친환경 농업 - UV-C처리에 따른 마늘의 기능성 성분 증진 효과

  • 일반적으로 식물은 생장 중에 많은 양의 생체 방어 물질이 자체적으로 생성되지만, 건실한 성장 후에 생체 방어 물질은 급격하게 소멸되거나 감소한다. 이에 식물체에 저농도 자외선(Ultraviolet(UV) Radiation)을 처리하면 스트레스 또는 DNA의 손상이 유도되며 이에 대응하여 식물체는 항상성(Homeoslasis)을 유지하기 위한 긍적적인 변화를 일으킨다. 농촌진흥청에서는 마늘에 저농도 자외선을 처리해 저장성 기능성 성분을 높이는 기술을 개발했다.

  • 수산화나트륨(Sodium hydroxide)의 직업적 노출에 대한 TLV-Ceiling은 $2mg/m^3$으로 권고하였다. 권고수준은 수산화나트륨 에어로졸이 눈, 점막, 피부에 심한 자극을 유발하고 수산화나트륨 분진이 상기도 기관에 자극을 유발하는 농도에 근거하였다. 노출기준은 눈과 상기도 기관에 자극을 최소화하기 위하여 권고하였다. 고농도의 수산화나트륨에 장기간 노출되면 비강 궤양과 눈과 피부에 심한 손상을 유발할 수 있다. "피부", "감작제(SEN)", "발암성"의 경고주석을 권고하기에는 유용한 자료가 충분하지 않다.

The Effect of Histamine on Polymorphonuclear Leukocyte-induced Pneumocyte Injury in Vitro (다형핵구에 의한 폐포세포 손상에 Histamine이 미치는 영향)

  • Background: Although polymorphonuclear leukocytes (PMN) are important in protecting the airways and alveolar surfaces, there is evidence that they can also injure the lung while exercising their defensive role. However it has been unclear whether PMN-induced pneumocyte injury is mediated by their direct cytotoxic effect on target cells or by PMN-derived cytotoxic mediators. On the other hand histamine was known not only to act as an important chemical mediator participated in the pathogenesis of some atotic and allegic disorders, but also to have an inhibitory effect on normal PMN functions. Method: To study the mechanism by which PMN induce pneumocyte injury, we cocultured PMN from four healthy nonsmokers or their PMN-derived supernatants (PMN-SPN) with monolayers of $^{51}Cr$-labeled human A549 pneumocytes and compared PMN-and PMN-SPN-mediated pneumocyte injuries measured by $^{51}Cr$ release assay. We also compared the effects of histamine on each pneumocyte injury. Results: 1) PMN-SPN showed more injurious effect on A549 pneumocytes than that of PMN itself regardless histamine pretreatment of PMN. 2) Pneumocyte injury by PMN with histamine pretreatment was increased or decreased compared with that by PMN without histamine pretreatment, according to histamine concentrations, and PMN stimulating agents and their concentrations. 3) Pneumocyte injury by PMN-SPN with histamine pretreatment tended to be decreased compared with that by PMN-SPN without histamine pretreatment. Conclusion: Our results suggest that PMN-SPN may play more important role in mediating pneumocyte injury than PMN itself and that histamine may partially play a protective role on PMN-induced pneumocyte injury. Alternatively we conclude that the effects of histamine on PMN-induced pneumocyte injury may be affected by microenvironment in vivo.

Changes of Glutamate and Polyamine Levels of Hippocampal Microdialysates in Response to Occlusion of Both Carotid Arteries in Mongolian Gerbils (뇌허혈 손상에 있어서 해마-세포외액내 Glutamate와 Polyamine 농도의 변동에 관한 연구)

  • Reversible brain ischemia was produced by occluding both common carotid arteries for 5 min, and the effects of aminoguanidine (AG), $DL-{\alpha}-difluoromethylornithine$ (DFMO), MK-801, and nimodipine (NM) on the ischemia induced changes of the polyamine, glutamate and acetylcholine levels in the hippocampus CA1 subfield and the specific $[^3H]\;MK-801$ binding to the hippocampus synaptosomal membranes were studied with a histological reference of the cresyl violet stained hippocampus. The basal putrescine level $(PT:\;74.4{\pm}8.8\;nM)$ showed a rapid increase (up to 1.7 fold) for 5 min of ischemia, remained significantly increased for 6 h, and then resumed the further increase to amount gradually up to about 3 fold 96 h after recirculation. However, the level of spermidine was little changed, and the spermine level showed a transient increase during ischemia followed by a sustained decrease to about 40% of the preischemic level after recirculation. The increase of PT level induced by brain ischemia was enhanced with AG or MK-801, but it was reduced by DFMO or NM. The basal glutamate level $(GT:\;0.90{\pm}0.l4\;{\mu}M)$ rapidly increased to a peak level of $8.19{\pm}1.14\;{\mu}M$ within 5 min after onset of the ischemia and then decreased to the preischemic level in about 25 min after recirculation. And NM reduced the ischemia induced increase of GT level by about 25%, but AG, DFMO and MK-801 did not affect the GT increase. The basal acetylcholine level $(ACh:\;118.0{\pm}10.5\;{\mu}M)$ did little change during/after brain ischemia and was little affected by AG or NM. But DFMO and MK-801, respectively, produced the moderate decrease of ACh level. The specific $[^3H]\;MK-801$ binding to the hippocampus synaptosomal membrane was little affected by brain ischemia for 5 min. The control value (78.9 fmole/mg protein) was moderately decreased by AG and MK-801, respectively but was little changed by DFMO or NM. The microscopic findings of the brains extirpated on day 7 after ischemia showed severe neuronal damage of the hippocampus, particularly CA1 subfield. NM and AG moderately attenuated the delayed neuronal damage, and DFMO, on the contrary, aggravated the ischemia induced damage. However, MK-801 did not protect the hippocampus from ischemic damage. These results suggest that unlike to the mode of anti-ischemic action of NM, AG might protect the hippocampus from ischemic injury as being negatively regulatory on the N-methyl-D-aspartate (NMDA) receptor function in the hippocampus.

The Effect of the Swimming Exercise by Load on Concentration of BDNF in Serum and Behavioral Change of CNS Injury in the Rats (부하유무에 따른 수영운동이 중추신경계 손상 흰쥐의 혈청 BDNF 농도 및 행동변화에 미치는 영향)

  • The purposes of the present investigation was to evaluate the effects of loaded and unloaded swimming stimulation after central nerve system injury in the rats. SCI model rats were damaged in L1-L2 injected with 6-OHDA. The twenty one Sprague-Dawley adult male rats weights($200\pm10g$) were randomly divided into control group and 2 swimming groups and then swimming groups divided into 15 minute unloaded swimming group and 15 minute loaded swimming group by swimming intensity. Behavioral Change was evaluated by the BBB(Basso, Brestti, Brenahan) scales test and the maximal angles of the inclined board on which the rat could maintain its intial position for the progressive locomotor recovery. Using enzyme-linked immunosolbent assays(ELISA), we measured concentrations of brain-delived growth factor(BDNF) in serum after swimming. There was significant change of BBB scores in control group as compared to unloaded swimming group and loaded swimming group(p<.05), and unloaded swimming group were significantly higher than loaded swimming group(p<.05). The maximal angles of the inclined plane test were higher in the unloaded swimming group and loaded swimming group than the control group(p<.05), and loaded swimming group were significantly lower than unloaded swimming group(p<.05). There were singnificant difference of concentration of BDNF in serum change in each group(p<.05). The results suggest that swimming applied from the early phase after spinal cord injury may be beneficial in the early recovery of motor function.

Effect of Damage to Medial Amygdaloid Nucleus on Pancreatic Exocrine Secretion Stimulated by Hydrochloric Acid in the Rat (흰쥐에서 내측 편도핵의 손상이 염산 자극에 의한 췌장 외분비에 비치는 영향)

  • This study was undertaken to investigate the effect of the medial amygdaloid nucleus on the pancreatic exocrine secretion and plasma secretin concentration in 44 male albino rats. Twenty-three rats in which the medial amygdaloid nucleus was damaged bilaterally by radio frequency a.c. through stereotaxically inserted electrodes (medical amygdaloid group, MA) and twenty-one rats which received the same operation without damage (operated control, OC), were prepared. Under urethan anesthesia, 0.01 N hydrochloric acid (HCl) or physiological saline (0.9% NaCl) was infused at a rate of 0.18 ml/min into the duodenum for 20 minutes. Pancreatic jucie was collected for the 20 min infusion period. After collection of pancreatic juice, blood was sampled from the abdominal aorta for the radioimmunoassay of plasma secretin concentration. In the MA group, the exocrine pancreatic secretory response to 0.01 N HCI as well as saline infusion was significantly inhibited compared with that in the OC group. The pancreatic protein output of the MA group significantly decreased after the saline infusion and tended to decrease after the 0.01 N HCI infusion, compared with that of the OC group. However, there was no significant difference in plasma secretin concentration between the two groups. Therefore it is strongly suggested that the rat medial amygdaloid nucleus has a facilitatory influence on both basal and acid-stimulated pancreatic exocrine secretion, but the releasing mechanism of secretin appears not to be involved in the influence.

Separation and Purification of Effective Components from the Alisma orientale and its Application as a Cosmeceutical Ingredient (택사추출물의 성분분리와 화장품 원료로서의 특성)

  • In this study, we performed anti-oxidation, whitening, cell recovery and anti-inflammation effects with Alisma orientale to evaluate the cosmeceutical properties. Alisma orientate extract (30, 70, 100 % MeOH) exhibited a significant tree radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and showed tyrosinase inhibition effect in a dose dependent manner (over 0.5% concentration). In cell proliferation assay using human fibroblast, it didn't show any proliferation effect but showed safety from cytotoxicity under 0.05% concentration. For whitening assay, we evaluated the melanin synthesis rate using B16 melanocyte and it showed a significant inhibitory effect (up to 40% under 0.05% concentration). After major screening assay, we separate 3 fractions from Alisma orientate extract by MPLC and performed cell recovery assay, melanin synthesis inhibition assay and anti-inflammatory assay. The third fraction showed a cell recovery effect over 30% against radical damage and remarkable repression in melanin synthesis and COX-2 synthesis.

Protective Effects of Branched-chain Amino Acid (BCAA)-enriched Corn Gluten Hydrolysates on Ethanol-induced Hepatic Injury in Rats (알코올성 간 손상을 유발한 흰쥐에 대한 고 분지아미노산 함유 옥수수 단백가수물의 간 기능 보호효과)

  • Hepatoprotective effects of corn gluten hydrolysates (CGH) were investigated in rats orally treated with ethanol (30%(v/v), 3 g/kg body weight/day) for 4 weeks. Six-week old Sprague-Dawley male rats were divided into four dietary groups: normal diet (N), alcohol diet (E), E+CGH 1% diet (CGH-1%), and E+CGH 3% diet (CGH-3%). Body weights and liver indices were not significantly different among the four groups. However, food intakes were lower in the CGH groups than in the normal group (p<0.05). The administration of CGH significantly reduced serum alkaline phosphatase activity by 30% compared to the alcohol diet group. Among the antioxidative enzymes assessed, catalase activity was significantly decreased by 79% in the CGH diet groups compared to the alcohol diet group. In comparison to the alcohol-treated group, aldehyde dehydrogenase activity was increased by 20%, while microsomal ethanol oxidizing system activity was decreased by 20% in the CGH-treated groups. Furthermore, the area under the curve of the blood acetaldehyde concentration versus time profile after the administration of ethanol was significantly lower for the CGH rats than for the ethanol or asparaginic acid treated groups. Thus, CGH seems to offer beneficial effects by protecting against ethanol-induced hepatotoxicity by improving the acetaldehyde-related metabolizing system.

Effect of an Oriental Herbal Composition, Jang Baek Union (JBU), on Alcohol-Induced Hangover and CCl4-Induced Liver Injury in Rats (한약재 조성물이 흰쥐의 알코올성 숙취 및 사염화탄소 유발 간 손상에 미치는 영향)

  • Effects of the extract of oriental herbal composition, Jang Baek Union (JBU), on hangover and liver function were studied in rats. To investigate anti-hangover effect, alcohol concentration and acetaldehyde concentration of blood were measured after oral ethanol treatment. Blood alcohol concentration was significantly reduced by pre-treatment of JBU extract. The effect of JBU extract on the blood acetaldehyde concentration was more significant than commercial product used as a positive control. To investigate hepato-protective effect, serum GOT and GPT levels and histological changes of liver tissue were analyzed in $CCl_4$-treated rats. JBU extract adminstration significantly inhibited the increase of serum GPT and GOT levels induced by $CCl_4$-treatment. Moreover, histological injuries of liver tissue such as fatty changes and sinusoidal leukocytosis were inhibited by JBU extract.

Synergistic Interaction of Radiation with Pesticide on DNA Damage in Human Lymphocytes as Biological Information for Prevention of Environmental Disaster (환경재해 방지를 위한 생물정보로서의 사람 림프구 DNA 손상에 대한 방사선과 살충제의 상승작용)

  • Agricultural pesticides may cause certain biological risks since they are widely used to eradicate pests. Agricultural disasters may arise even from the possibility of their synergistic interaction with other harmful enviromnetal factors. The effect of pesticide on radiation-induced DNA damage in human blood lymphocytes was evaluated by the single cell gel electrophoresis (SCGE) assay. The lymphocytes, with or without pretreatment of the pesticide, were exposed to 0-2.0 Gy of $^60 CO$ gamma ray. Significantly increased tail moment, which was a marker of DNA strand breaks in SCGE assay, showed an excellent dose-response relationship. The present study confirms that the pesticide has the cytotoxic effect on lymphocytes and that it shows the synergistic interaction with radiation on DNA damage as well. The results may have a role of providing biological information necessary for the prevention of environmental disaster.
