate that LRRK2 interacts with but does not phosphorylate MRS, and the stability of each partner is not affected by the other.

Anti-cancer Effect of Hot Water Extract from Mycelium in Germanium-enriched Cordyceps militaris (게르마늄 농도를 증가 시킨 동충하초 균사체 열수 추출액의 항암효과)

  • Cordyceps militaris has been used in traditional Chinese medicine owing to its anticancer and immunomodulatory activities. Germanium compounds have also been shown to be associated with many pharmacological functions, such as antimicrobial, antiviral, antitumor, antimutagenic, and immunomodulating effects. In this study, we examined the biological properties of hot water extract from mycelial liquid culture of germanium-enriched C. militaris (CMGe). CMGe displayed a concentration-dependent antiproliferation activity against four human cancer cell lines. The antiproliferative activity of CMGe was 2-4-fold lower than that of hot water extract from mycelial liquid culture in C. militaris (CM). However, CM had a concentration-dependent cytotoxicity to human bone marrow-derived mesenchymal stem cells (MSCs). Contrastingly, CMGe did not cause any cellular damage to MSCs. MSCs cultured with CMGe displayed an increased proliferative activity with no cytotoxic effect. The oral administration of CMGe inhibited increased tumor volume and weight compared with the control group. CMGe has the potential to be used as an industrial product in medicinal foods as well as in pharmaceutical products.

도공층의 공극 구조와 인쇄후 잉크의 잔류 거동에 관한 연구(II)

  • 종이의 공극은 도공용 원지 또는 인쇄 용지로 사용될 때 도공액과 잉크 조성분의 침 투 거동에 영향을 미침으로써 여러 가지 물리적 성질에 변화를 가져온다. 도공 공정의 경우, 도공액에 포함되어 있는 물과 수용성 고분자 물질의 원지 침투는 도공 작업성과 도공층의 구조에 변화를 가져 올 수 있다. 이러한 변화는 물을 포함한 수용성 물질의 원지 침투가 빨라질 때 도공액의 고형화점 상승에 의한 원지 피복력 저하에 따른 도공 지의 평활성 감소를 가져온다. 또한 높은 전단력이 가해지는 블레이드 도공에서는 도피 직전 도공액의 수분 이탈이 급격하게 일어날 경우 블레이드에서 도공액의 유동성 저하 가 발생됨으로써 여러가지 문제점을 나타낸다고 보고 된 바 있다. 인쇄 공정의 경우에 는 도공층이 잉크의 직접적인 기질(substrate)로 작용되어 잉크 성분의 잔류 및 침투 거동에 영향을 미친다. 즉 지나치게’발달된공극 특성을 지닌 도공층에서는 잉크 안료가 적절한 결합력을 가지고 도공층에 정착되기 전에 잉크 속의 레진과 용제가 도공층의 공극으로 소실됨으로써 인쇄 완료 후 작은 외력에도 잉크 층이 파괴되는 효킹 ( (chalking) 현상이 일어난다. 그러나 과도한 바인더의 적용과 미세한 안료의 사용에 따 라 도공층의 공극이 폐쇄될 경우에는 인쇄 후 잉크의 건조가 늦어져 인쇄 작업성의 저 해 요인으로써 작용한다. 종이의 공극성과 인쇄적성은 주지하는 바와 같이 불가분의 관계에 있으며, 본 연구에 서는 피인쇄체로서 공극성을 달리한 세 종류의 도공지를 제조하고 실험실적으로 제조 된 잉크를 사용하여 각각의 특성에 따른 잉크의 잔류 특성에 대하여 고찰해 보고자 하 였다. 특히 인쇄 전 피인쇄체의 공극 특성에 따른 인쇄 전후의 공극율 변화와 주요 인 쇄 적성간의 관계에 대해 초점을 맞추었다.시아노에틸화한 PYA용액의 점탄성 평가를 위하여 storage modulus와 loss modulus 를 분석하였다. 일반적 유변특성 평가 결과 PYA용액은 shear-thinning, pseudoplastic 한 특성을 나타내어 표면사이즈 공정에서의 적용 가능성을 확인할 수 있었다. 사용하는 통계기법 중의 하나인 주성분회귀분석을 실시하였다. 주성분 분석은 여러 개의 반응변수에 대하여 얻어진 다변량 자료의 다차원적인 변 수들을 축소, 요약하는 차원의 단순화와 더불어 서로 상관되어있는 반응변수들 상호간 의 복잡한 구조를 분석하는 기법이다. 본 발표에서는 공정 자료를 활용하여 인공신경망 과 주성분분석을 통해 공정 트러블의 발생에 영향 하는 인자들을 보다 현실적으로 추 정하고, 그 대책을 모색함으로써 이를 최소화할 수 있는 방안을 소개하고자 한다.금 빛 용사 둥과 같은 표면처리를 할 경우임의 소재 표면에 도금 및 용 사에 용이한 재료를 오버레이용접시킨 후 표면처리를 함으로써 보다 고품질의 표면층을 얻기위한 시도가 이루어지고 있다. 따라서 국내, 외의 오버레이 용접기술의 적용현황 및 대표적인 적용사례, 오버레이 용접기술 및 용접재료의 개발현황 둥을 중심으로 살펴봄으로서 아직 국내에서는 널리 알려지지 않은 본 기 술의 활용을 넓이고자 한다. within minimum time from beginning of the shutdown.및 12.36%, $101{\sim}200$일의 경우 12.78% 및 12.44%, 201일 이상의 경우 13.17% 및 11.30%로 201일 이상의 유기의 경우에만 대조구와 삭제 구간에 유의적인(p<0.05) 차이를 나타내었다.는 담수(淡水)에서 10%o의 해수(海水)로 이주된지 14일(日) 이후에 신장(腎臟)에서 수축된 것으로

Effects of Aralia elata, Acanthopanacis cortex and Ulmus davidiana Water Extracts on Plasma Biomarkers in Streptozotocin - Induced Diabetic Rats (두릅, 오가피 및 느릅 열수추출물이 Streptozotocin 투여 흰쥐의 혈장바이오마커에 미치는 영향)

  • This study was conducted to investigate the effects of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts on plasma glucose and biomarkers in the streptozotocin (STZ)-induced diabetic rats. Male Wistar rats were divided into normal and diabetic groups. The diabetic groups subdivided into the control group (DM), Aralia elata (DM-AE), Acanthopanacis cortex (DM-AC) and Ulmus davidiana (DM-UD). The extracts were supplemented in diet base on 11.42 g of raw materials/㎏ diet for 7 weeks. The diabetes was induced by injecting STZ (55 ㎎/㎏ B.W., i.p.) once 2 weeks before sacrifying. Plasma glucose level was significantly higher in the DM group than in the normal group, whereas insulin and C-peptide concentrations were significantly lowered in the DM groups compared to the normal group. These parameters were normalized in the DM-AE, DM-AC and DM-UD supplemented groups. Plasma albumin content was significantly lowered in the DM group compared to the normal group, yet it was significantly higher in the DM-AE group than in the DM group. Bilirubin and creatinine contents were elevated in the DM group, while the supplementation of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts ameliorate the change of these contents in STZ-induced diabetic rats. Plasma AST, ALT, ALP and LDH activities were significantly higher in the DM group than in the normal groups. The supplementation of Araliaceae family water extracts significantly lowered these parameters compared to the DM group. Accordingly, these results indicate that Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts would seem to improve the glucose and biomarker in STZ-induced diabetic rats.

Antioxidant Capacity and Protective Effects on Neuronal PC-12 Cells of Domestic Bred Kiwifruit (국내 육성 참다래의 항산화능 및 PC-12 신경세포 보호 효과)

  • This study was conducted to comparatively evaluate antioxidant capacity (AC) of seven cultivars of kiwifruit (Actinidia spp.) and their protective effects on neuronal PC-12 cells. The contents of total phenolics (TP) and total flavonoids (TF) of kiwifruits were also examined. Five cultivars of kiwifruit, Actinidia chinensis (cv. Haehyang and cv. Haegeum), A. eriantha (cv. Bidan), A. arguta ${\times}$ A. deliciosa (cv. Mansoo), and A. arguta (cv. Chiak), were bred in Korea, while two cultivars, A. deliciosa (cv. Hayward) and A. linguiensis (accession number 041AE), originated from New Zealand and China, respectively. Skin extracts of kiwifruit showed higher TP, TF, and AC than flesh extracts. The highest levels of TP and AC were found in cv. Bidan flesh extract among cultivars studied, but the TF content of cv. Bidan flesh extract was the lowest. The kiwifruit bred in Korea had higher AC than their counterparts. AC of kiwifruit had a highly positive linear correlation with TP and TF. The flesh extracts from cv. Hayward, cv. Haehyang, and cv. Haegeum significantly (p < 0.05) prevented PC-12 cells from oxidative stress induced using $H_2O_2$ compared to a control with $H_2O_2$ only. Overall, our results suggest that kiwifruit bred in Korea may offer a good source of antioxidants and serve as functional materials.

Overview of Autophagy in Plant Cells (식물 세포의 자식작용에 대한 개요)

  • In a variety of eukaryotic cells, autophagy sequesters a portion of the cytoplasm and targets it to a lytic compartment for degradation in bulk. Autophagy is a dynamic process for degrading cytoplasmic cargoes with various degrees of selectivity, and its activity is tightly regulated in a nutrient- and development-dependent manner. Autophagy research has drawn much attention since autophagy not only is an interesting cell biological phenomenon but also has great potential for medical and agricultural applications. For example, autophagy is associated with cancers and neurodegenerative diseases in human and mammalian cells and is also suggested in remobilization of nutrients during the senescence of plant leaves. In this general review, we describe genetic components of the core autophagic machinery conserved among yeast, animals, and plants and briefly explain how these components are responsible for major steps in plant autophagy. We discuss four common features of autophagic processes: (i) autophagy as a degradation pathway, (ii) the concept of flux in autophagy research, (iii) dependency on developmental and nutritional cues, and (iv) diversity of autophagy, focusing on selective types of autophagy. We also summarize cell biological and physiological functions of plant autophagy. Our intention is to provide a quick guide to autophagy for those who are new to autophagy research.

Gardenia jasminoides Exerts Anti-inflammatory Activity via Akt and p38-dependent Heme Oxygenase-1 Upregulation in Microglial Cells (소교세포에서 heme oxygenase-1 발현 유도를 통한 치자(Gardenia jasminoides)의 항염증 효과)

  • Died Gardenia jasminoides fruit is used as a dye in the food and clothes industries in Asia. The present study investigated the anti-inflammatory effects of aqueous extract of G. jasminoides fruits (GJ) in BV-2 microglial cells. GJ inhibited lipopolysaccharide-induced nitric oxide (NO) secretion, inducible nitric oxide synthase (iNOS) expression, and reactive oxygen species production, without affecting cell viability. Furthermore, GJ increased the expression of heme oxygenase-1 (HO-1) in a dose-dependent manner. Moreover, the inhibitory effect of GJ on iNOS expression was abrogated by small interfering RNA-mediated knock-down of HO-1. In addition, GJ induced nuclear translocation of nuclear factor E2-related factor 2 (Nrf2), a transcription factor that regulates HO-1 expression. GJ-mediated expression of HO-1 was suppressed by LY294002, a phosphoinositide 3-kinase (PI-3K) inhibitor, and SB203580, a p38 kinase inhibitor, but not by the extracellular signal-regulated kinase (ERK) inhibitor PD98059 or c-Jun N-terminal kinase (JNK) inhibitor SP600125. GJ also enhanced the phosphorylation of Akt and p38. These results suggest that GJ suppresses the production of NO, a pro-inflammatory mediator, by inducing HO-1 expression via PI-3K/Akt/p38 signaling. These findings illustrate a novel molecular mechanism by which extract from G. jasminoides fruits inhibits neuroinflammation.

Prediction and analysis of acute fish toxicity of pesticides to the rainbow trout using 2D-QSAR (2D-QSAR방법을 이용한 농약류의 무지개 송어 급성 어독성 분석 및 예측)

  • The acute toxicity in the rainbow trout (Oncorhynchus mykiss) was analyzed and predicted using quantitative structure-activity relationships (QSAR). The aquatic toxicity, 96h $LC_{50}$ (median lethal concentration) of 275 organic pesticides, was obtained from EU-funded project DEMETRA. Prediction models were derived from 558 2D molecular descriptors, calculated in PreADMET. The linear (multiple linear regression) and nonlinear (support vector machine and artificial neural network) learning methods were optimized by taking into account the statistical parameters between the experimental and predicted p$LC_{50}$. After preprocessing, population based forward selection were used to select the best subsets of descriptors in the learning methods including 5-fold cross-validation procedure. The support vector machine model was used as the best model ($R^2_{CV}$=0.677, RMSECV=0.887, MSECV=0.674) and also correctly classified 87% for the training set according to EU regulation criteria. The MLR model could describe the structural characteristics of toxic chemicals and interaction with lipid membrane of fish. All the developed models were validated by 5 fold cross-validation and Y-scrambling test.
