Effect of Chamaecyparis obtusa tree Phytoncide on Candida albicans (편백 피톤치드가 Candida albicans에 미치는 영향에 대한 연구)

  • Phytoncide, essential oil of trees, has microbicidal, insecticidal, acaricidal, and deodorizing effect. The present study was performed to examine the effect of phytoncide on Candida albicans, which is a commensal colonizer of the mucous membranes but has become an opportunistic pathogen. C. albicans was incubated with or without phytoncide extracted from Hinoki (Chamaecyparis obtusa Sieb. et Zucc.; Japanese cypress) and then changes were observed in its optical density, cell viability and morphology. As concentrations of phytoncide added to the culture medium increased, optical density and cell viability of C. albicans decreased. Minimum inhibitory concentration of phytoncide for C. albicans was observed to be 0.25%, and minimum fungicidal concentration was 0.5%. Numbers of morphologically atypical cells with electron-dense cytoplasm and granules and increased with increasing concentration of the phytoncide. At higher concentrations of phytoncide, compartments and organelles in the cytoplasm became indistinguishable. The overall results indicate that the phytoncide used for this study has a strong antimicrobial activity against C. albicans. Therefore, the phytoncide may be used as a candidate for prevention and therapeutic agent against oral candidiasis.

Effects of low dose $\gamma$-ray on the early growth of tomato and the resistance to subsequent high doses of radiation (저선량 $\gamma$선 조사가 토마토의 초기생육과 후속고선량 $\gamma$선 저항성에 미치는 영향)

  • Tomato (Lycopericum esculentum $M_{ILL}$ cv. Seokwang and cv. Housemomotaro) seeds were irradiated with the doses of $1{\sim}20$ Gy from $^{60}Co$ $\gamma$-ray source to investigate the effect of the low dose $\gamma$-ray radiation on the early growth and resistance to subsequent high dose of radiation. Germination rate of seeds irradiated with low dose $\gamma$-ray was enhanced in Seokwang cultivar but not in Housemomotaro cultivar. Seedling height increased in 4 Gy and 8 Gy irradiation group of both cultivars. Plant height of Seokwang cultivar was depressed in low dose irradiation group but fresh weight was increased in 2 Gy and 4 Gy irradiation group. In Housemomotaro cultivar, plant height increased in 12 Gy and 20 Gy irradiation group and fresh weight increased in 4 Gy and 20 Gy irradiation group. Growth inhibition of tomato plants by high dose radiation was noticeably reduced by pre-irradiation of low dose radiation. Resistance to subsequent high dose of radiation was enhanced in 2 Gy and 8 Gy Irradiation group of Seokwang cultivar and in 2 Gy and 12 Gy irradiation group of Housemomotaro cultivar.

  • For the purpose of establishment of Reference Korean and estimation of internal and external exposure doses in the Reference Korean, we have surveyed reference values for Koreans, such as physical standards including height, weight and body surface area, food consumption rate of daily intake of radioactive substances and exposure dose from natural radiation. The results obtained are as follows: 1) The age group of the Reference Korean ranged from 20 to 30 years old in both sexes. The height, weight and surface area of the body of the Reference Korean are 167cm, 61kg and $1.67m^2$ in male and 155cm, 51kg and $1.51m^2$, respectively in female. 2) The food consumption of the Korean is 812.8g (669.6g of vegetable food and 143.2g of animal food) per capita per day. 3) Koreans are taken about 1,200 pCi of radioactive substances(${\beta}$-ray) per capita per day. 4) The external and internal radiation exposure doses of the Korean are estimated to be 127 mrem and 8 mrem per year, respectively. However, it is believed that these values will be modified upon the addition of data collection.

Comparison of Measured and Predicted $^3H$ Concentrations in Environmental Media around the Wolsung Site for the Validation of INDAC Code (주면피폭선량 평가코드(INDAC)의 검증을 위한 월성원전 주면 삼중수소 농도 실측치와 예측치의 비교 평가)

  • The predicted results of INDAC code were compared with measured $^3H$ concentrations in air and pine-needle around the Wolsung site. The optimal sets of input data to INDAC were in addition selected by comparing the measured values with the predicted values of INDAC based on various conditions such as the release modes of effluents into the environment, the classification of wind classes, and the consideration of terrain. The predicted $^3H$ concentrations in air and pine-needle were shown to have good agreement with measured values, although there are some limitations such as uncertainties in measured values, complex topology around the site, and the land-sea breeze effects. The assumption on the $^3H$ behavior in vegetables or plants that the ratio of $^3H$ concentration in plant water to $^3H$ concentration in atmospheric water is 1/2 was shown to be conservative in terms of the audit calculation performed by the regulator. It was also found that data sets based on mixed mode and no terrain data were not appropriate for the audit calculation ensuring the compliance with regulations. Thus, if the mixed mode is considered as the release mode of effluents into the environment, meteorological data measured at 58 m height and terrain data should be used to evaluate the atmospheric dispersion factor.

Physiological Activities of Extracts of Cedrela sinensis leaves (참죽나무 잎 추출물의 생리활성)

  • The purpose of this study was to confirm the content of total polyphenol, antioxidative and immune activities of the extracts of Cedrela sinensis leaf. The content of total polyphenol of water extracts ranged from 46.5-59.6 mg/100 g, which was higher than other extracts using organic solvents such as EtOAc, $CH_2Cl_2$ and $C_6H_{14}$. The antioxidant activity of the water and organic solvents extracts showed 6-33% in terms of 2,2-diphenyl-picryl-hydrazyl (DPPH) scavenging activity. To analyze the immuno-stimulation activity of C. sinensis leaf extract, we investigated the effect of the extracts on NO synthesis which is important in host defense against bacterial infection. Hot water extracts significantly increased NO generation by RAW 264.7, macrophage cell line, while organic solvent extract has no significant effect on NO production. To further analyzed the anti-inflammatory effect of the extracts, we investigated the effects of the extracts on lipopolysaccharide(LPS)-induced NO generation. Organic solvent extracts of C. sinensis leaves showed strong inhibitory effect on NO production in LPS-stimulated RAW 264.7 cells. These results suggest that C. sinensis leaf extract may represent a useful immune stimulating agent and anti-inflammatory agent.

Identification of Cold Stress-related Proteins in Rice Leaf Tissue (벼의 잎 조직에서 발현되는 저온 스트레스 관련 단백질의 분리 동정)

  • To investigation protein expression pattern in rice leaves exposed to cold stress, the soluble proteins extracted from leaf tissue were fractionated with $15\%$ PEG and separated by two-dimensional polyacrylamide gel electrophoresis (2-DE). Differentially expressed proteins were identified by peptide mass fingerprinting using matrix assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS). Eight proteins up-regulated and 10 down-regulated were found in $15\%$ PEG supernatant fraction. In addition, 13 proteins up-regulated and 14 down-regulated were found in $15\%$ PEG pellet fraction. It was identified the differentially expressed proteins in $15\%$ PEG supernatant fraction as pimerase/dehydratase fructokinase, ribose-5-phosphate isomerase (Rpi), chaperonin 21 precursor, probable photosystem II oxygen-envolving complex (PS II OEC) protein 2 precursor and thioredoxin h-type (Trx-h) and those in $15\%$ PEG pellet fraction as OSINBb0059K02.15, hypothetical protein, putative mitogen-activated protein kinase kinase (MAPKK), beta 7 subunit of 205 proteasome, ribulose-1, 5-bisphosphate carboxylase/oxygenase (Rubisco) small subunit. These proteins are involved in metabolism, energy, protein synthesis, disease/defense and signal transduction-related proteins.

Changes of antioxidant enzyme activities subjected to water stress in soybean leaves (대두(大豆)에서 분석(水分)스트레스에 의(依)한 항산화효소(抗酸化酵素)의 활성도(活性度) 변화(變化))

  • This experiment was carried out to elucidate and study about plant defense mechanism subjected to water stress(drought, flooding). We measured water content, total soluble protein content as stress marker and superoxide dismutase(SOD), catalase (CAT) as antioxidant enzymes subjected to water stress(drought, flooding) and recovery in soybean leaves. The results obtained were as follows; Two soybean lines(keonolkong, euhakong)leaves exposed to water stress (drought, flooding) showed premature senescence as evidence by the decrease in water content, and total soluble protein content, but those of soybean leaves subject to water stress recovery for 3 days were recovered. Visual damage was much worse at drought stress than flooding stress and was worse keunolkong than enhakong. The activity of superoxide dismutase, catalase subjected to water stress(drought, flooding)was on the decrease, but degree of decrease was different from a sort of soybean lines, drought and flooding stress.

Enhancement of Photosynthetic Characteristics and Antioxidant Enzyme Activities on Chili Pepper Plants by Salicylic Acid Foliar Application under High Temperature and Drought Stress Conditions (고온 및 건조 스트레스 조건 하에서 살리실산 경엽처리에 의한 고추의 광합성 특성 및 항산화효소 활성 증대)

  • Salicylic acid (SA), a phenolic compound, plays a pivotal role in regulating a wide range of physiological and metabolic processes in plants such as antioxidant cellular defense, photosynthesis, and biotic and abiotic stress responses during the growth and development. We examined the effect of exogenous SA application (100 mg·L-1) on the growth, yield, photosynthetic characteristics, lipid peroxidation, and antioxidant enzyme activity of chili pepper plants under high temperature and drought stress conditions. SA treatment induced increases of net photosynthetic rate (Pn), stomatal conductance (Gs) and transpiration rate (Tr) under the stress condition with the highest level after the third treatment. The contents of malondialdehyde and H2O2 were significantly lower in the third treatment of SA compared to the control. The activity of ascorbate peroxidase, catalase, peroxidase and superoxide dismutase, increased in treated plants by up to 247, 318, 55 and 54%, respectively compared to the nontreated control. There was no significant difference in the growth characteristics between SA-treated and nontreated plants, while the SA treatment increased marketable yield (kg/10a) by about 15% compared to the nontreated control. Taken together, these results suggest that foliar application of SA alleviates physiological damages caused by the combination of drought and heat stress, and enhances the photosynthetic capacity and antioxidant enzyme activities, thereby improving tolerance to a combination of water deficit and heat stress in chili pepper plants.

Anti-melanogenic Effects of Cnidium japonicum in B16F10 Murine Melanoma Cells (B16F10 피부 흑색종세포에서 갯사상자 추출물의 멜라닌 합성 저해 효과)

  • Melanin is a pigment produced by melanocytes to protect the skin from external stimuli, mainly ultraviolet (UV) rays. However, abnormal and excessive production of melanin causes hyperpigmentation disorders, such as freckles, age spots, and discoloration. Natural cosmeceuticals are a new trend for treating or preventing hyperpigmentation due to fewer side effects and biocompatibility. In this context, the current study focused on Cnidium japonicum, a halophyte with several uses in folk medicine, to evaluate its potential as a skin-whitening agent. The effect of C. japonicum extract (CJE) on melanin production was analyzed in melanogenesis-stimulated B16F10 melanoma cells. The results showed that CJE successfully inhibited the oxidation of tyrosine and L-DOPA by tyrosinase and subsequently decreased the production of the key enzymes responsible for melanin production: tyrosinase, tyrosinase-related protein-1, and protein-2. This effect was confirmed by decreased intracellular and extracellular melanin levels in B16F10 melanoma cells after CJE treatment. Further experiments to elucidate the action mechanism revealed that CJE treatment suppressed melanin production by inhibiting the activation of glycogen synthase kinase 3 β (GSKβ)/β-catenin and protein kinase A (PKA)/cAMP-response element binding protein (CREB) pathways, which are the upstream activators of melanogenesis. In conclusion, the present study suggests that C. japonicum is a potential natural source of bioactive substances for the development of novel cosmeceuticals that can act against hyperpigmentation.

Factors Affecting Community Resilience in the Process of Environmental Pollution Purification: Focusing on the Restoration of Soil Pollution around the Janghang Smelter (환경오염 정화 과정에 나타난 지역공동체 회복력 영향 요인: 장항제련소 토양오염 복구를 중심으로)

  • The objective of this study was to evaluate the community's capacity and hindrance factors affecting the community's activities by exploring the process of restoring the community's environmental pollution. This study examined the community activities related to the restoration of pollution that occurred at the former Janghang smelter in Seocheon-gun. The results of this study showed that leaders, cooperative resources, and the media were elements positively influencing the recovery of the community. The starting point that became the driving force for community activities was the presence of a leader who had a small stake in it. Moreover, it required cooperation such as the continuous interest of local active groups and residents to continue this effort. Above all, the media publicized the incident and amplified the activities of members. Meanwhile, there were various conflicts hindering the activities of local communities in the process of restoring the pollution such as the internal conflicts of community groups, conflicts between residents and active groups, and conflicts with the government. The behaviors that appeared in the process of restoring the pollution revealed a form of united governance. Moreover, they changed from an initial "supportive" character to "resistant" and "defensive" activities. Later, it was transformed into "subjective" and "creative" activities.
