Antioxidant Activities of the Mycelium and Culture Broth of Phellinus igniarius and Agrocybe cylindracea (말똥진흙버섯 및 버들송이버섯의 균사체 및 배양액의 항산화활성)

  • The fractions obtained from the mycelia and their culture broth of Agrocybe cylindracea and Phellinus igniarius were tested for the antioxidation activities. In electron donating ability test, strong activities more than 70% were observed in ethyl acetate and butanol fractions of A. cylindracea mycelium, butanol fraction of A. cylindracea culture broth, diethyl ether fraction of P. igniarius mycelium, and diethyl ether and ethyl acetate fractions of P. igniarius culture broth. Lipid peroxidation inhibition was observed in diethyl ether, butanol and water fractions of A. cylindracea mycelium, and water fraction of A. cylindracea induced the decrease in uric acid production. Activities of $50{\sim}60\;%$ of nitrite scavenging ability were observed in diethyl ether fraction of A. cylindracea mycelium, ethyl acetate and water fractions of A. cylindracea culture broth, diethyl ether fraction of P. igniarius mycelium, and diethyl ether, chloroform, butanol and water fractions of P. igniarius culture broth. In melanin biosynthesis inhibition activity measurement, water fraction of A. cylindracea mycelium and butanol fraction of P. igniarius mycelium showed twice higher activities than control. From these results, the fungi of P. igniarius and A. cylindracea had strong antioxidant activities.

  • Some agaric fungi were collected at Chink from Sep. 10, 1988 and Aug. 12, 1990. some agaric fungi were recognized to the species. Among them six species were confirmed new to Korea and registered here with descriptions. They are Calocybe ionides (Bull.: Fr.) Donk ; Tricholoma psammopus (Kalchbr.) Quel.; Collybia cirrhatta (Per.) Quel.; Marasmius aurantioferrugineus Hongo; Amanita abrupta Peck and Bull.; Amanita castanopsidis Hongo.

The 18s rDNA Sequences of the Basidiocarps of Tricholoma matsutake in Korea (한국산 송이버섯에서의 18s ribosomal DNA 서열)

  • The 18S rDNA sequences of Tricholoma matsutake (TM=T. caligatum var. nauseoum) collected in Korea were analyzed for the ectomycorrhizal fungi in the roots of Pinus densiflora. The 514 base pairs of rDNA region were synthesized by UF-5 and UR-6 primers, and double checked in the base pair. The sequence of four strains synthesized were all identical in this work, but different from those done by the previous workers. The basidiocarps collected in this work. were identified to T. matstake after searching the 18s rDNA by the BLAST in NCBI. Only several base pairs of 18S rDNA analyzed from other related basidiocarps were different from our analyses of 18S rDNA. The dendrogram were made based on the sequences of the 514 bp 18S rDNA by CLUSTAL-X alignment program. The groupings of the species at the level of genus in the dendrogram were well constructed.

  • 표고(Lentinus edodes (Berk.) Sing.)는 송이과 Tricholomataceae에 속하는 식용 버섯으로서, 동북아시아 지역에서 가장 인기 있는 재배 버섯이다. 표고의 수요와 생산량을 매년 꾸준히 증가하고 있고, 1985년 표고 생산량은 880톤, 87년 1,080톤, 89년 1,530톤, 91년 1,761톤, 92년의 표고의 재배면적은 397,340평이고, 생산량은 2,253톤이며 금액으로 375억에 이르고 있다. (중략)

  • Fresh fungi were collected during five field trips to Ullung Island from October of 1989 to September of 1991. Through the observation of some agaric fungi, thirty mushrooms were identified to the species and are listed as below. Among them, three species, Crepidotus geophilus, Mycena elegans, ana Mycena carolinensis were confirmed new to Korea and are registered here with descriptions.

Comparison on the fatty acid Composition of Korean edible mushrooms (한국산 식용버섯의 지방산 조성에 관한 비교)

  • A Comparison on the fatty acid composition of 7 species of Korean edible mushroom(Agaricus bisporus, Tricholoma matsutake, Lentinus edodes, Pleurotus ostreatus, Ramaria botrytis, Sarcodon asparatus, Calocybe gambosa) were investigated by gasliquid Chromatography. The results were obtained as follows: 1) The major fatty acid in all samples were linoleic $(77.33-35.53\%)$, oleic $(39.69-1.58\%)$ and palmitic$(22.51-7.31\%)$ acid, 31 2) The content of linoleic acid was the highest in Agaricus Bisporus, Pleurotus ostreatus and Lentinus edodes meanwhile, the content of oleic acid of these mushrooms was significantly low compared with others. 3) Quantity of myristic acid was higher in comparison with fatty acid composition from common lipid source.

Optimization of Hot Water Extraction Conditions for Tricholoma matsutake by Response Surface Methodology (반응표면분석을 이용한 송이버섯 열수추출조건의 최적화)

  • This study was performed to establish optimum extraction condition of Tricholoma matsutake. A central composite design was applied to investigate the effects of independent variables, extraction temperature ($X_1$), extraction time ($X_2$) and water per sample ($X_3$) on dependent variables such as soluble solids contents ($Y_1$), total phenolics contents ($Y_2$), reducing sugar contents ($Y_3$), electron donating ability ($Y_4$) and nitrite scavenging ability ($Y_5$). The optimum extraction conditions were predicted and monitored by response surface methodology using SAS program based regression analysis. Soluble solids content, electron donating ability and nitrite scavenging ability were highly affected by water per sample. However, the contents of total phenolics and reducing sugar were affected by water per sample and extraction temperature as well. The optimum extraction conditions for soluble solids were 34.84 mL/g (water/sample) at $78.85^{\circ}C$, for 3.33 hr. In contrast, the optimum extraction conditions of electron donating ability were temperature of $91.00^{\circ}C$, time of 1.62 hr and water per sample of 39.42 mL/g. Taken together, the optimum ranges for hot water extraction of Tricholoma matsutake were $70{\sim}90^{\circ}C$, 2~4 hr and 30~50 mL/g.

Mycelial Growth Properties of Domestically Collected Ectomycorrhizal Tricholoma Mushrooms in Various Culture Conditions (다양한 배양 환경에 따른 국내 수집 외생균근성 Tricholoma속 종의 균사생장 특성)

  • The ectomycorrhizal basidiomycete Tricholoma is one of mushroom groups that cannot be cultivated artificially. To use this mushroom as applicable resource for food production, it is necessary to obtain information about their mycelial growth properties in various environmental conditions. This study investigated the mycelial growth of four domestic isolates of Tricholoma species (T. bakamatsutake, T. fulvocastaneum, T. matsutake, T. terreum) at different physical and chemical conditions. The optimal physical conditions for their mycelia growth were found to be a temperature range of $20{\sim}25^{\circ}C$ and a pH range of 4.0~7.0 in dark condition. The growth of T. matsutake was retarded at high temperature ($30^{\circ}C$). Tests to determine the chemical factors that affected mycelial growth showed that the four Tricholoma spp. grew 1% saline. T. matsutake grew in up to 2% saline. In the presence of various heavy metals (50 ppm) and pesticides (suppliers' recommended concentration), mycelial growth was inhibited the most by cadmium and emamectin benzoate, respectively. However, all the four Tricholoma spp. grew with $Cu^+$. The growth of T. matsutake was not inhibited by abamectin, acetamiprid, and thiacloprid. Extracellular enzyme activities of amylase and ${\beta}$-glucosidase were detected only in T. bakamatsutake and T. fulvocastaneum. The results of the present study allowed us to determine suitable or harmful environmental conditions for the mycelial cultivation of the Tricholoma spp.

Characterization of a new variety "Backryeon" developed by crossing in Tricholoma giganteum (왕송이(Tricholoma giganteum) 신품종 '백련'의 특성)

  • Tricholoma giganteum, belongs to Tricholomataceae of Tricholoma, is also well-known as the medicinal mushroom in Taiwan. "Backryon" was the first variety developed by intra-specific crossing in Korea. It was improved with hybridization between monokaryotic strain derived from MKACC50852 and MKACC 50853. The optimum temperature of mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $20{\sim}25^{\circ}C$, respectively. The color of fruitingbody was pure white and cap type was umbrella. It suggested that 'Backryon' was new commercial variety for small-sized cultivator during the summer season.
